    IRF7 mediates MCP-1 in adipocyte

    Hypertrophy, associated with adipocyte dysfunction, causes increased pro-inflammatory adipokine, and abnormal glucose and lipid metabolism, leading to insulin resistance and obesity-related-health problems. By combining DNA microarray and genomic data analyses to predict DNA binding motifs, we identified the transcription factor Interferon Regulatory Factor 7 (IRF7) as a possible regulator of genes related to adipocyte hypertrophy. To investigate the role of IRF7 in adipocytes, we examined gene expression patterns in 3T3-L1 cells infected with a retrovirus carrying the IRF7 gene and found that enforced IRF7 expression induced the expression of monocyte chemoattractant protein-1 (MCP-1), a key initial adipokine in the chronic inflammation of obesity. CRISPR/Cas9 mediated-suppression of IRF7 significantly reduced MCP-1 mRNA. Luciferase assays, chromatin immunoprecipitation PCR analysis and gel shift assay showed that IRF7 transactivates the MCP-1 gene by binding to its proximal Interferon Stimulation Response Element (ISRE), a putative IRF7 binding motif. IRF7 knockout mice exhibited lower expression of MCP-1 in epidydimal white adipose tissue under high-fat feeding conditions, suggesting the transcription factor is physiologically important for inducing MCP-1. Taken together, our results suggest that IRF7 transactivates MCP-1 mRNA in adipocytes, and it may be involved in the adipose tissue inflammation associated with obesity

    A Shock-Induced Pair of Superbubbles in the High-Redshift Powerful Radio Galaxy MRC 0406-244

    We present new optical spectroscopy of the high-redshift powerful radio galaxy MRC 0406-244 at redshift of 2.429. We find that the two extensions toward NW and SE probed in the rest-frame ultraviolet image are heated mainly by the nonthermal continuum of the active galactic nucleus. However, each extension shows a shell-like morphology, suggesting that they are a pair of superbubbles induced by the superwind activity rather than by the interaction between the radio jet and the ambient gas clouds. If this is the case, the intense starburst responsible for the formation of superbubbles could occur 1×109\sim 1 \times 10^9 yr ago. On the other hand, the age of the radio jets may be of the order of 106\sim 10^6 yr, being much shorter than the starburst age. Therefore, the two events, i.e., the starburst and the radio-jet activities, are independent phenomena. However, their directions of the expanding motions could be governed by the rotational motion of the gaseous component in the host galaxy.     An overview of the current status of the 8.2 m Subaru Telescope constructed and operated at Mauna Kea, Hawaii, by the National Astronomical Observatory of Japan is presented. The basic design concept and the verified performance of the telescope system are described. Also given are the status of the instrument package offered to the astronomical community, the status of operation, and some of the future plans. The status of the telescope reported in a number of SPIE papers as of the summer of 2002 are incorporated with some updates included as of 2004 February.     筆者らはこれまで、大学における全学科学生を対象とした帰属感高揚プログラムを開発してきた。本研究では、それまでのVTR を改善し、かつ受講生の所属する学科教員が登壇した2014 年度以降のプログラムの方が、2013 年度のプログラムに比べてより効果的であるかを再検証し、さらにその効果について「教員の対談」に注目して分析し検討することを目的とした。質問紙調査の結果、2014 年度以降のプログラムの方が効果的であり、特に教員の対談の効果が高まっていることが示された。さらに教員の対談の狙いを語りの内容から分析したところ、①フランクに自身の学生時代を示すことで、受講生の教員に対する親近感を高める、②自身の体験を踏まえて、大学・学科に関して語りかけ、受講生の大学生活の内省と帰属意識を促す、③個としての成長の取り組みを示し、呼びかけることで、成長の場として今の大学生活への自己関与意識を高め、帰属感につなげる、といった内容に分類され、受講者にとって親近性の高い教員がそれらの狙いに沿って語ることで、プログラムの効果が高まることが示唆された。At our university, we developed “University and I”-a program for students from all departments to foster a greater sense of identification with the university. We have improved the program we developed in 2013 by remaking video clips and boosting familiarity with faculty through our Talk of the Teachers approach. We incorporated those improvements in the 2014 and 2015 programs. This study analyzes the effectiveness of those programs compared with that in 2013. Questionnaire results showed that the programs and Talk of the Teachers were more effective in 2014 and 2015. Faculty members’ speaking objectives in Talk of the Teachers were classified as follows: (1) by frankly sharing their own university experiences, they could increase student familiarity with their teachers; (2) their talking about university and department matters based on their own experiences encouraged student self reflection on university life and a sense of belonging to the university; (3) by sharing their own actions as individuals, they could enhance students’ awareness of university life as a stage of personal development, thereby promoting a sense of belonging to the university. We found that when faculty with whom the students felt a sense of familiarity spoke with these intentions, the program was more effective in its aims

    Cross-National Differences in Victimization : Disentangling the Impact of Composition and Context

    Varying rates of criminal victimization across countries are assumed to be the outcome of countrylevel structural constraints that determine the supply ofmotivated o¡enders, as well as the differential composition within countries of suitable targets and capable guardianship. However, previous empirical tests of these ‘compositional’ and ‘contextual’ explanations of cross-national di¡erences have been performed upon macro-level crime data due to the unavailability of comparable individual-level data across countries. This limitation has had two important consequences for cross-national crime research. First, micro-/meso-level mechanisms underlying cross-national differences cannot be truly inferred from macro-level data. Secondly, the e¡ects of contextual measures (e.g. income inequality) on crime are uncontrolled for compositional heterogeneity. In this paper, these limitations are overcome by analysing individual-level victimization data across 18 countries from the International CrimeVictims Survey. Results from multi-level analyses on theft and violent victimization indicate that the national level of income inequality is positively related to risk, independent of compositional (i.e. micro- and meso-level) di¡erences. Furthermore, crossnational variation in victimization rates is not only shaped by di¡erences in national context, but also by varying composition. More speci¢cally, countries had higher crime rates the more they consisted of urban residents and regions with lowaverage social cohesion.

    Magnetic Fields toward Ophiuchus-B Derived from SCUBA-2 Polarization Measurements

    We present the results of dust emission polarization measurements of Ophiuchus-B (Oph-B) carried out using the Submillimetre Common-User Bolometer Array 2 (SCUBA-2) camera with its associated polarimeter (POL-2) on the James Clerk Maxwell Telescope in Hawaii. This work is part of the B-fields in Star-forming Region Observations survey initiated to understand the role of magnetic fields in star formation for nearby star-forming molecular clouds. We present a first look at the geometry and strength of magnetic fields in Oph-B. The field geometry is traced over ~0.2 pc, with clear detection of both of the sub-clumps of Oph-B. The field pattern appears significantly disordered in sub-clump Oph-B1. The field geometry in Oph-B2 is more ordered, with a tendency to be along the major axis of the clump, parallel to the filamentary structure within which it lies. The degree of polarization decreases systematically toward the dense core material in the two sub-clumps. The field lines in the lower density material along the periphery are smoothly joined to the large-scale magnetic fields probed by NIR polarization observations. We estimated a magnetic field strength of 630 ± 410 μG in the Oph-B2 sub-clump using a Davis–Chandrasekhar–Fermi analysis. With this magnetic field strength, we find a mass-to-flux ratio λ = 1.6 ± 1.1, which suggests that the Oph-B2 clump is slightly magnetically supercritical

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target

    The JCMT BISTRO Survey: Revealing the Diverse Magnetic Field Morphologies in Taurus Dense Cores with Sensitive Submillimeter Polarimetry

    Abstract: We have obtained sensitive dust continuum polarization observations at 850 μm in the B213 region of Taurus using POL-2 on SCUBA-2 at the James Clerk Maxwell Telescope as part of the B-fields in STar-forming Region Observations (BISTRO) survey. These observations allow us to probe magnetic field (B-field) at high spatial resolution (∼2000 au or ∼0.01 pc at 140 pc) in two protostellar cores (K04166 and K04169) and one prestellar core (Miz-8b) that lie within the B213 filament. Using the Davis–Chandrasekhar–Fermi method, we estimate the B-field strengths in K04166, K04169, and Miz-8b to be 38 ± 14, 44 ± 16, and 12 ± 5 μG, respectively. These cores show distinct mean B-field orientations. The B-field in K04166 is well ordered and aligned parallel to the orientations of the core minor axis, outflows, core rotation axis, and large-scale uniform B-field, in accordance with magnetically regulated star formation via ambipolar diffusion taking place in K04166. The B-field in K04169 is found to be ordered but oriented nearly perpendicular to the core minor axis and large-scale B-field and not well correlated with other axes. In contrast, Miz-8b exhibits a disordered B-field that shows no preferred alignment with the core minor axis or large-scale field. We found that only one core, K04166, retains a memory of the large-scale uniform B-field. The other two cores, K04169 and Miz-8b, are decoupled from the large-scale field. Such a complex B-field configuration could be caused by gas inflow onto the filament, even in the presence of a substantial magnetic flux

    JCMT BISTRO Survey: Magnetic Fields within the Hub-filament Structure in IC 5146

    We present the 850 μm polarization observations toward the IC 5146 filamentary cloud taken using the Submillimetre Common-User Bolometer Array 2 (SCUBA-2) and its associated polarimeter (POL-2), mounted on the James Clerk Maxwell Telescope, as part of the B-fields In STar forming Regions Observations. This work is aimed at revealing the magnetic field morphology within a core-scale (lesssim1.0 pc) hub-filament structure (HFS) located at the end of a parsec-scale filament. To investigate whether the observed polarization traces the magnetic field in the HFS, we analyze the dependence between the observed polarization fraction and total intensity using a Bayesian approach with the polarization fraction described by the Rice likelihood function, which can correctly describe the probability density function of the observed polarization fraction for low signal-to-noise ratio data. We find a power-law dependence between the polarization fraction and total intensity with an index of 0.56 in A V ~ 20–300 mag regions, suggesting that the dust grains in these dense regions can still be aligned with magnetic fields in the IC 5146 regions. Our polarization maps reveal a curved magnetic field, possibly dragged by the contraction along the parsec-scale filament. We further obtain a magnetic field strength of 0.5 ± 0.2 mG toward the central hub using the Davis–Chandrasekhar–Fermi method, corresponding to a mass-to-flux criticality of ~1.3 ± 0.4 and an Alfvénic Mach number of <0.6. These results suggest that gravity and magnetic field are currently of comparable importance in the HFS and that turbulence is less important
