    IMMUNOPROFILE IN EFFUSION CYTOLOGY

    BACKGROUND: Cytology has a crucial role for diagnosing pleural and abdominal effusions. A prompt accurate diagnosis has both prognostic and therapeutic significance. However, cell morphology alone is not always sufficient to formulate such a diagnosis. In human medicine, immunocytochemistry of effusion cytology has now standardized procedures that provide reliable insights into various diagnostic dilemmas. OBJECTIVE: To describe the method of immunocytochemistry in effusion cytology and to estimate the value of a panel of markers in identifying cells in canine and feline effusions. MATERIALS AND METHODS: Human, feline and canine mesothelial cells were isolated in culture. Western\u2010blot (WB) analysis was used to ascertain antibody cross\u2010reactivity for all the markers, with the exception of HBME\u20101. Forty\u2010four cytospined or smeared effusion specimens from dogs and cats with a cytological diagnosis of reactive effusion or malignancy of non\u2010hematopoietic origin were stained with a standard panel of Vimentin, Cytokeratin (CK) AE1/AE3, CK 5/6 and HBME\u20101 as mesothelial cell markers; desmin as mesothelial cell malignancy marker; and CK7/CK20 as a marker of metastasis. Malignancy was confirmed by histologic evaluation; non\u2010malignant conditions were confirmed by follow\u2010up. Sensitivities, specificities and predictive values were calculated. RESULTS: The WB analysis confirmed the specific crossreactivity of the human antibodies for canine and feline proteins in mesothelial tissue. No significant differences were found between canine and feline results. Vimentin/cytokeratin coexpression had a sensitivity of 79% and a specificity of 92%, HBME\u20101 had 89% sensitivity and 23% specificity, and CK5/6 had 26% sensitivity and 100% specificity for mesothelial cells. Desmin had only 20% specificity for benign mesothelial cells, while CK7\u2010/CK20+ had a specificity of 79% and sensitivity of 30% for metastatic cells on effusions. CONCLUSION: Immunocytochemistry can be applied in effusion samples, and valuable results can be obtained. The most useful marker, with the highest overall accuracy for the identification of mesothelial cells in effusion, is the Vim/CK coexpression, being CK5/6 the more specific and HBME\u20101 the more sensitive antibody. Desmin is not useful for discriminating between benign and malignant mesothelial cells. The coordinate expression of CK7\u2010/CK20+ has not proved to be useful on the identification of metastatic cells on effusions

    Composição química e atividade antimicrobiana do óleo essencial de folhas e flores de Aloysia gratissima

    Volatile oils from leaves and flowers of Aloysia gratissima were investigated for their chemical composition and antimicrobial activity against the bacteria Bacilus subtilis, Staphylococcus aureus, Salmonella choleraesuis, Pseudomonas aeruginosa, Streptococcus pneumoniae and the Candida albicans yeast. The minimum inhibitory concentrations (MIC) of the oils were determined by the micro-dilution method, while the chemical composition was determined by GC-MS (gas chromatography mass spectrometry). The fresh leaves and inflorescence were subjected to hydrodistillation for 120 min using a Clevenger-type apparatus, and the essential oil was tested against microorganisms. High concentrations of sesquiterpenes were observed for the inflorescence, and monoterpenes were observed for the leaves. The main compounds of the inflorescence essential oil were E-caryophyllene, germacrene B, guaiol and bulnesol, while in the leaves the main compounds were trans-pinocamphone, trans-pinocarveyl acetate, and guaiol. The essential oil from the leaves showed an effect against P. aeruginosa and S. pneumonia, and the essential oil of the inflorescence showed an effect against P. aeruginosa, S. pneumonia, and Candida albicans.O óleo essencial de folhas e de flores de Aloysia gratissima foi avaliado quanto à composição química e ação antimicrobiana contra as bactérias Bacilus subtilis, Staphylococcus aureus, Salmonella choleraesuis, Pseudomonas aeruginosa, Streptococcus pneumoniae, e a levedura Candida albicans. A concentração mínima inibitória (MIC) dos óleos essenciais foi determinada pelo método da microdiluição e a composição química determinada por CG-EM (Cromatografia Gasosa acoplada a Espectrômetro de Massas). Folhas e inflorescências frescas foram hidrodestiladas por 120 minutos em aparelho Clevenger sendo o óleo essencial testado contra microorganismos. Para as flores foi observada maior concentração de sesquiterpenos, enquanto que as folhas apresentaram maior concentração de monoterpenos. Os principais constituintes do óleo essencial da flor foram: E-cariofileno, germacreno B, guaiol e bulnesol; e das folhas foram: trans-pinocamfona, acetato de trans-pinocarveol e guaiol.     Evaluation of the almond shell oxypropylation process trough the surface response methodology

    The efficiency of an oxypropylation process depends on several variables and operating conditions, which may not be generalized due to the diverse nature of subtracts. In lignocellulosic biomasses, the content of each fraction (lignin, cellulose and hemicellulose) can differ, as well as, the crystalline organization, which may limit reagent’s access to biomass. Also, the hydroxyl content can vary among biomasses; high values demand higher amounts of reactants, namely catalyst content, and more severe reaction conditions. Therefore, owing to biomass variability, selection of the operating conditions for oxypropylation their optimization is a key issue. In this context, one-factor-at-a-time approaches are commonly used to optimize processes; but it is well-known that optimal operating conditions or interactions between variables cannot be predicted by this simplistic method.     Self-Similar Factor Approximants

    The problem of reconstructing functions from their asymptotic expansions in powers of a small variable is addressed by deriving a novel type of approximants. The derivation is based on the self-similar approximation theory, which presents the passage from one approximant to another as the motion realized by a dynamical system with the property of group self-similarity. The derived approximants, because of their form, are named the self-similar factor approximants. These complement the obtained earlier self-similar exponential approximants and self-similar root approximants. The specific feature of the self-similar factor approximants is that their control functions, providing convergence of the computational algorithm, are completely defined from the accuracy-through-order conditions. These approximants contain the Pade approximants as a particular case, and in some limit they can be reduced to the self-similar exponential approximants previously introduced by two of us. It is proved that the self-similar factor approximants are able to reproduce exactly a wide class of functions which include a variety of transcendental functions. For other functions, not pertaining to this exactly reproducible class, the factor approximants provide very accurate approximations, whose accuracy surpasses significantly that of the most accurate Pade approximants. This is illustrated by a number of examples showing the generality and accuracy of the factor approximants even when conventional techniques meet serious difficulties.Comment: 22 pages + 11 ps figure

    Distribution of black flies (Diptera: Simuliidae) in the State of Espírito Santo, Brazil

    Entre os anos de 2004 e 2008 foram realizadas coletas de Simuliidae em várias localidades no Estado do Espírito Santo, Brasil. No total, 66 córregos e rios das doze principais bacias hidrográficas do estado foram amostrados. Dezessete espécies foram coletadas, sendo que nove delas representam ocorrências novas para o estado (Simulium pertinax, S. jujuyense, S. rubrithorax, S. subnigrum, S. travassosi, S. spinibranchium, S. hirtipupa, S. lutzianum e S. anamariae), duplicando o número de espécies conhecidas no estado.We conducted a freshwater survey from 2004 to 2008 in several localities of Espírito Santo, Brazil. In total, 66 streams and rivers were sampled in the 12 hydrographic basins of the state. We collected and identified 17 species of which nine represent new records (Simulium pertinax, S. jujuyense, S. rubrithorax, S. subnigrum, S. travassosi, S. spinibranchium, S. hirtipupa, S. lutzianum e S. anamariae). These findings doubled the number of records of black fly species to the State of Espírito Santo, Brazil

    The First Magnetic Fields

    We review current ideas on the origin of galactic and extragalactic magnetic fields. We begin by summarizing observations of magnetic fields at cosmological redshifts and on cosmological scales. These observations translate into constraints on the strength and scale magnetic fields must have during the early stages of galaxy formation in order to seed the galactic dynamo. We examine mechanisms for the generation of magnetic fields that operate prior during inflation and during subsequent phase transitions such as electroweak symmetry breaking and the quark-hadron phase transition. The implications of strong primordial magnetic fields for the reionization epoch as well as the first generation of stars is discussed in detail. The exotic, early-Universe mechanisms are contrasted with astrophysical processes that generate fields after recombination. For example, a Biermann-type battery can operate in a proto-galaxy during the early stages of structure formation. Moreover, magnetic fields in either an early generation of stars or active galactic nuclei can be dispersed into the intergalactic medium.Comment: Accepted for publication in Space Science Reviews.     Anisotropy studies around the galactic centre at EeV energies with the Auger Observatory

    Data from the Pierre Auger Observatory are analyzed to search for anisotropies near the direction of the Galactic Centre at EeV energies. The exposure of the surface array in this part of the sky is already significantly larger than that of the fore-runner experiments. Our results do not support previous findings of localized excesses in the AGASA and SUGAR data. We set an upper bound on a point-like flux of cosmic rays arriving from the Galactic Centre which excludes several scenarios predicting sources of EeV neutrons from Sagittarius AA. Also the events detected simultaneously by the surface and fluorescence detectors (the `hybrid' data set), which have better pointing accuracy but are less numerous than those of the surface array alone, do not show any significant localized excess from this direction.Comment: Matches published versio
