    Mode waters and subduction rates in a high-resolution South Atlantic simulation

    Water mass production and destruction in the subtropical South Atlantic gyre is studied. A high resolution numerical model is used to examine regional mode water formation and estimate the associated instantaneous and mean subduction rates. Primitive equation dynamics expressed in depth following (sigma) coordinates are employed. The main hydrographic and kinematic features of the South Atlantic are faithfully reproduced by the model. In particular, the principle current systems appear and the model exhibits a sequence of ventilated potential vorticity minima on density surfaces coinciding with those of observed South Atlantic mode waters. The formation sequence within the model of these mode waters is described. Net formation rates are estimated using a pseudo-Lagrangian method and by diagnosing the time history of subsurface water mass volumes. Maximum formation rates occur in the density bands of the mode waters. It is argued that the roots of the model mode waters are found along open ocean late winter outcrops, rather than in the waters entering the gyre from the Brazil Current/Malvinas Current Confluence region. Eddies generate interannual variability in mode water formation and precondition the waters in the outcrop regions for convection. On the other hand, the eddy kinetic energy of the Confluence region is too intense to permit a direct connection between deep convection cells in the western boundary current and those in the open South Atlantic that directly form mode water

    Educators and staff adapted to work-from-home setup amidst the covid-19 pandemic.  The transition to full-online classes and services leads to poor mental health. The current study explored the association of educator and staff personal characteristics, well-being, and mental health.  326 university employees completed the demographic profile, mental health, and well-being scales. Various hierarchical regression was conducted to determine if personal characteristics and well-being predict common mental health symptoms (depression, anxiety, and stress). Series of multivariate analyses of variance (MANOVA) was conducted to determine the difference between the levels of mental health symptoms according to mental health category, and personal characteristics. The results support the hypothesis with psychological and emotional well-being inversely predicting depression, anxiety, and stress. However, social well-being failed to serve as a significant determinant of common mental health symptoms. MANOVA obtained a significant difference with common mental health symptoms and mental health category and personal characteristics

    Congenital Hypogonadotropic Hypogonadism Due to GNRH Receptor Mutations in Three Brothers Reveal Sites Affecting Conformation and Coupling

    Congenital hypogonadotropic hypogonadism (CHH) is characterized by low gonadotropins and failure to progress normally through puberty. Mutations in the gene encoding the GnRH receptor (GNRHR1) result in CHH when present as compound heterozygous or homozygous inactivating mutations. This study identifies and characterizes the properties of two novel GNRHR1 mutations in a family in which three brothers display normosmic CHH while their sister was unaffected. Molecular analysis in the proband and the affected brothers revealed two novel non-synonymous missense GNRHR1 mutations, present in a compound heterozygous state, whereas their unaffected parents possessed only one inactivating mutation, demonstrating the autosomal recessive transmission in this kindred and excluding X-linked inheritance equivocally suggested by the initial pedigree analysis. The first mutation at c.845 C>G introduces an Arg substitution for the conserved Pro 282 in transmembrane domain (TMD) 6. The Pro282Arg mutant is unable to bind radiolabeled GnRH analogue. As this conserved residue is important in receptor conformation, it is likely that the mutation perturbs the binding pocket and affects trafficking to the cell surface. The second mutation at c.968 A>G introduces a Cys substitution for Tyr 323 in the functionally crucial N/DPxxY motif in TMD 7. The Tyr323Cys mutant has an increased GnRH binding affinity but reduced receptor expression at the plasma membrane and impaired G protein-coupling. Inositol phosphate accumulation assays demonstrated absent and impaired Gαq/11 signal transduction by Pro282Arg and Tyr323Cys mutants, respectively. Pretreatment with the membrane permeant GnRHR antagonist NBI-42902, which rescues cell surface expression of many GNRHR1 mutants, significantly increased the levels of radioligand binding and intracellular signaling of the Tyr323Cys mutant but not Pro282Arg. Immunocytochemistry confirmed that both mutants are present on the cell membrane albeit at low levels. Together these molecular deficiencies of the two novel GNRHR1 mutations lead to the CHH phenotype when present as a compound heterozygote

This article presents the updated taxonomy of the family Arenaviridae and the order Bunyavirales as now accepted by the International Committee on Taxonomy of Viruses (ICTV) and summarizes additional taxonomic proposals that may affect the order in the near future.Peer reviewe

    Global fine-resolution data on springtail abundance and community structure

    Springtails (Collembola) inhabit soils from the Arctic to the Antarctic and comprise an estimated ~32% of all terrestrial arthropods on Earth. Here, we present a global, spatially-explicit database on springtail communities that includes 249,912 occurrences from 44,999 samples and 2,990 sites. These data are mainly raw sample-level records at the species level collected predominantly from private archives of the authors that were quality-controlled and taxonomically-standardised. Despite covering all continents, most of the sample-level data come from the European continent (82.5% of all samples) and represent four habitats: woodlands (57.4%), grasslands (14.0%), agrosystems (13.7%) and scrublands (9.0%). We included sampling by soil layers, and across seasons and years, representing temporal and spatial within-site variation in springtail communities. We also provided data use and sharing guidelines and R code to facilitate the use of the database by other researchers. This data paper describes a static version of the database at the publication date, but the database will be further expanded to include underrepresented regions and linked with trait data.</p
