    Development of boron calibration via hybrid comparator method in prompt gamma activation analysis

    The prompt gamma activation analysis (PGAA) facility at the Nuclear Engineering Teaching Laboratory at The University of Texas at Austin was utilized to quantify boron concentrations in boron carbide semiconductor films deposited on silicon substrates. Calibration was complicated by the unique and varying sample geometries analyzed. In addition, there was a dearth of solid materials available with quantified boron concentrations having comparable or readily modifiable dimensions to exploit for calibration purposes. Therefore, a novel hybrid comparator method was developed for the quantification of boron utilizing aluminum as an inexpensive and easily machinable reference material. Aluminum samples were manufactured with high tolerances to match the geometry of each sample of interest. Each boron carbide film sample and its congruent aluminum sample were measured in the PGAA system. The measured aluminum responses and relevant nuclear parameters were used to standardize the measurements. A boron standard was created using a procedure derived from a similar approach used by the National Institute of Standards and Technology. Quality control measurements using this standard show that the method provided accuracy to within 5% for boron quantification

    Ecological changes in historically polluted soils: Metal(loid) bioaccumulation in microarthropods and their impact on community structure

    International audienceSoil pollution by persistent metal(loid)s present environmental and sanitary risks. While the effects of metal(loid)s on vegetation and macrofauna have been widely studied, their impact on microarthropods (millimetre scale) and their bioaccumulation capacity have been less investigated. However, microarthropods provide important ecosystem services, contributing in particular to soil organic matter dynamics. This study focussed on the impact of metal(loid) pollution on the structure and distribution of microarthropod communities and their potential to bioaccumulate lead (Pb). Soil samples were collected from a contaminated historical site with a strong horizontal and vertical gradient of Pb concentrations. Microarthropods were extracted using the Berlese method. The field experiments showed that microarthropods were present even in extremely polluted soils (30,000 mg Pb kg− 1). However, while microarthropod abundance increased with increasing soil C/N content (R2 = 0.79), richness decreased with increasing pollution. A shift in the community structure from an oribatid-to a springtail-dominated community was observed in less polluted soils (R2 = 0.68). In addition, Pb bioamplification occurred in microarthropods, with higher Pb concentrations in predators than in detritivorous microarthropods. Finally, the importance of feeding and reproductive ecological traits as potentially relevant descriptors of springtail community structures was highlighted.     Effect of sitagliptin on cardiovascular outcomes in type 2 diabetes

    BACKGROUND: Data are lacking on the long-term effect on cardiovascular events of adding sitagliptin, a dipeptidyl peptidase 4 inhibitor, to usual care in patients with type 2 diabetes and cardiovascular disease. METHODS: In this randomized, double-blind study, we assigned 14,671 patients to add either sitagliptin or placebo to their existing therapy. Open-label use of antihyperglycemic therapy was encouraged as required, aimed at reaching individually appropriate glycemic targets in all patients. To determine whether sitagliptin was noninferior to placebo, we used a relative risk of 1.3 as the marginal upper boundary. The primary cardiovascular outcome was a composite of cardiovascular death, nonfatal myocardial infarction, nonfatal stroke, or hospitalization for unstable angina. RESULTS: During a median follow-up of 3.0 years, there was a small difference in glycated hemoglobin levels (least-squares mean difference for sitagliptin vs. placebo, -0.29 percentage points; 95% confidence interval [CI], -0.32 to -0.27). Overall, the primary outcome occurred in 839 patients in the sitagliptin group (11.4%; 4.06 per 100 person-years) and 851 patients in the placebo group (11.6%; 4.17 per 100 person-years). Sitagliptin was noninferior to placebo for the primary composite cardiovascular outcome (hazard ratio, 0.98; 95% CI, 0.88 to 1.09; P<0.001). Rates of hospitalization for heart failure did not differ between the two groups (hazard ratio, 1.00; 95% CI, 0.83 to 1.20; P = 0.98). There were no significant between-group differences in rates of acute pancreatitis (P = 0.07) or pancreatic cancer (P = 0.32). CONCLUSIONS: Among patients with type 2 diabetes and established cardiovascular disease, adding sitagliptin to usual care did not appear to increase the risk of major adverse cardiovascular events, hospitalization for heart failure, or other adverse events

    Evacetrapib and Cardiovascular Outcomes in High-Risk Vascular Disease

    BACKGROUND: The cholesteryl ester transfer protein inhibitor evacetrapib substantially raises the high-density lipoprotein (HDL) cholesterol level, reduces the low-density lipoprotein (LDL) cholesterol level, and enhances cellular cholesterol efflux capacity. We sought to determine the effect of evacetrapib on major adverse cardiovascular outcomes in patients with high-risk vascular disease. METHODS: In a multicenter, randomized, double-blind, placebo-controlled phase 3 trial, we enrolled 12,092 patients who had at least one of the following conditions: an acute coronary syndrome within the previous 30 to 365 days, cerebrovascular atherosclerotic disease, peripheral vascular arterial disease, or diabetes mellitus with coronary artery disease. Patients were randomly assigned to receive either evacetrapib at a dose of 130 mg or matching placebo, administered daily, in addition to standard medical therapy. The primary efficacy end point was the first occurrence of any component of the composite of death from cardiovascular causes, myocardial infarction, stroke, coronary revascularization, or hospitalization for unstable angina. RESULTS: At 3 months, a 31.1% decrease in the mean LDL cholesterol level was observed with evacetrapib versus a 6.0% increase with placebo, and a 133.2% increase in the mean HDL cholesterol level was seen with evacetrapib versus a 1.6% increase with placebo. After 1363 of the planned 1670 primary end-point events had occurred, the data and safety monitoring board recommended that the trial be terminated early because of a lack of efficacy. After a median of 26 months of evacetrapib or placebo, a primary end-point event occurred in 12.9% of the patients in the evacetrapib group and in 12.8% of those in the placebo group (hazard ratio, 1.01; 95% confidence interval, 0.91 to 1.11; P=0.91). CONCLUSIONS: Although the cholesteryl ester transfer protein inhibitor evacetrapib had favorable effects on established lipid biomarkers, treatment with evacetrapib did not result in a lower rate of cardiovascular events than placebo among patients with high-risk vascular disease.     &lt;p&gt;The fate and effects of toxic trace metals in soil freshly spiked soluble metal salts do not mimic those of metals in the field. This study was set up to test the magnitude of effects of salinity, acidification, and ageing on toxicity of lead (Pb) to plants, invertebrates and soil microbial processes. Three soils were spiked with Pb2+ salts up to a concentration of 8000 mg Pb/kg and were tested either after spiking, after soil leaching followed by pH correction, or after a 5-year outdoor ageing period with free drainage followed by pH correction. Soil solution ionic strength exceeded 150 mmol/L in soils tested directly after spiking and this decreased partially after leaching and returned back to background values after 5-year outdoor equilibration. Chronic toxicity to two plants, two invertebrates, and three microbial endpoints was consistently found in all spiked soils that were not leached. This toxicity significantly decreased or became absent after 5 years of ageing in 19 of the 20 toxicity tests by a factor 8 (median factor; range: 1.4-&amp;gt;50), measured by the factor increase of total soil Pb dose required to induce 10% inhibition. The toxicity of Pb in leached soils was intermediate between the other two treatments. The lowest detectable chronic thresholds (EC10) in aged soils ranged 350-5300 mg Pb/kg. Correlation analysis, including data of Pb2+ speciation in soil solution, suggests that reduced ionic strength rather than acidification or true ageing is the main factor explaining the soil treatment effects after spiking. It is suggested that future toxicity studies should test fine PbO powder as a relevant source for Pb in soils to exclude the confounding salt effects.&lt;/p&gt;</p

    OBJECTIVES: The aim of this study was to detect the onset, evolution, and meaning of extraluminal free fluid in a rat model of small-bowel obstruction using high-frequency microsonography. METHODS: Small-bowel obstruction was surgically created in 8 rats divided into 2 groups of 4 rats each. All rats were examined by high-frequency microsonography to monitor the evolution of small-bowel obstruction and the abdominal sonographic findings. In group 2 rats, the obstruction was resolved 2 hours after surgery. RESULTS: In all rats, free peritoneal fluid was detected just near the obstructed loop after 1 hour and in the hepatorenal recess after 2 hours. These features progressively increased in the following hours in group 1 rats. In group 2, the amount of free fluid decreased shortly after removing the obstruction. CONCLUSIONS: Free fluid is an early finding in small-bowel obstruction, and the increase or decrease of its amount is correlated with the worsening or resolution of the obstruction
