    FIB/FESEM experimental and analytical assessment of R-curve behavior of WC-Co cemented carbides

    Exceptional fracture toughness levels exhibited by WC-Co cemented carbides (hardmetals) are due mainly to toughening derived from plastic stretching of crack-bridging ductile enclaves. This takes place due to the development of a multiligament zone at the wake of cracks growing in a stable manner. As a result, hardmetals exhibit crack growth resistance (R-curve) behavior. In this work, the toughening mechanics and mechanisms of these materials are investigated by combining experimental and analytical approaches. Focused Ion Beam technique (FIB) and Field-Emission Scanning Electron Microscopy ( FESEM) are implemented to obtain serial sectioning and imaging of crack-microstructure interaction in cracks arrested after stable extension under monotonic loading. The micrographs obtained provide experimental proof of the developing multiligament zone, including failure micromechanisms within individual bridging ligaments. Analytical assessment of the multiligament zone is then conducted on the basis of experimental information attained from FIB/FESEM images, and a model for the description of R-curve behavior of hardmetals is proposed. It was found that, due to the large stresses supported by the highly constrained and strongly bonded bridging ligaments, WC-Co cemented carbides exhibit quite steep but short R-curve behavior.     Recycling of tailings from the Barruecopardo tungsten deposit for the production of glass

    Tailings from tungsten mining activities in the vicinity of Barruecopardo (Salamanca) represent high environmental pollution. In this paper we present a study of the use of these wastes as raw materials for the manufacture of glass. This procedure aims to contribute to environmental remediation of mining areas through vitrification, a process which offers an alternative for stabilization of hazardous wastes. In addition, the marketing of the obtained product would provide an additional income to the mining areas. The chemical composition of the tailings to be used as raw materials was determined by X-ray fluorescence and their mineralogy by X-ray diffraction. Wastes are of granitic composition enriched in potentially toxic elements. For this study, a representative sample of mining wastes of sandy grain size was used to make the glass. On the basis of its composition, glass was formulated by adding 29.28 mass% of CaCO3 and 14.03 mass% of Na2CO3 and a green glass was produced. Crystallisation temperatures, obtained by DTA, were 875 and 1022 °C and the melting temperature was 1175 °C. The transition temperature of glass was of 644 °C. The temperatures for the fixed viscosity points, and the working temperatures were obtained. A thermal treatment induced devitrification to produce a glass-ceramic made of nepheline and wollastonite. Leaching tests of the obtained glass confirm its capacity to retain potentially toxic elements

    Complementariedad entre las relaciones diacrónicas de los T-Patterns y los patrones de conducta en acciones de esgrima de espada masculina de élite

    El objetivo de este artículo es establecer las posibles relacionesdiacrónicas detectadas en el análisis táctico de los asaltos de esgrima deespada masculina, desde tres técnicas analíticas complementarias. Se utilizóun diseño observacional nomotético, puntual y multidimensional enel registro de ocho asaltos de dos esgrimistas finalistas en los mundialesde Budapest 2013 y Kazán 2014. Se utilizó el instrumento de observaciónESGRIMOBS. En el muestreo observacional realizado con LINCE v.1.1 seregistraron 407 frases de armas. El análisis metodológico se realizó con losprogramas THEME v.6 Edu, GSEQ5 y HOISAN. Los resultados concluyenel interés de este análisis complementario a través de dos claras orientaciones.En primer lugar, un análisis táctico (intra-frase de armas), que vinculael pensamiento táctico con la ejecución técnica específica y su eficacia.Este análisis intra-frase de armas se lleva a cabo mediante el análisis secuencialdel Retardo 0 (análisis de coocurrencias) y las configuraciones de losT-Patterns. En segundo lugar, un análisis estratégico (inter-frases de armas)que nos informa de la relación bidireccional, prospectiva y retrospectiva, delas acciones de esgrima en relación a su eficacia. Este análisis inter-frases dearmas se realiza mediante el análisis secuencial en los retardos -1 a -5 y +1a +5, las agrupaciones de clusters de los T-Patterns y el análisis de coordenadaspolares. Estas herramientas nos permiten establecer relaciones directasentre las distintas acciones tácticas y su eficacia en el marcador, así comosobre las operaciones estratégicas de los tiradores durante la competición,pudiendo constituir un excelente complemento para la mejora de la tomade decisiones de los esgrimistas

    Compositional data supports decentralized model of production and circulation of artifacts in the pre-Columbian south-central Andes

The final version is available from National Academy of Sciences via the DOI in this record.The circulation and exchange of goods and resources at various scales have long been considered central to the understanding of complex societies, and the Andes have provided a fertile ground for investigating this process. However, long-standing archaeological emphasis on typological analysis, although helpful to hypothesize the direction of contacts, has left important aspects of ancient exchange open to speculation. To improve understanding of ancient exchange practices and their potential role in structuring alliances, we examine material exchanges in northwest Argentina (part of the south-central Andes) during 400 BC to AD 1000 (part of the regional Formative Period), with a multianalytical approach (petrography, instrumental neutron activation analysis, laser ablation inductively coupled plasma mass spectrometry) to artifacts previously studied separately. We assess the standard centralized model of interaction vs. a decentralized model through the largest provenance database available to date in the region. The results show: (i) intervalley heterogeneity of clays and fabrics for ordinary wares; (ii) intervalley homogeneity of clays and fabrics for a wide range of decorated wares (e.g., painted Ciénaga); (iii) selective circulation of two distinct polychrome wares (Vaquerías and Condorhuasi); (iv) generalized access to obsidian from one major source and various minor sources; and (v) selective circulation of volcanic rock tools from a single source. These trends reflect the multiple and conflicting demands experienced by people in small-scale societies, which may be difficult to capitalize by aspiring elites. The study undermines centralized narratives of exchange for this period, offering a new platform for understanding ancient exchange based on actual material transfers, both in the Andes and beyond.We thank the former directors of Museo Etnográfico (University of Buenos Aires), M.     Análisis del fenómeno de la expansividad de materiales en túneles construidos en anhidrita. Consecuencias y experiencias

    Los graves problemas geotécnicos sucedidos en numerosos túneles europeos excavados en materiales que generan problemas de estabilidad, como consecuencia de su expansividad, los convierten en canalizadores de aguas. Estos materiales son fácilmente lavables al disolverse los sulfatos con el agua, provocando una decompresión y remoldeo del macizo, agravando y trasladando el problema más allá del inmediato entorno del túnel. En el caso particular de presencia de capas de anhidrita entre argilitas, además de la adopción de medidas de sostenimiento y revestimiento especiales, el sostenimiento primario y el revestimiento posterior deben ser excepcionalmente resistentes a las presiones que pueden llegar a transmitir el terreno a medio y largo plazo. En este artículo se describen los aspectos más relevantes de comportamiento hidromecánico de los suelos y las rocas sulfatadas y se presentan algunas experiencias, concluyendo sobre aspectos asociados al fenómeno de hinchamiento en túneles y su posible tratamiento

    Molecular epidemiology of enterovirus 71, coxsackievirus A16 and A6 associated with hand, foot and mouth disease in Spain

    Producción CientíficaHand, foot and mouth disease (HFMD) is a childhood illness frequently caused by genotypes belonging to the enterovirus A species, including coxsackievirus (CV)-A16 and enterovirus (EV)-71. Between 2010 and 2012, several outbreaks and sporadic cases of HFMD occurred in different regions of Spain. The objective of the present study was to describe the enterovirus epidemiology associated with HFMD in the country. A total of 80 patients with HFMD or atypical rash were included. Detection and typing of the enteroviruses were performed directly in clinical samples using molecular methods. Enteroviruses were detected in 53 of the patients (66%). CV-A6 was the most frequent genotype, followed by CV-A16 and EV-71, but other minority types were also identified. Interestingly, during almost all of 2010, CV-A16 was the only causative agent of HFMD but by the end of the year and during 2011, CV-A6 became predominant, while CV-A16 was not detected. In 2012, however, both CV-A6 and CV-A16 circulated. EV-71 was associated with HFMD symptoms only in three cases during 2012. All Spanish CV-A6 sequences segregated into one major genetic cluster together with other European and Asian strains isolated between 2008 and 2011, most forming a particular clade. Spanish EV-71 strains belonged to subgenogroup C2, as did most of the European sequences circulated. In conclusion, the recent increase of HFMD cases in Spain and other European countries has been due to a larger incidence of circulating species A enteroviruses, mainly CV-A6 and CV-A16, and the emergence of new genetic variants of these viruses

    Structural Analysis of Prolyl Oligopeptidases Using Molecular Docking and Dynamics: Insights into Conformational Changes and Ligand Binding

    Prolyl oligopeptidase (POP) is considered as an important pharmaceutical target for the treatment of numerous diseases. Despite enormous studies on various aspects of POPs structure and function still some of the questions are intriguing like conformational dynamics of the protein and interplay between ligand entry/egress. Here, we have used molecular modeling and docking based approaches to unravel questions like differences in ligand binding affinities in three POP species (porcine, human and A. thaliana). Despite high sequence and structural similarity, they possess different affinities for the ligands. Interestingly, human POP was found to be more specific, selective and incapable of binding to a few planar ligands which showed extrapolation of porcine POP in human context is more complicated. Possible routes for substrate entry and product egress were also investigated by detailed analyses of molecular dynamics (MD) simulations for the three proteins. Trajectory analysis of bound and unbound forms of three species showed differences in conformational dynamics, especially variations in β-propeller pore size, which was found to be hidden by five lysine residues present on blades one and seven. During simulation, β-propeller pore size was increased by ∼2 Å in porcine ligand-bound form which might act as a passage for smaller product movement as free energy barrier was reduced, while there were no significant changes in human and A. thaliana POPs. We also suggest that these differences in pore size could lead to fundamental differences in mode of product egress among three species. This analysis also showed some functionally important residues which can be used further for in vitro mutagenesis and inhibitor design. This study can help us in better understanding of the etiology of POPs in several neurodegenerative diseases

    Increase in serotype 19A prevalence and amoxicillin non-susceptibility among paediatric Streptococcus pneumoniae isolates from middle ear fluid in a passive laboratory-based surveillance in Spain, 1997-2009

    BACKGROUND: Conjugate vaccines, such as the 7-valent conjugate vaccine (PCV7), alter serotype nasopharyngeal carriage, potentially increasing cases of otitis media by non-vaccine serotypes. METHODS: All paediatric middle ear fluid (MEF) isolates received in the Spanish Reference Laboratory for Pneumococci through a passive, laboratory-based surveillance system from January 1997 to June 2009 were analysed. Data from 1997 to 2000 were pooled as pre-vaccination period. Trends over time were explored by linear regression analysis. RESULTS: A total of 2,077 isolates were analysed: 855 belonging to PCV7 serotypes, 466 to serotype 19A, 215 to serotype 3, 89 to serotype 6A and 452 to other serotypes ( 35% isolates) since PCV7 strains represented < 11% of total clinical isolates. CONCLUSIONS: In contrast to reports on invasive pneumococcal strains, in MEF isolates the reduction in the prevalence of PCV7 serotypes was not associated with decreases in penicillin/erythromycin non-susceptibility. The high prevalence of serotype 19A among paediatric MEF isolates and the amoxicillin non-susceptibility found in this serotype are worrisome since amoxicillin is the most common antibiotic used in the treatment of acute otitis media. These data suggest that non-PCV7 serotypes (mainly serotype 19A followed by serotypes 3 and 6A) are important etiological agents of acute otitis media and support the added value of the broader coverage of the new 13-valent conjugate vaccine.This study was supported in part by an unrestricted grant from Pfizer S.A., Madrid, Spain and PRISM-AG, Madrid, Spain.     A mechanically active heterotypic E-cadherin/N-cadherin adhesion enables fibroblasts to drive cancer cell invasion

    Cancer-associated fibroblasts (CAFs) promote tumour invasion and metastasis. We show that CAFs exert a physical force on cancer cells that enables their collective invasion. Force transmission is mediated by a heterophilic adhesion involving N-cadherin at the CAF membrane and E-cadherin at the cancer cell membrane. This adhesion is mechanically active; when subjected to force it triggers β-catenin recruitment and adhesion reinforcement dependent on α-catenin/vinculin interaction. Impairment of E-cadherin/N-cadherin adhesion abrogates the ability of CAFs to guide collective cell migration and blocks cancer cell invasion. N-cadherin also mediates repolarization of the CAFs away from the cancer cells. In parallel, nectins and afadin are recruited to the cancer cell/CAF interface and CAF repolarization is afadin dependent. Heterotypic junctions between CAFs and cancer cells are observed in patient-derived material. Together, our findings show that a mechanically active heterophilic adhesion between CAFs and cancer cells enables cooperative tumour invasion
