    Lead Speciation and Bioavailability in Apatite-Amended Sediments

    The in situ sequestration of lead (Pb) in sediment with a phosphate amendment was investigated by Pb speciation and bioavailability. Sediment Pb in preamendment samples was identified as galena (PbS) with trace amounts of absorbed Pb. Sediment exposed to atmospheric conditions underwent conversion to hydrocerussite and anglesite. Sediments mixed with apatite exhibited limited conversion to pyromorphite, the hypothesized end product. Conversion of PbS to pyromorphite is inhibited under reducing conditions, and pyromorphite formation appears limited to reaction with pore water Pb and PbS oxidation products. Porewater Pb values were decreased by 94% or more when sediment was amended with apatite. The acute toxicity of the sediment Pb was evaluated with Hyalella azteca and bioaccumulation of Pb with Lumbriculus variegatus. The growth of H. azteca may be mildly inhibited in contaminated sediment, with apatite-amended sediments exhibiting on average a higher growth weight by approximately 20%. The bioaccumulation of Pb in L. variegatus tissue decreased with increased phosphate loading in contaminated sediment. The study indicates limited effectiveness of apatite in sequestering Pb if present as PbS under reducing conditions, but sequestration of porewater Pb and stabilization of near-surface sediment may be a feasible and alternative approach to decreasing potential toxicity of Pb

    Confronting herbicide resistance with cooperative management

    Resistance of pathogens and pests to antibiotics and pesticides worldwide is rapidly reaching critical levels. The common-pool-resource nature of this problem (i.e., whereby the susceptibility to treatment of target organisms is a shared resource) has been largely overlooked. Using herbicide-resistant weeds as a model system, we developed a discrete-time landscape-scale simulation to investigate how aggregating herbicide management strategies at different spatial scales from individual farms to larger cooperative structures affects the evolution of glyphosate resistance in common waterhemp (Amaranthus tuberculatus).Our findings indicate that high-efficacy herbicide management strategies practiced at the farm scale are insufficient to slow resistance evolution in A. tuberculatus. When best practices were aggregated at large spatial scales, resistance evolution was hindered; conversely, when poor management practices were aggregated, resistance was exacerbated. Tank mixture-based strategies were more effective than rotation-based strategies in most circumstances, while applying glyphosate alone resulted in the poorest outcomes.Our findings highlight the importance of landscape-scale cooperative management for confronting common-pool-resource resistance problems in weeds and other analogous systems.     The impact of intraoperative fluid management during laparoscopic donor nephrectomy on donor and recipient outcomes

    BackgroundIntraoperative fluid management during laparoscopic donor nephrectomy (LDN) may have a significant effect on donor and recipient outcomes. We sought to quantify variability in fluid management and investigate its impact on donor and recipient outcomes.MethodsA retrospective review of patients who underwent LDN from July 2011 to January 2016 with paired kidney recipients at a single center was performed. Patients were divided into tertiles of intraoperative fluid management (standard, high, and aggressive). Donor and recipient demographics, intraoperative data, and postoperative outcomes were analyzed.ResultsOverall, 413 paired kidney donors and recipients were identified. Intraoperative fluid management (mL/h) was highly variable with no correlation to donor weight (kg) (R = 0.017). The aggressive fluid management group had significantly lower recipient creatinine levels on postoperative day 1. However, no significant differences were noted in creatinine levels out to 6 months between groups. No significant differences were noted in recipient postoperative complications, graft loss, and death.     The human ACC2 CT-domain C-terminus is required for full functionality and has a novel twist

    Inhibition of acetyl-CoA carboxylase (ACC) may prevent lipid-induced insulin resistance and type 2 diabetes, making the enzyme an attractive pharmaceutical target. Although the enzyme is highly conserved amongst animals, only the yeast enzyme structure is available for rational drug design. The use of biophysical assays has permitted the identification of a specific C-terminal truncation of the 826-residue human ACC2 carboxyl transferase (CT) domain that is both functionally competent to bind inhibitors and crystallizes in their presence. This C-terminal truncation led to the determination of the human ACC2 CT domain–CP-640186 complex crystal structure, which revealed distinctions from the yeast-enzyme complex. The human ACC2 CT-domain C-terminus is com­prised of three intertwined α-helices that extend outwards from the enzyme on the opposite side to the ligand-binding site. Differences in the observed inhibitor conformation between the yeast and human structures are caused by differing residues in the binding pocket

    An evaluation of different meta-analysis approaches in the presence of allelic heterogeneity

    Meta-analysis has proven a useful tool in genetic association studies. Allelic heterogeneity can arise from ethnic background differences across populations being meta-analyzed (for example, in search of common frequency variants through genome-wide association studies), and through the presence of multiple low frequency and rare associated variants in the same functional unit of interest (for example, within a gene or a regulatory region). The latter challenge will be increasingly relevant in whole-genome and whole-exome sequencing studies investigating association with complex traits. Here, we evaluate the performance of different approaches to meta-analysis in the presence of allelic heterogeneity. We simulate allelic heterogeneity scenarios in three populations and examine the performance of current approaches to the analysis of these data. We show that current approaches can detect only a small fraction of common frequency causal variants. We also find that for low-frequency variants with large effects (odds ratios 2–3), single-point tests have high power, but also high false-positive rates. P-value based meta-analysis of summary results from allele-matching locus-wide tests outperforms collapsing approaches. We conclude that current strategies for the combination of genetic association data in the presence of allelic heterogeneity are insufficiently powered

    Climate warming, marine protected areas and the ocean-scale integrity of coral reef ecosystems

    Coral reefs have emerged as one of the ecosystems most vulnerable to climate variation and change. While the contribution of a warming climate to the loss of live coral cover has been well documented across large spatial and temporal scales, the associated effects on fish have not. Here, we respond to recent and repeated calls to assess the importance of local management in conserving coral reefs in the context of global climate change. Such information is important, as coral reef fish assemblages are the most species dense vertebrate communities on earth, contributing critical ecosystem functions and providing crucial ecosystem services to human societies in tropical countries. Our assessment of the impacts of the 1998 mass bleaching event on coral cover, reef structural complexity, and reef associated fishes spans 7 countries, 66 sites and 26 degrees of latitude in the Indian Ocean. Using Bayesian meta-analysis we show that changes in the size structure, diversity and trophic composition of the reef fish community have followed coral declines. Although the ocean scale integrity of these coral reef ecosystems has been lost, it is positive to see the effects are spatially variable at multiple scales, with impacts and vulnerability affected by geography but not management regime. Existing no-take marine protected areas still support high biomass of fish, however they had no positive affect on the ecosystem response to large-scale disturbance. This suggests a need for future conservation and management efforts to identify and protect regional refugia, which should be integrated into existing management frameworks and combined with policies to improve system-wide resilience to climate variation and change

    Measurement of three-jet differential cross sections d sigma-3jet / d M-3jet in p anti-p collisions at sqrt(s)=1.96 TeV

    We present the first measurement of the inclusive three-jet differential cross section as a function of the invariant mass of the three jets with the largest transverse momenta in an event in p anti-p collisions at sqrt(s) = 1.96 TeV. The measurement is made in different rapidity regions and for different jet transverse momentum requirements and is based on a data set corresponding to an integrated luminosity of 0.7 fb^{-1} collected with the D0 detector at the Fermilab Tevatron Collider. The results are used to test the three-jet matrix elements in perturbative QCD calculations at next-to-leading order in the strong coupling constant. The data allow discrimination between parametrizations of the parton distribution functions of the proton.Comment: 10 pages, 4 figures, 2 tables, submitted to Phys. Lett.     Measurement of the production of a W boson in association with a charm quark in pp collisions at √s = 7 TeV with the ATLAS detector

    The production of a W boson in association with a single charm quark is studied using 4.6 fb−1 of pp collision data at s√ = 7 TeV collected with the ATLAS detector at the Large Hadron Collider. In events in which a W boson decays to an electron or muon, the charm quark is tagged either by its semileptonic decay to a muon or by the presence of a charmed meson. The integrated and differential cross sections as a function of the pseudorapidity of the lepton from the W-boson decay are measured. Results are compared to the predictions of next-to-leading-order QCD calculations obtained from various parton distribution function parameterisations. The ratio of the strange-to-down sea-quark distributions is determined to be 0.96+0.26−0.30 at Q 2 = 1.9 GeV2, which supports the hypothesis of an SU(3)-symmetric composition of the light-quark sea. Additionally, the cross-section ratio σ(W + +cÂŻÂŻ)/σ(W − + c) is compared to the predictions obtained using parton distribution function parameterisations with different assumptions about the s−sÂŻÂŻÂŻ quark asymmetry
