    Determination of Bulk Magnetic Volume Properties by Neutron Dark-Field Imaging

    For the production of high-class electrical steel grades a deeper understanding of the magnetic domain interaction with induced mechanical stresses is strongly required. This holds for non-oriented (NO) as well as grain-oriented (GO) steels. In the case of non-oriented steels the magnetic property degeneration after punching or laser cutting is essential for selecting correct obstructing material grades and designing efficient electrical machines. Until now these effects stay undiscovered due to the lack of adequate investigation methods that reveal local bulk information on processed laminations. Here we show how the use of a non-destructive testing method based on a neutron grating interferometry providing the dark-field image contrast delivers spatially-resolved transmission information about the local bulk domain arrangement and domain wall density. With the help of this technique it is possible to visualize magnetization processes within the NO laminations. Different representative manufacturing techniques are compared in terms of magnetic flux density deterioration such as punching, mechanically cutting by guillotine as well as laser fusion cutting using industrial high power laser beam sources. For GO steel laminations the method is applicable on the one hand to visualize the internal domain structure without being hindered by the coating layer. On the other hand, we can show the influence of the coating layer onto the underlying domain structure

    Bayesian analysis to identify new star candidates in nearby young stellar kinematic groups

    We present a new method based on a Bayesian analysis to identify new members of nearby young kinematic groups. The analysis minimally takes into account the position, proper motion, magnitude and color of a star, but other observables can be readily added (e.g. radial velocity, distance). We use this method to find new young low-mass stars in the \beta Pictoris (\beta PMG) and AB Doradus (ABDMG) moving groups and in the TW Hydrae (TWA), Tucana-Horologium (THA), Columba, Carina and Argus associations. Starting from a sample of 758 mid-KM (K5V-M5V) stars showing youth indicators such as H\alpha\ and X-ray emission, our analysis yields 215 new highly probable low-mass members of the kinematic groups analyzed. One is in TWA, 37 in \beta PMG, 17 in THA, 20 in Columba, 6 in Carina, 50 in Argus, 33 in ABDMG, and the remaining 51 candidates are likely young but have an ambiguous membership to more than one association. The false alarm rate for new candidates is estimated to be 5% for \beta PMG and TWA, 10% for THA, Columba, Carina and Argus, and 14% for ABDMG. Our analysis confirms the membership of 58 stars proposed in the literature. Firm membership confirmation of our new candidates will require measurement of their radial velocity (predicted by our analysis), parallax and lithium 6708 {\AA} equivalent width. We have initiated these follow-up observations for a number of candidates and we have identified two stars (2MASSJ0111+1526, 2MASSJ0524-1601) as very strong candidate members of the \beta PMG and one strong candidate member (2MASSJ0533-5117) of the THA; these three stars have radial velocity measurements confirming their membership and lithium detections consistent with young age. Finally, we proposed that six stars should be considered as new bona fide members of \beta PMG and ABDMG, one of which being first identified in this work, the others being known candidates from the literature.Comment: Accepted for publication in Ap

    Lenalidomide in combination with dexamethasone at first relapse in comparison with its use as later salvage therapy in relapsed or refractory multiple myeloma

    This subset analysis of data from two phase III studies in patients with relapsed or refractory multiple myeloma (MM) evaluated the benefit of initiating lenalidomide plus dexamethasone at first relapse. Multivariate analysis showed that fewer prior therapies, along with ÎČ2-microglobulin (≀2.5 mg/L), predicted a better time to progression (TTP; study end-point) with lenalidomide plus dexamethasone treatment. Patients with one prior therapy showed a significant improvement in benefit after first relapse compared with those who received two or more therapies. Patients with one prior therapy had significantly prolonged median TTP (17.1 vs. 10.6 months; P=0.026) and progression-free survival (14.1 vs. 9.5 months, P=0.047) compared with patients treated in later lines. Overall response rates were higher (66.9% vs. 56.8%, P=0.06), and the complete response plus very good partial response rate was significantly higher in first relapse (39.8% vs. 27.7%, P=0.025). Importantly, overall survival was significantly prolonged for patients treated with lenalidomide plus dexamethasone with one prior therapy, compared with patients treated later in salvage (median of 42.0 vs. 35.8 months, P=0.041), with no differences in toxicity, dose reductions, or discontinuations despite longer treatment. Therefore, lenalidomide plus dexamethasone is both effective and tolerable for second-line MM therapy and the data suggest that the greatest benefit occurs with earlier use

    Prevalence and incidence of iron deficiency in European community-dwelling older adults : An observational analysis of the DO-HEALTH trial

    Background and aim Iron deficiency is associated with increased morbidity and mortality in older adults. However, data on its prevalence and incidence among older adults is limited. The aim of this study was to investigate the prevalence and incidence of iron deficiency in European community-dwelling older adults aged ≄ 70 years. Methods Secondary analysis of the DO-HEALTH trial, a 3-year clinical trial including 2157 community-dwelling adults aged ≄ 70 years from Austria, France, Germany, Portugal and Switzerland. Iron deficiency was defined as soluble transferrin receptor (sTfR) > 28.1 nmol/L. Prevalence and incidence rate (IR) of iron deficiency per 100 person-years were examined overall and stratified by sex, age group, and country. Sensitivity analysis for three commonly used definitions of iron deficiency (ferritin  1.5) were also performed. Results Out of 2157 participants, 2141 had sTfR measured at baseline (mean age 74.9 years; 61.5% women). The prevalence of iron deficiency at baseline was 26.8%, and did not differ by sex, but by age (35.6% in age group ≄ 80, 29.3% in age group 75–79, 23.2% in age group 70–74); P  1.5. Occurrences of iron deficiency were observed with IR per 100 person-years of 9.2 (95% CI 8.3–10.1) and did not significantly differ by sex or age group. The highest IR per 100 person-years was observed in Austria (20.8, 95% CI 16.1–26.9), the lowest in Germany (6.1, 95% CI 4.7–8.0). Regarding the other definitions of iron deficiency, the IR per 100 person-years was 4.5 (95% CI 4.0–4.9) for ferritin  1.5. Conclusions Iron deficiency is frequent among relatively healthy European older adults, with people aged ≄ 80 years and residence in Austria and Portugal associated with the highest risk

    Die Erfindung betrifft ein Verfahren zur Ausbildung und Detektion von Sicherheitselementen auf der OberflĂ€che eines Bauteils und/oder in einem Bauteil, bei dem auf der OberflĂ€che des Bauteils und/oder in das Bauteil, das aus einem magnetischen oder einem nichtmagnetischen Werkstoff gebildet ist, mindestens eine Schicht oder mindestens ein Bereich, der bevorzugt mit einem magnetischen Werkstoff oder einem sich vom Bauteilwerkstoff unterscheidenden Werkstoff gebildet ist, lokal und geometrisch definiert ausgebildet wird. Durch einen lokal und geometrisch definierten Werkstoffabtrag oder Energieeintrag wird auf/an der OberflĂ€che eines Bauteils entlang einer vorgegebenen dem jeweiligen Sicherheitsmerkmal entsprechenden Kontur mindestens eine Abtragsspur, mindestens ein wĂ€rmebeeinflusster Bereich und/oder mindestens eine umgeschmolzene Behandlungsspur ausgebildet. Bei der Herstellung des Bauteils mittels eines generativen Fertigungsverfahrens wird lokal und geometrisch definiert ein sich vom Werkstoff des Bauteils unterscheidender magnetischer Werkstoff in das Bauteil an mindestens einer vorgegebenen Position eingebracht. Zur Detektion des Sicherheitselements mittels einer Magnetisierungseinheit wird mindestens ein in das Bauteil eindringendes Magnetfeld generiert oder ein magnetischer Fluss innerhalb der Magnetisierungseinheit erzeugt, der in das Bauteil eindringt. Zur ÜberprĂŒfung eines so ausgebildeten Sicherheitselements werden mit einer Detektionseinheit, die an dem Sicherheitselement infolge des mindestens einen Magnetfeldes auftretenden magnetischen Streufelder detektiert und die mit der Detektionseinheit erfassten Messsignale an eine Auswerteeinheit mit Bildverarbeitungs- oder Mustererkennungssystem ĂŒbermittelt. Mit der Auswerteeinheit mit Bildverarbeitungs- oder Mustererkennungssystem wird mittels der detektierten magnetischen Streufelder ĂŒberprĂŒft, ob eine Übereinstimmung des detektierten Sicherheitselements mit einer Vorgabe vorliegt oder nicht

    SAMHD1 is recurrently mutated in T-cell prolymphocytic leukemia

    T-cell prolymphocytic leukemia (T-PLL) is an aggressive malignancy with a median survival of the patients of less than two years. Besides characteristic chromosomal translocations, frequent mutations affect the ATM gene, JAK/STAT pathway members, and epigenetic regulators. We here performed a targeted mutation analysis for 40 genes selected from a RNA sequencing of 10 T-PLL in a collection of 28 T-PLL, and an exome analysis of five further cases. Nonsynonymous mutations were identified in 30 of the 40 genes, 18 being recurrently mutated. We identified recurrently mutated genes previously unknown to be mutated in T-PLL, which are SAMHD1, HERC1, HERC2, PRDM2, PARP10, PTPRC, and FOXP1. SAMHD1 regulates cellular deoxynucleotide levels and acts as a potential tumor suppressor in other leukemias. We observed destructive mutations in 18% of cases as well as deletions in two further cases. Taken together, we identified additional genes involved in JAK/STAT signaling (PTPRC), epigenetic regulation (PRDM2), or DNA damage repair (SAMHD1, PARP10, HERC1, and HERC2) as being recurrently mutated in T-PLL. Thus, our study considerably extends the picture of pathways involved in molecular pathogenesis of T-PLL and identifies the tumor suppressor gene SAMHD1 with ~20% of T-PLL affected by destructive lesions likely as major player in T-PLL pathogenesis
