Portfolio decision analysis for pandemic sentiment assessment based on finance and web queries
COVID-19 has spread worldwide, affecting peopleās health and the socio-economic environment. Such a pandemic is responsible for peopleās deteriorated mood, pessimism, and lack of trust in the future. This paper presents a portfolio decision analysis framework for policymakers aiming at recovering the population from psychological distress. Specifically, we explore the relative relevance of a country to the overall āmood of the worldā in light of pursuing predefined targets through optimization criteria. Toward this aim, we design a statistical indicator for measuring the mood by considering the financial marketsā outcomes and the peopleās online searches about COVID-19. Then, we adapt existing portfolio selection models to evaluate the role of an extensive collection of countries and stock markets based on different criteria. More precisely, such criteria are established assuming ārationalā goals of a policymaker, namely to aspire to a general and stable optimism and avoid waves of opposite moods or excess pessimism. Empirical experiments validate the theoretical proposal. The employed dataset contains 39 countries selected on the basis of data reliability and relevance in the context of COVID-19. Data on daily Google Trends searches of the term ācoronavirusā (and its translations) and closing prices of relevant domestic stock indexes are considered for 2020 to develop the statistical mood indicator. Results offer different insights based on the selected optimization criteria. The practical implications of the proposed models have been illustrated through arguments based on a National Recovery and Resilience Plan-type normative framework

Effect of Strain Magnitude on the Tissue Properties of Engineered Cardiovascular Constructs
Mechanical loading is a powerful regulator of tissue properties in engineered cardiovascular tissues. To ultimately regulate the biochemical processes, it is essential to quantify the effect of mechanical loading on the properties of engineered cardiovascular constructs. In this study the Flexercell FX-4000T (Flexcell Int. Corp., USA) straining system was modified to simultaneously apply various strain magnitudes to individual samples during one experiment. In addition, porous polyglycolic acid (PGA) scaffolds, coated with poly-4-hydroxybutyrate (P4HB), were partially embedded in a silicone layer to allow long-term uniaxial cyclic mechanical straining of cardiovascular engineered constructs. The constructs were subjected to two different strain magnitudes and showed differences in biochemical properties, mechanical properties and organization of the microstructure compared to the unstrained constructs. The results suggest that when the tissues are exposed to prolonged mechanical stimulation, the production of collagen with a higher fraction of crosslinks is induced. However, straining with a large strain magnitude resulted in a negative effect on the mechanical properties of the tissue. In addition, dynamic straining induced a different alignment of cells and collagen in the superficial layers compared to the deeper layers of the construct. The presented model system can be used to systematically optimize culture protocols for engineered cardiovascular tissues
Mechanisms and strategies for effective delivery of antisense and siRNA oligonucleotides
The potential use of antisense and siRNA oligonucleotides as therapeutic agents has elicited a great deal of interest. However, a major issue for oligonucleotide-based therapeutics involves effective intracellular delivery of the active molecules. In this Survey and Summary, we review recent reports on delivery strategies, including conjugates of oligonucleotides with various ligands, as well as use of nanocarrier approaches. These are discussed in the context of intracellular trafficking pathways and issues regarding in vivo biodistribution of molecules and nanoparticles. Molecular-sized chemical conjugates and supramolecular nanocarriers each display advantages and disadvantages in terms of effective and nontoxic delivery. Thus, choice of an optimal delivery modality will likely depend on the therapeutic context
