    Open archives and instruments of quality management as resources for disseminating scientific information in occupational health and safety

    The objective of this study is to present the philosophy of open archives as an instrument for occupational health and safety (OHS) management. The survey method stands for the bibliography research, together with an interview session with the 23 OSH management professionals that enabled the further understanding about access level and the perception on scientific information by these professionals. Qualitative and quantitative questionnaire analysis showed the open archives initiative, through digital information repositories as an instrument for the occupational health and safety management, as well as need of developing policies on incentive to generation and sharing of intellectual capital among the community in question, encouraging the construction of an OSH digital repository, for maintaining most of all the worker's physical and mental integrity. Lack of available information in this area is still a barrier to the occupational knowledge management for promoting the safety and healthy work development in the organizations.O objetivo do estudo foi apresentar a utilização da filosofia de arquivos abertos, aliada aos instrumentos de gestão da qualidade, PDCA e 5S, como um dos pilares para a gestão da segurança e saúde no trabalho (SST).     A Base de Exportação do Oeste Paranaense 2000/2010/2020

    Diversifying the export base of a region can be a way to be followed for regional development. This study seeks to identify the export profile of Western Paraná in terms of international trade in goods between 2000, 2010 and 2020. Classified as a quantitative research, descriptive case study and using secondary data, from the analysis of data provided by the Ministry of Economy, Foreign Trade Statistics in Open Data (Comex Stat). Data on the economy of the Paraná State and its regional and national importance are presented.     Dietary Cholesterol-Induced Post-Testicular Infertility

    This work shows that an overload of dietary cholesterol causes complete infertility in dyslipidemic male mice (the Liver X Receptor-deficient mouse model). Infertility resulted from post-testicular defects affecting the fertilizing potential of spermatozoa. Spermatozoa of cholesterol-fed lxr−/− animals were found to be dramatically less viable and motile, and highly susceptible to undergo a premature acrosome reaction. We also provide evidence, that this lipid-induced infertility is associated with the accelerated appearance of a highly regionalized epididymal phenotype in segments 1 and 2 of the caput epididymidis that was otherwise only observed in aged LXR-deficient males. The epididymal epithelial phenotype is characterized by peritubular accumulation of cholesteryl ester lipid droplets in smooth muscle cells lining the epididymal duct, leading to their transdifferentiation into foam cells that eventually migrate through the duct wall, a situation that resembles the inflammatory atherosclerotic process. These findings establish the high level of susceptibility of epididymal sperm maturation to dietary cholesterol overload and could partly explain reproductive failures encountered by young dyslipidemic men as well as ageing males wishing to reproduce

    A EFICÁCIA DO TREINO MUSCULAR RESPIRATÓRIO NA FASE I DE REABILITAÇÃO CARDÍACA: UM ESTUDO DE CASO

    A cirurgia cardíaca está entre a terapêutica atual das cardiopatias, e apesar de seus benefícios pode propiciar redução da força muscular respiratória, dos volumes e capacidades pulmonares. A aplicação de diferentes modalidades da fisioterapia respiratória tem sido utilizada, entre elas o Treino Muscular Respiratório (TMR). O objetivo do presente estudo avalia a eficácia do TMR associado a protocolo de reabilitação cardíaca na qualidade de vida e função pulmonar de um paciente submetido à Cirurgia de Revascularização do Miocárdio (CRM). Para isso foi realizado um estudo de caso com um paciente submetido a CRM, onde o mesmo foi submetido a um protocolo de reabilitação cardíaca associada ao TMR com Threshold®, as medidas foram avaliadas no pré-operatório, 1° dia do pós operatório (PO) e 6°PO. Foi observado que o paciente apresentava uma percepção negativa da qualidade de vida já no pré-operatório, mas apresentou aumento de alguns domínios no pós-operatório, apontando uma melhora da percepção de saúde, com piora dos domínios “dor” e “aspectos sociais”. Em relação a função pulmonar, o paciente apresentou queda de todas as variáveis no 1°PO, confirmando que o procedimento cirúrgico repercute na função pulmonar, melhorando no 6°PO, com aumento mais expressivo para volume corrente (VC), que retornou ao valor inicial, e pressão inspiratória máxima (PImáx), que foi maior que no pré-operatório. Constatou-se que o TMR associado a um protocolo convencional de reabilitação cardíaca pode proporcionar melhora ao paciente submetido a cirurgia cardíaca. Acredita-se que estudos randomizados e com diferentes amostras possam ilustrar melhor os benefícios dessa terapia

    Expression of TIGIT, PD-1 and HLA-DR/CD38 markers on CD8-T cells of children and adolescents infected with HIV and uninfected controls

    Immune exhaustion and senescence are scarcely studied in HIV-pediatric patients. We studied the circulatory CD8 T cells activation/exhaustion and senescent phenotype of children and adolescents vertically infected with HIV or uninfected controls based on the expression of human leukocyte antigen (HLA-DR), CD38, T cell immunoglobulin and immunoreceptor tyrosine-based inhibitory motif (ITIM) domain (TIGIT), programmed death 1 (PD-1) and CD57 by flow cytometry, during approximately one year. Eleven HIV-infected (HI) and nine HIV-uninfected (HU) children/adolescents who received two doses or one dose of meningococcal C conjugate vaccine (MenC), respectively, were involved in this study. Blood samples were collected before the immunization (T0), 1–2 months after the first dose (T1), and 1–2 months after the second dose (T2), which was administered approximately one year after the first one. HI patients not receiving combined antiretroviral therapy (cART) showed a higher frequency of CD8 T cells TIGIT+, PD-1+ or CD57+, as well as a higher frequency of CD8 T cells co-expressing CD38/HLA-DR/TIGIT or CD38/HLA-DR/PD-1 when compared to HI treated or HU individuals, at all times that they were assessed. CD8 T cells co-expressing CD38/DR/TIGIT were inversely correlated with the CD4/CD8 ratio but positively associated with viral load. The co-expression of CD38/DR/TIGIT or CD38/DR/PD-1 on CD8 T cells was also inversely associated with the CD4 T cells expressing co-stimulatory molecules CD127/CD28. The results showed a higher expression of exhaustion/senescence markers on CD8 T cells of untreated HI children/adolescents and its correlations with viral load

    Structural and functional stabilization of bacteriophage particles within the aqueous core of a W/O/W multiple emulsion: a potential biotherapeutic system for the inhalational treatment of bacterial pneumonia

    The increase of antibiotic-resistant bacteria is growing every day, most likely associated with the indiscriminate use of these antimicrobials or even with evolutionary adaptability of bacteria to their environment. This situation brings a need to develop new alternatives to conventional antibiotics, and thus the application of strictly lytic bacteriophages has been proposed as an alternative (or complement) to the former, allowing release of the natural predators of bacteria directly where they are needed the most: the infection site. The main advantages of bacteriophages to treat infections is the maintenance of a high concentration of bacteriophage particles in the action site while any viable target bacteria still exist, coupled to the production of enzymes that hydrolyze the polymeric matrix of bacterial biofilms promoting penetration and antibacterial action. In the research effort entertained herein, the potential for protection and stabilization of strictly lytic bacteriophages with broad spectrum capable of infecting Pseudomonas aeruginosa, so as to maintain their structure and functionality, was investigated via encapsulation within the aqueous-core of lipid nanodroplets integrating a W/O/W multiple emulsion system, aiming at developing isotonic derivative solutions thereof for administration by nebulization.Project funding by FAPESP (São Paulo, Brazil; Refs.     Genome-wide association and HLA fine-mapping studies identify risk loci and genetic pathways underlying allergic rhinitis

    Allergic rhinitis is the most common clinical presentation of allergy, affecting 400 million people worldwide, with increasing incidence in westernized countries1,2. To elucidate the genetic architecture and understand the underlying disease mechanisms, we carried out a meta-analysis of allergic rhinitis in 59,762 cases and 152,358 controls of European ancestry and identified a total of 41 risk loci for allergic rhinitis, including 20 loci not previously associated with allergic rhinitis, which were confirmed in a replication phase of 60,720 cases and 618,527 controls. Functional annotation implicated genes involved in various immune pathways, and fine mapping of the HLA region suggested amino acid variants important for antigen binding. We further performed genome-wide association study (GWAS) analyses of allergic sensitization against inhalant allergens and nonallergic rhinitis, which suggested shared genetic mechanisms across rhinitis-related traits. Future studies of the identified loci and genes might identify novel targets for treatment and prevention of allergic rhinitis
