    A population of hematopoietic stem cells derives from GATA4-expressing progenitors located in the placenta and lateral mesoderm of mice

    GATA transcription factors are expressed in the mesoderm and endoderm during development. GATA1-3, but not GATA4, are critically involved in hematopoiesis. An enhancer (G2) of the mouse Gata4 gene directs its expression throughout the lateral mesoderm and the allantois, beginning at embryonic day 7.5, becoming restricted to the septum transversum by embryonic day 10.5, and disappearing by midgestation. We have studied the developmental fate of the G2-Gata4 cell lineage using a G2-Gata4Cre;R26REYFP mouse line. We found a substantial number of YFP+ hematopoietic cells of lymphoid, myeloid and erythroid lineages in embryos. Fetal CD41+ /cKit+ /CD34+ and Lin– /cKit+ /CD31+ YFP+ hematopoietic progenitors were much more abundant in the placenta than in the aorta-gonad-mesonephros area. They were clonogenic in the MethoCult assay and fully reconstituted hematopoiesis in myeloablated mice. YFP+ cells represented about 20% of the hematopoietic system of adult mice. Adult YFP+ hematopoietic stem cells constituted a long-term repopulating, transplantable population. Thus, a lineage of adult hematopoietic stem cells is characterized by the expression of GATA4 in their embryonic progenitors and probably by its extraembryonic (placental) origin, although GATA4 appeared not to be required for hematopoietic stem cell differentiation.     Carbonation rate and reinforcing steel corrosion of concretes with recycled concrete aggregates and supplementary cementing materials

    Because of the significant environmental impact that the concrete production causes, and as sustainability contribution for this industry; presently some improvements are being implemented in its durability and its components are been replaced for alternative recyclable materials. Concrete carbonation is one of the main causes of reinforcement electrochemical corrosion. Coarse Recycled Concrete Aggregates (RCA) and Supplementary Cementing Materials (SCM) such as fly ash and silica fume were used to produce durable and environmentally friendly concrete and for the evaluation of its carbonation depth and carbonation-induced reinforcement corrosion when it is exposed to accelerated conditions. The depth of carbonation was observed by spraying a phenolphthalein solution on the fresh broken concrete surface. The corrosion rates of steel embedded in these concretes have been measured by the lineal polarization resistance technique. The results indicate that to use RCA and SCM increase the carbonation depth and corrosion current density.Postprint (published version

    Como estrategias para contribuir en la sustentabilidad de la industria del concreto se está proyectando mejorar la durabilidad de las estructuras de concreto reforzado y el reemplazo parcial o total de sus ingredientes por materiales reciclables. En este estudio se evaluó la resistividad eléctrica y su relación con la microestructura y resistencia a la corrosión electroquímica de concretos reforzados fabricados con características de sustentabilidad, es decir, con agregados reciclados y materiales cementantes suplementarios. La resistividad eléctrica del concreto está íntimamente relacionada con la microestructura de la matriz cementante y con la estructura y distribución de poros. Por la naturaleza de sus ingredientes, es de suponerse que los concretos sustentables propuestos presentan diferente microestructura y porosidad que el concreto convencional, motivo por el cual se analizó la respuesta eléctrica y electroquímica de dichos sistemas mediante la técnica de espectroscopía de impedancia electroquímica.Peer ReviewedPostprint (published version

    Resumen de la tarea Rest-Mex en IberLEF 2023: Investigaci´on sobre An´alisis de Sentimiento para Textos Tur´ısticos Mexicanos

    This paper presents the framework and results of the Rest-Mex task at IberLEF 2023, focusing on sentiment analysis and text clustering of tourist texts. The study primarily focuses on texts related to tourist destinations in Mexico, although this edition included data from Cuba and Colombia for the first time. The sentiment analysis task aims to predict the polarity of opinions expressed by tourists, classifying the type of place visited, whether it’s a tourist attraction, hotel, or restaurant, as well as the country it is located in. On the other hand, the text clustering task aims to classify news articles related to tourism in Mexico. For both tasks, corpora were built using Spanish opinions extracted from TripAdvisor and news articles from Mexican media. This article compares and discusses the results obtained by the participants in both sub-tasks. Additionally, a method is proposed to measure the easiness of a multi-class text classification corpus, along with an approach for system selection in a possible late fusion scheme.Este artículo presenta el marco y los resultados de la tarea Rest-Mex en IberLEF 2023, que se enfoca en el análisis de sentimiento y agrupamiento de textos turísticos. El estudio se centra principalmente en textos relacionados con destinos turísticos en México, aunque esta edición incluyó datos de Cuba y Colombia por primera vez. La tarea de análisis de sentimiento tiene como objetivo predecir la polaridad de opiniones expresadas por turistas, clasificando el tipo de lugar visitado, ya sea un atractivo turístico, un hotel o un restaurante, así como el país en el que se encuentra. Por otro lado, la tarea de agrupamiento de textos busca clasificar noticias relacionadas con el turismo en México. Para ambas tareas, se construyeron corpus utilizando opiniones en español extraídas de TripAdvisor y noticias de medios mexicanos. En este artículo, se comparan y discuten los resultados obtenidos por los participantes en ambas sub tareas.     En los últimos años se han materializado algunas modificaciones normativas que han supuesto una clara apuesta por la contratación en régimen laboral en el ámbito de la investigación en detrimento de la política de becas y de los contratos de naturaleza administrativa, dando carta de naturaleza también a la contratación por tiempo indefinido. En este contexto el objeto de estudio del presente proyecto de investigación consiste en analizar el estatuto profesional del personal investigador contratado en régimen laboral por los organismos públicos de investigación de la administración general del estado y por las universidades públicas. A partir de aquí, sucede que muchas de las reflexiones y análisis que se van a efectuar en el trabajo son extrapolables a otros centros y organismos, tales como las universidades privadas y de la iglesia católica o ciertas entidades privadas de investigación sin ánimo de lucro.Realizado en el marco del Proyecto de investigación sobre «El Estatuto profesional del personal investigador contratado en régimen laboral» concedido por el Instituto Nacional de Administración Pública (INAP) con fecha 17 de febrero de 2015.     The Emissivity Of Foam-Covered Water Surface At L-Band: Theoretical Modeling And Experimental Results From The FROG 2003 Field Experiment

    Sea surface salinity can be measured by microwave radiometry at L-band (1400-1427 MHz). This frequency is a compromise between sensitivity to the salinity, small atmospheric perturbation, and reasonable pixel resolution. The description of the ocean emission depends on two main factors: (1) the sea water permittivity, which is a function of salinity, temperature, and frequency, and (2) the sea surface state, which depends on the wind-induced wave spectrum, swell, and rain-induced roughness spectrum, and by the foam coverage and its emissivity. This study presents a simplified two-layer emission model for foam-covered water and the results of a controlled experiment to measure the foam emissivity as a function of salinity, foam thickness, incidence angle, and polarization. Experimental results are presented, and then compared to the two-layer foam emission model with the measured foam parameters used as input model parameters. At 37 psu salt water the foam-induced emissivity increase is /spl sim/0.007 per millimeter of foam thickness (extrapolated to nadir), increasing with increasing incidence angles at vertical polarization, and decreasing with increasing incidence angles at horizontal polarization.Peer Reviewe

    Met signaling in cardiomyocytes is required for normal cardiac function in adult mice.

    Hepatocyte growth factor (HGF) and its receptor, Met, are key determinants of distinct developmental processes. Although HGF exerts cardio-protective effects in a number of cardiac pathologies, it remains unknown whether HGF/Met signaling is essential for myocardial development and/or physiological function in adulthood. We therefore investigated the requirement of HGF/Met signaling in cardiomyocyte for embryonic and postnatal heart development and function by conditional inactivation of the Met receptor in cardiomyocytes using the Cre-α-MHC mouse line (referred to as α-MHCMet-KO). Although α-MHCMet-KO mice showed normal heart development and were viable and fertile, by 6 months of age, males developed cardiomyocyte hypertrophy, associated with interstitial fibrosis. A significant upregulation in markers of myocardial damage, such as β-MHC and ANF, was also observed. By the age of 9 months, α-MHCMet-KO males displayed systolic cardiac dysfunction. Mechanistically, we provide evidence of a severe imbalance in the antioxidant defenses in α-MHCMet-KO hearts involving a reduced expression and activity of catalase and superoxide dismutase, with consequent reactive oxygen species accumulation. Similar anomalies were observed in females, although with a slower kinetics. We also found that Met signaling down-regulation leads to an increase in TGF-β production and a decrease in p38MAPK activation, which may contribute to phenotypic alterations displayed in α-MHCMet-KO mice. Consistently, we show that HGF acts through p38α to upregulate antioxidant enzymes in cardiomyocytes.     Environmental sensitivity differs between rabbit lines selectedfor reproductive intensity and longevity

    To better understand the mechanisms that allow some animals to sustain their productive effort in harsh environmental conditions, rabbit does from two selection lines (LP and V) were housed in normal (NC), nutritional (NF) or heat (HC) challenging environmental conditions from first to third partum. The LP line (n=85) was founded on reproductive longevity criteria by selecting does from commercial farms that had a minimum of 25 partum with more than 7.5 kits born alive per parity. Line V (n=79) was constituted from four specialised maternal lines into a composite synthetic line and then selected by litter size at weaning for 36 generations. Female rabbits in NC and NF environments were housed at normal room temperature (18 degrees C to 24 degrees C) and fed with control [11.6 MJ digestible energy (DE)/kg dry matter (DM)] or low-energy diets (9.1 MJ DE/kg DM). HC does were housed at high room temperatures (25 degrees C to 35 degrees C) and fed the control diet. Female rabbits in the HC and NF environments ingested 11.5% and 6% less DE than NC does, respectively (P<0.05). These differences between environments occurred in both lines, with the differences being higher for LP than for V does (+6%; P<0.05). Milk yield responses followed those of energy intake also being higher for LP does (+21.3 g/day; P<0.05). The environmental conditions did not affect the perirenal fat thickness (PFT), but a genotype by environment interaction was observed. In NC and HC, the PFT was higher for line V (+0.23 and +0.35 mm, respectively; P<0.05) than for LP does, but this was not the case at NF (-0.01 mm). Moreover, the PFT evolution was different between them. In the NC environment, LP does used the accreted PFT in late lactation (-0.29 mm), whereas V does did not (-0.08 mm). Conversely, in the HC environment, LP does showed a flat PFT evolution in late lactation, whereas V does accumulated PFT. In the NF environment, LP and V does had a similar PFT evolution. There was also a litter size reduction for V does of -2.59 kits total born in HC and -1.78 kits total born in NF environments, whereas this was not observed for LP does.     Detectable clonal mosaicism in blood as a biomarker of cancer risk in Fanconi anemia

    Detectable clonal mosaicism for large chromosomal events has been associated with aging and an increased risk of hematological and some solid cancers. We hypothesized that genetic cancer predisposition disorders, such as Fanconi anemia (FA), could manifest a high rate of chromosomal mosaic events (CMEs) in peripheral blood, which could be used as early biomarkers of cancer risk. We studied the prevalence of CMEs by single-nucleotide polymorphism (SNP) array in 130 FA patients' blood DNA and their impact on cancer risk. We detected 51 CMEs (4.4-159 Mb in size) in 16 out of 130 patients (12.3%), of which 9 had multiple CMEs. The most frequent events were gains at 3q (n = 6) and 1q (n = 5), both previously associated with leukemia, as well as rearrangements with breakpoint clustering within the major histocompatibility complex locus (P = 7.3 x 10(-9)). Compared with 15 743 age-matched population controls, FA patients had a 126 to 140 times higher risk of detectable CMEs in blood (P < 2.2 x 10(-16)). Prevalent and incident hematologic and solid cancers were more common in CME carriers (odds ratio [OR] = 11.6, 95% confidence interval [CI] = 3.4-39.3, P = 2.8 x 10(-5)), leading to poorer prognosis. The age-adjusted hazard risk (HR) of having cancer was almost 5 times higher in FA individuals with CMEs than in those without CMEs. Regarding survival, the HR of dying was 4 times higher in FA individuals having CMEs (HR = 4.0, 95% CI = 2.0-7.9, P = 5.7 x 10(-5)). Therefore, our data suggest that molecular karyotyping with SNP arrays in easy-to-obtain blood samples could be used for better monitoring of bone marrow clonal events, cancer risk, and overall survival of FA patients
