Epstein-Barr Virus–Associated Gastric Carcinoma: The Americas’ Perspective
Epstein-Barr virus (EBV) infection has been associated with different malignancies, and compelling evidence has shown that it may have a causative or at least contributing role in gastric carcinogenesis. EBV-associated gastric cancers have a unique molecular signature, which has defined this group of tumors as a distinctive molecular subtype of gastric cancer. This subtype has shown a greater incidence in the Americas than in the Asian countries. This chapter discusses about possible factors underlying these differences and the emerging roles of epigenetics in the pathogenesis of Epstein-Barr virus–associated gastric cancer
Stabilizing bubble and droplet interfaces using dipeptide hydrogels
Hydrophobic dipeptide molecules can be used to create interfacial films covering bubbles and droplets made from a range of oils. At high pH, the dipeptide molecules form micelles which transform into a hydrogel of fibres in response to the addition of salt. We characterize the properties of the hydrogel for two different salt (MgSO4) concentrations and then we use these gels to stabilize interfaces. Under high shear, the hydrogel is disrupted and will reform around bubbles or droplets. Here, we reveal that at low dipeptide concentration, the gel is too weak to prevent ripening of the bubbles; this then reduces the long-term stability of the foam. Under the same conditions, emulsions prepared from some oils are highly stable. We examine the wetting properties of the oil droplets at a hydrogel surface as a guide to the resulting emulsions
Validation of the Women’s Views of Birth Labor Satisfaction Questionnaire (WOMBLSQ4) in the Spanish Population
The satisfaction of women with the birth experience has implications for the health and
wellness of the women themselves and also of their newborn baby. The objectives of this study were
to determine the factor structure of the Women’s Views of Birth Labor Satisfaction Questionnaire
(WOMBLSQ4) questionnaire on satisfaction with the attention received during birth delivery in
Spanish women and to compare the level of satisfaction of pregnant women during the birth process
with that in other studies that validated this instrument. A cross-sectional study using a self-completed
questionnaire of 385 Spanish-speaking puerperal women who gave birth in the Public University
Hospitals of Granada (Spain) was conducted. An exploratory factor analysis of the WOMBLSQ4
questionnaire was performed to identify the best fit model. Those items that showed commonalities
higher than 0.50 were kept in the questionnaire. Using the principal components method, nine factors
with eigenvalues greater than one were extracted after merging pain-related factors into a single item.
These factors explain 90% of the global variance, indicating the high internal consistency of the full
scale. In the model resulting from the WOMBLSQ4 questionnaire, its nine dimensions measure the
levels of satisfaction of puerperal women with childbirth care. Average scores somewhat higher than
those of the original questionnaire and close to those achieved in the study carried out in Madrid
(Spain) were obtained. In clinical practice, this scale may be relevant for measuring the levels of
satisfaction during childbirth of Spanish-speaking women
The effect of l -thymidine, acyclic thymine and 8-bromoguanine on the stability of model G-quadruplex structures
Sensitive and label-free biosensing of RNA with predicted secondary structures by a triplex affinity capture method
A novel biosensing approach for the label-free detection of nucleic acid sequences of short and large lengths has been implemented, with special emphasis on targeting RNA sequences with secondary structures. The approach is based on selecting 8-aminoadenine-modified parallel-stranded DNA tail-clamps as affinity bioreceptors. These receptors have the ability of creating a stable triplex-stranded helix at neutral pH upon hybridization with the nucleic acid target. A surface plasmon resonance biosensor has been used for the detection. With this strategy, we have detected short DNA sequences (32-mer) and purified RNA (103-mer) at the femtomol level in a few minutes in an easy and level-free way. This approach is particularly suitable for the detection of RNA molecules with predicted secondary structures, reaching a limit of detection of 50 fmol without any label or amplification steps. Our methodology has shown a marked enhancement for the detection (18% for short DNA and 54% for RNA), when compared with the conventional duplex approach, highlighting the large difficulty of the duplex approach to detect nucleic acid sequences, especially those exhibiting stable secondary structures. We believe that our strategy could be of great interest to the RNA field
Gis1 and Rph1 Regulate Glycerol and Acetate Metabolism in Glucose Depleted Yeast Cells
Aging in organisms as diverse as yeast, nematodes, and mammals is delayed by caloric restriction, an effect mediated by the nutrient sensing TOR, RAS/cAMP, and AKT/Sch9 pathways. The transcription factor Gis1 functions downstream of these pathways in extending the lifespan of nutrient restricted yeast cells, but the mechanisms involved are still poorly understood. We have used gene expression microarrays to study the targets of Gis1 and the related protein Rph1 in different growth phases. Our results show that Gis1 and Rph1 act both as repressors and activators, on overlapping sets of genes as well as on distinct targets. Interestingly, both the activities and the target specificities of Gis1 and Rph1 depend on the growth phase. Thus, both proteins are associated with repression during exponential growth, targeting genes with STRE or PDS motifs in their promoters. After the diauxic shift, both become involved in activation, with Gis1 acting primarily on genes with PDS motifs, and Rph1 on genes with STRE motifs. Significantly, Gis1 and Rph1 control a number of genes involved in acetate and glycerol formation, metabolites that have been implicated in aging. Furthermore, several genes involved in acetyl-CoA metabolism are downregulated by Gis1
