    Measuring the inertial properties of a tennis racket

    Simple and bifilar pendulum were used to measure the mass moments of inertia of three tennis rackets. The pendulum setups were filmed using an off-the-shelf camcorder, with a stopwatch in view to provide timing data. The measurement accuracy was assessed using calibration rods of known mass moment of inertia. The simple pendulum method was found to be most accurate (<1.0 % difference to theoretical value) when a square profile rod was used as a pivot. The bifilar pendulum was found to be very accurate (0.0% difference to theoretical value) but sensitive to non-parallel support wires. A Babolat Racket Diagnostic Centre (RDC) was assessed using four calibration rods, of known mass moment of inertia. Measurement agreement was greater than 99.0% for mass moments of inertias within the range of 220 – 380 kg·cm2

    Biodegradation of thiocyanate by a native groundwater microbial consortium

    Gold ore processing typically generates large amounts of thiocyanate (SCN−)-contaminated effluent. When this effluent is stored in unlined tailings dams, contamination of the underlying aquifer can occur. The potential for bioremediation of SCN−-contaminated groundwater, either in situ or ex situ, remains largely unexplored. This study aimed to enrich and characterise SCN−-degrading microorganisms from mining-contaminated groundwater under a range of culturing conditions. Mildly acidic and suboxic groundwater, containing ∌135 mg L−1 SCN−, was collected from an aquifer below an unlined tailings dam. An SCN−-degrading consortium was enriched from contaminated groundwater using combinatory amendments of air, glucose and phosphate. Biodegradation occurred in all oxic cultures, except with the sole addition of glucose, but was inhibited by NH4+ and did not occur under anoxic conditions. The SCN−-degrading consortium was characterised using 16S and 18S rRNA gene sequencing, identifying a variety of heterotrophic taxa in addition to sulphur-oxidising bacteria. Interestingly, few recognised SCN−-degrading taxa were identified in significant abundance. These results provide both proof-of-concept and the required conditions for biostimulation of SCN− degradation in groundwater by native aquifer microorganisms

    Using five-minute bird counts to study magpie (Gymnorhina tibicen) impacts on other birds in New Zealand

    We used five-minute bird counts to investigate whether introduced Australian magpies (Gymnorhina tibicen) influence the abundance of other birds in rural New Zealand. Over 3 years, magpies were removed from five c. 900-ha study blocks, one in each of Northland, Waikato, Bay of Plenty, Wellington and Southland. Birds were counted in both the treatment blocks and paired non-treatment blocks for the 3 years of removal and also 1 year before. To minimise problems raised elsewhere with index counts we (1) selected treatment blocks and count stations using randomisation procedures, (2) used trained observers who spent equal time in paired treatment and non-treatment blocks, and (3) counted all blocks at the same time of year and only in good weather. On average, 548 magpies were removed from each treatment block each year, with magpie counts reduced by 76% relative to non-treatment blocks. Our results suggest magpies may restrict the movements of some birds (including kererƫ and tƫī) in rural areas, but are less important than pest mammals at limiting population abundance at a landscape scale. We submit that five-minute bird counts were appropriate for our objectives, but that more research to examine their relationship to absolute densities is needed

    Comparative bio-accessibility, bioavailability and bioequivalence of quercetin, apigenin, glucoraphanin and carotenoids from freeze-dried vegetables incorporated into a baked snack versus minimally processed vegetables:Evidence from in vitro models and a human bioavailability study

    The aim was to incorporate vegetables containing the phytochemicals quercetin, apigenin, glucoraphanin and carotenoids into a processed potato-based snack and assess their bioaccessibility and bioavailability. Three different processing routes were tested for incorporation and retention of phytochemicals in snacks using individually quick frozen or freeze-dried vegetables. No significant differences in the uptake or transport of quercetin or apigenin between a vegetable mix or snacks were observed using the CaCo-2 transwell model. Simulated in vitro digestions predicted a substantial release of quercetin and apigenin, some release of glucoraphanin but none for carotenes from either the snack or equivalent steamed vegetables. In humans, there were no significant differences in the bioavailability of quercetin, apigenin or glucoraphanin from the snack or equivalent steamed vegetables. We have shown that significant quantities of freeze-dried vegetables can be incorporated into snacks with good retention of phytochemicals and with similar bioavailability to equivalent steamed vegetables

    Retrieval, Inter-Comparison, and Validation of Above-Cloud Aerosol Optical Depth from A-train Sensors

    Absorbing aerosols produced from biomass burning and dust outbreaks are often found to overlay lower level cloud decks and pose greater potentials of exerting positive radiative effects (warming) whose magnitude directly depends on the aerosol loading above cloud, optical properties of clouds and aerosols, and cloud fraction. Recent development of a 'color ratio' (CR) algorithm applied to observations made by the Aura/OMI and Aqua/MODIS constitutes a major breakthrough and has provided unprecedented maps of above-cloud aerosol optical depth (ACAOD). The CR technique employs reflectance measurements at TOA in two channels (354 and 388 nm for OMI; 470 and 860 nm for MODIS) to retrieve ACAOD in near-UV and visible regions and aerosol-corrected cloud optical depth, simultaneously. An inter-satellite comparison of ACAOD retrieved from NASA's A-train sensors reveals a good level of agreement between the passive sensors over the homogeneous cloud fields. Direct measurements of ACA such as carried out by the NASA Ames Airborne Tracking Sunphotometer (AATS) and Spectrometer for Sky-Scanning, Sun-Tracking Atmospheric Research (4STAR) can be of immense help in validating ACA retrievals. We validate the ACA optical depth retrieved using the CR method applied to the MODIS cloudy-sky reflectance against the airborne AATS and 4STAR measurements. A thorough search of the historic AATS-4STAR database collected during different field campaigns revealed five events where biomass burning, dust, and wildfire-emitted aerosols were found to overlay lower level cloud decks observed during SAFARI-2000, ACE-ASIA 2001, and SEAC4RS- 2013, respectively. The co-located satellite-airborne measurements revealed a good agreement (RMSE less than 0.1 for AOD at 500 nm) with most matchups falling within the estimated uncertainties in the MODIS retrievals. An extensive validation of satellite-based ACA retrievals requires equivalent field measurements particularly over the regions where ACA are often observed from satellites, i.e., south-eastern Atlantic Ocean, tropical Atlantic Ocean, northern Arabian Sea, South-East and North-East Asia

    Three Novel Pigmentation Mutants Generated by Genome-Wide Random ENU Mutagenesis in the Mouse

    Three mutant mice with pigmentation phenotypes were recovered from a genomewide random mouse chemical mutagenesis study. White toes (Whto; MGI:1861986), Belly spot and white toes (Bswt; MGI:2152776) and Dark footpads 2 (Dfp2; MGI:1861991) were identified following visual inspection of progeny from a male exposed to the point mutagen ethylnitrosourea (ENU). In order to rapidly localize the causative mutations, genome-wide linkage scans were performed on pooled DNA samples from backcross animals for each mutant line. Whto was mapped to proximal mouse chromosome (Mmu) 7 between Cen (the centromere) and D7Mit112 (8.0 cM from the centromere), Bswt was mapped to centric Mmul between D1Mit214 (32.1 cM) and D1Mit480 (32.8 cM) and Dfp2 was mapped to proximalMmu4 between Cen and D4Mit18 (5.2 cM). Whto, Bswt and Dfp2 may provide novel starting points in furthering the elucidation of genetic and biochemical pathways relevant to pigmentation and associated biological processes

    Multifactorial Analysis of Differences Between Sporadic Breast Cancers and Cancers Involving BRCA1 and BRCA2 Mutations

    Background: We have previously demonstrated that breast cancers associated with inherited BRCA1 and BRCA2 gene mutations differ from each other in their histopathologic appearances and that each of these types differs from breast cancers in patients unselected for family history (i.e., sporadic cancers). We have now conducted a more detailed examination of cytologic and architectural features of these tumors. Methods: Specimens of tumor tissue (5-”m-thick sections) were examined independently by two pathologists, who were unaware of the case or control subject status, for the presence of cell mitosis, lymphocytic infiltration, continuous pushing margins, and solid sheets of cancer cells; cell nuclei, cell nucleoli, cell necrosis, and cell borders were also evaluated. The resulting data were combined with previously available information on tumor type and tumor grade and further evaluated by multifactorial analysis. All statistical tests are two-sided. Results: Cancers associated with BRCA1 mutations exhibited higher mitotic counts (P = .001), a greater proportion of the tumor with a continuous pushing margin (P<.0001), and more lymphocytic infiltration (P = .002) than sporadic (i.e., control) cancers. Cancers associated with BRCA2 mutations exhibited a higher score for tubule formation (fewer tubules) (P = .0002), a higher proportion of the tumor perimeter with a continuous pushing margin (P<.0001), and a lower mitotic count (P = .003) than control cancers. Conclusions: Our study has identified key features of the histologic phenotypes of breast cancers in carriers of mutant BRCA1 and BRCA2 genes. This information may improve the classification of breast cancers in individuals with a family history of the disease and may ultimately aid in the clinical management of patients.     Comparative in situ analysis reveals the dynamic nature of sclerenchyma cell walls of the fern Asplenium rutifolium

    Background and Aims: A key structural adaptation of vascular plants was the evolution of specialized vascular and mechanical tissues, innovations likely to have generated novel cell wall architectures. While collenchyma is a strengthening tissue typically found in growing organs of angiosperms, a similar tissue occurs in the petiole of the fern Asplenium rutifolium. Methods: The in situ cell wall (ultra)structure and composition of this tissue was investigated and characterized mechanically as well as structurally through nano-indentation and wide-angle X-ray diffraction, respectively. Key Results: Structurally the mechanical tissue resembles sclerenchyma, while its biomechanical properties and molecular composition both share more characteristics with angiosperm collenchyma. Cell wall thickening only occurs late during cell expansion or after cell expansion has ceased. Conclusions: If the term collenchyma is reserved for walls that thicken during expansive growth, the mechanical tissue in A. rutifolium represents sclerenchyma that mimics the properties of collenchyma and has the ability to modify its mechanical properties through sclerification. These results support the view that collenchyma does not occur in ferns and most probably evolved in angiosperms
