    Creation of silicon vacancy color centers with a narrow emission line in nanodiamonds by ion implantation

    Nanodiamonds containing silicon-vacancy (SiV) centers with high brightness, high photo-stability, and a narrow zero phonon line (ZPL) have attracted attention for bioimaging, nanoscale thermometry, and quantum technologies. One method to create such nanodiamonds is the milling of diamond films synthesized by chemical vapor deposition (CVD). However, this requires post-processing such as acid treatment and centrifugation after the milling process. Therefore, the number of SiV center-containing nanodiamonds made from an initial CVD diamond is small. An alternative method without these problems is the implantation of Si ions into preselected nanodiamonds. This method, however, has an issue regarding the ZPL linewidths, which are more than twice as broad as those in nanodiamonds synthesized by CVD. In order to reduce the linewidth, we employed annealing treatment at high temperatures (up to 1100°C) and high vacuum after the implantation. For an ion fluence of 1013 ions/cm2, a ZPL with a linewidth of about 7 nm at room temperature was observed for a nanodiamond with a median size of 29.9 ± 16.0 nm. This was close to the linewidth for nanodiamonds created by CVD

    Current Performance and On-Going Improvements of the 8.2 m Subaru Telescope

    An overview of the current status of the 8.2 m Subaru Telescope constructed and operated at Mauna Kea, Hawaii, by the National Astronomical Observatory of Japan is presented. The basic design concept and the verified performance of the telescope system are described. Also given are the status of the instrument package offered to the astronomical community, the status of operation, and some of the future plans. The status of the telescope reported in a number of SPIE papers as of the summer of 2002 are incorporated with some updates included as of 2004 February.     Biodistribution and Pharmacokinetics of O-Palmitoyl Tilisolol, a Lipophilic Prodrug of Tilisolol, after Intravenous Administration in Rats

    The purpose of this study was to modify the biodistribution and pharmacokinetics of tilisolol, a β-blocker, using the palmitoyl prodrug approach. After intravenous administration of tilisolol and O-palmitoyl tilisolol in rats, drug concentrations were determined in blood, bile, urine, and several tissues. The concentration-time profiles of tilisolol and O-palmitoyl tilisolol were analyzed pharmacokinetically. The blood concentrations of O-palmitoyl tilisolol after intravenous administration of O-palmitoyl tilisolol were about 10-fold higher than those of tilisolol after intravenous administration of tilisolol. The biliary excretion rates of O-palmitoyl tilisolol and tilisolol after intravenous administration of O-palmitoyl tilisolol were about 10- to 100-fold larger than those of tilisolol after intravenous administration of tilisolol. In addition, the hepatic uptake clearance of O-palmitoyl tilisolol after intravenous administration of O-palmitoyl tilisolol was 3.6-fold higher than that of tilisolol after the intravenous administration of tilisolol. In the in vitro experiments, it was demonstrated that the distribution ratios between blood cells and plasma (blood/plasma) of O-palmitoyl tilisolol and tilisolol was 95.7 and 55.5%, respectively. These findings suggest that O-palmitoyl tilisolol exists as a binding form with biological components, especially blood cells, in systemic circulation. In conclusion, the palmitoyl prodrug approach is useful as a drug delivery system to deliver the parent drug to the liver

    The Constrained Maximal Expression Level Owing to Haploidy Shapes Gene Content on the Mammalian X Chromosome.

    X chromosomes are unusual in many regards, not least of which is their nonrandom gene content. The causes of this bias are commonly discussed in the context of sexual antagonism and the avoidance of activity in the male germline. Here, we examine the notion that, at least in some taxa, functionally biased gene content may more profoundly be shaped by limits imposed on gene expression owing to haploid expression of the X chromosome. Notably, if the X, as in primates, is transcribed at rates comparable to the ancestral rate (per promoter) prior to the X chromosome formation, then the X is not a tolerable environment for genes with very high maximal net levels of expression, owing to transcriptional traffic jams. We test this hypothesis using The Encyclopedia of DNA Elements (ENCODE) and data from the Functional Annotation of the Mammalian Genome (FANTOM5) project. As predicted, the maximal expression of human X-linked genes is much lower than that of genes on autosomes: on average, maximal expression is three times lower on the X chromosome than on autosomes. Similarly, autosome-to-X retroposition events are associated with lower maximal expression of retrogenes on the X than seen for X-to-autosome retrogenes on autosomes. Also as expected, X-linked genes have a lesser degree of increase in gene expression than autosomal ones (compared to the human/Chimpanzee common ancestor) if highly expressed, but not if lowly expressed. The traffic jam model also explains the known lower breadth of expression for genes on the X (and the Z of birds), as genes with broad expression are, on average, those with high maximal expression. As then further predicted, highly expressed tissue-specific genes are also rare on the X and broadly expressed genes on the X tend to be lowly expressed, both indicating that the trend is shaped by the maximal expression level not the breadth of expression per se. Importantly, a limit to the maximal expression level explains biased tissue of expression profiles of X-linked genes. Tissues whose tissue-specific genes are very highly expressed (e.g., secretory tissues, tissues abundant in structural proteins) are also tissues in which gene expression is relatively rare on the X chromosome. These trends cannot be fully accounted for in terms of alternative models of biased expression. In conclusion, the notion that it is hard for genes on the Therian X to be highly expressed, owing to transcriptional traffic jams, provides a simple yet robustly supported rationale of many peculiar features of X's gene content, gene expression, and evolution

    Thickness distribution, texture and stratigraphy, and a simple probabilistic model for dynamical thickening of sea ice in the southern Sea of Okhotsk

    Sea ice thickness data and sea ice samples were analyzed to examine the characteristics of the ice thickness distribution and ice texture, and to understand ice growth processes in the southern Sea of Okhotsk. Ice thickness data and samples were obtained aboard the icebreaker Soya in early February, the ice growth season. Ice thickness data, which were obtained with a video monitoring system installed on the side deck of the ship each winter from 1991 to 2000 except 1995, show that the average thickness ranges from 19±7 to 55±23 cm and that it matches the characteristics of a Poisson distribution. Ice structure analysis reveals that granular texture occupies about three quarters of the total ice thickness and that the ice exhibits a layered structure with unit thickness averaging 5 to 10 cm. Stratigraphy and stable isotopic composition of the ice indicate that snow ice accounts for 10% and frazil ice accounts for 64% of the total ice thickness. This suggests that dynamic ice thickening processes such as frazil ice growth and piling up are more significant than congelation growth. On the basis of these characteristics, which resemble more those of Antarctic than Arctic sea ice, we propose a conceptual model for the ice thickening process in this region. It is shown that this model can explain the shape of the ice thickness distribution well, and is analogous with the concept of the “pancake cycle” and multiple rafting of Antarctic sea ice growth and thickening.An edited version of this paper was published by AGU.
