    Taking the pulse of Earth's tropical forests using networks of highly distributed plots

    Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science. In the future, humanity can benefit greatly by nurturing the grassroots communities now collectively capable of generating unique, long-term understanding of Earth's most precious forests. Resumen Los bosques tropicales son los ecosistemas más diversos y productivos del mundo y entender su funcionamiento es crítico para nuestro futuro colectivo. Sin embargo, hasta hace muy poco, los esfuerzos para medirlos y monitorearlos han estado muy desconectados. El trabajo en redes es esencial para descubrir las respuestas a preguntas que trascienden las fronteras y los plazos de las agencias de financiamiento. Aquí mostramos cómo una comunidad global está respondiendo a los desafíos de la investigación en ecosistemas tropicales a través de diversos equipos realizando mediciones árbol por árbol en miles de parcelas permanentes de largo plazo. Revisamos los descubrimientos más importantes de este trabajo y discutimos cómo este proceso está cambiando la ciencia relacionada a los bosques tropicales. Apalancando lo que realmente es una tecnología antigua, las redes de parcelas están generando una verdadera y moderna revolución en la ciencia tropical. En el futuro, la humanidad puede beneficiarse enormemente si se nutren y cultivan comunidades de investigadores de base, actualmente con la capacidad de generar información única y de largo plazo para entender los que probablemente son los bosques más preciados de la tierra. Resumo Florestas tropicais são os ecossistemas mais diversos e produtivos da Terra. Embora uma boa compreensão destas florestas seja crucial para o nosso futuro coletivo, até muito recentemente os esforços de medições e monitoramento foram amplamente desconexos. É essencial formarmos redes para obtermos respostas que transcendem fronteiras e horizontes de agências financiadoras. Neste estudo nós mostramos como uma comunidade global está respondendo aos desafios da pesquisa de ecossistemas tropicais, com equipes diversas medindo florestas, árvore por árvore, em milhares de parcelas monitoradas à longo prazo. Nós revisamos as maiores descobertas científicas deste trabalho, e mostramos também como este processo está mudando a ciência de florestas tropicais. Nossa abordagem principal envolve unir iniciativas de base a protocolos padronizados e gerenciamento de dados a fim de gerar resultados robustos em escalas ampliadas. Si une meilleure compréhension de ces forêts est essentielle pour notre avenir collectif, jusqu'à tout récemment, les efforts déployés pour les mesurer et les surveiller ont été largement déconnectés. La mise en réseau est essentielle pour découvrir les réponses à des questions qui dépassent les frontières et les horizons des organismes de financement. Nous montrons ici comment une communauté mondiale relève les défis de la recherche sur les écosystèmes tropicaux avec diverses équipes qui mesurent les forêts arbre après arbre dans de milliers de parcelles permanentes. Nous passons en revue les principales découvertes scientifiques de ces travaux et montrons comment ce processus modifie la science des forêts tropicales. Notre approche principale consiste à relier les initiatives de base à long terme à des protocoles standardisés et une gestion de données afin de générer des résultats solides à grande échelle. Walaupun pemahaman mengenai hutan tropika amat penting untuk masa depan kita, usaha-usaha untuk mengkaji dan mengawas hutah-hutan tersebut baru sekarang menjadi lebih diperhubungkan. Perangkaian adalah sangat penting untuk mencari jawapan kepada soalan-soalan yang menjangkaui sempadan dan batasan agensi pendanaan. Di sini kami menunjukkan bagaimana sebuah komuniti global bertindak balas terhadap cabaran penyelidikan ekosistem tropika melalui penglibatan pelbagai kumpulan yang mengukur hutan secara pokok demi pokok dalam beribu-ribu plot jangka panjang.     Mortality from gastrointestinal congenital anomalies at 264 hospitals in 74 low-income, middle-income, and high-income countries: a multicentre, international, prospective cohort study

    Summary Background Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. Methods We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung’s disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. Findings We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung’s disease) from 264 hospitals (89 in high-income countries, 166 in middleincome countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Mortality among all patients was 37 (39·8%) of 93 in low-income countries, 583 (20·4%) of 2860 in middle-income countries, and 50 (5·6%) of 896 in high-income countries (p<0·0001 between all country income groups). Gastroschisis had the greatest difference in mortality between country income strata (nine [90·0%] of ten in lowincome countries, 97 [31·9%] of 304 in middle-income countries, and two [1·4%] of 139 in high-income countries; p≤0·0001 between all country income groups). Factors significantly associated with higher mortality for all patients combined included country income status (low-income vs high-income countries, risk ratio 2·78 [95% CI 1·88–4·11], p<0·0001; middle-income vs high-income countries, 2·11 [1·59–2·79], p<0·0001), sepsis at presentation (1·20 [1·04–1·40], p=0·016), higher American Society of Anesthesiologists (ASA) score at primary intervention (ASA 4–5 vs ASA 1–2, 1·82 [1·40–2·35], p<0·0001; ASA 3 vs ASA 1–2, 1·58, [1·30–1·92], p<0·0001]), surgical safety checklist not used (1·39 [1·02–1·90], p=0·035), and ventilation or parenteral nutrition unavailable when needed (ventilation 1·96, [1·41–2·71], p=0·0001; parenteral nutrition 1·35, [1·05–1·74], p=0·018). Administration of parenteral nutrition (0·61, [0·47–0·79], p=0·0002) and use of a peripherally inserted central catheter (0·65 [0·50–0·86], p=0·0024) or percutaneous central line (0·69 [0·48–1·00], p=0·049) were associated with lower mortality. Interpretation Unacceptable differences in mortality exist for gastrointestinal congenital anomalies between lowincome, middle-income, and high-income countries. Improving access to quality neonatal surgical care in LMICs will be vital to achieve Sustainable Development Goal 3.2 of ending preventable deaths in neonates and children younger than 5 years by 2030

    The use of techniques of cryopreservation of gametes in domestic cats are considered important in enhancing procedures in assisted reproduction. Vitrification is based on the use of high concentrations of cryoprotectants in order to inhibit the formation of ice crystals. It is possible that cryopreservation induces the production of reactive oxygen species (ROS) or change the enzymatic antioxidant potential of the oocyte, so one might speculate that the addition of antioxidants to the means of vitrification could reduce the oxidative stress caused by cryoprotectants. In the first experiment are compared four protocols vitrification using as base medium (MB) the TCM199 (tissue culture medium 199) more antibiotic glucose plus cryoprotectants: G1 (EG + 3M ethylene glycol + 2M dimethyl-DMSO), G2 (3M EG + 3M 1,2 propanediol - PrOH) , G3 (1.5 M EG + 1M DMSO) and G4 (1.5 M EG + 1.5 M PrOH) . After thawing, the material was assessed for viability and morphological changes (staining cFDA / trypan blue). In this , there was no statistical difference (p = 0.2857) among the four groups thawed oocytes studied with respect to morphology, but in relation to oocyte viability G1 and G2 groups had a higher percentage of viable oocytes , but did not differ ( 35.71% and 36.84 %, respectively , p = 0.018). In the second experiment three concentrations of vitamin E (0.4 , 0.6 and 0.8 mM) were added to the protocol MB with 3M EG + 3M PrOH , L forming the groups G 0.4 mM, G 0.6 mM, G 0.8 mM and G 0 (no added vitamin E). The choice of the base protocol was in accordance with the results obtained in experiment I. We observed improvement in percentage (p = 0.0032) of oocytes with normal morphology in groups G 0.6 mM (88.6 %) and G 0.8 mM (62.5 %). Experiment II oocytes that maintained their morphology after thawing were subjected to in vitro maturation for a period of 36 hours. When evaluating the resumption of meiosis, we observed no statistical difference ...O emprego de técnicas de criopreservação de gametas em felinos domésticos são consideradas importantes no aprimoramento de procedimentos na reprodução assistida. A vitrificação se baseia no uso de altas concentrações de crioprotetores com o intuito de inibir a formação de cristais de gelo. É possível que a criopreservação induza a produção de espécies reativas de oxigênio (EROs) ou altere o potencial antioxidante enzimático do oócito, sendo assim, aventa-se que a adição de antioxidantes aos meios de vitrificação poderia reduzir o estresse oxidativo causado pelos crioprotetores. No experimento I comparam-se quatro protocolos de vitrificação utilizando-se como meio base (MB) o TCM199 (Meio de cultura de tecidos 199) com glicose mais antibiótico, acrescido de crioprotetores: G1 (3M etilenoglicol–EG + 2M de dimetilsulfóxido-DMSO), G2 (3M EG + 3M 1,2 propanediol-PrOH), G3 (1,5M EG + 1M DMSO) e G4 (1,5M EG + 1,5M PrOH). Após descongelamento, este material foi avaliado quanto à alteração morfológica e viabilidade (coloração cFDA/trypan blue). Neste, não houve diferença estatística (p=0,2857) entre os oócitos descongelados dos quatro grupos estudados em relação à morfologia, mas em relação a viabilidade oocitária os grupos G1 e G2 obtiveram maior porcentagem de oócitos viáveis, porém não diferiram entre si (35,71% e 36,84%, respectivamente; p=0,018). No experimento II três concentrações de vitamina E (0,4, 0,6 e 0,8mM) foram acrescidas ao protocolo de MB com 3M EG + 3M PrOH, formando os grupos G 0,4mM, G 0,6mM, G 0,8mM e o G 0 (sem adição de vitamina E).     Canine Pyometra is a uterine disease that occurs in sexually mature bitches, with higher incidence in nulliparous and animals over 4 years and is characterized by presenting an accumulation of pus in the uterine lumen, usually occurring in diestrus. Laboratory tests are important tools for the detection of metabolic abnormalities associated with sepsis and renal function, which are serious consequences of pyometra. In blood the main findings are normochromic non-regenerative anemia, presence of dehydration, and sometimes thrombocytopenia. The WBC count may be normal but most often occurs a neutrophilic leukocytosis with a left shift, monocytosis and the presence of toxic neutrophils. In less than 1 / 3 of the animals the presence of azotemia is present and a density lower than 1035 is detected in the urine of almost 90% of bitches which may be in normal range at the onset of the desease. Urinary protein loss is rare but the protein may be elevated in the reagent strip due to urinary contamination by uterine secretion. The increase of gamma-glutamyltransferase (GGT), alkaline phosphatase (ALP), aspartate aminotransferase (AST) and creatine kinase (CK) may be present, indicating disorders in the liver. Currently, additional laboratory tests are being studied for the diagnosis of pyometra and its prognosis, such as the measurement of C-reactive protein and fibrinogen for monitoring the recovery of the inflammatory process and the urine electrophoresis to characterize the origin of proteinuria in these animals . The aim of this work is to review the literature on the main laboratory tests that aid the diagnosis of PyometraPiometra canina é uma doença uterina que ocorre em cadelas sexualmente maduras, com maior incidência em nulíparas e em animais maiores de 4 anos; se caracteriza por apresentar um acumulo de material purulento no lúmen uterino, ocorrendo geralmente no diestro. Os exames laboratoriais são importantes ferramentas para detecção de anormalidades metabólicas, associadas à sepse e avaliação da função renal, as quais são graves consequências da piometra. No hemograma os principais achados são de anemia normocítica normocrômica arregenerativa, presença de desidratação, e as vezes trombocitopenia. A leucometria pode estar normal mas na maioria das vezes ocorre leucocitose por neutrofilia com desvio à esquerda, além de monocitose e presença de neutrófilos tóxicos. Em menos de 1/3 dos animais há observação de azotemia e em praticamente 90% das cadelas é detectada uma densidade inferior a 1.035 na urina, podendo estar no limite normal no início da doença. Perda da proteína urinária é rara porém a proteína pode estar elevada no teste da fita urinária devido a contaminação pela secreção uterina. O aumento de gama-glutamiltransferase (GGT), fosfatase alcalina (FA), aspartato aminotransferase (AST) e da creatina kinase (CK) podem estar presentes, indicando desordens no fígado. Atualmente novos exames laboratoriais estão sendo pesquisados para o diagnóstico da piometra e do seu prognóstico, como a dosagem de proteína C reativa (PCR) e fibrinogênio para monitoramento da recuperação do processo inflamatório e a realização de eletroforese da urina para caracterizar a origem da proteinúria nesses animais.     The possibilities of using the sperm collected from the epididymis have been widely used because the fertilizing capacity sperm preservation and the possibility of using it for wild cats. But in the process of cryopreservation, some studies show a decrease in the quality of the sperm when left under cooling before frozen for some time. This study aimed to assess the quality of the epididymal sperm obtained from domestic cats after cryopreservation using a diluent based on egg yolk and glycerol (Botu-crio®), comparing the morphofunctional characteristics after cooling for 24 hours in a container of semen transport (Botu-tainer®). We use eight cats submitted to elective orchiectomy, aging from eight months, without racial determination, and good nutritional status. These sperm characteristics were: motility, vigor, concentration, membrane integrity and morphology. It has been found, after statistical analysis, that the container of semen was able to maintain sperm viability, even for 24h. We also observed a significant decrease on all parameters after frozen, consequential, probably to thermal stress that occurs in processing. However, the percentage of membrane integrity after thawing shows good employability of the Botu-crio®, which viability is possible to perform in vitro fertilization, requiring higher ratings

    Pub. 42 Resection of Vaginal Neoplasms by Video-vaginoscopy in Bitches

    ABSTRACT Background: Vaginal neoplasms usually represent a challenge for veterinary surgeons. Surgical resection often requires episiotomy or even laparotomy and pubic osteotomy, increasing the risk for intra and postoperative complications, such as severe pain, bleeding, wound infection or dehiscence and vaginal stenosis. Endoscopic treatment of neoplastic lesions is routinely used in human patients. However, the information about its use in small animals is sparse. Thus, the aim of the current study was to report two cases of successful vaginoscopic treatment of vaginal neoplasms in bitches. Case: A female Labrador weighing 26 kg (patient 1) and a mongrel bitch weighting 10 kg (patient 2) were attended due to vaginal bleeding. Physical examination revealed a pendunculated hard nodular mass in the caudal third of the vaginal dorsal fl oor in patient 1. In patient 2, two nodular, fi brous, infi ltrated masses of different dimension were touched on the mucosa of the caudal third of the vagina. The vaginal cytology revealed erythrocytes, cellular debris and anestrus in both cases. Given the clinical suspicion of vaginal neoplasia, the endoscopic approach by vaginoscopy was chosen in order for diagnostic investigation and surgical treatment. Under general anesthesia, a rigid 10-mm telescope with 6-mm working channel was employed for initial examination. A 10-mmHg CO2 pneumovagina was estabilished using an automatic insuffl ator. A pedunculated spherical neoplastic mass of approximately 2 cm of diameter was excised from patient 1 using a simultaneous bipolar coagulation and cut forceps, inserted through the working channel of the telescope. In patient 2, the two infi ltrated masses were resected using cup type biopsy forceps, followed by cauterization of the neoplastic underlining area with bipolar forceps, also through the working channel. The sampled specimen were sent for histopathological examination, which revealed leiomyosarcoma and fi broepithelial hyperplasia in patients 1 and 2, respectively. Moreover, no other therapy was prescribed and the patients convalesced uneventfully. Discussion: The surgical procedures lasted 23 and 38 min in patients 1 and 2, respectively. Patients recovered uneventfully in the early postoperative period, with no signs of pain. The operative-telescope proved to be versatile and effective for vaginoscopic resection of neoplasms, which was effi cient for both diagnostic and therapeutic purposes. The patients presented excellent recovery with minimally invasive surgical trauma, especially compared with conventional techniques. The majority of tumors found in the vagina of dogs are benign, mostly from smooth muscle or fi brous tissue origin (leiomyoma, fi broma and leiomyosarcoma). Surgical excision of the tumor combined with ovariohysterectomy is usually effective to prevent recurrence. After 3 months, patient 1 was submitted to clinical and laboratory tests, including a second look vaginoscopy. Vaginal endoscopic examination revealed only small area of approximately 3 mm scar on the resection site, showing no evidence of recurrence and/or metastasis. Moreover, vaginoscopy was able to avoid the need for episiotomy or laparotomy for resection of the vaginal masses. In conclusion, the resection of vaginal neoplasia by video-vaginoscopy approach was feasible, effective and minimally invasive, which can be an alternative to conventional surgical approaches

Os efeitos colaterais após a administração da PGF2a, como taquicardia, salivação, emese, diarréia e convulsões geralmente são relacionadas com a dose utilizada.
