    William John Howey was a land speculator who came to Florida in 1908 and devised a unique land sales and development program on nearly 60,000 acres of land in Lake County. Howey’s plan, based on the sale of undeveloped citrus land, led to the creation of Howey-in-the-Hills, a town he envisioned as the “City Inevitable.“ In addition to this visionary enterprise Howey ran as the Republican candidate for governor of Florida in 1928, and again in 1932

    The effects of symmetrical recombination site hixC on Hin recombinase function

    An artificial recombination site hixC composed of two identical half- sites that bind the Hin recombinase served as a better operator in vivo than the wild type site hixL (Hughes, K. T., Youderian, P., and Simon, M. I (1988) Genes & Dev. 2, 937-948). In vitro binding assays such as gel retardation assay and methylation protection assay demonstrated that Hin binds to hixC as tightly as it binds to hixL, even when the sites are located in negatively supercoiled plasmids. However, hixC served as a poor recombination site when it was subjected to the standard inversion assay in vitro. hixC showed a 16-fold slower inversion rate than the wild type. A series of biochemical assays designed to probe different stages of the Hin-mediated inversion reaction, demonstrated that Hin dimers bound to hixC have difficulty in forming paired hix site intermediates. KMnO4 and S1 nuclease assays detected an anomalous structure of the center of hixC only when the site was in negatively supercoiled plasmids. Mutational analysis in the central region of hixC and assays of paired hix site formation with topoisomers of the hixC substrate plasmid suggested that Hin is not able to pair hixC sites because of the presence of the anomalous structure in the center of the site. The structure does not behave like a DNA "cruciform" since Hin dimers still bind efficiently to the site. It is thought to consist of a short denatured "bubble" encompassing 2 base pairs. During the study of mutations in the center of hixC, it was found that Hin is not able to cleave DNA if a guanine residue is one of the two central nucleotides close to the cleavage site. Furthermore, Hin acts in a concerted fashion and cannot cleave any DNA strand if one of the four strands in the inversion intermediate is not cleavable

    Identity and integration of Russian speakers in the Baltic states: a framework for analysis

    Following a review of current scholarship on identity and integration patterns of Russian speakers in the Baltic states, this article proposes an analytical framework to help understand current trends. Rogers Brubaker's widely employed triadic nexus is expanded to demonstrate why a form of Russian-speaking identity has been emerging, but has failed to become fully consolidated, and why significant integration has occurred structurally but not identificationally. By enumerating the subfields of political, economic, and cultural ‘stances’ and ‘representations’ the model helps to understand the complicated integration processes of minority groups that possess complex relationships with ‘external homelands’, ‘nationalizing states’ and ‘international organizations’. Ultimately, it is argued that socio-economic factors largely reduce the capacity for a consolidated identity; political factors have a moderate tendency to reduce this capacity, whereas cultural factors generally increase the potential for a consolidated group identity

    Chromatin and oxygen sensing in the context of JmjC histone demethylases

    Responding appropriately to changes in oxygen availability is essential for multicellular organism survival. Molecularly, cells have evolved intricate gene expression programmes to handle this stressful condition. Although it is appreciated that gene expression is co-ordinated by changes in transcription and translation in hypoxia, much less is known about how chromatin changes allow for transcription to take place. The missing link between co-ordinating chromatin structure and the hypoxia-induced transcriptional programme could be in the form of a class of dioxygenases called JmjC (Jumonji C) enzymes, the majority of which are histone demethylases. In the present review, we will focus on the function of JmjC histone demethylases, and how these could act as oxygen sensors for chromatin in hypoxia. The current knowledge concerning the role of JmjC histone demethylases in the process of organism development and human disease will also be reviewed

    Radial Growth of Qilian Juniper on the Northeast Tibetan Plateau and Potential Climate Associations

    There is controversy regarding the limiting climatic factor for tree radial growth at the alpine treeline on the northeastern Tibetan Plateau. In this study, we collected 594 increment cores from 331 trees, grouped within four altitude belts spanning the range 3550 to 4020 m.a.s.l. on a single hillside. We have developed four equivalent ring-width chronologies and shown that there are no significant differences in their growth-climate responses during 1956 to 2011 or in their longer-term growth patterns during the period AD 1110–2011. The main climate influence on radial growth is shown to be precipitation variability. Missing ring analysis shows that tree radial growth at the uppermost treeline location is more sensitive to climate variation than that at other elevations, and poor tree radial growth is particularly linked to the occurrence of serious drought events. Hence water limitation, rather than temperature stress, plays the pivotal role in controlling the radial growth of Sabina przewalskii Kom. at the treeline in this region. This finding contradicts any generalisation that tree-ring chronologies from high-elevation treeline environments are mostly indicators of temperature changes

    Interventions for promoting smoking cessation during pregnancy.

    BACKGROUND: Smoking remains one of the few potentially preventable factors associated with low birthweight, preterm birth and perinatal death. OBJECTIVES: To assess the effects of smoking cessation programs implemented during pregnancy on the health of the fetus, infant, mother, and family. SEARCH STRATEGY: We searched the Cochrane Pregnancy and Childbirth Group trials register and the Cochrane Tobacco Addiction Group trials register (July 2003), MEDLINE (January 2002 to July 2003), EMBASE (January 2002 to July 2003), PsychLIT (January 2002 to July 2003), CINAHL (January 2002 to July 2003), and AUSTHEALTH (January 2002 to 2003). We contacted trial authors to locate additional unpublished data. We handsearched references of identified trials and recent obstetric journals. SELECTION CRITERIA: Randomised and quasi-randomised trials of smoking cessation programs implemented during pregnancy. DATA COLLECTION AND ANALYSIS: Four reviewers assessed trial quality and extracted data independently. MAIN RESULTS: This review included 64 trials. Fifty-one randomised controlled trials (20,931 women) and six cluster-randomised trials (over 7500 women) provided data on smoking cessation and/or perinatal outcomes. Despite substantial variation in the intensity of the intervention and the extent of reminders and reinforcement through pregnancy, there was an increase in the median intensity of both 'usual care' and interventions over time. There was a significant reduction in smoking in the intervention groups of the 48 trials included: (relative risk (RR) 0.94, 95% confidence interval (CI) 0.93 to 0.95), an absolute difference of six in 100 women continuing to smoke. The 36 trials with validated smoking cessation had a similar reduction (RR 0.94, 95% CI 0.92 to 0.95). Smoking cessation interventions reduced low birthweight (RR 0.81, 95% CI 0.70 to 0.94) and preterm birth (RR 0.84, 95% CI 0.72 to 0.98), and there was a 33 g (95% CI 11 g to 55 g) increase in mean birthweight. There were no statistically significant differences in very low birthweight, stillbirths, perinatal or neonatal mortality but these analyses had very limited power. One intervention strategy, rewards plus social support (two trials), resulted in a significantly greater smoking reduction than other strategies (RR 0.77, 95% CI 0.72 to 0.82). Five trials of smoking relapse prevention (over 800 women) showed no statistically significant reduction in relapse. REVIEWERS' CONCLUSIONS: Smoking cessation programs in pregnancy reduce the proportion of women who continue to smoke, and reduce low birthweight and preterm birth. The pooled trials have inadequate power to detect reductions in perinatal mortality or very low birthweight

    Electrocardiographic Deep Learning for Predicting Post-Procedural Mortality

    Background. Pre-operative risk assessments used in clinical practice are limited in their ability to identify risk for post-operative mortality. We hypothesize that electrocardiograms contain hidden risk markers that can help prognosticate post-operative mortality. Methods. In a derivation cohort of 45,969 pre-operative patients (age 59+- 19 years, 55 percent women), a deep learning algorithm was developed to leverage waveform signals from pre-operative ECGs to discriminate post-operative mortality. Model performance was assessed in a holdout internal test dataset and in two external hospital cohorts and compared with the Revised Cardiac Risk Index (RCRI) score. Results. In the derivation cohort, there were 1,452 deaths. The algorithm discriminates mortality with an AUC of 0.83 (95% CI 0.79-0.87) surpassing the discrimination of the RCRI score with an AUC of 0.67 (CI 0.61-0.72) in the held out test cohort. Patients determined to be high risk by the deep learning model's risk prediction had an unadjusted odds ratio (OR) of 8.83 (5.57-13.20) for post-operative mortality as compared to an unadjusted OR of 2.08 (CI 0.77-3.50) for post-operative mortality for RCRI greater than 2. The deep learning algorithm performed similarly for patients undergoing cardiac surgery with an AUC of 0.85 (CI 0.77-0.92), non-cardiac surgery with an AUC of 0.83 (0.79-0.88), and catherization or endoscopy suite procedures with an AUC of 0.76 (0.72-0.81). The algorithm similarly discriminated risk for mortality in two separate external validation cohorts from independent healthcare systems with AUCs of 0.79 (0.75-0.83) and 0.75 (0.74-0.76) respectively. Conclusion. The findings demonstrate how a novel deep learning algorithm, applied to pre-operative ECGs, can improve discrimination of post-operative mortality

    Is eastern Mongolia drying? A long-term perspective of a multidecadal trend

    Temperatures in semiarid Mongolia have rapidly risen over the past few decades, and increases in drought, urban development, mining, and agriculture have intensified demands on limited water resources. Understanding long-term streamflow variation is critical for Mongolia, particularly if alterations in streamflow are being considered and because of the potential negative impacts of drought on the animal agriculture sector. Here, we present a temporally and spatially improved streamflow reconstruction for the Kherlen River. We have added 11 new records in comparison with two in the original 2001 reconstruction. This new reconstruction extends from 1630 to 2007 and places the most recent droughts in a multicentennial perspective. We find that variations in streamflow have been much greater in the past than in the original study. There was higher variability in the mid to late 1700s, ranging from severe and extended drought conditions from 1723 to 1739 and again in 1768–1778 to two decadal length episodes of very wet conditions in the mid 1700s and late 1700s. Reduced amplitude is seen in the mid-1800s, and several pluvial events are reconstructed for the 1900s. Although recent droughts are severe and disturbing economic and ecological systems in Mongolia and it appears that eastern Mongolia is drying, the drying trend since the late 1900s might in fact be accentuated by a change from a particularly wet era in Mongolia. The recent drought might be a return to more characteristic hydroclimatic conditions of the past four centuries in Mongolia
