Contribution of the Twin-Arginine Translocation System to the Intracellular Survival of Salmonella Typhimurium in Dictyostelium discoideum

The twin-arginine translocation (Tat) system is a specialized secretion pathway required for bacteria to export fully folded proteins through the cytoplasmic membrane. This system is crucial during Salmonella infection of animal hosts. In this study, we show that Salmonella enterica serovar Typhimurium (S. Typhimurium) requires the Tat system to survive and proliferate intracellularly in the social amoeba Dictyostelium discoideum. To achieve this, we developed a new infection assay to assess intracellular bacterial loads in amoeba by direct enumeration of colony forming units (CFU) at different times of infection. Using this assay we observed that a ΔtatABC mutant was internalized in higher numbers than the wild type, and was defective for intracellular survival in the amoeba at all times post infection evaluated. In addition, we assessed the effect of the ΔtatABC mutant in the social development of D. discoideum. In contrast to the wild-type strain, we observed that the mutant was unable to delay the social development of the amoeba at 2 days of co-incubation. This phenotype correlated with defects in intracellular proliferation presented by the ΔtatABC mutant in D. discoideum after 24 h of infection. All phenotypes described for the mutant were reverted by the presence of a plasmid carrying tatABC genes, indicating that abrogation of Tat system attenuates S. Typhimurium in this model organism. Overall, our results indicate that the Tat system is crucial for S. Typhimurium to survive and proliferate intracellularly in D. discoideum and for virulence in this host. To the best of our knowledge, this is the first report on the relevance of the Tat system in the interaction of any bacterial pathogen with the social amoeba D. discoideum

Diabetes and neuropathy have been linked to postural instability. Aims: The purpose of this study is to determine how each system involved in balance is affected when challenged in a static standing posture. The goal was to identify postural instability by measuring balance through the amount of sway and weight bearing distribution in non-neuropathic controlled type II diabetics. Methods: Twelve participants (five males and seven females) with controlled type II diabetes mellitus and no history of peripheral neuropathy (Non-PN cDMII) formed the diabetic group, whereas eighteen participants (7 males and 11 females) without type II diabetes formed the control group. The exclusion criteria was applied via a series of screening tests (Berg Balance Scale, Five Times Sit To Stand Test, Functional Reach Test and Monofilament Test). Postural stability and weight distribution during quiet standing were measured using a Tekscan Matscan pressure mat, which measured the amount of sway and weight distribution. Static postural control was evaluated during eight sensory conditions that perturbed or stimulated the visual, proprioceptive, and vestibular systems. Results: Postural control was found to be significantly deficient when the vestibular system was stimulated, whereas the proprioceptive system was perturbed. After the data analysis, there was a significant difference in antero-posterior sway (P=0.05) with the following tasks: eyes open with head movements on an unstable surface, and eyes closed with head movements on a firm surface. Conclusions: The results revealed that the experimental group with type II diabetes had greater postural instability when compared to the control group during tasks with the vestibular system on an unstable surface. This demonstrates that, due to their deficits, individuals with non-PN cDMII are unable to maintain their balance when the vestibular and proprioceptive systems are challenged simultaneously

Absence of molecular evidence of Leptospira spp. in urine samples collected from rodents captured in Yucatán, México

Leptospira spp. is a spirochete bacteria, causal agent of leptospirosis, zoonotic disease endemic in México that represents a serious public health and veterinary problem. Rodents are recognised as the most important reservoirs of this bacteria, which is transmitted mainly through direct or indirect contact with the Leptospira spp. excreted in the urine of infected individuals. Theaim of this study was to evaluate the circulation of Leptospira spp. in urine samples of wild and synanthropic rodents from Yucatán, México. Eighty-four rodents were captured in the community of Cenotillo, Yucatán. Twenty-six urine samples were collected from the bladder and were used in the total DNA extraction. The identification of Leptospira spp. was intended through the polymerase chain reaction test in its endpoint variant. No evidence of Leptospira spp. was found in the urine samples. It is necessary to use other tissues for the identification of Leptospira spp., before concluding that the rodents used in the present study are not reservoirs of this bacteri

Molecular detection of pathogenic Leptospira in synanthropic and wild rodents captured in Yucatán, México

Introduction: Leptospirosis is a zoonotic disease caused by bacteria of the genus Leptospira, which is endemic in México and considered a public and veterinary health problem. Rodents are the most relevant reservoirs of Leptospira spp. because the bacteria establish and reproduce in its renal tissue and are excreted through the urine. Objective: To identify the presence of Leptospira spp. in renal tissue from rodents captured in Yucatán, México. Materials and methods: Synanthropic and wild rodents were captured in the rural municipality of Cenotillo, Yucatán, México. We collected one kidney from each rodent and extracted the total DNA. The identification of Leptospira spp. was done by detecting two fragments of the 16S rRNA gene using end-point polymerase chain reaction (PCR). We sequenced and analyzed positive products using alignment tools. Results: A total of 92 rodents belonging to seven different species were captured. The PCR yielded a global positivity of 5.4% (5/92). The alignment analysis of the sequenced products demonstrated a 100% of coverage and identity with Leptospira interrogans. This is the first molecular evidence of Leptospira spp. circulation in Heteromys gaumeri captured in Yucatán, México. Conclusion: Our results evidenced that rodents of Yucatán are reservoirs of Leptospira spp. and participate in the infection cycle of leptospirosis in the region
