    Therapeutic role of MSCs-derived exosomes for Alzheimer's disease

    The discovery of mesenchymal stem cell-derived exosomes (MSCs-derived exosomes) has shed a new light in the development of disease-modifying treatments for Alzheimer (AD). The present study is an attempt to investigate the influence of these extracellular vesicles (exosomes) in Alz therapy. A total of 38 adult female albino Wistar rats were randomly assigned to the following groups: Control group (C) of 8 rats given saline; Alzheimer’s group (Alz) of 15 ovariectomized animals inoculated orally with AlCl3 (17mg/Kg b.wt/day) for 2 months after 6 weeks of surgical operation and 15 Alzheimer’s disease-induced rats treated (i.v) with (107 MSCs-derived exosomes/rat/week). After 2 and 4 weeks animals were sacrificed by ether inhalation anesthesia where brains were removed and processed for histological investigation by Hx&E and biochemical analysis for measuring β-Amyloid 1-42 and Brain derived neurotrophic factor (BDNF) levels

    Heterotic grouping of maize inbred lines using line x tester analysis

    The efficiency of the assignment of germplasm lines into heterotic groups is a prerequisite for obtaining useful heterotic patterns among germplasm lines. Therefore, this study was conducted to group lines into heterotic group besides assess the effects of both general and specific abilities. Fifteen white maize parental lines were crossed with two testers, i.e. parental line SK-12 and Single cross 120 at Agricultural Research Station in Sakha, Egypt during 2021 summer season. The resultant 30 crosses were evaluated along with four commercial hybrids in three locations during 2022. The parental inbred lines Sk5001/42, Sk5003/48, Sk5003/53, Sk5004/55 and Sk5004/56 recorded positive GCA effect on grain yield while Sk5001/41, Sk5002/46 and Sk5004/56 exhibited desirable GCA effects for earliness. Five single crosses viz. (Sk5003/53 × Sk12), (Sk5001/42 × Sk12), (Sk5004/55 × Sk12), (Sk5003/48 × Sk12) and (Sk5004/56 × Sk12) and two three way crosses (Sk5001/42 × SC.120) and (Sk5003/53 × SC.120) did not differ significantly against their respective check hybrids (SC 10 and TWC 321) though they outyield their respective checks. These hybrids could be subjected to further multi-location evaluation to assess the yield stability for commercial exploitation. Two groups of parental lines were created based on Heterotic Group Specific and General Combining Ability (HSGCA) method for grain yield trait. These groups might be employed in a breeding program to pick the best parents to make crosses in other combinations

    Structural and biochemical impact of C8-aryl-guanine adducts within the NarI recognition DNA sequence: influence of aryl ring size on targeted and semi-targeted mutagenicity

    Chemical mutagens with an aromatic ring system may be enzymatically transformed to afford aryl radical species that preferentially react at the C8-site of 2′-deoxyguanosine (dG). The resulting carbon-linked C8-aryl-dG adduct possesses altered biophysical and genetic coding properties compared to the precursor nucleoside. Described herein are structural and in vitro mutagenicity studies of a series of fluorescent C8-aryl-dG analogues that differ in aryl ring size and are representative of authentic DNA adducts. These structural mimics have been inserted into a hotspot sequence for frameshift mutations, namely, the reiterated G3-position of the NarI sequence within 12mer (NarI(12)) and 22mer (NarI(22)) oligonucleotides. In the NarI(12) duplexes, the C8-aryl-dG adducts display a preference for adopting an anti-conformation opposite C, despite the strong syn preference of the free nucleoside. Using the NarI(22) sequence as a template for DNA synthesis in vitro, mutagenicity of the C8-aryl-dG adducts was assayed with representative high-fidelity replicative versus lesion bypass Y-family DNA polymerases, namely, Escherichia coli pol I Klenow fragment exo− (Kf−) and Sulfolobus solfataricus P2 DNA polymerase IV (Dpo4). Our experiments provide a basis for a model involving a two-base slippage and subsequent realignment process to relate the miscoding properties of C-linked C8-aryl-dG adducts with their chemical structure

    Restoration of heart function using transplantation of human umbilical cord matrix-derived cardiomyocytes and vascular endothelial growth factor

    Objectives: In the previous study, although it has been shown that intramyocardial injection of human umbilical cord matrix stem cell (hUCM) improved cardiac function 4 weeks post MI, but angiogenesis has not been observed. Angiogenesis and replacing lost cardiomyocytes with new, live cardiomyocytes are considered as two key agents in cardiac repair. To achieve the above two factors we examined the effects of combination of stem cell and angiogenic therapy approaches by simultaneously injection of hUCM-derived cardiomyocytes with vascular endothelial growth factor (VEGF) in cardiac repair. Methods: MI-induced animals(by ligation of LAD) received 50 μl PBS, 5 � 106 differentiated hUCM cells (dhUCM), 5μg VEGF in normal saline and 5 � 106 dhUCM cells combined with 5μg VEGF in normal saline, intramyocardialy. MI group, with no other intervention, served as a control group. We were assessed survival, migration and integration of dhUCM cells, as well as angiogenesis eight weeks post MI induction. Results: Eight weeks post MI, although dhUCM and VEGF groups have shown that LVEF and LVFS improved significantly, but animals in dhUCM+VEGF group have the highest rise in LVEF and LVFS in comparison to the other MI-induced groups (p<0.05). Histological and morphological analysis have revealed that myocardium of animals in dhUCM+VEGF group have the highest vascular density and the lowest fibrosis tissue in comparison to the other MI-induced groups (p<0.05). Immunohistological assessments revealed that transplanted dhUCM cells have survived, migrated to infarcted area and integrated with recipient cardiac tissue. Conclusion: we have found that intramyocardial administration of dhUCM cells combined with VEGF improved cardiac function, enhanced angiogenesis and reduced fibrosis tissue formation after MI, eight weeks post MI. © Kaveh et al

    Evidence of Color Coherence Effects in W+jets Events from ppbar Collisions at sqrt(s) = 1.8 TeV

    We report the results of a study of color coherence effects in ppbar collisions based on data collected by the D0 detector during the 1994-1995 run of the Fermilab Tevatron Collider, at a center of mass energy sqrt(s) = 1.8 TeV. Initial-to-final state color interference effects are studied by examining particle distribution patterns in events with a W boson and at least one jet. The data are compared to Monte Carlo simulations with different color coherence implementations and to an analytic modified-leading-logarithm perturbative calculation based on the local parton-hadron duality hypothesis.Comment: 13 pages, 6 figures.     Advanced functionality for radio analysis in the Offline software framework of the Pierre Auger Observatory

    The advent of the Auger Engineering Radio Array (AERA) necessitates the development of a powerful framework for the analysis of radio measurements of cosmic ray air showers. As AERA performs "radio-hybrid" measurements of air shower radio emission in coincidence with the surface particle detectors and fluorescence telescopes of the Pierre Auger Observatory, the radio analysis functionality had to be incorporated in the existing hybrid analysis solutions for fluoresence and surface detector data. This goal has been achieved in a natural way by extending the existing Auger Offline software framework with radio functionality. In this article, we lay out the design, highlights and features of the radio extension implemented in the Auger Offline framework. Its functionality has achieved a high degree of sophistication and offers advanced features such as vectorial reconstruction of the electric field, advanced signal processing algorithms, a transparent and efficient handling of FFTs, a very detailed simulation of detector effects, and the read-in of multiple data formats including data from various radio simulation codes.     Search for First Harmonic Modulation in the Right Ascension Distribution of Cosmic Rays Detected at the Pierre Auger Observatory

    We present the results of searches for dipolar-type anisotropies in different energy ranges above 2.5×10172.5\times 10^{17} eV with the surface detector array of the Pierre Auger Observatory, reporting on both the phase and the amplitude measurements of the first harmonic modulation in the right-ascension distribution. Upper limits on the amplitudes are obtained, which provide the most stringent bounds at present, being below 2% at 99% C.L.C.L. for EeV energies. We also compare our results to those of previous experiments as well as with some theoretical expectations.Comment: 28 pages, 11 figure
