    Antifungal effect of volatile organic compounds produced by Bacillus amyloliquefaciens CPA-8 against fruit pathogen decays of cherry

    The present work focuses on the antifungal effect of volatile organic compounds (VOCs) produced by Bacillus amyloliquefaciens CPA-8 against Monilinia laxa, M. fructicola and Botrytis cinera, three postharvest fruit pathogens of sweet cherry fruit. VOCs were evaluated with a double petri dish assay against mycelial and colony growth of target pathogens. For this purpose, CPA-8 was grown on different media and cultured for 24 and 48 h at 30 °C before assays. Data showed that mycelial growth inhibition was higher when CPA-8 was grown on Tryptone Soya Agar (TSA) while no differences were generally observed when CPA-8 was cultured for either, 24 and 48 h. Moreover, no effects were observed on colony growth. The main volatile compounds emitted by CPA-8 were identified by solid-phase microextraction (SPME)-gas chromatography as 1,3 pentadiene, acetoin (3-hydroxy-2-butanone) and thiophene. Pure compounds were also tested in vitro on mycelial growth inhibition and their EC50 values against the three pathogens were estimated. Thiophene was the most effective VOC, showing more than 82% suppression of mycelial growth at the highest concentration (1.35 μL/mL headspace) and EC50 values ranging from 0.06 to 6.67 μL/mL headspace. Finally, the effectiveness of thiophene and CPA-8 VOCs was evaluated against artificially inoculated cherry fruits. Among the target pathogens, M. fructicola was clearly controlled by CPA-8 with less than 25% of rotten fruits compared to the control (65% disease incidence) and for all pathogens, less than 37.5% of CPA-8 treated decayed fruits produced spores (disease sporulation). Otherwise, pure thiophene showed no effect against any pathogen on disease incidence and disease sporulation. The results indicated that VOCs produced by B. amyloliquefaciens CPA-8 could develop an additive antifungal effect against postharvest fruit pathogens on stone fruit.This research was supported by the European project BIOCOMES FP7-612713 and by the Catalan government (Generalitat de Catalunya) for the PhD grant 2016-FI-B2 00143 (Amparo M.     Efectos de la entrevista motivacional en la salud oral de pacientes pediátricos

    Objetivo: determinar la efectividad a corto plazo de una intervención basada en la entrevista motivacional (EM). en la reducción del índice de placa bacteriana en niños de entre 7 y 12 años (segunda infancia), en comparación con el modelo informativo tradicional (MIT). Métodos y diseño: con este objetivo fue realizado un ensayo clínico aleatorizado a doble ciego, con 30 pacientes y dos grupos activos de intervención. Al inicio del estudio se examinaron todos los participantes en relación con el índice de placa bacteriana (O'Leary) y. posteriormente, fueron asignados de fomia aleatoria al grupo experimental (una sola sesión EM) y al gnipo control (una sola sesión MIT). ambas con una duración aproximada de 15 minutos. A excepción de esta sesión, todos los participantes siguieron el mismo protocolo de tratamiento. Resultados: a los 3 meses postintervención, la placa bacteriana determinada por el índice de O'Leary disminuyó en ambos grupos (76. l frente a 69.8^f). con una diferencia altamente significativa para p 0,05 (t = 0,78; p = 0,441). Conclusiones: desde una perspectiva a corto plazo, una sesión breve de EM logró reducir los índices de placa bacteriana en pacientes infantiles, pero no demostró ser más eficaz que el MIT

    Mechanisms of resistance in postharvest fruit-pathogen interaction

    Abstract T he objective of this review was to bring together concepts related to studies aimed at elucidating defense mechanisms against disease-causing agents, mainly in postharvest. Like plants, fruits are exposed to attack by pathogens that cause rot during postharvest storage, resulting in considerable losses. To control these pathogens, synthetic chemicals are used; however, since they are toxic, genetic resistance is regarded as a viable alternative. Fruits can withstand pathogens by means of physical barriers (presence of thick cuticular or trichome layers) and chemical ones, or through induced defenses that are activated once the host detects the presence of the pathogen, triggering the oxidative burst during the early hours of interaction. This burst entails the generation of reactive oxygen species (ROS), such as superoxide (O 2ˉ) , hydroxyl radical (OHˉ) or hydrogen peroxide (H 2 O 2 ), and the activation of genes involved in several metabolic pathways. The study of such mechanisms may allow detecting disease-resistant genetic materials, thus reducing the use of toxic products. Resumen E l objetivo de esta revisión fue conjuntar conceptos relacionados con estudios dirigidos a elucidar los mecanismos de defensa contra agentes causantes de enfermedades, principalmente en poscosecha. Al igual que las plantas, los frutos se encuentran expuestos al ataque por patógenos que producen podredumbres durante su almacenamiento en poscosecha, causando considerables pérdidas. Para el control de dichos patógenos, se emplean productos químicos de síntesis que son tóxicos, y la resistencia genética se considera una alternativa viable. Los frutos pueden tolerar a los patógenos mediante barreras físicas (presencia de capas gruesas de cutícula o de tricomas) y químicas, o bien, a través de defensas inducidas que se activan una vez que el huésped detecta la presencia del patógeno, desencadenando la explosión oxidativa durante las primeras horas de la interacción. Esta explosión conlleva la generación de especies reactivas de oxígeno (ROS) como el superóxido (O 2ˉ) , el radical hidroxilo (OHˉ) o el peróxido de hidrógeno (H 2 O 2 ), y la activación de genes involucrados en diversas rutas metabólicas. El estudio de tales mecanismos puede permitir detectar materiales genéticos resistentes a enfermedades, reduciendo así el uso de productos tóxicos

    Severity-Related Changes of Bronchial Microbiome in Chronic Obstructive Pulmonary Disease

    Bronchial colonization by potentially pathogenic microorganisms (PPMs) is often demonstrated in chronic obstructive pulmonary disease (COPD), but culture-based techniques identify only a portion of the bacteria in mucosal surfaces. The aim of the study was to determine changes in the bronchial microbiome of COPD associated with the severity of the disease. The bronchial microbiome of COPD patients was analyzed by 16S rRNA gene amplification and pyrosequencing in sputum samples obtained during stable disease. Seventeen COPD patients were studied (forced expiratory volume in the first second expressed as a percentage of the forced vital capacity [FEV1%] median, 35.0%; interquartile range [IQR], 31.5 to 52.0), providing a mean of 4,493 (standard deviation [SD], 2,598) sequences corresponding to 47 operational taxonomic units (OTUs) (SD, 17) at a 97% identity level. Patients were dichotomized according to their lung function as moderate to severe when their FEV1% values were over the median and as advanced when FEV1% values were lower. The most prevalent phyla in sputum were Proteobacteria (44%) and Firmicutes (16%), followed by Actinobacteria (13%). A greater microbial diversity was found in patients with moderate-to-severe disease, and alpha diversity showed a statistically significant decrease in patients with advanced disease when assessed by Shannon (ρ = 0.528; P = 0.029, Spearman correlation coefficient) and Chao1 (ρ = 0.53; P = 0.028, Spearman correlation coefficient) alpha-diversity indexes. The higher severity that characterizes advanced COPD is paralleled by a decrease in the diversity of the bronchial microbiome, with a loss of part of the resident flora that is replaced by a more restricted microbiota that includes PPMs

    Terminological challenges in the translation of science documentaries: a case-study

    This article aims to describe some of the main terminological problems audiovisual translators have to face when dealing with the translation of science documentaries, specifically in the English-Catalan combination. The first section of the article presents some theoretical concepts which underlie this research and which are taken, for the most part, from Cabré's Communicative Theory of Terminology. Then, specific terminological problems audiovisual translators have to solve are described using the data provided by a corpus of four science documentaries lasting approximately 50 minutes each. These challenges include identifying a term, understanding a term, finding the right equivalent, dealing with the absence of an adequate equivalent, solving denominative variations, choosing between in vivo and in vitro terminology, and overcoming mistranscriptions

    The challenge of the laboratory diagnosis in a confirmed congenital Zika virus syndrome in utero: A case report

    Introduction: Zika virus (ZIKV) has caused one of the most challenging global infectious epidemics in recent years because of its causal association with severe microcephaly and other congenital malformations. The diagnosis of viral infections usually relies on the detection of virus proteins or genetic material in clinical samples as well as on the infected host immune responses. Serial serologic testing is required for the diagnosis of congenital infection when diagnostic molecular biology is not possible. Patient concerns: A 2-year-old girl, born to a mother with confirmed ZIKV infection during pregnancy, with a confirmed ZIKV infection in utero, showed at birth a severe microcephaly and clinical characteristics of fetal brain disruption sequence compatible with a congenital ZIKV syndrome (CZS). Diagnosis: ZIKV-RNA and ZIKV-IgM serological response performed at birth and during the follow-up time tested always negative. Serial serologic ZIKV-IgG tests were performed to assess the laboratory ZIKV diagnosis, ZIKV-IgG seroreversion was observed at 21 months of age. ZIKV diagnosis of this baby had to be relied on her clinical and radiological characteristics that were compatible with a CZS. Interventions: The patient was followed-up as per protocol at approximately 1, 4, 9, 12, 18–21, and 24 months of age. Neurological, radiological, audiological, and ophthalmological assessment were performed during this period of time. Prompt rehabilitation was initiated to prevent potential adverse long-term neurological outcomes. Outcomes: The growth of this girl showed a great restriction at 24 months of age with a weight of 8.5 kg (−2.5 z-score) and a head circumference of 40.5 cm (−4.8 z-score). She also had a great neurodevelopmental delay at the time of this report. Conclusion: We presume that as a consequence of prenatal ZIKV infection, the fetal brain and other organs are damaged before birth through direct injury. Following this, active infection ends during intrauterine life, and as a consequence the immune system of the infant is unable to build up a consistent immune response thereafter. Further understanding of the mechanisms taking part in the pathogenesis of ZIKV congenital infection is needed. This finding might change our paradigm regarding serological response in the ZIKV congenital infection

    Adjuvant dabrafenib and trametinib for patients with resected BRAF-mutated melanoma: DESCRIBE-AD real-world retrospective observational study

    BRAF and MEK inhibitor, dabrafenib plus trametinib, adjuvant therapy is effective for high-risk resected melanoma patients with BRAF-V600 mutations. However, real-world evidence is limited. We aimed to determine the feasibility of this therapy in routine clinical practice. DESCRIBE-AD, a retrospective observational study, collected real-world data from 25 hospitals in Spain. Histologically confirmed and resected BRAF-mutated melanoma patients aged & GE;18 years who were previously treated with dabrafenib plus trametinib adjuvant therapy, were included. The primary objectives were treatment discontinuation rate and time to discontinuation. The secondary objectives included safety and efficacy. From October 2020 to March 2021, 65 patients were included. Dabrafenib and trametinib discontinuation rate due to treatment-related adverse events (TRAEs) of any grade was 9%. Other reasons for discontinuation included patients' decisions (6%), physician decisions (6%), unrelated adverse events (3%), disease progression (5%), and others (5%). The median time to treatment discontinuation was 9 months [95% confidence interval (CI), 5-11]. G3-4 TRAEs occurred in 21.5% of patients, the most common being pyrexia (3%), asthenia (3%), and diarrhoea (3%). Unscheduled hospitalisations and clinical tests occurred in 6 and 22% of patients, respectively. After 20-month median follow-up (95% CI, 18-22), 9% of patients had exitus due to disease progression, with a 12-month relapse-free survival and overall survival rates of 95.3% and 100%, respectively. Dabrafenib and trametinib adjuvant therapy proved effective for melanoma patients in a real-world setting, with a manageable toxicity profile. Toxicity frequencies were low leading to low incidence of unscheduled medical visits, tests, and treatment discontinuations

    Does Banque de France control inflation and unemployment?

    We re-estimate statistical properties and predictive power of a set of Phillips curves, which are expressed as linear and lagged relationships between the rates of inflation, unemployment, and change in labour force. For France, several relationships were estimated eight years ago. The change rate of labour force was used as a driving force of inflation and unemployment within the Phillips curve framework. The set of nested models starts with a simplistic version without autoregressive terms and one lagged term of explanatory variable. The lag is determined empirically together with all coefficients. The model is estimated using the Boundary Element Method (BEM) with the least squares method applied to the integral solutions of the differential equations. All models include one structural break might be associated with revisions to definitions and measurement procedures in the 1980s and 1990s as well as with the change in monetary policy in 1994-1995. For the GDP deflator, our original model provided a root mean squared forecast error (RMSFE) of 1.0% per year at a four-year horizon for the period between 1971 and 2004. The rate of CPI inflation is predicted with RMSFE=1.5% per year. For the naive (no change) forecast, RMSFE at the same time horizon is 2.95% and 3.3% per year, respectively. Our model outperforms the naive one by a factor of 2 to 3. The relationships for inflation were successfully tested for cointegration. We have formally estimated several vector error correction (VEC) models for two measures of inflation. At a four year horizon, the estimated VECMs provide significant statistical improvements on the results obtained by the BEM: RMSFE=0.8% per year for the GDP deflator and ~1.2% per year for CPI. For a two year horizon, the VECMs improve RMSFEs by a factor of 2, with the smallest RMSFE=0.5% per year for the GDP deflator.Comment: 25 pages, 12 figure
