    Experimental vaccination of sheep and cattle against tick infestation using recombinant 5′-nucleotidase

    Limited prior evidence suggests that 5′-nucleotidase, an ectoenzyme principally located in the Malpighian tubules of the tick Rhipicephalus (Boophilus) microplus, could be an effective antigen in an anti-tick vaccine. To assess this, recombinant 5′-nucleotidase was expressed in Escherichia coli and used in vaccination trials with both sheep and cattle. Vaccinated sheep were challenged with freshly moulted adult ticks. Those with high titres of anti-nucleotidase antibodies showed significant protection against tick infestation, although protection was less than that found with the previously characterized antigen, Bm86. Cattle were vaccinated, in separate groups, with 5′-nucleotidase, Bm86 and both antigens combined. Cattle, as the natural host, were challenged with larval ticks. Although Bm86 showed typical efficacy, no significant protection was seen in cattle vaccinated with 5′-nucleotidase. Cattle receiving a dual antigen formulation were no better protected than those receiving Bm86 alone. One possible reason for the difference between host species, namely antibody titre, was examined and shown to be an unlikely explanation. This demonstrates a limitation of using a model host like sheep in vaccine studies

    Fluency Training the Correct Use of to Lie and to Lay

    Proper English is an important skill, but using to lie and to lay correctly appears challenging, as even college students misuse these verbs. The current project assessed a treatment package that included fluency-based software to train three participants to correctly use to lie and to lay. For each verb several meanings were identified. Model sentences were constructed from the definitions to form basic units. Each basic unit included examples (correct sentences) and non-examples (incorrect sentences) for one of the meanings of each verb. Participants practiced with each basic unit until fluent and then completed a cumulative unit based on all previous units before moving to a new basic unit. To assess whether the treatment package was effective, the units were embedded in a non-concurrent multiple-baseline design replicated across participants. The treatment package appeared to improve verb use for two participants and paradoxically worsened verb use for one participant. These findings and the implications for further research were discussed

    NSort/DB: an intra-nuclear compartment protein database

    Distinct substructures within the nucleus are associated with a wide variety of important nuclear processes. Structures such as chromatin and nuclear pores have specific roles, while others such as Cajal bodies are more functionally varied. Understanding the roles of these membraneless intra-nuclear compartments requires extensive data sets covering nuclear and compartment-associated proteins. NSort/DB is a database providing access to intra- or sub-nuclear compartment associations for the mouse nuclear proteome. Based on resources ranging from large-scale curated data sets to detailed experiments, this data set provides a high-quality set of annotations of non-exclusive association of nuclear proteins with structures such as promyelocytic leukaemia bodies and chromatin. The database is searchable by protein identifier or compartment, and has a documented web service API. The search interface, web service and data download are all freely available online at http://www.nsort.org/db/. Availability of this data set will enable systematic analyses of the protein complements of nuclear compartments, improving our understanding of the diverse functional repertoire of these structures

    A Foot in Both Worlds: Asian Americans' Perceptions of Asian, White, and Racially Ambiguous Faces

    Past research on racial perception has often focused on responses from White participants, making it difficult to determine the role of perceiver race in the perception of others. Similarly, studies examining perceptions of individuals whose racial category membership is unclear have not systematically examined responses from non-Whites. This was addressed by showing Asian participants pictures of Whites, Asians, and racially ambiguous White-Asian faces. Event-related potentials were recorded to measure early attention responses. Participants initially oriented more to outgroup White than ingroup Asian or racially ambiguous faces. Shortly after that, they showed sensitivity to the racial context in which the faces were presented, more deeply processing ingroup Asian and racially ambiguous faces when they were seeing lots of other Asians, but more deeply processing outgroup White and racially ambiguous faces when they were seeing lots of other Whites. Still later, responses were more sensitive to the objective physical properties of the faces, with racially ambiguous faces differentiated from both Whites and Asians. These results demonstrate the fluidity of racial processing, and when compared to responses obtained from White participants, show how perceiver race and racial context influences attention to racial cues

    Denne masteroppgaven presenterer og analyserer den svært omstridte fellingen av urskog i den australske øy-staten Tasmania. Avhandlingen er basert på materiale som ble samlet på feltarbeid i perioden fra januar til juli 2008. Avhandlingen søker å avdekke årsaker til at de tasmanske skogspraksisene er så omstridt, i tillegg til å undersøke hvilke strategier og teknikker som anvendes av miljøbevegelsen og skogsindustrien når de argumenterer for sine respektive synspunkter. Det blir funnet at konflikten utfolder seg på mange plan og omfatter ikke bare politiske, økonomiske og kulturelle parametere. Også emosjonelle og sågar eksistensielle dimensjoner berøres og flettes inn i samhandlingene. Ved å engasjere de symbolske praksisene i en dialog, kommer jeg fram til en erkjennelse av at aktørene i den tasmanske skogsdebatten konstruerer og forfekter visse fremstillinger av naturen ved retoriske virkemidler. Dette blir gjort slik at noen måter å forholde seg til naturen på blir favorisert, undertiden som andre relasjoner mellom natur og samfunn undergraves. Ved bruk av metaforiske assosiasjoner og oppskaleringsprosesser, knyttes konflikten til følelsesladede og normative narrativer av både religiøs og kulturell art

    Evaluation of glycoproteins purified from adult and larval camel ticks (Hyalomma dromedarii) as a candidate vaccine

    In order to identify antigens that can help prevent camel tick infestations, three major glycoproteins (GLPs) about 97, 66 and 40 kDa in size were purified from adult and larval Egyptian ticks, Hyalomma (H.) dromedarii, using a single-step purification method with Con-A sepharose. The purified GLPs were evaluated as vaccines against camel tick infestation in rabbits. The rabbits received three intramuscular inoculations of GLPs (20 µg/animal) on days 0, 14, and 28. In the immunoblot analysis, Sera from the immunized rabbits recognized the native GLPs and other proteins from larval and adult H. dromedarii ticks along with those from other tick species such as Rhipicephalus sanguineus but not Ornithodoros moubata. The effects of immunity induced by these GLPs were determined by exposing rabbits to adult H. dromedarii ticks. These results demonstrated that GLP immunization led to a slightly decreased reproductive index and significantly reduced rates of egg hatchability. These results demonstrated that immunization with the purified GLPs can provide protection against infestation by H. dromedarii and some other tick species. Further studies are needed to confirm the effectiveness of immunization with GLPs against other tick species

    Induction of humoral immune response to multiple recombinant Rhipicephalus appendiculatus antigens and their effect on tick feeding success and pathogen transmission

    BACKGROUND: Rhipicephalus appendiculatus is the primary vector of Theileria parva, the etiological agent of East Coast fever (ECF), a devastating disease of cattle in sub-Saharan Africa. We hypothesized that a vaccine targeting tick proteins that are involved in attachment and feeding might affect feeding success and possibly reduce tick-borne transmission of T. parva. Here we report the evaluation of a multivalent vaccine cocktail of tick antigens for their ability to reduce R. appendiculatus feeding success and possibly reduce tick-transmission of T. parva in a natural host-tick-parasite challenge model. METHODS: Cattle were inoculated with a multivalent antigen cocktail containing recombinant tick protective antigen subolesin as well as two additional R. appendiculatus saliva antigens: the cement protein TRP64, and three different histamine binding proteins. The cocktail also contained the T. parva sporozoite antigen p67C. The effect of vaccination on the feeding success of nymphal and adult R. appendiculatus ticks was evaluated together with the effect on transmission of T. parva using a tick challenge model. RESULTS: To our knowledge, this is the first evaluation of the anti-tick effects of these antigens in the natural host-tick-parasite combination. In spite of evidence of strong immune responses to all of the antigens in the cocktail, vaccination with this combination of tick and parasite antigens did not appear to effect tick feeding success or reduce transmission of T. parva. CONCLUSION: The results of this study highlight the importance of early evaluation of anti-tick vaccine candidates in biologically relevant challenge systems using the natural tick-host-parasite combination
