    The KATRIN experiment aims to determine the neutrino mass scale with a sensitivity of 200 meV/c^2 (90% C.L.) by a precision measurement of the shape of the tritium ÎČ\beta-spectrum in the endpoint region. The energy analysis of the decay electrons is achieved by a MAC-E filter spectrometer. To determine the transmission properties of the KATRIN main spectrometer, a mono-energetic and angular-selective electron source has been developed. In preparation for the second commissioning phase of the main spectrometer, a measurement phase was carried out at the KATRIN monitor spectrometer where the device was operated in a MAC-E filter setup for testing. The results of these measurements are compared with simulations using the particle-tracking software "Kassiopeia", which was developed in the KATRIN collaboration over recent years.Comment: 19 pages, 16 figures, submitted to European Physical Journal

    Eastern enlargement and differentiated integration: towards normalization

    Against the background of theoretical arguments and the historical record, enlargements of the European Union tend to increase differentiated integration. The actual development of differentiation of the 2004 and 2007 accession countries can, therefore, be seen as an indicator of the Union’s integration capacity. We map the newcomers’ differentiated integration since accession, and also distinguish between exemptions and discrimination. Furthermore, we approximate a counterfactual scenario of how their level of differentiation might have developed in the absence of recent accession through a comparison to the Southern member states. Finally, we explore correlations between exemptions, discriminations and two structural country characteristics, wealth and identity. Overall, the level, trajectory and patterns of differentiation point towards a normalization of the new member states’ integration in the Union

    Grand Theories, Differentiated Integration

    Even though differentiation has become a core feature of the EU, the grand theories have focused almost exclusively on uniform integration. In this paper, we derive hypotheses about differentiated integration from liberal intergovernmentalism, neofunctionalism, and postfunctionalism. In an analysis of EU treaty making between 1992 and 2016, we find evidence that heterogeneity of both wealth and identity, integration in the area of core state powers, and pre-existing differentiation drive differentiated integration. A comparison of the explanatory power of the grand theories shows that neo- and post-functionalism explain the differentiations that member states obtain in EU reform treaties more convincingly than liberal intergovernmentalism. A synthetic model performs best, however. The grand theories also leave noteworthy variation unexplained

    Explaining differentiation in European Union treaties

    Since the early 1990s, European integration has become increasingly differentiated. Analysing the conditions under which member states make use of the opportunity to opt out of, or exclude other countries from, European integration, we argue that different explanations apply to treaty and accession negotiations, respectively. Threatening to block deeper integration, member states with strong national identities secure differentiations in treaty reform. In enlargement, in turn, old member states fear economic disadvantages and low administrative capacity and therefore impose differentiation on poor newcomers. Opt-outs from treaty revisions are limited to the area of core state powers, whereas they also occur in the market in the context of enlargement

    Commissioning of the vacuum system of the KATRIN Main Spectrometer

    The KATRIN experiment will probe the neutrino mass by measuring the beta-electron energy spectrum near the endpoint of tritium beta-decay. An integral energy analysis will be performed by an electro-static spectrometer (Main Spectrometer), an ultra-high vacuum vessel with a length of 23.2 m, a volume of 1240 m^3, and a complex inner electrode system with about 120000 individual parts. The strong magnetic field that guides the beta-electrons is provided by super-conducting solenoids at both ends of the spectrometer. Its influence on turbo-molecular pumps and vacuum gauges had to be considered. A system consisting of 6 turbo-molecular pumps and 3 km of non-evaporable getter strips has been deployed and was tested during the commissioning of the spectrometer. In this paper the configuration, the commissioning with bake-out at 300{\deg}C, and the performance of this system are presented in detail. The vacuum system has to maintain a pressure in the 10^{-11} mbar range. It is demonstrated that the performance of the system is already close to these stringent functional requirements for the KATRIN experiment, which will start at the end of 2016.Comment: submitted for publication in JINST, 39 pages, 15 figure

    Introduction: Connecting with the Electorate? Parliamentary Communication in EU Affairs

    ational parliaments have often been described as latecomers to European integration, but there is little doubt that they have developed the institutional means to become more involved over the last few years – and especially since the Lisbon Treaty. Accordingly, the main focus of the literature has been on this institutional adaptation and thus on the relationship between the parliament and the government in European Union (EU) affairs. Other parliamentary functions, and in particular those that relate to their citizens such as the communication function, by contrast, have been largely neglected. Yet democracy depends on a viable public debate on policy choices and political alternatives to allow citizens to make informed political (electoral) choices and to exercise democratic control. This collection therefore investigates whether, and how, individual members of parliament, political parties, or legislatures as institutions ‘link’ with their electorates in EU politics. This introduction discusses why engaging with the public in EU affairs is – or at least should be – an important aspect of parliamentary work, introduces parliamentary means of communication and assesses parliamentary incentives and disincentives ‘to go public’ in EU politics.     Durable vesicles for reconstitution of membrane proteins in biotechnology

    The application of membrane proteins in biotechnology requires robust, durable reconstitution systems that enhance their stability and support their functionality in a range of working environments. Vesicular architectures are highly desirable to provide the compartmentalisation to utilise the functional transmembrane transport and signalling properties of membrane proteins. Proteoliposomes provide a native-like membrane environment to support membrane protein function, but can lack the required chemical and physical stability. Amphiphilic block copolymers can also self-assemble into polymersomes: tough vesicles with improved stability compared with liposomes. This review discusses the reconstitution of membrane proteins into polymersomes and the more recent development of hybrid vesicles, which blend the robust nature of block copolymers with the biofunctionality of lipids. These novel synthetic vesicles hold great promise for enabling membrane proteins within biotechnologies by supporting their enhanced in vitro performance and could also contribute to fundamental biochemical and biophysical research by improving the stability of membrane proteins that are challenging to work with

    miR-27b Targets KSRP to Coordinate TLR4-Mediated Epithelial Defense against Cryptosporidium parvum Infection

    Cryptosporidium is a protozoan parasite that infects the gastrointestinal epithelium and causes a diarrheal disease. Toll-like receptor (TLR)- and NF-ÎșB-mediated immune responses from epithelial cells, such as production of antimicrobial peptides and generation of reactive nitrogen species, are important components of the host's defense against cryptosporidial infection. Here we report data demonstrating a role for miR-27b in the regulation of TLR4/NF-ÎșB-mediated epithelial anti-Cryptosporidium parvum responses. We found that C. parvum infection induced nitric oxide (NO) production in host epithelial cells in a TLR4/NF-ÎșB-dependent manner, with the involvement of the stabilization of inducible NO synthase (iNOS) mRNA. C. parvum infection of epithelial cells activated NF-ÎșB signaling to increase transcription of the miR-27b gene. Meanwhile, downregulation of KH-type splicing regulatory protein (KSRP) was detected in epithelial cells following C. parvum infection. Importantly, miR-27b targeted the 3â€Č-untranslated region of KSRP, resulting in translational suppression. C. parvum infection decreased KSRP expression through upregulating miR-27b. Functional manipulation of KSRP or miR-27b caused reciprocal alterations in iNOS mRNA stability in infected cells. Forced expression of KSRP and inhibition of miR-27b resulted in an increased burden of C. parvum infection. Downregulation of KSRP through upregulating miR-27b was also detected in epithelial cells following LPS stimulation. These data suggest that miR-27b targets KSRP and modulates iNOS mRNA stability following C. parvum infection, a process that may be relevant to the regulation of epithelial anti-microbial defense in general

    Post-transcriptional control during chronic inflammation and cancer: a focus on AU-rich elements

    A considerable number of genes that code for AU-rich mRNAs including cytokines, growth factors, transcriptional factors, and certain receptors are involved in both chronic inflammation and cancer. Overexpression of these genes is affected by aberrations or by prolonged activation of several signaling pathways. AU-rich elements (ARE) are important cis-acting short sequences in the 3â€ČUTR that mediate recognition of an array of RNA-binding proteins and affect mRNA stability and translation. This review addresses the cellular and molecular mechanisms that are common between inflammation and cancer and that also govern ARE-mediated post-transcriptional control. The first part examines the role of the ARE-genes in inflammation and cancer and sequence characteristics of AU-rich elements. The second part addresses the common signaling pathways in inflammation and cancer that regulate the ARE-mediated pathways and how their deregulations affect ARE-gene regulation and disease outcome
