    The metabolic syndrome adds utility to the prediction of mortality over its components: The Vietnam Experience Study

    Background\ud The metabolic syndrome increases mortality risk. However, as “non-affected” individuals may still have up to two risk factors, the utility of using three or more components to identify the syndrome, and its predictive advantage over individual components have yet to be determined.\ud \ud Methods\ud Participants, male Vietnam-era veterans (n = 4265) from the USA, were followed-up from 1985/1986 for 14.7 years (61,498 person-years), and all-cause and cardiovascular disease deaths collated. Cox's proportional-hazards regression was used to assess the effect of the metabolic syndrome and its components on mortality adjusting for a wide range of potential confounders.\ud \ud Results\ud At baseline, 752 participants (17.9%) were identified as having metabolic syndrome. There were 231 (5.5%) deaths from all-causes, with 60 from cardiovascular disease. After adjustment for a range of covariates, the metabolic syndrome increased the risk of all-cause, HR 2.03, 95%CI 1.52, 2.71, and cardiovascular disease mortality, HR 1.92, 95%CI 1.10, 3.36. Risk increased dose-dependently with increasing numbers of components. The increased risk from possessing only one or two components was not statistically significant. The adjusted risk for four or more components was greater than for only three components for both all-cause, HR 2.30, 95%CI 1.45, 3.66 vs. HR 1.70, 95%CI 1.11, 2.61, and cardiovascular disease mortality, HR 3.34, 95%CI 1.19, 9.37 vs. HR 2.81, 95%CI 1.07, 7.35. The syndrome was more informative than the individual components for all-cause mortality, but could not be assessed for cardiovascular disease mortality due to multicollinearity. Hyperglycaemia was the individual strongest parameter associated with mortality.\ud \u

    Early postzygotic mutations contribute to de novo variation in a healthy monozygotic twin pair

    Cataloged from PDF version of article.Background: Human de novo single-nucleotide variation (SNV) rate is estimated to range between 0.82-1.70Ă—10-8 mutations per base per generation. However, contribution of early postzygotic mutations to the overall human de novo SNV rate is unknown. Methods: We performed deep whole-genome sequencing (more than 30-fold coverage per individual) of the whole-blood-derived DNA samples of a healthy monozygotic twin pair and their parents. We examined the genotypes of each individual simultaneously for each of the SNVs and discovered de novo SNVs regarding the timing of mutagenesis. Putative de novo SNVs were validated using Sanger-based capillary sequencing. Results: We conservatively characterised 23 de novo SNVs shared by the twin pair, 8 de novo SNVs specific to twin I and 1 de novo SNV specific to twin II. Based on the number of de novo SNVs validated by Sanger sequencing and the number of callable bases of each twin, we calculated the overall de novo SNV rate of 1.31Ă—10-8 and 1.01Ă—10-8 for twin I and twin II, respectively. Of these, rates of the early postzygotic de novo SNVs were estimated to be 0.34Ă—10-8 for twin I and 0.04Ă—10-8 for twin II. Conclusions: Early postzygotic mutations constitute a substantial proportion of de novo mutations in humans. Therefore, genome mosaicism resulting from early mitotic events during embryogenesis is common and could substantially contribute to the development of diseases

    Prevalence of metabolic syndrome-related disorders in a large adult population in Turkey

    BACKGROUND: There are few existing large population studies on the epidemiology of metabolic syndrome-related disorders of Turkey. The purpose of this study was to assess the prevalence of metabolic syndrome-related disorders in the Turkish adult population, to address sex, age, educational and geographical differences, and to examine blood pressure, body mass index, fasting blood glucose and serum lipids in Turkey. METHODS: This study was executed under the population study "The Healthy Nutrition for Healthy Heart Study" conducted between December 2000 and December 2002 by the Health Ministry of Turkey. Overall, 15,468 Caucasian inhabitants aged over 30 were recruited in 14 centers in the seven main different regions of Turkey. The data were analyzed with the Students' t, ANOVA or Chi-Square tests. RESULTS: Overall, more than one-third (35.08 %) of the participants was obese. The hypertensive people ratio in the population was 13.66 %, while these ratios for DM and metabolic syndrome were 4.16 % and 17.91 %, respectively. The prevalence of hypertension, metabolic syndrome and obesity were higher in females than males, whereas diabetes mellitus was higher in males than females. The prevalence of metabolic syndrome and related disorders were found to be significantly different across educational attainments for both men and women. The prevalence of hypertension increased with age, while it was remarkable that in the age group of 60–69 years, prevalence of diabetes mellitus and metabolic syndrome reached a peak value and than decreased. For obesity, the peak prevalence occurred in the 50–59 year old group. The prevalence of metabolic syndrome and related disorders were found to be significantly different according to geographical region. CONCLUSION: In conclusion, high prevalence of obesity and metabolic syndrome, particularly among women, is one of the major public health problems in Turkey. Interestingly, obesity prevalence is relatively high, but the prevalence of hypertension and hypercholesterolemia is relatively low in Turkish people. Future studies may focus on elucidating the reasons behind this controversy. Our findings may be helpful in formulating public health policy and prevention strategies on future health in Turkey

    Integrated modelling for sustainability assessment and decision making of alternative fuel buses

    In this paper, a hybrid life cycle sustainability assessment (LCSA) model integrating multi region input–output analysis with novel multi-criteria decision-making techniques is proposed to assess three different fuel alternatives: compressed natural gas (CNG), electric buses (EBs), and diesel buses (DBs). A global hybrid LCSA model first quantified the environmental, economic, and social impacts of alternative fuel buses. The results were investigated in terms of multiple combinations of manufacturing and end-of-life scenarios by encompassing impacts embedded in the global supply chains taking Qatar as a case applied to the proposed model. The Interval-Valued Neutrosophic Fuzzy (IVNF)-Analytic Hierarchy Process with the Combined Compromise Solution (CoCoSo) approach is used to rank the alternative fuel buses based on their corresponding sustainability performance. The proposed model will help in quantitatively capturing the macrolevel life cycle socioeconomic and environmental impacts along with optimally selecting alternatives to support sustainable urban transport policy towards a net-zero transportation system globally

    Eye movements as a window to cognitive processes

    Eye movement research is a highly active and productive research field. Here we focus on how the embodied nature of eye movements can act as a window to the brain and the mind. In particular, we discuss how conscious perception depends on the trajectory of fixated locations and consequently address how fixation locations are selected. Specifically, we argue that the selection of fixation points during visual exploration can be understood to a large degree based on retinotopically structured models. Yet, these models largely ignore spatiotemporal structure in eye-movement sequences. Explaining spatiotemporal structure in eye-movement trajectories requires an understanding of spatiotemporal properties of the visual sampling process. With this in mind, we discuss the availability of external information to internal inference about causes in the world. We demonstrate that visual foraging is a dynamic process that can be systematically modulated either towards exploration or exploitation. For an analysis at high temporal resolution, we suggest a new method: The renewal density allows the investigation of precise temporal relation of eye movements and other actions like a button press. We conclude with an outlook and propose that eye movement research has reached an appropriate stage and can easily be combined with other research methods to utilize this window to the brain and mind to its fullest

    Insights into the emerging networks of voids in simulated supercooled water

    The structural evolution of supercooled liquid water as we approach the glass transition temperature continues to be an active area of research. Here, we use molecular dynamics simulations of TIP4P/ice water to study the changes in the connected regions of empty space within the liquid, which we investigate using the Voronoi-voids network. We observe two important features: supercooling enhances the fraction of nonspherical voids and different sizes of voids tend to cluster forming a percolating network. By examining order parameters such as the local structure index (LSI), tetrahedrality and topological defects, we show that water molecules near large void clusters tend to be slightly more tetrahedral than those near small voids, with a lower population of under- and overcoordinated defects. We show further that the distribution of closed rings of water molecules around small and large void clusters maintain a balance between 6 and 7 membered rings. Our results highlight the changes of the dual voids and water network as a structural hallmark of supercooling and provide insights into the molecular origins of cooperative effects underlying density fluctuations on the subnanometer and nanometer length scale. In addition, the percolation of the voids and the hydrogen bond network around the voids may serve as useful order parameters to investigate density fluctuations in supercooled water

    How sustainable is liquefied natural gas supply chain? An integrated life cycle sustainability assessment model

    Integrating sustainability into the distribution network process is a significant problem for any industry hoping to prosper or survive in today's fast-paced environment. Since gas is one of the world's most important fuel sources, sustainability is more important for the gas industry. While such environmental and economic effects have been extensively researched in the literature, there is little emphasis on the full social sustainability of natural gas production and supply chains in terms of the triple bottom line. This research aims to perform the first hybrid life cycle sustainability assessment (LCSA) of liquefied natural gas and evaluate its performance from the natural gas extraction stage to LNG regasification after delivery through maritime transport carriers. LCSA is used for estimating the social, economic, and environmental impacts of processes, and our life cycle model included the multi-region input–output analysis, Aspen HYSYS, and LNG maritime transport operations sustainability assessment tools. The results spot the light on the most contributors of CO2-eq emission. It is found that LNG loading (export terminal) is the source that generated the highest carbon footprint, followed by the MDEA sweetening unit with the contribution of 40% and 24%, respectively. Socially, around 73% of human health impact comes from SRU and TGTU units which are the most contributors to the particulate matter emission. Based on the interpretation of life cycle results, the environmental indicators show better performance in the pre-separation unit and LNG receiving terminal representing a sustainability factor equal to 1. In terms of social and economic impacts, the natural gas extraction stage presents the best performance among all other stages, with a sustainability factor equal to 1. Based on this study's findings, an integrated framework model is proposed. Various suggestions for sustainability strategies and policies that consider business sustainability and geopolitics risk are presented
