Clinical markers for identifying cholinergic deficits in Parkinson's disease

BackgroundCholinergic projection systems degeneration is associated with dopamine nonresponsive features of Parkinson's disease (PD). Cholinergic deficits are variable in nondemented PD. Identification of cholinergic deficits in PD may help with selection of suitable patients for targeted cholinergic drug treatment in PD. The objective of this retrospective multivariate predictor analysis study was to identify clinical markers indicative of cholinergic deficits in PD patients, as assessed by acetylcholinesterase ([11C]PMP) positron emission tomography.MethodsOne hundred thirty‐seven PD patients (34 female) participated; median modified Hoehn and Yahr score was 2.5 (range, 1‐4), average age 65.6 ± 7.4 years, and average duration of motor disease symptoms of 6.0 ± 4.2 years. Subjects were dichotomized as “normocholinergic” or “hypocholinergic” based on a 5th percentile cutoff from normal for the basal forebrain‐cortical and pedunculopontine nucleus‐thalamic cholinergic projection systems. Previously identified clinical indices of cholinergic denervation were used for statistical prediction of cholinergic deficits. Logistic regression determined which risk factors predicted cholinergic deficits. Sensitivity, specificity, and accuracy were determined for the (combinations of) significant predictor variables.ResultsForty‐nine (35.8%) hypocholinergic PD subjects were identified. The combination of rapid eye movement (REM) sleep behavior disorder (RBD) symptoms and fall history showed highest diagnostic accuracy (81.1%) for predicting combined thalamic and cortical cholinergic deficits. A combined assessment of 8.5 m walk time and lower score on the Montreal cognitive assessment scale provided diagnostic accuracy of 80.7% for predicting isolated cortical cholinergic denervation.ConclusionAssessment of clinical indices of cholinergic denervation may be useful for identifying suitable subjects for trials of targeted cholinergic drug treatments in PD. © 2014 International Parkinson and Movement Disorder SocietyPeer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/110743/1/mds26061.pd

Long-term annual and monthly changes in mysids and caridean decapods in a macrotidal estuarine environment in relation to climate change and pollution

© 2018 Elsevier B.V. A 26-year time series of monthly samples from the water intake of a power station has been used to analyse the trends exhibited by number of species, total abundance, and composition of the mysids and caridean decapods in the inner Bristol Channel. During this period, annual water temperatures, salinities and the North Atlantic Oscillation Index (NAOI) in winter did not change significantly, whereas annual NAOI declined. Annual mean monthly values for the number of species and total abundance both increased over the 26 years, but these changes were not correlated with any of the measured physico-chemical/climatic factors. As previous studies demonstrated that, during a similar period, metal concentrations in the Severn Estuary and Bristol Channel (into which that estuary discharges) declined and water quality increased, it is proposed that the above changes are due to an improved environment. The fauna was dominated by the mysids Mesopodopsis slabberi and Schistomysis spiritus, which collectively contributed 94% to total abundance. Both species, which were represented by juveniles, males, non-brooding females and brooding females, underwent statistically-indistinguishable patterns of change in abundance over the 26 years. When analysis was based on the abundances of the various species, the overall species composition differed significantly among years and changed serially with year. When abundances were converted to percentage compositions, this pattern of seriation broke down, demonstrating that changes in abundance and not percentage composition were responsible for the seriation. As with the number and abundance of species, changes in composition over the 26 years were not related to any of the physico-chemical/climatic factors tested. Species composition changed monthly in a pronounced cyclical manner throughout the year, due to statistically different time-staggered changes in the abundance of each species. This cyclicity was related most strongly to salinity

A Sino-German 6cm polarization survey of the Galactic plane VII. Small supernova remnants

We study the spectral and polarization properties of supernova remnants (SNRs) based on our 6cm survey data. The observations were taken from the Sino-German 6cm polarization survey of the Galactic plane. By using the integrated flux densities at 6cm together with measurements at other wavelengths from the literature we derive the global spectra of 50 SNRs. In addition, we use the observations at 6cm to present the polarization images of 24 SNRs. We derived integrated flux densities at 6cm for 51 small SNRs with angular sizes less than 1 degree. Global radio spectral indices were obtained in all the cases except for Cas A. For SNRs G15.1-1.6, G16.2-2.7, G16.4-0.5, G17.4-2.3, G17.8-2.6, G20.4+0.1, G36.6+2.6, G43.9+1.6, G53.6-2.2, G55.7+3.4, G59.8+1.2, G68.6-1.2, and G113.0+0.2, the spectra have been significantly improved. From our analysis we argue that the object G16.8-1.1 is probably an HII region instead of a SNR. Cas A shows a secular decrease in total intensity, and we measured a flux density of 688+/-35 Jy at 6cm between 2004 and 2008. Polarized emission from 25 SNRs were detected. For G16.2-2.7, G69.7+1.0, G84.2-0.8 and G85.9-0.6, the polarized emission is detected for the first time confirming them as SNRs. High frequency observations of SNRs are rare but important to establish their spectra and trace them in polarization in particular towards the inner Galaxy where Faraday effects are important.Comment: 14 pages, 4 figures, 2 tables, accepted for publication in A&

Ecotoxicological effects of chemical contaminants adsorbed to microplastics in the clam Scrobicularia plana

Although microplastics (MPs) are distributed globally in the marine environment, a great deal of unknowns relating to their ecotoxicological effects on the marine biota remains. Due to their lipophilic nature, microplastics have the potential to adsorb persistent organic pollutants present in contaminated regions, which may increase their detrimental impact once assimilated by organisms. This study investigates the ecotoxicological effects of exposure to low-density polyethylene (LDPE) microplastics (11-13 beta m), with and without adsorbed contaminants (benzo[a]pyrene- BaP and perfluorooctane sulfonic acid-PFOS), in the peppery furrow shell clam, Scrobicularia plana. Environmentally relevant concentrations of contaminants (BaP-16.87 +/- 0.22 mu g g(-1) and PFOS-70.22 +/- 12.41 mu g g(-1)) were adsorbed to microplastics to evaluate the potential role of plastic particles as a source of chemical contamination once ingested. S. plana were exposed to microplastics, at a concentration of 1 mg L-1, in a water-sediment exposure setup for 14 days. Clams were sampled at the beginning of the experiment (day 0) and after 3, 7, and 14 days. BaP accumulation, in whole clam tissues, was analyzed. A multi-biomarker assessment was conducted in the gills, digestive gland, and haemolymph of clams to clarify the effects of exposure. This included the quantification of antioxidant (superoxide dismutase, catalase, glutathione peroxidase) and biotransformation (glutathione-Stransferases) enzyme activities, oxidative damage (lipid peroxidation levels), genotoxicity (single and double strand DNA breaks), and neurotoxicity (acetylcholinesterase activity). Results suggest a potential mechanical injury of gills caused by ingestion of microplastics that may also affect the analyzed biomarkers. The digestive gland seems less affected by mechanical damage caused by virgin microplastic exposure, with the MPs-adsorbed BaP and PFOS exerting a negative influence over the assessed biomarkers in this tissue.JPI Oceans FCT JPIOCEANS/0005/2015 JPI Oceans (BELSPO) JPI Oceans (FWO) JPI Oceans (FORMAS) JPI Oceans (SwAM) FCT UID/MAR/00350/2013info:eu-repo/semantics/publishedVersio

Association Between Parkinsonism and Participation in Agriculture in Korea

    Get PDF
BACKGROUND AND PURPOSE: Environmental factors might influence the pathogenesis of Parkinson's disease (PD) or multiple-system atrophy (MSA), and previous examinations of pesticide exposure, well-water drinking, and farming have produced inconclusive results. Because agriculture has been of considerable importance to Korean society, and hence the risk of exposure to pesticides was high in Korea, this study investigated whether such exposure is associated with elevated risks of developing PD and MSA. METHODS: Two hundred and thirty-five PD patients, 133 MSA patients, and 77 normal control subjects were examined. Data concerning environmental factors were collected by face-to-face interviews using a structured questionnaire. Odds ratios (ORs) were calculated by binary logistic regression. RESULTS: ORs for environmental risk factors for developing PD were 1.06 [95% confidence interval (CI) = 1.02-1.10] for age and 2.37 (95% CI = 1.32-4.27) for rural well-water drinking for >10 years. Smoking >10 pack-years (OR = 0.31; 95% CI = 0.11-0.64) was a preventable factor for developing PD in this study. However, no significant risk factors were identified for MSA. CONCLUSIONS: These results suggest that exposure to certain environmental risk factors plays a role in the development of PD. However, the development of MSA appears to be independent of environmental risk factors in Korean patients

Web of Scienc

Search for sub-mm, mm and radio continuum emission from Extremely Red Objects

    Get PDF
We present the results of sub-mm, mm (850 um, 450 um and 1250 um) and radio (1.4 and 4.8 GHz) continuum observations of a sample of 27 K-selected Extremely Red Objects, or EROs, (14 of which form a complete sample with K 5) aimed at detecting dusty starbursts, deriving the fraction of UltraLuminous Infrared Galaxies (ULIGs) in ERO samples, and constraining their redshifts using the radio-FIR correlation. One ERO was tentatively detected at 1250 um and two were detected at 1.4 GHz, one of which has a less secure identification as an ERO counterpart. Limits on their redshifts and their star forming properties are derived and discussed. We stacked the observations of the undetected objects at 850 um, 1250 um and 4.8 GHz in order to search for possible statistical emission from the ERO population as a whole, but no significant detections were derived either for the whole sample or as a function of the average NIR colours. These results strongly suggest that the dominant population of EROs with K < 20 is not comprised of ULIGs like HR 10, but is probably made of radio-quiet ellipticals and weaker starburst galaxies with L < 10^{12} L_sun and SFR < 100 M_sun/yr.Comment: 10 pages, 1 figure, 3 tables; accepted for publication in Astronomy & Astrophysic
