Calcium Influx Rescues Adenylate Cyclase-Hemolysin from Rapid Cell Membrane Removal and Enables Phagocyte Permeabilization by Toxin Pores
Bordetella adenylate cyclase toxin-hemolysin (CyaA) penetrates the cytoplasmic membrane of phagocytes and employs two distinct conformers to exert its multiple activities. One conformer forms cation-selective pores that permeabilize phagocyte membrane for efflux of cytosolic potassium. The other conformer conducts extracellular calcium ions across cytoplasmic membrane of cells, relocates into lipid rafts, translocates the adenylate cyclase enzyme (AC) domain into cells and converts cytosolic ATP to cAMP. We show that the calcium-conducting activity of CyaA controls the path and kinetics of endocytic removal of toxin pores from phagocyte membrane. The enzymatically inactive but calcium-conducting CyaA-ACâ toxoid was endocytosed via a clathrin-dependent pathway. In contrast, a doubly mutated (E570K+E581P) toxoid, unable to conduct Ca2+ into cells, was rapidly internalized by membrane macropinocytosis, unless rescued by Ca2+ influx promoted in trans by ionomycin or intact toxoid. Moreover, a fully pore-forming CyaA-ÎAC hemolysin failed to permeabilize phagocytes, unless endocytic removal of its pores from cell membrane was decelerated through Ca2+ influx promoted by molecules locked in a Ca2+-conducting conformation by the 3D1 antibody. Inhibition of endocytosis also enabled the native B. pertussis-produced CyaA to induce lysis of J774A.1 macrophages at concentrations starting from 100 ng/ml. Hence, by mediating calcium influx into cells, the translocating conformer of CyaA controls the removal of bystander toxin pores from phagocyte membrane. This triggers a positive feedback loop of exacerbated cell permeabilization, where the efflux of cellular potassium yields further decreased toxin pore removal from cell membrane and this further enhances cell permeabilization and potassium efflux
Supramolecular assembly of gelatin and inorganic polyanions: Fine-tuning the mechanical properties of nanocomposites by varying their composition and microstructure
A series of bionanocomposites has been synthesized through a complex coacervation process inducing the assembly of gelatin with a wide range of inorganic polyanions (IPyAs) differing by their diameter and charge and including polyoxometalates (POMs) and a polythiomolybdate cluster. The microstructure and stoichiometry of these hybrid coacervates, which are strongly dependent on the charge matching between both components, have been studied by combining Fourier transform infrared (FT-IR) spectroscopy, solid-state nuclear magnetic resonance (NMR), thermogravimetric analysis (TGA), elemental analysis, differential scanning calorimetry (DSC), scanning electron microscopy (SEM), and energy-dispersive X-ray (EDX) elemental mapping. The mechanical properties of these materials were deeply characterized by tensile measurements at large deformation, revealing different behaviors (i.e., elastomer and ductile), depending on the nature of the IPyA. It is noteworthy that the mechanical properties of these bionanocomposites are strongly enhanced, compared to pure gelatin hydrogels. When attempting to connect structure and properties in these bionanocomposites, we have demonstrated that the density of cross-links (gelatin triple helices and IPyA) is the key parameter to control the extensibility of these materials
Methanol along the path from envelope to protoplanetary disc
Interstellar methanol is considered to be a parent species of larger, more complex organic molecules. A physicochemical simulation of infalling parcels of matter is performed for a low-mass star-forming system to trace the chemical evolution from cloud to disc. An axisymmetric 2D semi-analytic model generates the time-dependent density and velocity distributions, and full continuum radiative transfer is performed to calculate the dust temperature and the UV radiation field at each position as a function of time. A comprehensive gasâgrain chemical network is employed to compute the chemical abundances along infall trajectories. Two physical scenarios are studied, one in which the dominant disc growth mechanism is viscous spreading, and another in which continuous infall of matter prevails. The results show that the infall path influences the abundance of methanol entering each type of disc, ranging from complete loss of methanol to an enhancement by a factor of >1 relative to the prestellar phase. Critical chemical processes and parameters for the methanol chemistry under different physical conditions are identified. The exact abundance and distribution of methanol is important for the budget of complex organic molecules in discs, which will be incorporated into forming planetary system objects such as protoplanets and comets. These simulations show that the comet-forming zone contains less methanol than in the precollapse phase, which is dominantly of prestellar origin, but also with additional layers built up in the envelope during infall. Such intriguing links will soon be tested by upcoming data from the Rosetta mission

Using time perception to explore implicit sensitivity to emotional stimuli in autism spectrum disorder
Establishing whether implicit responses to emotional cues are intact in autism spectrum disorder (ASD) is fundamental to ascertaining why their emotional understanding is compromised. We used a temporal bisection task to assess for responsiveness to face and wildlife images that varied in emotional salience. There were no significant differences between an adult ASD and comparison group, with both showing implicit overestimation of emotional stimuli. Further, there was no correlation between overestimation of emotional stimuli and autistic traits in undergraduate students. These data do not suggest a fundamental insensitivity to the arousing content of emotional images in ASD, or in individuals with a high degree of autistic traits. The findings have implications for understanding how emotional stimuli are processed in ASD
Probing Episodic Accretion in Very Low Luminosity Objects
Episodic accretion has been proposed as a solution to the long-standing luminosity problem in star formation; however, the process remains poorly understood. We present observations of line emission from N2H+ and CO isotopologues using the Atacama Large Millimeter/submillimeter Array (ALMA) in the envelopes of eight very low luminosity objects (VeLLOs). In five of the sources the spatial distribution of emission from N2H+ and CO isotopologues shows a clear anticorrelation. It is proposed that this is tracing the CO snow line in the envelopes: N2H+ emission is depleted toward the center of these sources, in contrast to the CO isotopologue emission, which exhibits a peak. The positions of the CO snow lines traced by the N2H+ emission are located at much larger radii than those calculated using the current luminosities of the central sources. This implies that these five sources have experienced a recent accretion burst because the CO snow line would have been pushed outward during the burst because of the increased luminosity of the central star. The N2H+ and CO isotopologue emission from DCE161, one of the other three sources, is most likely tracing a transition disk at a later evolutionary stage. Excluding DCE161, five out of seven sources (i.e., ~70%) show signatures of a recent accretion burst. This fraction is larger than that of the Class 0/I sources studied by JĂžrgensen et al. and Frimann et al., suggesting that the interval between accretion episodes in VeLLOs is shorter than that in Class 0/I sources

APPENDIX S3: Restoration project cycle.The global movement for ecosystem restoration has gained momentum in response to the Bonn Challenge (2010) and the UN Decade on Ecosystem Restoration (UNDER, 2021â2030). While several science-based guidelines exist to aid in achieving successful restoration outcomes, significant variation remains in the outcomes of restoration projects. Some of this disparity can be attributed to unexpected responses of ecosystem components to planned interventions.
Given the complex nature of ecosystems, we propose that concepts from Complex Systems Science (CSS) that are linked to non-linearity, such as regime shifts, ecological resilience and ecological feedbacks, should be employed to help explain this variation in restoration outcomes from an ecological perspective.
Our framework, Explore Before You Restore, illustrates how these concepts impact restoration outcomes by influencing degradation and recovery trajectories. Additionally, we propose incorporating CSS concepts into the typical restoration project cycle through a CSS assessment phase and suggest that the need for such assessment is explicitly included in the guidelines to improve restoration outcomes.
To facilitate this inclusion and make it workable by practitioners, we describe indicators and methods available for restoration teams to answer key questions that should make up such CSS assessment. In doing so, we identify key outstanding science and policy tasks that are needed to further operationalize CSS assessment in restoration.
Synthesis and applications. By illustrating how key Complex Systems Science (CSS) concepts linked to non-linear threshold behaviour can impact restoration outcomes through influencing recovery trajectories, our framework Explore Before You Restore demonstrates the need to incorporate Complex Systems thinking in ecosystem restoration. We argue that inclusion of CSS assessment into restoration project cycles, and more broadly, into international restoration guidelines, may significantly improve restoration outcomes.UK Centre for Ecology and Hydrology;

DMTs and Covid-19 severity in MS: a pooled analysis from Italy and France
We evaluated the effect of DMTs on Covid-19 severity in patients with MS, with a pooled-analysis of two large cohorts from Italy and France. The association of baseline characteristics and DMTs with Covid-19 severity was assessed by multivariate ordinal-logistic models and pooled by a fixed-effect meta-analysis. 1066 patients with MS from Italy and 721 from France were included. In the multivariate model, anti-CD20 therapies were significantly associated (OR = 2.05, 95%CI = 1.39â3.02, p < 0.001) with Covid-19 severity, whereas interferon indicated a decreased risk (OR = 0.42, 95%CI = 0.18â0.99, p = 0.047). This pooled-analysis confirms an increased risk of severe Covid-19 in patients on anti-CD20 therapies and supports the protective role of interferon
