    To examine the mechanisms of carbon mobilization and biodegradation during permafrost thawing and to establish a link between organic carbon (OC) and other chemical and microbiological parameters in forming thermokarst (thaw) lakes, we studied the biogeochemistry of OC and trace elements (TEs) in a chronosequence of small lakes that are being formed due to permafrost thawing in the northern part of western Siberia. Twenty lakes and small ponds of various sizes and ages were sampled for dissolved and colloidal organic carbon, metals and culturable heterotrophic bacterial cell number. We observed a sequence of ecosystems from peat thawing and palsa degradation due to permafrost subsidence in small ponds to large, km-size lakes that are subject to drainage to, finally, the khasyrey (drained lake) formation. There is a systematic evolution of both total dissolved and colloidal concentration of OC and TEs in the lake water along with the chronosequence of lake development that may be directly linked to the microbial mineralization of dissolved organic matter and the liberation of the inorganic components (Fe, Al, and TEs) from the organo-mineral colloids. <br><br> In this chronosequence of lake development, we observed an apparent decrease in the relative proportion of low molecular weight <1 kDa (1 kDa ~ 1 nm) OC concentration along with a decrease in the concentration of total dissolved (<0.45 μm) OC. This decrease was accompanied by an increase in the small size organic ligands (probably autochthonous exometabolites produced by the phytoplankton) and a simultaneous decrease in the proportion of large-size organic (humic) complexes of allochthonous (soil) origin. This evolution may be due to the activity of heterotrophic bacterioplankton that use allochthonous organic matter and dissolved nutrients originating from peat lixiviation. Most insoluble TEs demonstrate a systematic decrease in concentration during filtration (5 μm, 0.45 μm) exhibiting a similar pattern among different samples. At the same time, there is an increase in the relative proportion of large size particles over the <1 kDa fraction for most insoluble elements along the chronosequence of lake evolution. TEs are likely to be bound to colloidal OC and coprecipitate with the mineral (Fe, Al) part of the colloids. Upon progressive consumption of dissolved OC by the heterotrophic bacteria, there is liberation of Fe, Al, and insoluble TEs in the water column that may be subjected to coagulation in the form of particles or large-size mineral colloids

    Ectodysplasin/NF-kappa B Promotes Mammary Cell Fate via Wnt/beta-catenin Pathway

    Mammary gland development commences during embryogenesis with the establishment of a species typical number of mammary primordia on each flank of the embryo. It is thought that mammary cell fate can only be induced along the mammary line, a narrow region of the ventro-lateral skin running from the axilla to the groin. Ectodysplasin (Eda) is a tumor necrosis factor family ligand that regulates morphogenesis of several ectodermal appendages. We have previously shown that transgenic overexpression of Eda (K14-Eda mice) induces formation of supernumerary mammary placodes along the mammary line. Here, we investigate in more detail the role of Eda and its downstream mediator transcription factor NF-kappa B in mammary cell fate specification. We report that K14-Eda mice harbor accessory mammary glands also in the neck region indicating wider epidermal cell plasticity that previously appreciated. We show that even though NF-kappa B is not required for formation of endogenous mammary placodes, it is indispensable for the ability of Eda to induce supernumerary placodes. A genome-wide profiling of Eda-induced genes in mammary buds identified several Wnt pathway components as potential transcriptional targets of Eda. Using an ex vivo culture system, we show that suppression of canonical Wnt signalling leads to a dose-dependent inhibition of supernumerary placodes in K14-Eda tissue explants.Peer reviewe

    Riverine particulate C and N generated at the permafrost thaw front: case study of western Siberian rivers across a 1700 km latitudinal transect

    In contrast to numerous studies on the dynamics of dissolved ( &lt; 0.45&thinsp;µm) elements in permafrost-affected high-latitude rivers, very little is known of the behavior of river suspended ( &gt; 0.45&thinsp;µm) matter (RSM) in these regions. In order to test the effect of climate, permafrost and physio-geographical landscape parameters (bogs, forest and lake coverage of the watershed) on RSM and particulate C, N and P concentrations in river water, we sampled 33 small and medium-sized rivers (10–100&thinsp;000&thinsp;km2 watershed) along a 1700&thinsp;km N–S transect including both permafrost-affected and permafrost-free zones of the Western Siberian Lowland (WSL). The concentrations of C and N in RSM decreased with the increase in river watershed size, illustrating (i) the importance of organic debris in small rivers which drain peatlands and (ii) the role of mineral matter from bank abrasion in larger rivers. The presence of lakes in the watershed increased C and N but decreased P concentrations in the RSM. The C : N ratio in the RSM reflected the source from the deep soil horizon rather than surface soil horizon, similar to that of other Arctic rivers. This suggests the export of peat and mineral particles through suprapermafrost flow occurring at the base of the active layer. There was a maximum of both particulate C and N concentrations and export fluxes at the beginning of permafrost appearance, in the sporadic and discontinuous zone (62–64°&thinsp;N). This presumably reflected the organic matter mobilization from newly thawed organic horizons in soils at the active latitudinal thawing front. The results suggest that a northward shift of permafrost boundaries and an increase in active layer thickness may increase particulate C and N export by WSL rivers to the Arctic Ocean by a factor of 2, while P export may remain unchanged. In contrast, within a long-term climate warming scenario, the disappearance of permafrost in the north, the drainage of lakes and transformation of bogs to forest may decrease C and N concentrations in RSM by 2 to 3 times.</p

    The detergents used to solubilize GPCRs can make crystal growth the rate-limiting step in determining their structure. The Kobilka laboratory showed that insertion of T4-lysozyme (T4L) in the 3rd intracellular loop is a promising strategy towards increasing the solvent-exposed receptor area, and hence the number of possible lattice-forming contacts. The potential to use T4L with the olfactory-related receptors hOR17-4 and hVN1R1 was thus tested. The structure and function of native and T4L-variants were compared. Both receptors localized to the cell membrane, and could initiate ligand-activated signaling. Purified receptors not only had the predicted alpha-helical structures, but also bound their ligands canthoxal (MW = 178.23) and myrtenal (MW = 150.22). Interestingly, the T4L variants had higher percentages of soluble monomers compared to protein aggregates, effectively increasing the protein yield that could be used for structural and function studies. They also bound their ligands for longer times, suggesting higher receptor stability. Our results indicate that a T4L insertion may be a general method for obtaining GPCRs suitable for structural studies
