    Development of a new method for D-xylose detection and quantification in urine, based on the use of recombinant xylose dehydrogenase from Caulobacter crescentus.

    The gene xylB from Caulobacter crescentus has been cloned and expressed in Escherichia coli providing a high yield of xylose dehydrogenase (XylB) production and excellent purity (97%). Purified recombinant XylB showed an absolute dependence on the cofactor NAD+ and a strong preference for d-xylose against other assayed mono and disaccharides. Additionally, XylB showed strong stability when stored as freeze-dried powder at least 250 days both at 4 °C and room temperature. In addition, more than 80% of the initial activity of rehydrated freeze-dried enzyme remained after 150 days of incubation at 4 °C. Based on these characteristics, the capability of XylB in d-xylose detection and quantification was studied. The linearity of the method was maintained up to concentrations of d-xylose of 10 mg/dL and the calculated limits of detection (LoD) and quantification (LoQ) of xylose in buffer were 0.568 mg/dL and 1.89 mg/dL respectively. Thus, enzymatic detection was found to be an excellent method for quantification of d-xylose in both buffer and urine samples.     Computational Study of the Phosphoryl Donor Activity of Dihydroxyacetone Kinase from ATP to Inorganic Polyphosphate

    Adenosine triphosphate (ATP) is the main biological phosphoryl donor required in many enzymes including dihydroxyacetone kinases (DHAKs) that convert dihydroxyacetone (Dha) into dihydroxyacetone phosphate (Dha-P), a key species with potential applications in synthesis. Herein, we present a theoretical study of the molecular mechanism for the phosphoryl transfer reaction from an inorganic polyphosphate to Dha catalyzed by DHAK from C. freundii. This is part of a project devoted to modify the phosphoryl donor specificity of this enzyme avoiding the use of the problematic direct addition of ATP. Based on the use of hybrid QM/MM potentials, with the QM region described by semiempirical and DFT methods, the reaction mechanism of the wild-type enzyme and the most active experimentally measured mutant (Glu526Lys) with poly-P as phosphoryl donor has been explored to elucidate the origin of the activity of this mutant. The similar energy barriers obtained in both systems confirm our previous studies on the binding step (Sánchez-Moreno et al., Int. J. Mol. Sci. 2015, 16, 27835) suggesting that this mutation favors a more adequate position of the poly-P in the active site for the following step, the chemical reaction, to take place

This fusion enzyme consists of monomeric fructose-1,6-bisphosphate aldolase (FBPA) from Staphylococcus carnosus and the homodimeric dihydroxyacetone kinase (DHAK) from Citrobacter freundii CECT 4626 with an intervening five amino acid linker. The fusion protein was expressed soluble and retained both kinase and aldolase activities. The secondary structure of the bifuctional enzyme has been analysed by CD spectroscopy, as well as that of the parental enzymes, in order to study the effect of the covalent coupling of the two parent proteins on the structure of the fused enzyme. Since S. carnosus FBPA is a thermostable protein, the effect of the fusion on the thermal stability of the bifunctional enzyme has also been studied. The proximity of the active centres in the fused enzyme promotes a kinetic advantage as the 20-fold increment in the initial velocity of the overall aldol reaction indicates.     Fibromialgia en Trabajo: explorando su Impacto en el Desempeño Laboral

    Esta investigación cualitativa exploratoria pretende examinar la experiencia de la fibromialgia en el ámbito laboral y cómo la misma incide en el desempeño de las personas. Utilizamos el Modelo de Demandas y Recursos Laborales para entender cómo los recursos (laborales y personales) y las demandas afectan la efectividad en el trabajo de las personas con fibromialgia. Siete pacientes con fibromialgia residentes en Puerto Rico participaron en entrevistas semiestructuradas. Los resultados apuntan que la fibromialgia tiene un efecto negativo en el desempeño de tareas, principalmente si estas requieren de esfuerzo físico o continuidad en una misma postura por largos periodos. Además, el ambiente laboral incide en la sintomatología y está a su vez en el desempeño de las funciones. Por último, se presentan una serie de recomendaciones enmarcadas en la flexibilidad, recursos personales y la educación, las cuales buscan minimizar los efectos que tiene la fibromialgia en el trabajo

    Cardiac primary angiosarcoma is an uncommon malignant primary tumor of mesenchymal origin with very poor prognosis because it tends to infiltrate the heart and invades the surrounding structures. The diagnosis is made by cardiac imaging; transthoracic echocardiogram and transesophageal echocardiogram provide information about extension, implantation site, size and hemodynamic parameters related to the presence of the mass. Evaluation of the mass can include cardiac magnetic resonance and computerized cardiac tomography.El angiosarcoma cardíaco es un tumor primario maligno infrecuente de origen mesenquimal con muy mal pronóstico, que tiende a infiltrar el corazón e invadir las estructuras circundantes. We investigated -fructose-6-phosphate aldolase from E. coli (FSA) D6X variants as catalysts for the aldol additions of ethanal and nonfunctionalized linear and cyclic aliphatic ketones as nucleophiles to nonphosphorylated hydroxyaldehydes. Thus, addition of propanone, cyclobutanone, cyclopentanone, or ethanal to 3-hydroxypropanal or (S)- or (R)-3-hydroxybutanal catalyzed by FSA D6H and D6Q variants furnished rare deoxysugars in 8-77% isolated yields with high stereoselectivity (97:3 dr and >95% ee)

    Changes in malaria epidemiology in a rural area of Cubal, Angola

    Scarce information about malaria epidemiology in Angola has been published. The objective of this study is to describe the epidemiology of malaria at the Hospital Nossa Senhora da Paz (Cubal, Angola) and the fatality rate due to malaria (total and in children under five years) in the last five years. A retrospective, observational study was performed at the Hospital Nossa Senhora da Paz, a 400-bed rural hospital located in Benguela Province of Angola. The study population included all patients who attended the hospital from January 2009 to December 2013. Outcome variables were calculated as follows: the percentage of malaria cases (number of positive thick blood films, divided by the total thick blood films performed); the percentage of in-patients for malaria (number of in-patients diagnosed with malaria, divided by the total number of in-patients); and, the fatality rate (number of deaths due to malaria divided by the number of positive thick blood films). Overall, 23,106 thick blood films were performed, of which 3,279 (14.2%) were positive for Plasmodium falciparum infection. During this five-year period, a reduction of 40% (95% CI 37-43%, p < 0.001) in the malaria-positive slides was detected. Distribution of positive-malaria slides showed a seasonal distribution with a peak from December to March (rainy season). An average annual reduction of 52% (95% CI 50-54%, p < 0.001) in the admissions due to malaria was observed. The overall fatality rate due to malaria was 8.3%, and no significant differences in the annual fatality rate were found (p = 0.553). A reduction in the number of malaria cases and the number of admissions due to malaria has been observed at the Hospital Nossa Senhora da Paz, during the last five years, and incidence along the study period showed a seasonal distribution. All this information could be useful when deciding which malaria control strategies have to be implemented in this area

Muchos docentes e instituciones están incorporado las redes sociales a sus estrategias de enseñanza y difusión de contenidos al tratarse de un espacio idóneo para intercambiar información y el conocimiento de una forma rápida, sencilla y cómoda. Este trabajo presenta el proyecto docente llevado a cabo en el Grado de Lingüística y Lenguas Aplicadas de la Universidad de Cádiz durante el curso 2017‐2018 con la finalidad de mejorar el conocimiento, expectativas y motivación del alumnado mediante el uso de nuevos medios de comunicación social: twitter, Facebook, Instagram, youtube y mailing lists
