pessoas engajadas nessa actividade.
As
nativa.
poderão contribuir para a melhor compreensão do aparecimento e progressão
de algumas patologias, nomeadamente a DM. Os novos compostos e
estruturas modificadas que iremos identificar e caracterizar, poderão no futuro,
servir como marcadores destes mecanismos.
ABSTRACT: The modifications that occur in phospholipids due to oxidative stress have a
very important role in atherogenesis, contributing for an increase of
predisposition for strokes and thrombosis. Oxidative stress is implied in
diabetes mellitus (DM) complications, neurodegenerative disease as Alzheimer
and aging.
The phosphatidyletanolamine (PE) is a cellular membranes essential
phospholipid, present in red blood cells and mitochondrias. The PE are also
present in low density lipoprotein (LDL). The structural modifications in PE,
particularly those from oxidative stress, have been related with atherosclerosis,
contributing in an indirect way for the occurrence of cerebral vascular accidents
or strokes and acute myocardial infarction. Some studies relate that PE, in
presence of high glucose concentrations, reacts with those, suffering a process
called glycation. In diabetic, the high plasmatic glucose concentration, can
lead to phosphatidyletanolamines glycation. Moreover, when PE is linked to
glucose, it seems to undergo oxidation process easily, that in the native form.
The oxidized PE, seems to lead to platelet aggregation, fact that can explain
high tax of atherosclerosis in diabetic and consequently the thrombus
formation.
Although the importance that these alterations have in PE, few studies have
been carried through, with an intention to deepen this subject. This work is
intended to create a synthesis model between glucose and phospholipid, to get
PE-Glucose product. This product will undergo oxidative stress and its
modification will be evaluated by mass spectrometry. Comparative studies with
the PE not modified will be carried through, using the same experimental
conditions.
The study of these oxidation products from glucose and aminophospholipids,
will contribute for a better understanding of the beginning and progression of
some pathologies, as DM. The new compounds and modified structures that
we will identify and characterize can be used in the future as markers of these
mechanisms
The second gamma-H2AX assay inter-comparison exercise carried out in the framework of the European biodosimetry network (RENEB)
Purpose: Within the EU RENEB project, seven laboratories have taken part in training and harmonisation activities to strengthen triage gamma H2AX-based radiation exposure assessment. This has culminated in a second triage biodosimetry exercise.
Materials and methods: Whole blood and separated lymphocyte samples were homogenously irradiated with 60Co gamma rays at 0.5, 2.5 (blind samples), 0 and 2 Gy (reference samples). Following post-exposure incubations of 4 and 24 h, 16 samples were shipped on ice packs to each partner. The samples were stained and scored for gamma-H2AX foci, using manual and/or automated fluorescence microscope scoring strategies. Dose estimates were obtained and used to assign triage categories to the samples.
Results: Average dose estimates across all the laboratories correlated well with true doses. The most accurate assignment of triage category was achieved by manual scoring of the 4-h blood and lymphocyte samples. Only three samples out of a total of 46 were miscategorized in a way that could have adversely effected the clinical management of a radiation casualty.
Conclusions: This inter-comparison exercise has demonstrated that following a recent acute radiation exposure, the gamma-H2AX assay could be a useful triage tool that can be successfully applied across a network of laboratories

Development of a functional dark chocolate with baobab pulp
In recent years, cocoa and dark chocolate have attracted the interest of consumers not only for their sensory characteristics but also for their nutritional properties and positive impact on health. The baobab is a fruit of African origin with a sour and slightly sweet flavour, widely consumed by local communities due to its unique nutritional features. The aim of this work was to evaluate the impact of the concentration of baobab flour in the development of functional dark chocolate, including physical, chemical, nutritional and sensory evaluations. The results presented a positive correlation between the incorporation of baobab flour and the antioxidant activity (up to 2297 mmol TE/100 g), vitamin C content (up to 49.7 mg/100 g), calcium (up to 1052 mg/kg), potassium (up to 10,175 mg/kg), phosphorus (up to 795.9 mg/kg), chlorine (up to 235.4 mg/kg) and sulphur (up to 1158 mg/kg). The sensory evaluation of dark chocolate with 3% baobab presented the highest evaluation on the parameters “texture” and “overall flavour”, while the parameter “overall flavour” presented the lowest evaluation on chocolate with 9% baobab. No influence was observed on fatty acid profile, protein, fat and hardness
Nutritional Properties of Baobab Pulp from Different Angolan Origins
Publisher Copyright:
© 2022 by the authors. The baobab tree (Adansonia digitata L.) is found widely in the forests and savannas of sub-Saharan Africa. The baobab fruit has a sour and slightly sweet taste and is widely consumed by the natives, thus containing a high nutritional value and providing a source of income for rural people. This study aimed to compare the nutritional composition of baobab fruit pulp from different localities in the Namibe province (Angola). Twenty samples of baobab pulp were collected in markets of the four municipalities of Namibe. The results obtained showed that there is some geographic location dependence on nutritional and functional composition. The municipality of Camucuio showed samples with higher fibre content (56.62 g/100 g) and vitamin C (288.9 mg/100 g). Samples from the Virei municipality stood out for their antioxidant activity (1936 mmol TE/100 g), high K content (42.4 mg/g) and higher values of protein (2.42 g/100 g). The samples collected in the municipality of Bibala stood out for their high contents of carbohydrates (28.1 g/100 g), total phenolic compounds (972 mg GAE/100 g) and Ca (3.80 mg/g). Despite the differences in origin, the high nutritional value of baobab fruit has the potential to improve the diet of thousands of people in Africa qualitatively.publishersversionpublishe
High resolution melting : improvements in the genetic diagnosis of hypertrophic cardiomyopathy in a Portuguese cohort

heterogeneity. The elevated number of genes and mutations involved in HCM limits a gene-based diagnosis that should be considered of most importance for basic research and clinical medicine.
Methodology: In this report, we evaluated High Resolution Melting (HRM) robustness, regarding HCM genetic
testing, by means of analyzing 28 HCM-associated genes, including the most frequent 4 HCM-associated sarcomere genes, as well as 24 genes with lower reported HCM-phenotype association. We analyzed 80 Portuguese individuals with clinical phenotype of HCM allowing simultaneously a better characterization of this disease in the Portuguese population.
Results: HRM technology allowed us to identify 60 mutated alleles in 72 HCM patients: 49 missense mutations, 3 nonsense mutations, one 1-bp deletion, one 5-bp deletion, one in frame 3-bp deletion, one insertion/deletion, 3 splice mutations, one 5’UTR mutation in MYH7, MYBPC3, TNNT2, TNNI3, CSRP3, MYH6 and MYL2 genes. Significantly 22 are novel gene mutations.
Conclusions: HRM was proven to be a technique with high sensitivity and a low false positive ratio allowing a
rapid, innovative and low cost genotyping of HCM. In a short return, HRM as a gene scanning technique could be a cost-effective gene-based diagnosis for an accurate HCM genetic diagnosis and hopefully providing new insights into genotype/phenotype correlations.info:eu-repo/semantics/publishedVersio
Co-design of a marine protected area zoning and the lessons learned from it
Marine Protected Areas (MPAs) are a tool to safeguard marine natural systems,
yet their effectiveness depends on how well they are integrated into the existing
socioeconomic context. Stakeholder engagement in MPA design can
contribute to increasing integration. This study focuses on the co-design of
an MPA between researchers, public administration, the private sector, and
non-governmental organizations. The proposed MPA is in Portugal and
includes an area that is a hotspot for biodiversity and economic activities.
This is the first MPA proposal in mainland Portugal co-designed using a
participatory approach. This study highlights the steps of the zoning process
and synthesizes the eight main lessons learned, useful for other cases,
particularly for relatively small coastal MPAs with multiple socioeconomic
activities. Three zoning proposals were developed and discussed within the
participatory process. The proposals considered the best scientific and local
knowledge available and were defined using ecological, socioeconomic, and
shape-area guiding principles. In an iterative manner and following a
participatory approach, compromises with stakeholders were achieved, and a
final proposal, scientifically sound and socially accepted by most stakeholders,
was delivered to the government. The final zoning plan will achieve ambitious
conservation goals, including the largest fully protected area to be declared in
mainland Portugal, while minimizing the impacts on the existing economic activities and promoting its sustainability. This process resulted in valuable
lessons that may be applied elsewhere and guide future MPA implementation
or rezoning of existing ones. These transdisciplinary and participatory
processes can be time and resource-consuming but are vital for ensuring
MPA effectiveness.Fundação para a Ciência e Tecnologia - FCTinfo:eu-repo/semantics/publishedVersio
