    Restless Legs Syndrome-associated intronic common variant in Meis1 alters enhancer function in the developing telencephalon

    This article, published in Genome Research, is available under a Creative Commons License (Attribution-NonCommercial 3.0 Unported).-- et al.Genome-wide association studies (GWAS) identified the MEIS1 locus for Restless Legs Syndrome (RLS), but causal single nucleotide polymorphisms (SNPs) and their functional relevance remain unknown. This locus contains a large number of highly conserved noncoding regions (HCNRs) potentially functioning as cis-regulatory modules. We analyzed these HCNRs for allele-dependent enhancer activity in zebrafish and mice and found that the risk allele of the lead SNP rs12469063 reduces enhancer activity in the Meis1 expression domain of the murine embryonic ganglionic eminences (GE). CREB1 binds this enhancer and rs12469063 affects its binding in vitro. In addition, MEIS1 target genes suggest a role in the specification of neuronal progenitors in the GE, and heterozygous Meis1-deficient mice exhibit hyperactivity, resembling the RLS phenotype.     Synthesis, Photochemical, and Redox Properties of Gold(I) and Gold(III) Pincer Complexes Incorporating a 2,2′:6′,2″-Terpyridine Ligand Framework

    Reaction of [Au(C6F5)(tht)] (tht = tetrahydrothiophene) with 2,2′:6′,2″-terpyridine (terpy) leads to complex [Au(C6F5)(η1-terpy)] (1). The chemical oxidation of complex (1) with 2 equiv of [N(C6H4Br-4)3](PF6) or using electrosynthetic techniques affords the Au(III) complex [Au(C6F5)(η3-terpy)](PF6)2 (2). The X-ray diffraction study of complex 2 reveals that the terpyridine acts as tridentate chelate ligand, which leads to a slightly distorted square-planar geometry. Complex 1 displays fluorescence in the solid state at 77 K due to a metal (gold) to ligand (terpy) charge transfer transition, whereas complex 2 displays fluorescence in acetonitrile due to excimer or exciplex formation. Time-dependent density functional theory calculations match the experimental absorption spectra of the synthesized complexes. In order to further probe the frontier orbitals of both complexes and study their redox behavior, each compound was separately characterized using cyclic voltammetry.     Aerosol optical characteristics from a summer campaign in an urban coastal Mediterranean area

    The authors present a preliminary study of some optical properties of atmospheric aerosols over the area of Valencia, Spain, a coastal Mediterranean city. Measurements of spectral direct irradiance in the 300-1100 nm range were taken simultaneously at three sites: rural-continental, rural-coastal, and urban-coastal, all located within a 50 km radius of the city of Valencia. The irradiance measurements were obtained using three Li-cor 1800 spectroradiometers provided with radiance limiting tubes with field of views (FOVs) of 4.7/spl deg/. The measurements were made under clear sky conditions during a field campaign carried out in the summer of 1998. In order to avoid the uncertainties associated with the determination of the water vapor content and the other atmospheric constituents, the analysis of the spectral aerosol optical thickness (AOT) values was limited to the 400-670 mm spectral band. From the values of the spectral AOT, both the Angstrom coefficients and the aerosol size distributions were obtained. The results show the great dependence of the optical aerosol characteristic on the direction of the prevailing winds (maritime or continental) in this area

    Consumption of the Dietary Flavonoids Quercetin, Luteolin and Kaempferol and Overall Risk of Cancer - A Review and Meta-Analysis of the Epidemiological Data

    Numerous epidemiological and preclinical studies suggest that flavonoids may play an important role in the decreased risk of cancer associated with a diet rich in plant-derived foods. In this article, we have reviewed the epidemiological studies assessing the relationship between the consumption of three of the most common flavonoids, i.e. quercetin, luteolin and kaempferol, and the risk of developing cancer. We have also performed a meta-analysis on the consumption of these three flavonoids (alone and combined) and overall risk of cancer. The analysis of data from 18 case-control studies (8585 cases with cancer and 9975 control subjects) revealed that a high consumption of these three flavonoids (combined) was associated with a statistically significant reduction of overall cancer risk (OR: 0.73; 95% CI: 0.63, 0.84; p<0.01). A reduction of overall cancer risk was also observed for quercetin (OR: 0.73; 95% CI: 0.62, 0.86; p<0.01), kaempferol (OR: 0.86; 95% CI: 0.73, 1.11; p>0.05) and luteolin (OR: 0.90; 95% CI: 0.69, 1.18; p>0.05), which was statistically significant for quercetin. A high intake of these three flavonoids (combined) was also associated with a statistically significant reduction of lung cancer risk (OR: 0.67; 95% CI: 0.49, 0.91; p<0.05) and colon cancer risk (OR: 0.75; 95% CI: 0.57, 0.98; p<0.05). The analysis of data from 14 cohort studies (385033 individuals and 10809 cancer cases) showed a statistically significant reduction of overall cancer risk for the three flavonoids combined (RR: 0.89; 95% CI: 0.80, 1.00; p<0.05), for quercetin (RR: 0.82; 95% CI: 0.71, 0.96; p<0.05) and for kaempferol (RR: 0.88; 95% CI: 0.78, 0.99; p<0.05), and a non-statistically significant reduction for luteolin (RR: 0.95; 95% CI: 0.67, 1.34; p>0.05).     Scope for growth and dietary needs of Mediteranean Pinnids maintained in captivity

    Abstract Background: The measurement of the energy available for growth (scope of growth, SFG) can be used in bivalves to make a long-term prediction in a short-term experiment of the condition of the individual. In order to tackle the best conditions for captive maintenance of Mediterranean Pinnids, a SFG study was conducted using Pinna rudis as a model species. Three diets were examined to test the viability of live microalgae and commercial products: i) a control diet using 100% of live microalgae based on the species Isochrysis galbana (t-ISO), ii) a 100% of commercial microalgae diet based on the product Shellfish Diet 1800®, and iii) a 50/50% mix diet of I. galbana (t-ISO) and Shellfish Diet 1800®. Results: SFG results showed significant differences among diets in the physiological functions measured and suggested lower acceptability and digestibility of the commercial product. Negative SFG values were obtained for the commercial diet which indicates that it should be rejected for both Pinnid maintenance. The mixed diet showed improved physiological performance compared to the commercial diet, resulting in a higher SFG that had no significant differences with the control diet. However, in the long-term, the lower digestibility of the mixed diet compared to the control diet could lead to a deterioration of individuals’ conditions and should be considered cautiously. Conclusions: This work represents the first case study of SFG in Pinna spp. and provides fundamental data on dietary needs for the critically endangered species, P. nobilis.En prens

    RoboSTEAM - A Challenge Based Learning Approach for integrating STEAM and develop Computational Thinking

    We live in a digital society that needs new better prepared professionals for the new challenges and opportunities provided by the ICT. Students must learn how to deal with all the issues that emerge in this new context. They hsould caquire computational thinking skills by integrating STEAM, however this needs for changes in current learning curricula and also new learning approaches. RoboSTEAM project deals with this issue by the application of a Challenge Based Learning approach that uses Robotics and Physical Devices. One of the problems found during the project is the complexity of the application of a Challenge Based Learning approach due to the special needs of each educational institution. Given this situation the present work presents provides a flexible definition of challenge and describes also samples regarding how to use them

    Obesity-associated variants within FTO form long-range functional connections with IRX3

    PMCID: PMC4113484.-- et al.Genome-wide association studies (GWAS) have reproducibly associated variants within introns of FTO with increased risk for obesity and type 2 diabetes (T2D). Although the molecular mechanisms linking these noncoding variants with obesity are not immediately obvious, subsequent studies in mice demonstrated that FTO expression levels influence body mass and composition phenotypes. However, no direct connection between the obesity-associated variants and FTO expression or function has been made. Here we show that the obesity-associated noncoding sequences within FTO are functionally connected, at megabase distances, with the homeobox gene IRX3. The obesity-associated FTO region directly interacts with the promoters of IRX3 as well as FTO in the human, mouse and zebrafish genomes. Furthermore, long-range enhancers within this region recapitulate aspects of IRX3 expression, suggesting that the obesity-associated interval belongs to the regulatory landscape of IRX3. Consistent with this, obesity-associated single nucleotide polymorphisms are associated with expression of IRX3, but not FTO, in human brains. A direct link between IRX3 expression and regulation of body mass and composition is demonstrated by a reduction in body weight of 25 to 30% in Irx3-deficient mice, primarily through the loss of fat mass and increase in basal metabolic rate with browning of white adipose tissue. Finally, hypothalamic expression of a dominant-negative form of Irx3 reproduces the metabolic phenotypes of Irx3-deficient mice.
