Heteronuclear bimetallic complexes with 3d and 4f elements
Three heteronuclear bimetallic complexes [Cu(MeOH)(L)Ln(NO3)3] ( 1-Ce ; Ln = Ce, 1-Pr ; Ln = Pr, and 1-Nd ; Ln = Nd) were prepared using H2L (1,3-bis[(3-methoxysalicylidene)amino]-2,2-dimethylpropane) in methanol, affording the complexes as green crystalline materials. These can be prepared in a one-pot synthesis from 2,2-dimethylpropan-1,3-diamine, o-vanillin, copper(II) nitrate, and Ln(III) nitrate (Ln = Ce, Pr, Nd). X-ray crystallography, high-resolution mass spectrometry, and UV-vis spectroscopy were used to characterize the bimetallic complexes. All three complexes showed the copper center adopting a five-coordinate square pyramidal geometry and the lanthanoid cation adopting a ten-coordinate geometry.Publisher PDFPeer reviewe
Anomalies in T cell function are associated with individuals at risk of mycobacterium abscessus complex infection
The increasing global incidence and prevalence of non-tuberculous mycobacteria (NTM) infection is of growing concern. New evidence of person-to-person transmission of multidrug-resistant NTM adds to the global concern. The reason why certain individuals are at risk of NTM infections is unknown. Using high definition flow cytometry, we studied the immune profiles of two groups that are at risk of Mycobacterium abscessus complex infection and matched controls. The first group was cystic fibrosis (CF) patients and the second group was elderly individuals. CF individuals with active M. abscessus complex infection or a history of M. abscessus complex infection exhibited a unique surface T cell phenotype with a marked global deficiency in TNFa production during mitogen stimulation. Importantly, immune-based signatures were identified that appeared to predict at baseline the subset of CF individuals who were at risk of M. abscessus complex infection. In contrast, elderly individuals with M. abscessus complex infection exhibited a separate T cell phenotype underlined by the presence of exhaustion markers and dysregulation in type 1 cytokine release during mitogen stimulation. Collectively, these data suggest an association between T cell signatures and individuals at risk of M. abscessus complex infection, however, validation of these immune anomalies as robust biomarkers will require analysis on larger patient cohorts
Key Science Goals for the Next Generation Very Large Array (ngVLA): Report from the ngVLA Science Advisory Council
This document describes some of the fundamental astrophysical problems that
require observing capabilities at millimeter- and centimeter wavelengths well
beyond those of existing, or already planned, telescopes. The results
summarized in this report follow a solicitation from the National Radio
Astronomy Observatory to develop key science cases for a future U. S.-led radio
telescope, the "next generation Very Large Array" (ngVLA). The ngVLA will have
roughly 10 times the collecting area of the Jansky VLA, operate at frequencies
from 1 GHz to 116 GHz with up to 20 GHz of bandwidth, possess a compact core
for high surface-brightness sensitivity, and extended baselines of at least
hundreds of kilometers and ultimately across the continent to provide
high-resolution imaging. The ngVLA builds on the scientific and technical
legacy of the Jansky VLA and ALMA, and will be designed to provide the next
leap forward in our understanding of planets, galaxies, and black holes.Comment: ngVLA memo 1
Using airborne LiDAR Survey to explore historic-era archaeological landscapes of Montserrat in the eastern Caribbean
This article describes what appears to be the first archaeological application of airborne LiDAR survey to historic-era landscapes in the Caribbean archipelago, on the island of Montserrat. LiDAR is proving invaluable in extending the reach of traditional pedestrian survey into less favorable areas, such as those covered by dense neotropical forest and by ashfall from the past two decades of active eruptions by the Soufrière Hills volcano, and to sites in localities that are inaccessible on account of volcanic dangers. Emphasis is placed on two aspects of the research: first, the importance of ongoing, real-time interaction between the LiDAR analyst and the archaeological team in the field; and second, the advantages of exploiting the full potential of the three-dimensional LiDAR point cloud data for purposes of the visualization of archaeological sites and features
Genomic insights from the first chromosome-scale assemblies of oat (Avena spp.) diploid species
A phase II single-arm study of pembrolizumab with enzalutamide in men with metastatic castration-resistant prostate cancer progressing on enzalutamide alone.
BACKGROUND: Checkpoint inhibitors can induce profound anticancer responses, but programmed cell death protein-1 (PD-1) inhibition monotherapy has shown minimal activity in prostate cancer. A published report showed that men with prostate cancer who were resistant to the second-generation androgen receptor inhibitor enzalutamide had increased programmed death-ligand 1 (PD-L1) expression on circulating antigen-presenting cells. We hypothesized that the addition of PD-1 inhibition in these patients could induce a meaningful cancer response.
METHODS: We evaluated enzalutamide plus the PD-1 inhibitor pembrolizumab in a single-arm phase II study of 28 men with metastatic castration-resistant prostate cancer (mprogressing on enzalutamide alone. Pembrolizumab 200 mg intravenous was given every 3 weeks for four doses with enzalutamide. The primary endpoint was prostate-specific antigen (PSA) decline of ≥50%. Secondary endpoints were objective response, PSA progression-free survival (PFS), time to subsequent treatment, and time to death. Baseline tumor biopsies were obtained when feasible, and samples were sequenced and evaluated for the expression of PD-L1, microsatellite instability (MSI), mutational and neoepitope burdens.
RESULTS: Five (18%) of 28 patients had a PSA decline of ≥50%. Three (25%) of 12 patients with measurable disease at baseline achieved an objective response. Of the five responders, two continue with PSA and radiographic response after 39.3 and 37.8 months. For the entire cohort, median follow-up was 37 months, and median PSA PFS time was 3.8 months (95% CI: 2.8 to 9.9 months). Time to subsequent treatment was 7.21 months (95% CI: 5.1 to 11.1 months). Median overall survival for all patients was 21.9 months (95% CI: 14.7 to 28 .4 months), versus 41.7 months (95% CI: 22.16 to not reached (NR)) in the responders. Of the three responders with baseline biopsies, one had MSI high disease with mutations consistent with DNA-repair defects. None had detectable PD-L1 expression.
CONCLUSIONS: Pembrolizumab has activity in mCRPC when added to enzalutamide. Responses were deep and durable and did not require tumor PD-L1 expression or DNA-repair defects.

Impaired Brain Dopamine and Serotonin Release and Uptake in Wistar Rats Following Treatment with Carbotplatin
Chemotherapy-induced cognitive impairment, known also as “chemobrain”, is a medical complication of cancer treatment that is characterized by a general decline in cognition affecting visual and verbal memory, attention, complex problem solving skills, and motor function. It is estimated that one-third of patients who undergo chemotherapy treatment will experience cognitive impairment. Alterations in the release and uptake of dopamine and serotonin, central nervous system neurotransmitters that play important roles in cognition, could potentially contribute to impaired intellectual performance in those impacted by chemobrain. To investigate how chemotherapy treatment affects these systems, fast-scan cyclic voltammetry (FSCV) at carbon-fiber microelectrodes was used to measure dopamine and serotonin release and uptake in coronal brain slices containing the striatum and dorsal raphe nucleus, respectively. Measurements were taken from rats treated weekly with selected doses of carboplatin and from control rats treated with saline. Modeling the stimulated dopamine release plots revealed an impairment of dopamine release per stimulus pulse (80% of saline control at 5 mg/kg and 58% at 20 mg/kg) after 4 weeks of carboplatin treatment. Moreover, Vmax, the maximum uptake rate of dopamine, was also decreased (55% of saline control at 5 mg/kg and 57% at 20 mg/kg). Nevertheless, overall dopamine content, measured in striatal brain lysates by high performance liquid chromatography, and reserve pool dopamine, measured by FSCV after pharmacological manipulation, did not significantly change, suggesting that chemotherapy treatment selectively impairs the dopamine release and uptake processes. Similarly, serotonin release upon electrical stimulation was impaired (45% of saline control at 20 mg/kg). Measurements of spatial learning discrimination were taken throughout the treatment period and carboplatin was found to alter cognition. These studies support the need for additional neurochemical and behavioral analyses to identify the underlying mechanisms of chemotherapy-induced cognitive disorders

The clustering of galaxies in the SDSS-III Baryon Oscillation Spectroscopic Survey: Baryon Acoustic Oscillations in the Data Release 9 Spectroscopic Galaxy Sample
We present measurements of galaxy clustering from the Baryon Oscillation
Spectroscopic Survey (BOSS), which is part of the Sloan Digital Sky Survey III
(SDSS-III). These use the Data Release 9 (DR9) CMASS sample, which contains
264,283 massive galaxies covering 3275 square degrees with an effective
redshift z=0.57 and redshift range 0.43 < z < 0.7. Assuming a concordance
Lambda-CDM cosmological model, this sample covers an effective volume of 2.2
Gpc^3, and represents the largest sample of the Universe ever surveyed at this
density, n = 3 x 10^-4 h^-3 Mpc^3. We measure the angle-averaged galaxy
correlation function and power spectrum, including density-field reconstruction
of the baryon acoustic oscillation (BAO) feature. The acoustic features are
detected at a significance of 5\sigma in both the correlation function and
power spectrum. Combining with the SDSS-II Luminous Red Galaxy Sample, the
detection significance increases to 6.7\sigma. Fitting for the position of the
acoustic features measures the distance to z=0.57 relative to the sound horizon
DV /rs = 13.67 +/- 0.22 at z=0.57. Assuming a fiducial sound horizon of 153.19
Mpc, which matches cosmic microwave background constraints, this corresponds to
a distance DV(z=0.57) = 2094 +/- 34 Mpc. At 1.7 per cent, this is the most
precise distance constraint ever obtained from a galaxy survey. We place this
result alongside previous BAO measurements in a cosmological distance ladder
and find excellent agreement with the current supernova measurements. We use
these distance measurements to constrain various cosmological models, finding
continuing support for a flat Universe with a cosmological constant.Comment: 33 page
