    Wake interaction and power production of variable height model wind farms

    Understanding wake dynamics is an ongoing research topic in wind energy, since wakes have considerable effects on the power production when wind turbines are placed in a wind farm. Wind tunnel experiments have been conducted to study the wake to wake interaction in a model wind farm in tandem with measurements of the extracted power. The aim is to investigate how alternating mast height influences the interaction of the wakes and the power production. Via the use of stereo-particle image velocimetry, the flow field was obtained in the first and last rows of the wind turbine array as a basis of comparison. It was found that downstream of the exit row wind turbine, the power was increased by 25% in the case of a staggered height configuration. This is partly due to the fact that the taller turbines reach into a flow area with a softened velocity gradient. Another aspect is that the wake downstream of a tall wind turbine to some extent passes above the standard height wind turbine. Overall the experiments show that the velocity field downstream of the exit row changes considerably when the mast height is alternating

    Upwelling and outwelling effects on the benthic regime of the continental shelf off Galicia, NW Spain

    The benthic regime off the Galician coast of NW Spain was surveyed to assess biogenic enrichment from coastal upwelling and detritus outwelling from the rías, large coastal embayments: the Rías Altas along the northern and the Rías Bajas along the western coast, which have intense mussel aquaculture. Sediment samples were collected from 1984 to 1986 and used for geological, microbiological, and macrofaunal studies. Sub-bottom acoustic profiler records and grab and core samples identified two main mud deposits on the western shelf that were aligned north to south and parallel with the coastline. The major axis of the mud deposit, which extended south to the Portuguese border, is associated with the three most southern rías (Arosa, Pontevedra and Vigo) along the western shelf. Sediment particle size analysis showed that sediments on the western shelf were heterogeneous, and grain size increased from the inner shelf to the shelf break. On the northern shelf, sediments exhibited a more homogeneous textural distribution. Sediment organic matter followed a similar pattern with that of particle size. The highest organic matter values, 10%, occurred on the western shelf nearest the Rías Bajas, but these values decreased offshore to between 2 and 4%. On the northern shelf organic matter content was generally less than 4% but with patches of higher organic content. The composition and structure of macroinfauna on the northern shelf, where seasonal coastal upwelling results in benthic enrichment, showed mainly small, surface feeding, and fast growing polychaetes. In contrast, macroinfauna on the western shelf showed more subsurface, deposit-feeding polychaetes. A main difference between the two shelves is that off the rías Bajas, besides coastal upwelling, outwelling from the highly productive rías Bajas, with their intense mussel aquaculture, also enriches the coastal sediment regime with a steady source of organic matter. Although seasonal and interannual variations occurred in the benthic bacteria, their general density distribution followed the pattern of organic matter content and particle grain size seasonally and interannually. The highest numbers of bacteria occurred in the upwelling region off the northern shelf and nearest the Rías Bajas on the western shelf. Both coastal upwelling and organic outwelling from the rías Bajas support benthic production alongthe western Galician shelf. The main commercialdemersal finfishalongthis coast is hake, Merluccius merluccius and blue-whiting, Micomesistius poutassou. The norwegian lobster, Nephrops norvegicus is also an important crop, and is more abundant off the rías Bajas in the finer and organic-richsediments where there are abundant prey resources of benthic infauna

    Preservation of whole antibodies within ancient teeth

    Archaeological remains can preserve some proteins into deep time, offering remarkable opportunities for probing past events in human history. Recovering functional proteins from skeletal tissues could uncover a molecular memory related to the life-history of the associated remains. We demonstrate affinity purification of whole antibody molecules from medieval human teeth, dating to the 13th–15th centuries, from skeletons with different putative pathologies. Purified antibodies are intact retaining disulphide-linkages, are amenable to primary sequences analysis, and demonstrate apparent immunoreactivity against contemporary EBV antigen on western blot. Our observations highlight the potential of ancient antibodies to provide insights into the long-term association between host immune factors and ancient microbes, and more broadly retain a molecular memory related to the natural history of human health and immunity

    Enhanced chlorhexidine skin penetration with 1,8-cineole

    BACKGROUND: Chlorhexidine (CHG) penetrates poorly into skin. The purpose of this study was to compare the depth of CHG skin permeation from solutions containing either 2% (w/v) CHG and 70% (v/v) isopropyl alcohol (IPA) or 2% (w/v) CHG, 70% (v/v) IPA and 2% (v/v) 1,8-cineole. METHODS: An ex-vivo study using Franz diffusion cells was carried out. Full thickness human skin was mounted onto the cells and a CHG solution, with or without 2% (v/v) 1,8-cineole was applied to the skin surface. After twenty-four hours the skin was sectioned horizontally in 100 μm slices to a depth of 2000 μm and the concentration of CHG in each section quantified using high performance liquid chromatography (HPLC). The data were analysed with repeated measures analysis of variance. RESULTS: The concentration of CHG in the skin on average was significantly higher (33.3% [95%, CI 1.5% - 74.9%]) when a CHG solution which contained 1,8-cineole was applied to the skin compared to a CHG solution which did not contain this terpene (P = 0.042). CONCLUSIONS: Enhanced delivery of CHG can be achieved in the presence of 1,8-cineole, which is the major component of eucalyptus oil. This may reduce the numbers of microorganisms located in the deeper layers of the skin which potentially could decrease the risk of surgical site infection

    Representing complex data using localized principal components with application to astronomical data

    Often the relation between the variables constituting a multivariate data space might be characterized by one or more of the terms: ``nonlinear'', ``branched'', ``disconnected'', ``bended'', ``curved'', ``heterogeneous'', or, more general, ``complex''. In these cases, simple principal component analysis (PCA) as a tool for dimension reduction can fail badly. Of the many alternative approaches proposed so far, local approximations of PCA are among the most promising. This paper will give a short review of localized versions of PCA, focusing on local principal curves and local partitioning algorithms. Furthermore we discuss projections other than the local principal components. When performing local dimension reduction for regression or classification problems it is important to focus not only on the manifold structure of the covariates, but also on the response variable(s). Local principal components only achieve the former, whereas localized regression approaches concentrate on the latter. Local projection directions derived from the partial least squares (PLS) algorithm offer an interesting trade-off between these two objectives. We apply these methods to several real data sets.     Evolution of eddy viscosity in the wake of a wind turbine

    The eddy viscosity hypothesis is a popular method in wind turbine wake modeling for estimating turbulent Reynolds stresses. We document the downstream evolution of eddy viscosity in the wake of a wind turbine from experimental and large-eddy-simulation data. Wake eddy viscosity is isolated from its surroundings by subtracting the inflow profile, and the driving forces are identified in each wake region. Eddy viscosity varies in response to changes in turbine geometry and nacelle misalignment with larger turbines generating stronger velocity gradients and shear stresses. We propose a model for eddy viscosity based on a Rayleigh distribution. Model parameters are obtained from scaling the eddy viscosity hypothesis and demonstrate satisfactory agreement with the reference data. The model is implemented in the curled wake formulation in the FLOw Redirection and Induction in Steady State (FLORIS) framework and assessed through comparisons with the previous formulation. Our approach produced more accurate flow field estimates with lower total error for the majority of cases.</p

    Analysis of qPCR reference genes stability determination methods and a practical approach for efficiency calculation on a turbot (Scphthalmus maximus) gonad dataset

    Gene expression analysis by reverse transcription quantitative PCR (qPCR) is the most widely used method for analyzing the expression of a moderate number of genes and also for the validation of microarray results. Several issues are crucial for a successful qPCR study, particularly the selection of internal reference genes for normalization and efficiency determination. There is no agreement on which method is the best to detect the most stable genes neither on how to perform efficiency determination. In this study we offer a comprehensive evaluation of the characteristics of reference gene selection methods and how to decide which one is more reliable when they show discordant outcomes. Also, we analyze the current efficiency calculation controversy. Our dataset is composed by gonad samples of turbot at different development times reared at different temperatures. Turbot (Scophthalmus maximus) is a relevant marine aquaculture European species with increasing production in the incoming years.     Combination with tomatidine improves the potency of posaconazole against Trypanosoma cruzi

    Azoles such as posaconazole (Posa) are highly potent against Trypanosoma cruzi. However, when tested in chronic Chagas disease patients, a high rate of relapse after Posa treatment was observed. It appears that inhibition of T. cruzi cytochrome CYP51, the target of azoles, does not deliver sterile cure in monotherapy. Looking for suitable combination partners of azoles, we have selected a set of inhibitors of sterol and sphingolipid biosynthetic enzymes. A small-scale phenotypic screening was conducted in vitro against the proliferative forms of T. cruzi, extracellular epimastigotes and intracellular amastigotes. Against the intracellular, clinically relevant forms, four out of 15 tested compounds presented higher or equal activity as benznidazole (Bz), with EC50 values </=2.2 muM. Ro48-8071, an inhibitor of lanosterol synthase (ERG7), and the steroidal alkaloid tomatidine (TH), an inhibitor of C-24 sterol methyltransferase (ERG6), exhibited the highest potency and selectivity indices (SI = 12 and 115, respectively). Both were directed to combinatory assays using fixed-ratio protocols with Posa, Bz, and fexinidazole. The combination of TH with Posa displayed a synergistic profile against amastigotes, with a mean SigmaFICI value of 0.2. In vivo assays using an acute mouse model of T. cruzi infection demonstrated lack of antiparasitic activity of TH alone in doses ranging from 0.5 to 5 mg/kg. As observed in vitro, the best combo proportion in vivo was the ratio 3 TH:1 Posa. The combination of Posa at 1.25 mpk plus TH at 3.75 mpk displayed suppression of peak parasitemia of 80% and a survival rate of 60% in the acute infection model, as compared to 20% survival for Posa at 1.25 mpk alone and 40% for Posa at 10 mpk alone. These initial results indicate a potential for the combination of posaconazole with tomatidine against T. cruzi

    Within the DIVERSIFY project for the diversification of species in European aquaculture, wreckfish is a special case because of its late sexual maturation, which does not occur until 7-8 years of age (Peleteiro and Brunzón, 2014), besides existing very few data on its reproductive biology because it is a demersal species whose adult specimens tend to live at the range of 100-1000 meters of deep.
