    Effect of specific inhibition of gamma-glutamyl transpeptidase on amino acid uptake by mammary gland of the lactating rat

    AbstractWe showed [Biochem. J. (1981) 194, 99–102] that inhibition of γ-glutamyl transpeptidase in vivo with serine-borate decreases amino acid uptake by mammary gland. However, doubts arose about the validity of this inhibitor in metabolic studies because it must be used in very large amounts. New Inhibitors have been isolated, like anthglutin and acivicin, which are effective at low concentrations in vivo. Here, we show that treatment of lactating rats with these substances decreases the transpeptidase activity and the amino acid uptake by the gland. These results support the hypothesis that the γ-glutamyl cycle functions as an amino acid transport system in mammary gland

    Oxidative and nitrosative stress in the diaphragm of patients with COPD

    COPD is associated with an increased load on the diaphragm. Since chronic muscle loading results in changes in antioxidant capacity and formation of reactive oxygen and reactive nitrogen species, we hypothesized that COPD has a similar effect on the diaphragm, which is related to the severity of COPD. Catalase activity was determined spectrophotometrically. Levels of 4-hydroxy-2-nonenal (HNE)-protein adducts and 3-nitrotyrosine (NT) formation were measured using western blotting. Levels of malondialdehyde (MDA) were assessed by high-performance liquid chromatography. We found that catalase activity was ~89% higher in the diaphragm of severe COPD patients (FEV1 37 ± 5% predicted) compared with non-COPD patients. MDA levels, a marker for lipid peroxidation, were significantly lower in the diaphragm of COPD patients compared with non-COPD patients, whereas the level of HNE-protein adducts was equal in both groups. NT formation was not different between groups. However, increasing hyperinflation and NT formation were inversely correlated. These results indicate that in COPD the diaphragm adapts to a higher work load by increasing catalase activity, resulting in a reduction in oxidative damage to lipids and tyrosine nitration of proteins

    Id2 leaves the chromatin of the E2F4-p130-controlled c-myc promoter during hepatocyte priming for liver regeneration

    The Id (inhibitor of DNA binding or inhibitor of differentiation) helix-loop-helix proteins are involved in the regulation of cell growth, differentiation and cancer. The fact that the molecular mechanisms of liver regeneration are not completely understood prompted us to study the fate of Id2 in proliferating liver. Id2 increases in liver regeneration after partial hepatectomy, following the early induction of its gene. Co-immunoprecipitation shows that Id2 forms a complex with E2F4, p130 and mSin3A in quiescent liver and all these components are present at the c-myc promoter as shown using ChIP (chromatin immunoprecipitation). Activation of c-myc during hepatocyte priming (G0-G1 transition) correlates with the dissociation of Id2 and HDAC (histone deacetylase), albeit p130 remains bound at least until 6 h. Moreover, as the G0-G1 transition progresses, Id2 and HDAC again bind the c-myc promoter concomitantly with the repression of this gene. The time course of c-myc binding to the Id2 promoter, as determined by ChIP assays is compatible with a role of the oncoprotein as a transcriptional inducer of Id2 in liver regeneration. Immunohistochemical analysis shows that Id2 also increases in proliferating hepatocytes after bile duct ligation. In this case, the pattern of Id2 presence in the c-myc promoter parallels that found in regenerating liver.     ProspecciĂłn acĂșstico-pesquera y caracterizaciĂłn ambiental de algunos recursos de tipo demersal en el Caribe colombiano

    Within the framework of the fishing program Vecep (Inpa-Colombia) a pilot acoustic survey was carried out for the reconnaissance of bottoms and demersal resources, previous to the evaluation by the swept area method made by the same program. The survey was made in the strip between the outer limit of the artisanal fishing zone and the external edge of the continental shelf, and from Punta Gallinas to Cabo TiburĂłn, on the Colombian Caribbean.  Some fishing operations with bottom long lines also took place, as well as oceanographic stations in all the wurveyed area.  Although some resultos of interest were disclosed to the businessmen in the fishing sector through a technical bulletin and a workshop, the main results had not yet been formally published.  The present work rescues the most important aspects from the fishing and oceanographic point of view, making emphasis on the environmental conditions in the sites where groupers and snappers were caught.En el marco del programa de pesca Vecep (Inpa-Colombia) se llevĂł a cabo una campaña piloto de reconocimiento acustico de los fondos y de recursos demersales, previa a la evaluaciĂłn por ĂĄrea barrida realizada por ese mismo programa.  La campaña se realizĂł en la franja del Caribe Colombiano comprendida entre el lĂ­mite exterior de la pesca artesanal y al borde extremo de la plataforma  continental y desde Punta Gallinas  hasta Cabo TiburĂłn.  Se efectuaron tambien algunas pescas comprobatorias con palangres de fondo, asĂ­ como estaciones oceanogrĂĄficas en todo el area prospectada.     The in-plane paraconductivity in La_{2-x}Sr_xCuO_4 thin film superconductors at high reduced-temperatures: Independence of the normal-state pseudogap

    The in-plane resistivity has been measured in La2−xSrxCuO4La_{2-x}Sr_xCuO_4 (LSxCO) superconducting thin films of underdoped (x=0.10,0.12x=0.10,0.12), optimally-doped (x=0.15x=0.15) and overdoped (x=0.20,0.25x=0.20,0.25) compositions. These films were grown on (100)SrTiO3_3 substrates, and have about 150 nm thickness. The in-plane conductivity induced by superconducting fluctuations above the superconducting transition (the so-called in-plane paraconductivity, Δσab\Delta\sigma_{ab}) was extracted from these data in the reduced-temperature range 10^{-2}\lsim\epsilon\equiv\ln(T/\Tc)\lsim1. Such a Δσab(Ï”)\Delta\sigma_{ab}(\epsilon) was then analyzed in terms of the mean-field--like Gaussian-Ginzburg-Landau (GGL) approach extended to the high-Ï”\epsilon region by means of the introduction of a total-energy cutoff, which takes into account both the kinetic energy and the quantum localization energy of each fluctuating mode. Our results strongly suggest that at all temperatures above Tc, including the high reduced-temperature region, the doping mainly affects in LSxCO thin films the normal-state properties and that its influence on the superconducting fluctuations is relatively moderate: Even in the high-Ï”\epsilon region, the in-plane paraconductivity is found to be independent of the opening of a pseudogap in the normal state of the underdoped films.Comment: 35 pages including 10 figures and 1 tabl

    The Depletion of Nuclear Glutathione Impairs Cell Proliferation in 3t3 Fibroblasts

    BACKGROUND:Glutathione is considered essential for survival in mammalian cells and yeast but not in prokaryotic cells. The presence of a nuclear pool of glutathione has been demonstrated but its role in cellular proliferation and differentiation is still a matter of debate. PRINCIPAL FINDINGS:We have studied proliferation of 3T3 fibroblasts for a period of 5 days. Cells were treated with two well known depleting agents, diethyl maleate (DEM) and buthionine sulfoximine (BSO), and the cellular and nuclear glutathione levels were assessed by analytical and confocal microscopic techniques, respectively. Both agents decreased total cellular glutathione although depletion by BSO was more sustained. However, the nuclear glutathione pool resisted depletion by BSO but not with DEM. Interestingly, cell proliferation was impaired by DEM, but not by BSO. Treating the cells simultaneously with DEM and with glutathione ethyl ester to restore intracellular GSH levels completely prevented the effects of DEM on cell proliferation. CONCLUSIONS:Our results demonstrate the importance of nuclear glutathione in the control of cell proliferation in 3T3 fibroblasts and suggest that a reduced nuclear environment is necessary for cells to progress in the cell cycle

    The question whether taxonomic descriptions naming new animal species without type specimen(s) deposited in collections should be accepted for publication by scientific journals and allowed by the Code has already been discussed in Zootaxa (Dubois & NemĂ©sio 2007; Donegan 2008, 2009; NemĂ©sio 2009a–b; Dubois 2009; Gentile & Snell 2009; Minelli 2009; Cianferoni & Bartolozzi 2016; Amorim et al. 2016). This question was again raised in a letter supported by 35 signatories published in the journal Nature (Pape et al. 2016) on 15 September 2016. On 25 September 2016, the following rebuttal (strictly limited to 300 words as per the editorial rules of Nature) was submitted to Nature, which on 18 October 2016 refused to publish it. As we think this problem is a very important one for zoological taxonomy, this text is published here exactly as submitted to Nature, followed by the list of the 493 taxonomists and collection-based researchers who signed it in the short time span from 20 September to 6 October 2016
