Long-Term Soil Fertility Trials in Niger, West Africa
Soils of tropical Africa have been formed mainly from Pre-Cambrian materials and others from erosion process. Because of the type of parent material, the soils are characterized by poor native fertility surface crusting and low water holding capacity.
In Africa 65% of the agricultural land, 31% of the permanent pasture land, and 19% of the forest and woodland is affected by human-induced soil degradation. Nutrient depletion is the most important element in the land degradation equation.
With increasing population pressure on the land, the traditional farming system (natural fallow) to restore soil fertility is no longer possible. The present farming systems are unsustainable and destructive to the environment with negative plant nutrient balances resulting to mining of nutrients.
Many long term trials show that although application of mineral fertilizers is an effective mean of increasing yields in arable farming systems, mineral fertilizers alone cannot sustain yields in the long run. When mineral fertilizers are combined with crop residue or manure, added to cereal-legume intercrops or rotated, sustainable production can be obtained.
Any future interventions redressing distorted nutrient budgets, to be sustainable have to develop an integrated nutrient management, conceptualized as the judicious manipulation of the nutrient inputs and outputs processes
Integrating climate change mitigation and adaptation in agriculture and forestry: opportunities and trade-offs

In recent years, there has been a growing interest shown by practitioners in agriculture, forestry, and landscape management in the links between the two strategies. This review explores the opportunities and trade-offs when managing landscapes for both climate change mitigation and adaptation; different conceptua-lizations of the links between adaptation and mitigation are highlighted. Under a first conceptualization of 'joint outcomes,' several reviewed studies analyze how activities without climatic objectives deliver joint adaptation and mitigation outcomes. In a second conceptualization of 'unintended side effects,' the focus is on how activities aimed at only one climate objective—either adaptation or mitigation—can deliver outcomes for the other objective. A third conceptualization of 'joint objectives' highlights that associating both adaptation and mitigation objectives in a climate-related activity can influence its outcomes because of multiple possible interactions. The review reveals a diversity of reasons for mainstreaming adaptation and mitigation separately or jointly in landscape management. The three broad conceptualizations of the links between adaptation and mitigation suggest different implications for climate policy mainstreaming and integration
Feasibility of TEE-guided stroke risk assessment in atrial fibrillation—background, aims, design and baseline data of the TIARA pilot study

97916.pdf (publisher's version ) (Open Access)BACKGROUND: Antithrombotic management in atrial fibrillation (AF) is currently based on clinical characteristics, despite evidence of potential fine-tuning with transoesophageal echocardiography (TEE). This open, randomised, multicentre study addresses the hypothesis that a comprehensive strategy of TEE-based aspirin treatment in AF patients is feasible and safe. METHODS: Between 2005 and 2009, ten large hospitals in the Netherlands enrolled AF patients with a moderate risk of stroke. Patients without thrombogenic TEE characteristics were randomised to aspirin or vitamin K antagonists (VKA). The primary objective is to show that TEE-based aspirin treatment is safe compared with VKA therapy. The secondary objective tests feasibility of TEE as a tool to detect echocardiographic features of high stroke risk. This report compares randomised to non-randomised patients and describes the feasibility of a TEE-based approach. RESULTS: In total, 310 patients were included. Sixty-nine patients were not randomised because of non-visualisation (n = 6) or TEE risk factors (n = 63). Compared with non-randomised patients, randomised patients (n = 241) were younger (65 +/- 11 vs. 69 +/- 9 years, p = 0.004), had less coronary artery disease (9 vs. 20%, p = 0.018), previous TIA (1.7 vs. 7.2%, p = 0.029), AF during TEE (25 vs. 54%, p < 0.001), mitral incompetence (55 vs. 70%, p = 0.038), VKA use (69 vs. 82%, p = 0.032), had a lower mean CHADS(2) score (1.2 +/- 0.6 vs. 1.6 +/- 1.0, p = 0.004), and left ventricular ejection fraction (59 +/- 8 vs. 56 +/- 8%, p = 0.016). CONCLUSIONS: This study shows that a TEE-based approach for fine-tuning stroke risk in AF patients with a moderate risk for stroke is feasible. Follow-up data will address the safety of this TEE-based approach
Proteins with Complex Architecture as Potential Targets for Drug Design: A Case Study of Mycobacterium tuberculosis
Lengthy co-evolution of Homo sapiens and Mycobacterium tuberculosis, the main causative agent of tuberculosis, resulted in a dramatically successful pathogen species that presents considerable challenge for modern medicine. The continuous and ever increasing appearance of multi-drug resistant mycobacteria necessitates the identification of novel drug targets and drugs with new mechanisms of action. However, further insights are needed to establish automated protocols for target selection based on the available complete genome sequences. In the present study, we perform complete proteome level comparisons between M. tuberculosis, mycobacteria, other prokaryotes and available eukaryotes based on protein domains, local sequence similarities and protein disorder. We show that the enrichment of certain domains in the genome can indicate an important function specific to M. tuberculosis. We identified two families, termed pkn and PE/PPE that stand out in this respect. The common property of these two protein families is a complex domain organization that combines species-specific regions, commonly occurring domains and disordered segments. Besides highlighting promising novel drug target candidates in M. tuberculosis, the presented analysis can also be viewed as a general protocol to identify proteins involved in species-specific functions in a given organism. We conclude that target selection protocols should be extended to include proteins with complex domain architectures instead of focusing on sequentially unique and essential proteins only
Integrating livelihoods and conservation in protected areas: Understanding the role and stakeholder views on prospects for non-timber forest products, a Bangladesh case study
Protected areas (PAs) represent a key global strategy in biodiversity conservation. In tropical developing countries, the management of PAs is a great challenge as many contain resources on which local communities rely. Collection and trading of non-timber forest products (NTFPs) is a well-established forest-based livelihood strategy, which has been promoted as a potential means for enhanced conservation and improved rural livelihoods in recent years, even though the sustainability or ecological implications have rarely been tested. We conducted an exploratory survey to understand the role and stakeholder views on conservation prospects and perceived ecological feasibility of NTFPs and harvesting schemes in a northeastern PA of Bangladesh, namely the Satchari National Park. Households (n = 101) were interviewed from three different forest dependency categories, adopting a stratified random sampling approach and using a semi-structured questionnaire. The study identified 13 locally important NTFPs, with five being critically important to supporting local livelihoods. Our study suggests that collection, processing and trading in NTFPs constitutes the primary occupation for about 18% of local inhabitants and account for an estimated 19% of their cash annual income. The household consensus on issues relating to NTFPs and their prospective role in conservation was surprisingly high, with 48% of respondents believing that promotion of NTFPs in the PA could have positive conservation value. The majority (71%) of households also had some understanding of the ecological implications of NTFP harvesting, sustainability (53%) and possible management and monitoring regimes (100%). With little known about their real application in the field, our study suggests further investigations are required to understand the ecological compatibility of traditional NTFP harvesting patterns and management. © 2010 Taylor & Francis
Geometagenomics illuminates the impact of agriculture on the distribution and prevalence of plant viruses at the ecosystem scale
Disease emergence events regularly result from human activities such as agriculture, which
frequently brings large populations of genetically uniform hosts into contact with potential
pathogens. Although viruses cause nearly 50% of emerging plant diseases, there is little systematic
information about virus distribution across agro-ecological interfaces and large gaps in understanding
of virus diversity in nature. Here we applied a novel landscape-scale geometagenomics
approach to examine relationships between agricultural land use and distributions of plantassociated
viruses in two Mediterranean-climate biodiversity hotspots (Western Cape region of
South Africa and Rhône river delta region of France). In total, we analysed 1725 geo-referenced plant
samples collected over two years from 4.5 × 4.5 km2 grids spanning farmlands and adjacent
uncultivated vegetation. We found substantial virus prevalence (25.8–35.7%) in all ecosystems, but
prevalence and identified family-level virus diversity were greatest in cultivated areas, with some
virus families displaying strong agricultural associations. Our survey revealed 94 previously
unknown virus species, primarily from uncultivated plants. This is the first effort to systematically
evaluate plant-associated viromes across broad agro-ecological interfaces. Our findings indicate that
agriculture substantially influences plant virus distributions and highlight the extent of current
ignorance about the diversity and roles of viruses in nature
Excision of HIV-1 Proviral DNA by Recombinant Cell Permeable Tre-Recombinase
Over the previous years, comprehensive studies on antiretroviral drugs resulted in the successful introduction of highly active antiretroviral therapy (HAART) into clinical practice for treatment of HIV/AIDS. However, there is still need for new therapeutic approaches, since HAART cannot eradicate HIV-1 from the infected organism and, unfortunately, can be associated with long-term toxicity and the development of drug resistance. In contrast, novel gene therapy strategies may have the potential to reverse the infection by eradicating HIV-1. For example, expression of long terminal repeat (LTR)-specific recombinase (Tre-recombinase) has been shown to result in chromosomal excision of proviral DNA and, in consequence, in the eradication of HIV-1 from infected cell cultures. However, the delivery of Tre-recombinase currently depends on the genetic manipulation of target cells, a process that is complicating such therapeutic approaches and, thus, might be undesirable in a clinical setting. In this report we demonstrate that E.coli expressed Tre-recombinases, tagged either with the protein transduction domain (PTD) from the HIV-1 Tat trans-activator or the translocation motif (TLM) of the Hepatitis B virus PreS2 protein, were able to translocate efficiently into cells and showed significant recombination activity on HIV-1 LTR sequences. Tre activity was observed using episomal and stable integrated reporter constructs in transfected HeLa cells. Furthermore, the TLM-tagged enzyme was able to excise the full-length proviral DNA from chromosomal integration sites of HIV-1-infected HeLa and CEM-SS cells. The presented data confirm Tre-recombinase activity on integrated HIV-1 and provide the basis for the non-genetic transient application of engineered recombinases, which may be a valuable component of future HIV eradication strategies
