    Neurotoxin in a Louisiana estuary: quantitative analysis of domoic acid in gulf menhaden (Brevoortia patronus) and qualitative modeling of links in a shark nursery

    Harmful algal blooms are an increasing problem for coastal waters world-wide. The diatom genus, Pseudo-nitzschia, is of particular concern in Louisiana, due to the potential for several species to produce the neurotoxin domoic acid (DA). While trophic transfer of DA to consumers has repeatedly occurred along the California coast, little is known about trophic transfer of recently detected DA in the Gulf of Mexico. In this study, the presence of DA in gulf menhaden (Brevoortia patronus) and the potential for trophic transfer to higher order consumers was investigated. In addition, the effects of this transfer and other algal toxins that threaten Louisiana’s coastal food webs were evaluated. DA quantification in water and fish tissue samples was determined by Enzyme-Linked Immunosorbent Assay (ELISA). Food web effects of algal toxins were analyzed through the use of a qualitative modeling technique, loop analysis. The results of the toxin assay illustrated that low-levels of DA exist in both water and tissue samples, with a significant correlation between the two (n = 25, p = 0.025, significance level of 0.05). The effects of HABs on the entire food web showed the possibility of trophic cascades. This is the first documentation of a DA vector in the entire Gulf of Mexico and confirms DA contamination in food webs of coastal Louisiana. Through the use of qualitative modeling, present and future threats posed by phycotoxins to coastal food webs can be assessed, providing resource managers valuable information to aid in mitigation of their negative consequences

    Influence Of The Mississippi River On Pseudo-nitzschia spp. Abundance And Toxicity In Louisiana Coastal Waters

    The presence of domoic acid (DA) toxin from multiple species of Pseudo-nitzschia is a concern in the highly productive food webs of the northern 1,* 2,3 2 1 1,4 1 1 2 3 4 Gulf of Mexico. We documented the Pseudo-­nitzschia presence, abundance, blooms, and toxicity over 3 years along a transect ∼100 km west of the Mississippi River Delta on the continental shelf. Pseudo-nitzschia were present throughout the year and occurred in high abundances (\u3e10 cells l ) in the early spring months during high Mississippi River (MSR) flow (∼20,000 m s ) but were most abundant (\u3e10 cells l ) when MSR discharge was relatively lower among the spring months. A high particulate toxin production (maximum reaching 13 μg DA l ) was associated with the high cell abundances and exceeded, by an order of magnitude, prior reports of particulate DA concentrations in Louisiana coastal waters. Differences in Pseudo-­nitzschia peak times and its toxicity were correlated mainly with the timing and magnitude of MSR discharge and changes in associated parameters such as nutrient stoichiometry and salinity. A negative relationship between high MSR discharge and Pseudo-­nitzschia and particulate DA concentrations was documented. These riverine dynamics have the potential to influence DA contamination in pelagic and benthic food webs in the coastal waters of the northern Gulf of Mexico

Yet despite 50 years of sustained interest, its microscopic structure and its relation to durability remain open questions. We address the issue by archaeomimetism: engineering an archaeoinspired pigment, satisfactorily reproducing the colour and chemical stability of Maya Blue. By comparing and contrasting ancient pigment and the new analogue, we deduce a new explanation for this durability

    Whole grain consumption and human health: an umbrella review of observational studies

    Whole grains have been associated with a number of health benefits. We systematically reviewed existing meta-analyses of observational studies and evaluated the level of evidence for their putative effects based on pre-selected criteria. Of the 23 included studies, we found convincing evidence of an inverse association between whole grain consumption and risk of type-2 diabetes and colorectal cancer; possible evidence of decreased risk of colon cancer and cardiovascular mortality with increased whole grain intake, as well as increased risk of prostate cancer. Limited or insufficient evidence was available for all other outcomes investigated. Overall findings are encouraging for a positive effect of whole grain consumption on certain diseases, especially highly prevalent metabolic diseases, however, uncertainty of some negative associations deserves further attention

    Changes in Exposure to Neighborhood Characteristics are Associated with Sexual Network Characteristics in a Cohort of Adults Relocating from Public Housing

    Ecologic and cross-sectional multilevel analyses suggest that characteristics of the places where people live influence their vulnerability to HIV and other sexually-transmitted infections (STIs). Using data from a predominately substance-misusing cohort of African-American adults relocating from US public housing complexes, this multilevel longitudinal study tested the hypothesis that participants who experienced greater post-relocation improvements in economic disadvantage, violent crime, and male:female sex ratios would experience greater reductions in perceived partner risk and in the odds of having a partner who had another partner (i.e., indirect concurrency). Baseline data were collected from 172 public housing residents before relocations occurred; three waves of post-relocation data were collected every nine months. Participants who experienced greater improvements in community violence and in economic conditions experienced greater reductions in partner risk. Reduced community violence was associated with reduced indirect concurrency. Structural interventions that decrease exposure to violence and economic disadvantage may reduce vulnerability to HIV/STIs

    Meta-analysis of genome-wide association studies of asthma in ethnically diverse North American populations.

    Asthma is a common disease with a complex risk architecture including both genetic and environmental factors. We performed a meta-analysis of North American genome-wide association studies of asthma in 5,416 individuals with asthma (cases) including individuals of European American, African American or African Caribbean, and Latino ancestry, with replication in an additional 12,649 individuals from the same ethnic groups. We identified five susceptibility loci. Four were at previously reported loci on 17q21, near IL1RL1, TSLP and IL33, but we report for the first time, to our knowledge, that these loci are associated with asthma risk in three ethnic groups. In addition, we identified a new asthma susceptibility locus at PYHIN1, with the association being specific to individuals of African descent (P = 3.9 × 10(-9)). These results suggest that some asthma susceptibility loci are robust to differences in ancestry when sufficiently large samples sizes are investigated, and that ancestry-specific associations also contribute to the complex genetic architecture of asthma

    Colorectal cancer (CRC) develops and progresses through a systematic selection for (epi) genetic alterations that drive the transformation from normal colon epithelium to adenocarcinoma. These changes affect both noncoding RNAs and mRNAs and so define the clinical behaviour of cancer cells within a distinctive host genetic and environmental context. Although earlier diagnosis and more effective treatment modalities have decreased mortality from CRC, prognostic stratification and adjuvant therapy selection after surgery remain dependent on broad descriptive classifications, opportune histological markers of poor prognosis and chemotherapy efficacy data derived from diverse CRC populations. Crucially, there is significant inter- and intra-individual variability in response to, and tolerance of, chemotherapy treatments. These limitations explain the small clinical benefit of new agents studied in contemporary phase III trials. Molecular assays have the potential to address these constraints and there has been intense interest in the identification of clinically relevant molecular biomarkers. These must be easy to obtain and quantify and ideally represent steps in well-understood carcinogenic pathways or host-response mechanisms. Although some biomarkers can provide broad prognostic information based on CRC subtype (e.g. MSI status) or can somewhat predict response to targeted therapies (e.g. KRAS), no RNA-based biomarkers have entered routine clinical practice. This is due, in part, to the genetic heterogeneity of both patients and CRC. In addition, serious underlying issues with regards to study design, poor technical protocols, inadequate quality controls and inappropriate data analysis prevent successful translation of research results. Consequently, the identification of clinically relevant panels of biomarkers will depend not just on further advances in our understanding of CRC biology, but will need to be coupled with appropriate study designs and more suitable, standardised and transparent techniques
