    The thermal history of the Western Irish onshore

    We present here a low-temperature thermochronological study that combines the apatite fission-track and (U + Th)/He dating methods with a pseudo-vertical sampling approach to generate continuous and well-constrained temperature–time histories from the onshore Irish Atlantic margin. The apatite fission-track and (U + Th)/He ages range from the Late Jurassic to Early Cretaceous and the mean track lengths are relatively short. Thermal histories derived from inverse modelling show that following post-orogenic exhumation the sample profiles cooled to c. 75 °C. A rapid cooling event to surface temperatures occurred during the Late Jurassic to Early Cretaceous and was diachronous from north to south. It was most probably caused by c. 2.5 km of rift-shoulder related exhumation and can be temporally linked to the main stage of Mesozoic rifting in the offshore basins. A slow phase of reheating during the Late Cretaceous and Early Cenozoic is attributed to the deposition of a thick sedimentary sequence that resulted in c. 1.5 km of burial. Our data imply a final pulse of exhumation in Neogene times, probably related to compression of the margin. However, it is possible that an Early Cenozoic cooling event, compatible with our data but not seen in our inverse models, accounts for part of the Cenozoic exhumation

    Biological and radiological exploration and management of non-functioning pituitary adenoma

    AbstractNon-functioning pituitary adenoma may be totally asymptomatic and discovered “incidentally” during radiological examination for some other indication, or else induce tumoral signs with compression of the optic chiasm and pituitary dysfunction. Non-functioning adenomas are mainly gonadotroph, but may also be “silent”. Treatment strategy depends on initial clinical, biological, ophthalmological and radiological findings. The present French Society of Endocrinology Consensus work-group sought to update the pitfalls associated with hormone assay and outline a hormonal exploration strategy for diagnosis and follow-up, without overlooking the particularities of silent adenoma. We also drew up basic rules for initial exploration and radiological follow-up of both operated and non-operated pituitary adenomas

    Paleomagnetic Study of Mesozoic Continental Sediments Along the Northern Tien Shan (China) and Heterogeneous Strain in Central Asia

    A paleomagnetic study of rocks from the northern foot of the Tien Shan and the southern border of the Dzungar Basin, east of Urumqi (44.2°N, 86.0°E), spanning ages from middle Jurassic to early Tertiary was carried out to constrain the tectonic evolution in central Asia since Mesozoic time. Five middle Jurassic sites reveal a remagnetized direction close to the present Earth field in geographic coordinates: D = 6.6°, I = 72.6° (α_(95) = 7.4°). Thirteen out of 17 upper Jurassic and lower Cretaceous sites yield a characteristic direction (stratigraphic coordinates) of D = 12.7°, I = 48.6° (α_(95) = 5.5°). Nine of 16 upper Cretaceous and lower Tertiary sites provide a characteristic direction of D = 12.5°, I = 51.3° (α_(95) = 6.9°). The latter two directions pass fold and reversal tests. The pole positions are close to each other and to the Besse and Courtillot [1989, 1990] Eurasian apparent polar wander path, for ages ranging from 130 to 70 Ma. However, the difference in paleolatitudes amounts to about 5.9° ± 3.7°, which could indicate significant continental shortening in the Altai Mountains and perhaps further north, subsequent to India-Asia collision. The pole positions from the Dzungar Basin are close to those found for the Tarim [Li et al., 1988a], leading to an insignificant paleolatitude difference (3.0° ± 6.9°), but showing a larger difference in declination (8.6° ± 8.7°). These paleomagnetic results are compatible with a model of heterogeneous deformation in the western part of the collision zone between India and Siberia. A significant shortening in the Altai, a slight counterclockwise rotation of the Dzungar block, the westward-increasing shortening in the Tien Shan with attendant clockwise rotation of the Tarim block are all consistent with this model, in which Tibet, the Tien Shan and the Altai undergo differential strain along strike in a relay fashion, with the total India-Siberia convergence remaining approximately constant

    Genetic and phenotypic analysis of B-cell post-transplant lymphoproliferative disorders provides insights into disease biology

    B‐cell post‐transplant lymphoproliferative disorders (PTLD) are classified as early lesions, polymorphic lymphomas (P‐PTLD) and monomorphic lymphomas (M‐PTLD). These morphologic categories are thought to reflect a biologic continuum, although supporting genetic data are lacking. To gain better insights into PTLD pathogenesis, we characterized the phenotypes, immunoglobulin (Ig) gene alterations and non‐Ig gene (BCL6, RhoH/TTF, c‐MYC, PAX5, CIITA, BCL7A, PIM1) mutations of 21 PTLD, including an IM‐like lesion, 8 P‐PTLD and 12 M‐PTLD. Gene expression profile analysis was also performed in 12 cases. All PTLD with clonal Ig rearrangements showed evidence of germinal centre (GC) transit based on the analysis of Ig and BCL6 gene mutations, and 74% had a non‐GC phenotype (BCL6 ± MUM1+). Although surface Ig abnormalities were seen in 6/19 (32%) PTLD, only three showed ‘crippling’ Ig mutations indicating other etiologies for loss of the B‐cell receptor. Aberrant somatic hypermutation (ASHM) was almost exclusively observed in M‐PTLD (8/12 vs. 1/8 P‐PTLD) and all three recurrent cases analysed showed additional mutations in genes targeted by ASHM. Gene expression analysis showed distinct clustering of PTLD compared to B‐cell non‐Hodgkin lymphomas (B‐NHL) without segregation of P‐PTLD from non‐GC M‐PTLD or EBV+ from EBV− PTLD. The gene expression pattern of PTLD appeared more related to that of memory and activated B‐cells. Together, our results suggest that PTLD represent a distinct type of B‐NHL deriving from an antigen experienced B‐cell, whose evolution is associated with accrual of genetic lesions

    De <i>novo </i>transcriptomes of 14 gammarid individuals for proteogenomic analysis of seven taxonomic groups

    Gammarids are amphipods found worldwide distributed in fresh and marine waters. They play an important role in aquatic ecosystems and are well established sentinel species in ecotoxicology. In this study, we sequenced the transcriptomes of a male individual and a female individual for seven different taxonomic groups belonging to the two genera Gammarus and Echinogammarus: Gammarus fossarum A, G. fossarum B, G. fossarum C, Gammarus wautieri, Gammarus pulex, Echinogammarus berilloni, and Echinogammarus marinus. These taxa were chosen to explore the molecular diversity of transcribed genes of genotyped individuals from these groups. Transcriptomes were de novo assembled and annotated. High-quality assembly was confirmed by BUSCO comparison against the Arthropod dataset. The 14 RNA-Seq-derived protein sequence databases proposed here will be a significant resource for proteogenomics studies of these ecotoxicologically relevant non-model organisms. These transcriptomes represent reliable reference sequences for whole-transcriptome and proteome studies on other gammarids, for primer design to clone specific genes or monitor their specific expression, and for analyses of molecular differences between gammarid species

    <i>USP27X </i>variants underlying X-linked intellectual disability disrupt protein function via distinct mechanisms

    Neurodevelopmental disorders with intellectual disability (ND/ID) are a heterogeneous group of diseases driving lifelong deficits in cognition and behavior with no definitive cure. X-linked intellectual disability disorder 105 (XLID105, #300984; OMIM) is a ND/ID driven by hemizygous variants in the USP27X gene encoding a protein deubiquitylase with a role in cell proliferation and neural development. Currently, only four genetically diagnosed individuals from two unrelated families have been described with limited clinical data. Furthermore, the mechanisms underlying the disorder are unknown. Here, we report 10 new XLID105 individuals from nine families and determine the impact of gene variants on USP27X protein function. Using a combination of clinical genetics, bioinformatics, biochemical, and cell biology approaches, we determined that XLID105 variants alter USP27X protein biology via distinct mechanisms including changes in developmentally relevant protein-protein interactions and deubiquitylating activity. Our data better define the phenotypic spectrum of XLID105 and suggest that XLID105 is driven by USP27X functional disruption. Understanding the pathogenic mechanisms of XLID105 variants will provide molecular insight into USP27X biology and may create the potential for therapy development.</p
