    The Digestion, Absorption and Utilization of Refined Palm Oil, Palm Olein and Palm Stearin in the Rat

    A nutritional evaluation of refined, bleached and deodorised (RBD) palm oil and its fractionation products, RBD palm olein and RBD palm stearin in terms of their digestibility, rate of absorption and food efficiency in rats, shows that RBD palm olein is a better oil by these criteria than RBD palm stearin and the unfractionated RBD palm oil. Nevertheless, the nutritional indices reported for RBD palm oil and RBD palm stearin are well within the range reported for most cooking oils and fats. The digestibility and food efficiency of RBD palm olein are comparable to that of soybean oil, being 97% and 98% respectively of the values found for the latter oil, although the absorption rate of RED palm olein is 10% less than that obtained for soybean oil

    Chemically-interesterified (CIE) fats are trans-fat free and are increasingly being used as an alternative to hydrogenated oils for food manufacturing industries to optimize their products’ characteristics and nutrient compositions. The metabolic effects of CIE fats on insulin activity, lipids, and adiposity in humans are not well established. We investigated the effects of CIE fats rich in palmitic (C16:0, IEPalm) and stearic (C18:0, IEStear) acids on insulin resistance, serum lipids, apolipoprotein concentrations, and adiposity, using C16:0-rich natural palm olein (NatPO) as the control. We designed a parallel, double-blind clinical trial. Three test fats were used to prepare daily snacks for consumption with a standard background diet over a period of 8 weeks by three groups of a total of 85 healthy, overweight adult volunteers. We measured the outcome variables at weeks 0, 6, and at the endpoint of 8. After 8 weeks, there was no significant difference in surrogate biomarkers of insulin resistance in any of the IE fat diets (IEPalm and IEStear) compared to the NatPO diet. The change in serum triacylglycerol concentrations was significantly lower with the IEStear diet, and the changes in serum leptin and body fat percentages were significantly lower in the NatPO-diet compared to the IEPalm diet. We conclude that diets containing C16:0 and C18:0-rich CIE fats do not affect markers of insulin resistance compared to a natural C16:0-rich fat (NatPO) diet. Higher amounts of saturated fatty acids (SFAs) and longer chain SFAs situated at the sn-1,3 position of the triacylglycerol (TAG) backbones resulted in less weight gain and lower changes in body fat percentage and leptin concentration to those observed in NatPO and IEStear

    Case study of temporal changes in maternal dietary intake and the association with breast milk mineral contents

    Minerals play important roles in infant growth and development, even though they only make up to 0.2 g% of the mature breast milk contents. Limited studies examined the association between maternal dietary intake and breast milk mineral contents in a temporal manner. Twenty Malaysian Malay postpartum mothers were recruited by either convenience or snow balling sampling from the urban lower middle income residential areas. Dietary intake of the subjects was obtained by 24-hour recall during each breast milk collection. Adequacy of maternal mineral intake was compared with the latest Recommended Nutrient Intake for Malaysia. Each of the subject provided breast milk samples for three times (T1, T2, and T3) at consecutive 2-week intervals. Breast milk concentrations of selected macro- and micro-minerals, including Ca, P, K, Na, Mg, Fe, Zn, Cu, Mn, Se, I, Cr and Mo were determined by inductively coupled plasma mass spectrometry (ICP-MS). Subjects were aged 31.4 +/- 6.1 years with a majority (60 %) having post-secondary school/college education. While maternal intake of macro-minerals, Ca, P, K and Mg, did not display a significant temporal changes from T1 to T3, the intake of micro-minerals, Cu, Mn and I decreased significantly over time from T1 to T3. Breast milk K, Fe, Zn and Cu concentrations showed a significant decreased with the progression of lactation from transitional (2-3 weeks) to established stage (>8-12 weeks). Significant correlations were established between maternal intake of K, Na, Fe and Se and their respective concentrations in breast milk in the present study. This case study revealed an inadequate maternal intake of several key micro-minerals (Cu, Mn, I) among the postpartum Malay mothers and a decreasing concentrations of certain essential minerals (K, Fe, Zn and Cu) in breast milk with lactation stage

    Temporal Changes in Breast Milk Fatty Acids Contents:A Case Study of Malay Breastfeeding Women

    The composition of human breast milk changes in the first two months of life, adapting itself to the evolving needs of the growing new-born. Lipids in milk are a source of energy, essential fatty acids (FA), fat-soluble vitamins, and vital bioactive components. Information on breast milk FA of Malaysian lactating women is scarce. Based on convenience sampling, a total of 20 Malay breastfeeding women who fulfilled the inclusion criteria were recruited. Breast milk was collected three times from each subject at consecutive intervals of 2-3 weeks apart. A total of 60 breast milk samples were collected and classified into "transitional milk" (n = 8), "early milk" (n = 26) and "mature milk" (n = 26). All milk samples were air freighted to University of Groningen, Netherlands for analysis. The dominant breast milk FA were oleic acid, constituting 33% of total fatty acids, followed by palmitic acid (26%). Both these FA and the essential FA, linoleic acid (10%) and alpha-linolenic acid (0.4%), showed no significant changes from transitional to mature milk. Breast milk ratio of n-6:n-3 polyunsaturated fatty acids (PUFA) was comparatively high, exceeding 10 throughout the lactation period, suggesting a healthier balance of PUFA intake is needed in pregnancy and at postpartum

    Improving phenotyping in winter barley cultivars towards waterlogging tolerance by combining field trials under natural conditions with controlled growth condition experiments

    Additional rainfall in Northern Europe due to global climate change is increasing the incidences of field flooding. Flooding causes hypoxic stress that results in a reduced capacity for photosynthesis, reduction in nutrient availability and uptake, increased production of toxic metabolites by anaerobic bacteria in the soil, and ultimately yield losses and crop death. To overcome hypoxic environmental conditions, new cultivars need to be bred and tested for waterlogging tolerance. We scored 403 winter barley cultivars from the ‘Association Genetics of UK Elite Barley’ (AGOUEB) population, taking advantage of the phenotypic changes associated with hypoxic stress. This enabled us to identify an initial set of waterlogging sensitive and tolerant cultivars. Comparative analysis of a subset of 65 cultivars exposed to waterlogging stress under field and growth cabinet environments showed variability in scores due to varying sensitivity to waterlogging over multi-season field trials. In field trials, we observed waterlogging damage resulting in reductions in biomass, grain yield and crop height. However, the effects varied between seasons and the severity of waterlogging due to differences in the topography of the field and the amount of rainfall. To overcome the seasonal variations in environmental conditions in multi-season field trials, we developed in parallel, an enhanced phenotyping method by complementing field experiments with phenotyping under controlled growth conditions. The phenotyping scoring method allows for the grouping of cultivars by sensitivity and tolerance to waterlogging, with limited variance between cultivars scored in the field and controlled conditions. Together, these two complementary approaches maximise the data available to breeders, allowing for the reliable selection of more tolerant cultivars able to grow under flooding conditions

    Gradient Optics of subwavelength nanofilms

    Propagation and tunneling of light through subwavelength photonic barriers, formed by dielectric layers with continuous spatial variations of dielectric susceptibility across the film are considered. Effects of giant heterogeneity-induced non-local dispersion, both normal and anomalous, are examined by means of a series of exact analytical solutions of Maxwell equations for gradient media. Generalized Fresnel formulae, visualizing a profound influence of gradient and curvature of dielectric susceptibility profiles on reflectance/transmittance of periodical photonic heterostructures are presented. Depending on the cutoff frequency of the barrier, governed by technologically managed spatial profile of its refractive index, propagation or tunneling of light through these barriers are examined. Nonattenuative transfer of EM energy by evanescent waves, tunneling through dielectric gradient barriers, characterized by real values of refractive index, decreasing in the depth of medium, is shown. Scaling of the obtained results for different spectral ranges of visible, IR and THz waves is illustrated. Potential of gradient optical structures for design of miniaturized filters, polarizers and frequency-selective interfaces of subwavelength thickness is considered

    Predicting Alcohol Pre-Drinking in Australian Undergraduate Students Using an Integrated Theoretical Model

    Background: The aim of the present study was to examine the social-cognitive and motivational factors associated with pre-drinking based on a model integrating motivational constructs from self-determination theory and belief-based constructs from the theory of planned behaviour. Methods: A prospective correlational design was used. Participants (N = 286; 66.4% female) completed self-report measures of past alcohol consumption, autonomous and controlled forms of motivation from self-determination theory, and attitudes, subjective norms, perceived behavioural control, and behavioural intentions from the theory of planned behaviour at baseline. Participants reported pre-drinking frequency four weeks later. Results: Variance-based structural equation modelling showed that the hypothesised model predicted 54 per cent of the variance in pre-drinking intentions at baseline, and 20 per cent of the variance in pre-drinking behaviour at follow-up. Mediation analyses indicated strong, statistically significant effects of autonomous motivation on intentions to pre-drink, partially mediated by attitudes and subjective norms. Intention and perceived behavioural control significantly predicted pre-drinking frequency. Conclusions: Results provide support for the hypothesised model relationships. Autonomous motivation, attitude, subjective norm, and perceived behavioural control were influential in forming students' intentions to pre-drink. However, consistent with previous findings, the intention-behaviour relationship was relatively weak. Future research should look to non-intentional and volitional processes that may influence pre-drinking in undergraduates

    Extended Longevity of Reproductives Appears to be Common in Fukomys Mole-Rats (Rodentia, Bathyergidae)

    African mole-rats (Bathyergidae, Rodentia) contain several social, cooperatively breeding species with low extrinsic mortality and unusually high longevity. All social bathyergids live in multigenerational families where reproduction is skewed towards a few breeding individuals. Most of their offspring remain as reproductively inactive “helpers” in their natal families, often for several years. This “reproductive subdivision” of mole-rat societies might be of interest for ageing research, as in at least one social bathyergid (Ansell's mole-rats Fukomys anselli), breeders have been shown to age significantly slower than non-breeders. These animals thus provide excellent conditions for studying the epigenetics of senescence by comparing divergent longevities within the same genotypes without the inescapable short-comings of inter-species comparisons. It has been claimed that many if not all social mole-rat species may have evolved similar ageing patterns, too. However, this remains unclear on account of the scarcity of reliable datasets on the subject. We therefore analyzed a 20-year breeding record of Giant mole-rats Fukomys mechowii, another social bathyergid species. We found that breeders indeed lived significantly longer than helpers (ca. 1.5–2.2fold depending on the sex), irrespective of social rank or other potentially confounding factors. Considering the phylogenetic positions of F. mechowii and F. anselli and unpublished data on a third Fukomys-species (F. damarensis) showing essentially the same pattern, it seems probable that the reversal of the classic trade-off between somatic maintenance and sexual reproduction is characteristic of the whole genus and hence of the vast majority of social mole-rats

    Tissue engineering of functional articular cartilage: the current status

    Osteoarthritis is a degenerative joint disease characterized by pain and disability. It involves all ages and 70% of people aged >65 have some degree of osteoarthritis. Natural cartilage repair is limited because chondrocyte density and metabolism are low and cartilage has no blood supply. The results of joint-preserving treatment protocols such as debridement, mosaicplasty, perichondrium transplantation and autologous chondrocyte implantation vary largely and the average long-term result is unsatisfactory. One reason for limited clinical success is that most treatments require new cartilage to be formed at the site of a defect. However, the mechanical conditions at such sites are unfavorable for repair of the original damaged cartilage. Therefore, it is unlikely that healthy cartilage would form at these locations. The most promising method to circumvent this problem is to engineer mechanically stable cartilage ex vivo and to implant that into the damaged tissue area. This review outlines the issues related to the composition and functionality of tissue-engineered cartilage. In particular, the focus will be on the parameters cell source, signaling molecules, scaffolds and mechanical stimulation. In addition, the current status of tissue engineering of cartilage will be discussed, with the focus on extracellular matrix content, structure and its functionality
