    Harnessing Evaluation and Learning for Equity and Impact Insights for Foundation Executives

    Grantmaking foundations are increasingly using E&L functions in diverse ways. A 2019 survey from the Center for Evaluation Innovation found that 42 percent of foundations had a dedicated E&L unit or department that functioned separately from the program department, up from 34 percent in 2015.The survey also shows that E&L staff fill numerous and evolving roles including directing and managing evaluation work within the foundation; supporting broader team and organizational learning efforts, including equity work; supporting strategy development and review; and providing advice or coaching about evaluation to other staff. Beyond the foundation's walls, many E&L teams contribute to the fields of evaluation and philanthropy by sharing actionable knowledge or learning strategies with peer organizations. Field-building increases the influence and impact of a foundation by helping to advance philanthropic thought leadership and E&L practices more broadly.But leveraging the power of evaluation and learning is easier said than done. This guide is a resource for foundation executives interested in harnessing the power of evaluation and learning for impact. It was developed by Engage R+D with support from The James Irvine Foundation, Ewing Marion Kauffman Foundation, Center for Evaluation Innovation, and Kresge Foundation. It is based on our study of the E&L function across these three diverse foundations, all of which champion the Equitable Evaluation Initiative and are on their own equity journeys

    This brief grew out of conversations with evaluation and learning leaders working in foundations across the United States about both the value of evaluation and learning in philanthropy, and the challenges of implementing this function well across diverse institutional contexts. Our intent is to provide practical guidance that new and existing leaders can use to navigate their roles in support of more effective and equitable philanthropy. It is based on indepth case studies of the Irvine, Kauffman, and Kresge Foundations along with our own experience partnering with foundations on evaluation, strategy, and learning efforts

    How do graduate students approach college teaching? Influences of professional development, teaching assistantships, and Big Five personality traits

    Introduction: Graduate students engage in college teaching with varied attitudes and approaches. Their teaching practices may be influenced by professional development experiences related to pedagogy, and their personality traits. Methods: Through an online survey of graduate students teaching undergraduate courses (N = 109, 69.7% women, M age = 30 years, 59% psychology), we examined whether self-reported participation in professional development related to pedagogy, teaching assistantship (TA) experience, academic discipline (psychology vs. other), and Big Five personality traits were associated with variation in teaching practices. Results: Participation in professional development correlated positively with years of undergraduate teaching experience and with the trait of openness. Hierarchical regressions indicated that professional development served to promote student-focused practices and discouraged lecturing, while TA experience (mostly restricted to psychology instructors) promoted lecturing and discouraged a student-focused approach. Regarding academic discipline, psychology instructors gave higher endorsements to an information transmission, teacher-focused approach to teaching, lectured more, and were less likely to provoke debate than instructors of other disciplines. Such differences may be attributed in part to larger enrollments of psychology courses. Regarding personality traits, both openness and agreeableness were associated with some student-focused practices, while conscientiousness was associated with an information transmission, teacher-focused approach and with practices aligned with backward course design. Discussion: In light of previous evidence that personality traits are malleable, graduate training programs may want to cultivate traits like openness as a means of encouraging graduate students to reflect on their pedagogy and seek ways to improve their teaching through professional development. Relatedly, graduate programs should aim to support students’ participation in professional development related to pedagogy and, in doing so, communicate its value

    Kinase inhibitor pulldown assay identifies a chemotherapy response signature in triple-negative breast cancer based on purine-binding proteins

    UNLABELLED: Triple-negative breast cancer (TNBC) constitutes 10%-15% of all breast tumors. The current standard of care is multiagent chemotherapy, which is effective in only a subset of patients. The original objective of this study was to deploy a mass spectrometry (MS)-based kinase inhibitor pulldown assay (KIPA) to identify kinases elevated in non-pCR (pathologic complete response) cases for therapeutic targeting. Frozen optimal cutting temperature compound-embedded core needle biopsies were obtained from 43 patients with TNBC before docetaxel- and carboplatin-based neoadjuvant chemotherapy. KIPA was applied to the native tumor lysates that were extracted from samples with high tumor content. Seven percent of all identified proteins were kinases, and none were significantly associated with lack of pCR. However, among a large population of off-target purine-binding proteins (PBP) identified, seven were enriched in pCR-associated samples ( SIGNIFICANCE: The identification of pretreatment predictive biomarkers for pCR in response to neoadjuvant chemotherapy would advance precision treatment for TNBC. To complement standard proteogenomic discovery profiling, a KIPA was deployed and unexpectedly identified a seven-member non-kinase PBP pCR-associated signature.     Oxygen as a Driver of Early Arthropod Micro-Benthos Evolution

    BACKGROUND: We examine the physiological and lifestyle adaptations which facilitated the emergence of ostracods as the numerically dominant Phanerozoic bivalve arthropod micro-benthos. METHODOLOGY/PRINCIPAL FINDINGS: The PO(2) of modern normoxic seawater is 21 kPa (air-equilibrated water), a level that would cause cellular damage if found in the tissues of ostracods and much other marine fauna. The PO(2) of most aquatic breathers at the cellular level is much lower, between 1 and 3 kPa. Ostracods avoid oxygen toxicity by migrating to waters which are hypoxic, or by developing metabolisms which generate high consumption of O(2). Interrogation of the Cambrian record of bivalve arthropod micro-benthos suggests a strong control on ecosystem evolution exerted by changing seawater O(2) levels. The PO(2) of air-equilibrated Cambrian-seawater is predicted to have varied between 10 and 30 kPa. Three groups of marine shelf-dwelling bivalve arthropods adopted different responses to Cambrian seawater O(2). Bradoriida evolved cardiovascular systems that favoured colonization of oxygenated marine waters. Their biodiversity declined during intervals associated with black shale deposition and marine shelf anoxia and their diversity may also have been curtailed by elevated late Cambrian (Furongian) oxygen-levels that increased the PO(2) gradient between seawater and bradoriid tissues. Phosphatocopida responded to Cambrian anoxia differently, reaching their peak during widespread seabed dysoxia of the SPICE event. They lacked a cardiovascular system and appear to have been adapted to seawater hypoxia. As latest Cambrian marine shelf waters became well oxygenated, phosphatocopids went extinct. Changing seawater oxygen-levels and the demise of much of the seabed bradoriid micro-benthos favoured a third group of arthropod micro-benthos, the ostracods. These animals adopted lifestyles that made them tolerant of changes in seawater O(2). Ostracods became the numerically dominant arthropod micro-benthos of the Phanerozoic.     Anticoagulant selection in relation to the SAMe-TT2R2 score in patients with atrial fibrillation. the GLORIA-AF registry

    Aim: The SAMe-TT2R2 score helps identify patients with atrial fibrillation (AF) likely to have poor anticoagulation control during anticoagulation with vitamin K antagonists (VKA) and those with scores >2 might be better managed with a target-specific oral anticoagulant (NOAC). We hypothesized that in clinical practice, VKAs may be prescribed less frequently to patients with AF and SAMe-TT2R2 scores >2 than to patients with lower scores. Methods and results: We analyzed the Phase III dataset of the Global Registry on Long-Term Oral Antithrombotic Treatment in Patients with Atrial Fibrillation (GLORIA-AF), a large, global, prospective global registry of patients with newly diagnosed AF and ≥1 stroke risk factor. We compared baseline clinical characteristics and antithrombotic prescriptions to determine the probability of the VKA prescription among anticoagulated patients with the baseline SAMe-TT2R2 score >2 and ≤ 2. Among 17,465 anticoagulated patients with AF, 4,828 (27.6%) patients were prescribed VKA and 12,637 (72.4%) patients an NOAC: 11,884 (68.0%) patients had SAMe-TT2R2 scores 0-2 and 5,581 (32.0%) patients had scores >2. The proportion of patients prescribed VKA was 28.0% among patients with SAMe-TT2R2 scores >2 and 27.5% in those with scores ≤2. Conclusions: The lack of a clear association between the SAMe-TT2R2 score and anticoagulant selection may be attributed to the relative efficacy and safety profiles between NOACs and VKAs as well as to the absence of trial evidence that an SAMe-TT2R2-guided strategy for the selection of the type of anticoagulation in NVAF patients has an impact on clinical outcomes of efficacy and safety. The latter hypothesis is currently being tested in a randomized controlled trial.     Data Descriptor: An open resource for transdiagnostic research in pediatric mental health and learning disorders

    Technological and methodological innovations are equipping researchers with unprecedented capabilities for detecting and characterizing pathologic processes in the developing human brain. As a result, ambitions to achieve clinically useful tools to assist in the diagnosis and management of mental health and learning disorders are gaining momentum. To this end, it is critical to accrue large-scale multimodal datasets that capture a broad range of commonly encountered clinical psychopathology. The Child Mind Institute has launched the Healthy Brain Network (HBN), an ongoing initiative focused on creating and sharing a biobank of data from 10,000 New York area participants (ages 5–21). The HBN Biobank houses data about psychiatric, behavioral, cognitive, and lifestyle phenotypes, as well as multimodal brain imaging (resting and naturalistic viewing fMRI, diffusion MRI, morphometric MRI), electroencephalography, eyetracking, voice and video recordings, genetics and actigraphy. Here, we present the rationale, design and implementation of HBN protocols.     Formulation, stabilisation and encapsulation of bacteriophage for phage therapy

    Against a backdrop of global antibiotic resistance and increasing awareness of the importance of the human microbiota, there has been resurgent interest in the potential use of bacteriophages for therapeutic purposes, known as phage therapy. A number of phage therapy phase I and II clinical trials have concluded, and shown phages don’t present significant adverse safety concerns. These clinical trials used simple phage suspensions without any formulation and phage stability was of secondary concern. Phages have a limited stability in solution, and undergo a significant drop in phage titre during processing and storage which is unacceptable if phages are to become regulated pharmaceuticals, where stable dosage and well defined pharmacokinetics and pharmacodynamics are de rigueur. Animal studies have shown that the efficacy of phage therapy outcomes depend on the phage concentration (i.e. the dose) delivered at the site of infection, and their ability to target and kill bacteria, arresting bacterial growth and clearing the infection. In addition, in vitro and animal studies have shown the importance of using phage cocktails rather than single phage preparations to achieve better therapy outcomes. The in vivo reduction of phage concentration due to interactions with host antibodies or other clearance mechanisms may necessitate repeated dosing of phages, or sustained release approaches. Modelling of phage-bacterium population dynamics reinforces these points. Surprisingly little attention has been devoted to the effect of formulation on phage therapy outcomes, given the need for phage cocktails, where each phage within a cocktail may require significantly different formulation to retain a high enough infective dose. This review firstly looks at the clinical needs and challenges (informed through a review of key animal studies evaluating phage therapy) associated with treatment of acute and chronic infections and the drivers for phage encapsulation. An important driver for formulation and encapsulation is shelf life and storage of phage to ensure reproducible dosages. Other drivers include formulation of phage for encapsulation in micro- and nanoparticles for effective delivery, encapsulation in stimuli responsive systems for triggered controlled or sustained release at the targeted site of infection. Encapsulation of phage (e.g. in liposomes) may also be used to increase the circulation time of phage for treating systemic infections, for prophylactic treatment or to treat intracellular infections. We then proceed to document approaches used in the published literature on the formulation and stabilisation of phage for storage and encapsulation of bacteriophage in micro- and nanostructured materials using freeze drying (lyophilization), spray drying, in emulsions e.g. ointments, polymeric microparticles, nanoparticles and liposomes. As phage therapy moves forward towards Phase III clinical trials, the review concludes by looking at promising new approaches for micro- and nanoencapsulation of phages and how these may address gaps in the field
