    High-throughput generation of product profiles for arabinoxylan-active enzymes from metagenomes

    Metagenomics is an exciting alternative to seek carbohydrate-active enzymes from a range of sources. Typically, metagenomics reveals dozens of putative catalysts that require functional characterization for further application in industrial processes. High-throughput screening methods compatible with adequate natural substrates are crucial for an accurate functional elucidation of substrate preferences. Based on DNA sequencer-aided fluorophore-assisted carbohydrate electrophoresis (DSA-FACE) analysis of enzymatic-reaction products, we generated product profiles to consequently infer substrate cleavage positions, resulting in the generation of enzymatic-degradation maps. Product profiles were produced in high throughput for arabinoxylan (AX)-active enzymes belonging to the glycoside hydrolase families GH43 (subfamilies 2 [MG432], 7 [MG437], and 28 [MG4328]) and GH8 (MG8) starting from 12 (arabino)xylo-oligosaccharides. These enzymes were discovered through functional metagenomic studies of feces from the North American beaver (Castor canadensis). This work shows how enzyme loading alters the product profiles of all enzymes studied and gives insight into AX degradation patterns, revealing sequential substrate preferences of AX-active enzymes. IMPORTANCE: Arabinoxylan is mainly found in the hemicellulosic fractions of rice straw, corn cobs, and rice husk. Converting arabinoxylan into (arabino)xylooligosaccharides as added-value products that can be applied in food, feed, and cosmetics presents a sustainable and economic alternative for the biorefinery industries. Efficient and profitable AX degradation requires a set of enzymes with particular characteristics. Therefore, enzyme discovery and the study of substrate preferences are of utmost importance. Beavers, as consumers of woody biomass, are a promising source of a repertoire of enzymes able to deconstruct hemicelluloses into soluble oligosaccharides. High-throughput analysis of the oligosaccharide profiles produced by these enzymes will assist in the selection of the most appropriate enzymes for the biorefinery

    Immune system recognize and fight back foreign microorganisms and inner modifications that lead to deficient cell and tissue functions. During a dog's life, the immune system needs to adapt to different physiological conditions, assuring surveillance and protection in a careful and controlled way. Pregnancy alters normal homeostasis, requiring a balance between immunity and tolerance. The embryos and fetus should be protected from infections, while the female dog must tolerate the growing of semi-allografts in her uterus. After birth, newborn puppies are at great risk of developing infectious diseases, because their immune system is in development and immune memory is absent. Passive transfer of immunity through colostrum is fundamental for puppy survival in the first weeks of life, but hampers the development of an active immune response to vaccination. At the end of life, dogs experience a decline in the structure and functional competence of the immune system, compromising the immune responses to novel antigenic challenges, such as infections and vaccines. Therefore, the current article reviews the general processes related to the development of the dog´s immune system, providing an overview of immune activity throughout the dog's life and its implications in canine health, and highlighting priority research goals.publishersversionpublishe

    Behavioral responses of rural and urban greater white-toothed shrews (Crocidura russula) to sound disturbance

    The development of urban areas imposes challenges that wildlife must adapt to in order to persist in these new habitats. One of the greatest changes brought by urbanization has been an increase in anthropogenic noise, with negative consequences for the natural behavior of animals. Small mammals are particularly vulnerable to urbanization and noise, despite some species having successfully occupied urban environments. To understand some of the traits that have enabled small mammals to deal with the consequences of urbanization, we compared the behavioral responses of urban and rural greater white-toothed shrews, Crocidura russula, to different sound stimuli. A total of 32 shrews, 16 from each habitat, were exposed in captivity to four sound treatments: silence, tawny owl calls, traffic noise, and white noise. Urban and rural shrews showed different behaviors, with urban animals being more active, feeding more frequently, and using less torpor than rural individuals. However, responses to sound treatments were similar in both populations: urban and rural shrews exhibited a slight decrease in activity and feeding behavior, as well as more fleeing responses, when exposed to traffic noise or white noise, but not to owl calls. These results suggest urbanization induces long-term changes in the general activity of C. russula, but the short-term behavioral response to sound disturbance remains similar in rural and urban populations.info:eu-repo/semantics/publishedVersio

    Study of Biodiesel Production from Sunflower Oil Using Non Usual Basic Polymeric Resin as Catalyst

    Biodiesel is defined as a long-chain mono alkyl (methyl, ethyl or propyl) ester of fatty acids obtained from renewable sources by transesterification reaction using an acid or base. In this work, a basic resin, Amberlyst A26, was used to produce methyl sunflower biodiesel. The iodine value, corrosion to copper, oxidative stability, specific mass, water content, acid value, cloud point and ester composition were evaluated. The heterogeneous catalyst was also characterized to verify its efficiency and its ability to be reused. We used the following analytical techniques: scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), X-rays fluorescence (XRF) and differential scanning calorimetry (DSC). The results showed that the characteristics of biodiesel made from sunflower oil using resin A26 as a catalyst are in accordance with Brazilian biodiesel regulations, except the oxidative stability. The techniques used to characterize the resin showed that it is possible to reuse the resin after regeneration

    The chytrid fungus Batrachochytrium dendrobatidis (Bd) has been linked to the declines of more than 500 amphibian species globally. In Brazil, Bd has been identified in several regions but predominantly in the Atlantic Forest. Data on the occurrence of this amphibian pathogen in northeastern Brazil are scarce, specifically from the Caatinga ecoregion, where there is only one study that reports Bd in the region. This study is the first to show a high prevalence of Bd in Caatinga’s species and includes two new records of species infected by Bd: Rhinella granulosa and R. jimi.     Context-dependence of race self-classification : results from a highly mixed and unequal middle-income country

    Ethnic-racial classification criteria are widely recognized to vary according to historical, cultural and political contexts. In Brazil, the strong influence of individual socio-economic factors on race/colour self-classification is well known. With the expansion of genomic technologies, the use of genomic ancestry has been suggested as a substitute for classification procedures such as self-declaring race, as if they represented the same concept. We investigated the association between genomic ancestry, the racial composition of census tracts and individual socioeconomic factors and self-declared race/colour in a cohort of 15,105 Brazilians. Results show that the probability of self-declaring as black or brown increases according to the proportion of African ancestry and varies widely among cities. In Porto Alegre, where most of the population is white, with every 10% increase in the proportion of African ancestry, the odds of self-declaring as black increased 14 times (95%CI 6.08–32.81). In Salvador, where most of the population is black or brown, that increase was of 3.98 times (95%CI 2.96–5.35). The racial composition of the area of residence was also associated with the probability of selfdeclaring as black or brown. Every 10% increase in the proportion of black and brown inhabitants in the residential census tract increased the odds of self-declaring as black by 1.33 times (95%CI 1.24–1.42). Ancestry alone does not explain self-declared race/colour. An emphasis on multiple situational contexts (both individual and collective) provides a more comprehensive framework for the study of the predictors of self-declared race/colour, a highly relevant construct in many different scenarios, such as public policy, sociology and medicine
