    System Frequency Dynamic Response of a Novel, Self-Synchronizing Inverter in a High Renewable Penetration Grid

    In this thesis, a controller achieving current tracking objective without knowledge of the grid parameters is developed. An estimated rotating reference frame (γδ -frame) is utilized. Within the control scheme, adaptive compensation terms facilitate the current tracking objective and, simultaneously, accounts for the unavailable grid voltage magnitude, grid frequency, and grid phase, hence eliminating the need for an additional measurement and feedback system for synchronization, such as a Phase-Locked Loop (PLL). System frequency behavior using the controller in a high-renewable (low-inertia) grid indicates that the monitored performance metrics are significantly improved when compared to PLL-controlled, inverter-dominated grids

    The Listening Network and Cochlear Implant Benefits in Hearing-Impaired Adults

    Older adults with mild or no hearing loss make more errors and expend more effort listening to speech. Cochlear implants (CI) restore hearing to deaf patients but with limited fidelity. We hypothesized that patient-reported hearing and health-related quality of life in CI patients may similarly vary according to age. Speech Spatial Qualities (SSQ) of hearing scale and Health Utilities Index Mark III (HUI) questionnaires were administered to 543 unilaterally implanted adults across Europe, South Africa, and South America. Data were acquired before surgery and at 1, 2, and 3 years post-surgery. Data were analyzed using linear mixed models with visit, age group (18–34, 35–44, 45–54, 55–64, and 65+), and side of implant as main factors and adjusted for other covariates. Tinnitus and dizziness prevalence did not vary with age, but older groups had more preoperative hearing. Preoperatively and postoperatively, SSQ scores were significantly higher (¿0.75–0.82) for those aged <45 compared with those 55+. However, gains in SSQ scores were equivalent across age groups, although postoperative SSQ scores were higher in right-ear implanted subjects. All age groups benefited equally in terms of HUI gain (0.18), with no decrease in scores with age. Overall, younger adults appeared to cope better with a degraded hearing before and after CI, leading to better subjective hearing performance.

    This article presents an autobiographical account of an older woman’s lived experience of self-employment. Little is known about women who experience ongoing self-employment into their 50s and beyond. Shoshanna’s personal narrative describes her experiences and the challenges she has faced as she reflects upon her attempts to grow and sustain her business and the implications of ageism and gender inequality in laying a claim to entrepreneurship. The narrative proceeds to reflect on her activist work, as it is constructed through the creation of a social enterprise to support older people. Shoshanna’s narrative provides valuable insights into the intersection of age and gender in self-employment moving from discrimination to active support

    Serotonergic signalling suppresses ataxin 3 aggregation and neurotoxicity in animal models of Machado-Joseph disease

    Polyglutamine diseases are a class of dominantly inherited neurodegenerative disorders for which there is no effective treatment. Here we provide evidence that activation of serotonergic signalling is beneficial in animal models of Machado-Joseph disease. We identified citalopram, a selective serotonin reuptake inhibitor, in a small molecule screen of FDA-approved drugs that rescued neuronal dysfunction and reduced aggregation using a Caenorhabditis elegans model of mutant ataxin 3-induced neurotoxicity. MOD-5, the C. elegans orthologue of the serotonin transporter and cellular target of citalopram, and the serotonin receptors SER-1 and SER-4 were strong genetic modifiers of ataxin 3 neurotoxicity and necessary for therapeutic efficacy. Moreover, chronic treatment of CMVMJD135 mice with citalopram significantly reduced ataxin 3 neuronal inclusions and astrogliosis, rescued diminished body weight and strikingly ameliorated motor symptoms.     Business Owner-Managers’ Job Autonomy and Job Satisfaction: Up, Down or No Change?

    KingdomThe current study developed a dynamic model which identified a pattern of change in small business owner-managers’ job autonomy and job satisfaction separately through the trend analyses (linear, quadratic, and cubic trends). The current study then tested the associations between the growth models of job autonomy and job satisfaction. The study utilized data from an Australian sample over 9 years with a total sample of 1,044 self-employed individuals. In brief, the findings illustrate a curvilinear relationship (cubic and non-monotonic) between changes in job autonomy and job satisfaction. Further, the change rate of job satisfaction was faster among small business owner-managers who perceived greater fluctuation of job autonomy, compared to those who perceived lesser shifts in job autonomy

    Development of nanoelectrospray high resolution isotope dilution mass spectrometry for targeted quantitative analysis of urinary metabolites: Application to population profiling and clinical studies

    ABSTRACT An automated chip-based electrospray platform was used to develop a high-throughput nanoelectrospray high resolution mass spectrometry (nESI-HRMS) method for multiplexed parallel untargeted and targeted quantitative metabolic analysis of the urine samples. The method was demonstrated to be suitable for metabolic analysis of large sample numbers and can be applied to large-scale epidemiological and stratified medicine studies. The method requires a small amount of sample (5 μL of injectable volume containing 250 nL of original sample), and the analysis time for each sample is three minutes per sample to acquire data in both negative and positive ion modes. Identification of metabolites was based on the high resolution accurate mass and tandem mass spectrometry using authentic standards. The method was validated for 8 targeted metabolites and was shown to be precise and accurate. The mean accuracy of individual measurements being of 106% and the intra-and inter-day precision (expressed as relative standard deviations) were 9% and 14%, respectively. Selected metabolites were quantified by standard addition calibration using the stable isotope labelled internal standards in a pooled urine sample, to account for any matrix effect. The multiple point standard addition calibration curves yielded correlation coefficients greater than 0.99, and the linear dynamic range was more than three orders of magnitude. As a proof-of-concept the developed method was applied for targeted quantitative analysis of a set of 101 urine samples obtained from female participants with different pregnancy outcomes. In addition to the specifically targeted metabolites, several other metabolites were quantified relative to the internal standards. Based on the calculated concentrations, some metabolites showed significant differences according to different pregnancy outcomes. The acquired high resolution full-scan data were used for further untargeted fingerprinting and improved the differentiation of urine samples based on pregnancy outcome

    Lithium suppression of tau induces brain iron accumulation and neurodegeneration

    Lithium is a first-line therapy for bipolar affective disorder. However, various adverse effects, including a Parkinson-like hand tremor, often limit its use. The understanding of the neurobiological basis of these side effects is still very limited. Nigral iron elevation is also a feature of Parkinsonian degeneration that may be related to soluble tau reduction. We found that magnetic resonance imaging T2 relaxation time changes in subjects commenced on lithium therapy were consistent with iron elevation. In mice, lithium treatment lowers brain tau levels and increases nigral and cortical iron elevation that is closely associated with neurodegeneration, cognitive loss and parkinsonian features. In neuronal cultures lithium attenuates iron efflux by lowering tau protein that traffics amyloid precursor protein to facilitate iron efflux. Thus, tau- and amyloid protein precursor-knockout mice were protected against lithium-induced iron elevation and neurotoxicity. These findings challenge the appropriateness of lithium as a potential treatment for disorders where brain iron is elevated (for example, Alzheimer’s disease), and may explain lithium-associated motor symptoms in susceptible patients

    New approaches to youth in thrill society: sport participation as fuel to boost outlook on future and concepts on self

    The theory of “thrill-society” (Schulze 1992) conceptualizes that increased economic status that allows the focus of daily life to switch from providing for physical needs to searching for a meaningful life and self-fulfilment. Combined with the expansion of education, it causes a smooth transition from traditionally inherited social positions and class-based hierarchy in society to a higher degree of social mobility, increased individualisation and diversification of life styles. Noting that, the actualization of this concept in Hungarian society came into effect only along societal discrepancies; still, the uncertainties and insecurities that accompany the process of ‘thrill projects’ collection are substantial. The purpose of this study was to examine the role of sport participation in the lives of young people, how sport may assist young people in coping with the insecurities and uncertainties created in the societal scene that is characterised by the combination of Schulze’s “thrill-society” and the inheritance of political, economic and societal transition of Hungary. Specifically, this study aims to find differences between sport participants and non-participants in their perception of future on micro and macro level, their readiness to take risks and challenges, and their self-concept related to their own health, physical condition, and physical appearance. Stratified random sampling was applied to obtain an accurate representation of Hungarian youth population. Data were analysed by using cross tabulation, non-parametric and multidimensional statistical methods. The results showed that sport participants adopted a more positive image of the future, higher ability to assume risks and a more modern state of mind, as well as a more stable self-concept in comparison to non-participant youth. Also, it seems that the sporting contest may be as strong as sociodemographic positioning in the formulation of these life capabilities. It can be suggested that sport may assist youth with a stable and accountable value environment that reduces the variety of opportunities and provides resources to better deal with societal uncertainties; meanwhile it opens new avenues of personal freedom even in a “thrill society” that filled with deficits in transitioning societies
