The transfer of asymmetric molecular information to the lateral plate mesoderm is essential for the precise patterning of asymmetric internal organs, such as the heart. However, at the same developmental time, it is crucial to maintain symmetry at the somite level for correct musculature and vertebrae specification. We demonstrate how left-right signals affect the behavior of zebrafish somite cell precursors by using live imaging and fate mapping studies in dand5 homozygous mutants compared to wildtype embryos. We describe a population of cells in the vicinity of the LRO, named Non-KV Sox17:GFP+ Tailbud Cells (NKSTCs), which migrate anteriorly and contribute to future somites. We show that NKSTCs originate in a cluster of cells aligned with the midline, posterior to the LRO, and leave that cluster in a left-right alternating manner, primarily from the left side. Fate mapping revealed that more NKSTCs integrated somites on the left side of the embryo. We then abolished the asymmetric cues from the LRO using dand5−/− mutant embryos and verified that NKSTCs no longer displayed asymmetric patterns. Cell exit from the posterior cluster became bilaterally synchronous in dand5−/− mutants. Our study revealed a new link between somite specification and Dand5 function. The gene dand5 is well known as the first asymmetric gene involved in vertebrate LR development. This study revealed a new link for Dand5 as a player in cell exit from the maturation zone into the presomitic mesoderm, affecting the expression patterns of myogenic factors and tail size.publishersversionpublishe

    Adaptação dos estudantes com deficiência no ensino superior: a experiência de 6 estudantes com deficiência da FCS da UAN

    Após o alcance da paz em 2002 a melhoria do sistema de ensino surge na agenda do governo de forma expressiva, através da Estratégia Integrada para Melhoria do Sistema de Educação. Foram empreendidos esforços visando assegurar uma educação inclusiva, equitativa que se alinhassem aos Objectivos de Desenvolvimento do Milénio (ODM) assumidos por Angola. Os esforços empreendidos não significaram necessariamente melhorias em todos os subsistemas de ensino e em todas as modalidades de ensino. A realidade angolana mostra que, as necessidades das pessoas com deficiência nem sempre têm sido consideradas, visando a sua inclusão nas diversas instituições de ensino, com destaque para o ensino superior. No presente artigo, procura-se descrever e analisar experiências de 6 estudantes com deficiência visual e auditiva, matriculados na Faculdade de Ciências Sociais da Universidade Agostinho Neto (FCS-UAN), em Luanda. Os casos e depoimentos referidos apresentam-se como dados ilustrativos de uma realidade mais vasta. O estudo foi realizado durante o primeiro semestre do ano lectivo 2018, com base em entrevistas semiestruturadas. Observações e informações obtidas através das entrevistas, sugerem que o processo de inclusão de pessoas com deficiência em instituições do ensino superior público em Angola, coloca grandes e novos desafios, tanto para as instituições de ensino superior, como para os estudantes. A situação é agravada devido à falta de preparação dos docentes para lidar com estudantes com deficiência, falta de material de apoio e de estruturas físicas concebidas à altura dos novos desafios educacionais

    An extended set of PRDM1/BLIMP1 target genes links binding motif type to dynamic repression

    The transcriptional repressor B lymphocyte-induced maturation protein-1 (BLIMP1) regulates gene expression and cell fate. The DNA motif bound by BLIMP1 in vitro overlaps with that of interferon regulatory factors (IRFs), which respond to inflammatory/immune signals. At such sites, BLIMP1 and IRFs can antagonistically regulate promoter activity. In vitro motif selection predicts that only a subset of BLIMP1 or IRF sites is subject to antagonistic regulation, but the extent to which antagonism occurs is unknown, since an unbiased assessment of BLIMP1 occupancy in vivo is lacking. To address this, we identified an extended set of promoters occupied by BLIMP1. Motif discovery and enrichment analysis demonstrate that multiple motif variants are required to capture BLIMP1 binding specificity. These are differentially associated with CpG content, leading to the observation that BLIMP1 DNA-binding is methylation sensitive. In occupied promoters, only a subset of BLIMP1 motifs overlap with IRF motifs. Conversely, a distinct subset of IRF motifs is not enriched amongst occupied promoters. Genes linked to occupied promoters containing overlapping BLIMP1/IRF motifs (e.g. AIM2, SP110, BTN3A3) are shown to constitute a dynamic target set which is preferentially activated by BLIMP1 knock-down.     COOPEDU IV — Cooperação e Educação de Qualidade

    O quarto Congresso Internacional de Cooperação e Educação-IV COOPEDU, organizado pelo Centro de Estudos Internacionais (CEI) do Instituto Universitário de Lisboa e pela Escola Superior de Educação e Ciências Sociais do Instituto Politécnico de Leiria decorreu nos dias 8 e 9 de novembro de 2018, subordinado à temática Cooperação e Educação de Qualidade. Este congresso insere-se numa linha de continuidade de intervenção por parte das duas instituições organizadoras e dos elementos coordenadores e este ano beneficiou do financiamento do Instituto Camões, obtido através de um procedimento concursal, que nos permitiu contar com a participação presencial de elementos dos Países Africanos de Língua Portuguesa, fortemente implicados nas problemáticas da Educação e da Formação. Contou também com a participação do Instituto Camões e da Fundação Calouste Gulbenkian, entidades que sistematizaram a sua intervenção nos domínios da cooperação na área da educação nos últimos anos. A opção pela temática da qualidade pareceu aos organizadores pertinente e actual. Com efeito os sistemas educativos dos países que constituem a Comunidade de países de língua portuguesa têm implementado várias reformas mas em vários domínios mantem-se a insatisfação de responsáveis políticos, pedagogos, técnicos sociais face aos resultados obtidos. Aliás o caminho de procura da Qualidade é interminável porque vai a par da aposta na exigência e na promoção da cidadania e responsabilidade social. As comunicações que agora se publicam estão organizadas em dois eixos: o das Políticas da Educação e Formação e o das dimensões em que se traduzem essas políticas. Neste último eixo encontramos fios condutores para agregarmos as comunicações apresentadas

    Prevalence, associated factors and outcomes of pressure injuries in adult intensive care unit patients: the DecubICUs study

Epidemiologic data is however unavailable. We aimed to provide an international picture of the extent of pressure injuries and factors associated with ICU-acquired pressure injuries in adult ICU patients. Methods: International 1-day point-prevalence study; follow-up for outcome assessment until hospital discharge (maximum 12 weeks). Factors associated with ICU-acquired pressure injury and hospital mortality were assessed by generalised linear mixed-effects regression analysis. Results: Data from 13,254 patients in 1117 ICUs (90 countries) revealed 6747 pressure injuries; 3997 (59.2%) were ICU-acquired. Overall prevalence was 26.6% (95% confidence interval [CI] 25.9–27.3). ICU-acquired prevalence was 16.2% (95% CI 15.6–16.8). Sacrum (37%) and heels (19.5%) were most affected. Factors independently associated with ICU-acquired pressure injuries were older age, male sex, being underweight, emergency surgery, higher Simplified Acute Physiology Score II, Braden score 3 days, comorbidities (chronic obstructive pulmonary disease, immunodeficiency), organ support (renal replacement, mechanical ventilation on ICU admission), and being in a low or lower-middle income-economy. Gradually increasing associations with mortality were identified for increasing severity of pressure injury: stage I (odds ratio [OR] 1.5; 95% CI 1.2–1.8), stage II (OR 1.6; 95% CI 1.4–1.9), and stage III or worse (OR 2.8; 95% CI 2.3–3.3). Conclusion: Pressure injuries are common in adult ICU patients. ICU-acquired pressure injuries are associated with mainly intrinsic factors and mortality. Optimal care standards, increased awareness, appropriate resource allocation, and further research into optimal prevention are pivotal to tackle this important patient safety threat

    Fungal endophyte communities in above- and belowground olive tree organs and the effect of season and geographic location on their structures

    Studies comparing fungal endophytes between above- and belowground woody crop organs and the factors that may structure their communities are lacking. Due to its great impact on the Mediterranean Basin, the olive tree was chosen for the isolation of endophytic fungi from roots, leaves and twigs in two seasons in north-eastern Portugal. Nine hundred seventy-six isolates belonging to 38 fungal species were obtained. Phomopsis columnaris, Fusarium oxysporum and Trichoderma gamsii were the most frequently isolated, collectively representing 69% of the isolates. Fungal diversity in the roots was higher than in the aboveground organs and higher in spring than autumn. Endophyte community similarity between the above- and belowground organs and between seasons was low. Species composition also varied spatially, with the fungal composition of the roots varying more among locations than that of the aboveground organs.     Influencia dos hábitos de sucção na manifestação de dor temporo-mandibular: revisão sistemática

    Objective: To assess the attitudes and knowledge about hepatitis by scholars and dental practitioners from Recife and its metropolitan region. Method: The research protocol was submitted and approved by the Ethics and Human Research Committee of the Federal University of Pernambuco. There were included 230 undergraduate students in dentistry and 104 dentists who exercise their professional activity in Recife and its Metropolitan Region. They signed a consent form and answered a questionnaire, the evaluative instrument, with objective questions about biosafety, transmission of hepatitis and sexually transmitted diseases. Results: About the types of hepatitis 133 (57.8%) students and 61 (58.7%) professionals claimed to know the types A, B and C. Concerning transmission 31 (13.5%) students and 25 (24.0%) professionals said that the main route of transmission were sexual and bloodstream respectively. Relating to imunization 221 (96.1%) students and 99 (95.2%) professionals reported that have been vaccinated against hepatitis B, however, only 126 (54.8%) students and 55 (52.9%) professionals have followed the immunization’s schema. Regarding the cleaning of the office equipment, 18.7% of the students replied that they do not know who does it. 56.7% of professionals said that it was performed with alcohol 70 ° GL.Conclusions: The findings indicate that there is a concern for biosafety but the knowledge about the forms of hepatitis transmission and biosecurity measures need to be better settled.Este estudo teve como objetivo realizar uma revisão sistemática da literatura para avaliar os efeitos dos hábitos parafuncionais de sucção sobre a ATM e estruturas adjacentes em crianças. Os artigos incluí- dos foram identificados por meio de pesquisas bibliográficas da Bireme, Pubmed, ClinicalTrials.gov, The National Research Register e Pro-Quest Dissertation Abstracts and Thesis, usando os termos de busca temporomandibular joint, sucking behavior and fingersucking. As pesquisas sobre os hábitos atuais ou anteriores de chupar o dedo e/ou uso de mamadeiras e chupetas e a presença de dor na ATM com crian- ças até 12 anos foram incluídas. Oito estudos foram selecionados, e 4 apresentaram uma associação, mas sem significância estatística. Os achados científicos demonstram uma associação baixa ou ausente de DTM com hábitos de sucção parafuncionais

    Accuracy of radiographic examination in determining the depth of approximal carious lesions in primary molars

    The purpose of this study was to evaluate the accuracy of the depth of carious lesions on bitewing radiographs. Methods Recently extracted primary molars had their proximal surfaces evaluated visually (EC) and classified as healthy surface (0), signs that suggest the presence of carious lesions in enamel (1), signs of a superficial lesion in dentin (2) and carious lesions in deep dentin (3). Results The results were obtained by consensus between the investigators. The gold standard was determined by histological analysis. The values of sensitivity, specificity, accuracy and area under the ROC (Receiver Operating Characteristic) curve were evaluated. There was equilibrium between sensitivity (76.92% EC and 88.46% ER) and specificity (95.83% EC and 95.83% ER). Accuracy was 86.01% (EC) and 88.46% (ER). The Spearman correlation test was used to prove the correlation between clinical and radiographic examinations (0.886), for clinical and histological (0.736) and for radiographic and histological analysis (0.843).Avaliar a exatidão da profundidade da lesão de cárie em radiografias interproximais. Métodos Molares decíduos recentemente extraídos tiveram suas superfícies proximais avaliadas visualmente (exame clínico) e classificados como superfície hígida (0), sinais que sugerem presença de lesão de cárie em esmalte (1), sinais de lesão superficial em dentina (2) e cavidades sugerindo lesões profundas em dentina (3). As superfícies dentárias foram também examinadas em radiografias interproximais (exame radiográfico)e classificadas como ausência de radiolucidez (0), lesão de cárie em esmalte (1), lesão de cárie superficial em dentina (2) e lesão de cárie em dentina profunda (3). Resultados Os resultados foram obtidos por meio de consenso entre os examinadores. O padrão ouro foi determinado pela análise histológica. Foram avaliados os valores de sensibilidade, especificidade, acurácia e área sob a curva Receiver Operating Characteristic. Observou-se um equilíbrio entre os valores de sensibilidade (76,9% exame clínico e 88,4% exame radiográfico) e especificidade (95,8% exame clínico e 95,8% exame radiográfico). A acurácia foi de 86,0% (exame clínico) e 88,4% (exame radiográfico). O teste de correlação de Spearman foi utilizado para a comprovação da correlação entre os exames clínico e radiográfico (0,89), para clínico e histológico (0,7) e para os exames radiográfico e histológico (0,8). Conclusão O exame radiográfico apresentou melhor desempenho na detecção da lesão de cárie proximal em dentes decíduos
