    Anhydrous, medium/coarse-grained spinel bearing mantle xenoliths from the Subei Basin, Eastern China have mineral arrangements that reflect low energy geometry. Because of clinopyroxene modal contents, they are grouped into cpx-rich lherzolites (cpx ≥ 14percentage), lherzolites (8 5My, based on modelled H2O solid-solid diffusion rate) the occurrence of the last melting episode.     High H2O Content in Pyroxenes of Residual Mantle Peridotites at a Mid Atlantic Ridge Segment

    Global correlations of mid-ocean-ridges basalt chemistry, axial depth and crustal thickness have been ascribed to mantle temperature variations affecting degree of melting. However, mantle H2O content and elemental composition may also play a role. How H2O is distributed in the oceanic upper mantle remains poorly constrained. We tackled this problem by determining the H2O content of orthopyroxenes (opx) and clinopyroxenes (cpx) of peridotites from a continuous lithospheric section created during 26 Ma at a 11°N Mid-Atlantic Ridge segment, and exposed along the Vema Transform. The H2O content of opx ranges from 119 ppm to 383 ppm; that of cpx from 407 ppm to 1072 ppm. We found anomalous H2O-enriched peridotites with their H2O content not correlating inversely with their degree of melting, although H2O is assumed to be incompatible during melting. Inverse correlation of H2O with Ce, another highly incompatible component, suggests post-melting H2O enrichment. We attribute a major role to post-melting temperature-dependent diffusion of hydrogen occurring above the melting region, where water-rich melt flows faster than residual peridotites through dunitic conduits cross-cutting the uprising mantle. Accordingly, estimates of the H2O content of the MORB mantle source based on H2O in abyssal peridotites can be affected by strong uncertainties

    Buoyant hydrous mantle plume from the mantle transition zone

    Abstract Magmatism at some intraplate volcanoes and large igneous provinces (LIPs) in continental areas may originate from hydrous mantle upwelling (i.e. a plume) from the mantle transition zone (MTZ) at 410–660 km depths in the Earth’s deep interior. However, the ultimate origin of the magmatism, i.e. why mantle plumes could have been generated at the MTZ, remains unclear. Here, we study the buoyancy of a plume by investigating basalts from the Changbaishan volcano, beneath which a mantle plume from the hydrous MTZ is observed via seismology. Based on carefully determined water contents of the basalts, the potential temperature of the source mantle is estimated to be 1310–1400 °C, which is within the range of the normal upper mantle temperature. This observation suggests that the mantle plume did not have a significant excess heat, and that the plume upwelled because of buoyancy resulting from water supplied from the Pacific slab in the MTZ. Such a hydrous mantle plume can account for the formation of extremely hydrous LIP magmatism. The water was originally sourced from a stagnant slab and stored in the MTZ, and then upwelled irrespective of the presence or absence of a deep thermal plume

    Codonopsis pilosula Polysaccharide Attenuates Tau Hyperphosphorylation and Cognitive Impairments in hTau Infected Mice

    Codonopsis pilosula polysaccharide (CPPs), a natural products with potentially lower toxicity and better bioavailability has been used in traditional Chinese medicine for 1000s of years and a neuroprotective polysaccharide mitigates tau pathology in Alzheimer’s disease (AD) mouse model. However, whether CPPs can relieve AD pathology and cognitive defects remains poorly understood. Here we reported that CPPs remarkably increased the cell viability and PP2A activity, decreased tau phosphorylation in HEK 293/tau cells. Next, we employed an adeno-associated virus serotype 2 (AAV2)-induced expression of human full length tau (hTau) in C57/BL6 mice to mimic AD tau pathology. One month intragastric administration of CPPs significantly increased PP2A activity and reduced tau phosphorylation at Ser199, Ser202/Thr205 (AT8) and Thr231 in hippocampus of AAV2-hTau infected mice. Furthermore, behavioral tests revealed that CPPs rescued hTau overexpression induced cognitive defects while CPPs significantly increased the fEPSP slope and synaptic proteins including synaptotagmin and synaptophysin. Together, our data suggest that CPPs might prevent AD-like tau hyperphosphorylation via activation of PP2A and attenuates AD-like cognitive impairments through restoring the synaptic plasticity and synaptogenesis. In conclusion, our findings suggest that CPPs might be a potential candidate compound for the treatment of tau related diseases

    Biomarkers for Early Diagnostic of Mild Cognitive Impairment in Type-2 Diabetes Patients: A Multicentre, Retrospective, Nested Case–Control Study

    AbstractBackgroundBoth type 2 diabetes mellitus (T2DM) and Alzheimer's disease (AD) are common age-associated disorders and T2DM patients show an increased risk to suffer from AD, however, there is currently no marker to identify who in T2DM populations will develop AD. Since glycogen synthase kinase-3β (GSK-3β) activity, ApoE genotypes and olfactory function are involved in both T2DM and AD pathogenesis, we investigate whether alterations of these factors can identify cognitive impairment in T2DM patients.MethodsThe cognitive ability was evaluated using Minimum Mental State Examination (MMSE) and Clinical Dementia Rating (CDR), and the mild cognitive impairment (MCI) was diagnosed by Petersen's criteria. GSK-3β activity in platelet, ApoE genotypes in leucocytes and the olfactory function were detected by Western/dot blotting, the amplification refractory mutation system (ARMS) PCR and the Connecticut Chemosensory Clinical Research Center (CCCRC) test, respectively. The odds ratio (OR) and 95% confidence intervals (95% CI) of the biomarkers for MCI diagnosis were calculated by logistic regression. The diagnostic capability of the biomarkers was evaluated by receiver operating characteristics (ROC) analyses.FindingsWe recruited 694 T2DM patients from Jan. 2012 to May. 2015 in 5 hospitals (Wuhan), and 646 of them met the inclusion criteria and were included in this study. 345 patients in 2 hospitals were assigned to the training set, and 301 patients in another 3 hospitals assigned to the validation set. Patients in each set were randomly divided into two groups: T2DM without MCI (termed T2DM-nMCI) or with MCI (termed T2DM-MCI). There were no significant differences for sex, T2DM years, hypertension, hyperlipidemia, coronary disease, complications, insulin treatment, HbA1c, ApoE ε2, ApoE ε3, tGSK3β and pS9GSK3β between the two groups. Compared with the T2DM-nMCI group, T2DM-MCI group showed lower MMSE score with older age, ApoE ε4 allele, higher olfactory score and higher rGSK-3β (ratio of total GSK-3β to Ser9-phosphorylated GSK-3β) in the training set and the validation set. The OR values of age, ApoE ε4 gene, olfactory score and rGSK-3β were 1.09, 2.09, 1.51, 10.08 in the training set, and 1.06, 2.67, 1.47, 7.19 in the validation set, respectively. The diagnostic accuracy of age, ApoE ε4 gene, olfactory score and rGSK-3β were 0.76, 0.72, 0.66, 0.79 in the training set, and 0.70, 0.68, 0.73, 0.79 in the validation set, respectively. These four combined biomarkers had the area under the curve (AUC) of 82% and 86%, diagnostic accuracy of 83% and 81% in the training set and the validation set, respectively.InterpretationAging, activation of peripheral circulating GSK-3β, expression of ApoE ε4 and increase of olfactory score are diagnostic for the mild cognitive impairment in T2DM patients, and combination of these biomarkers can improve the diagnostic accuracy
