Aegilops tauschii Introgressions Improve Physio-Biochemical Traits and Metabolite Plasticity in Bread Wheat under Drought Stress

    Get PDF
With the aim of providing genetic materials for breeding drought-resistant wheat varieties, the physiological and metabolic plasticity of three drought-resistant wheat multiple synthetic derivative lines (MSDLs) and their backcross parent “Norin 61” (N61) were evaluated in response to drought stress. The results indicated that the D-genome introgressions from Aegilops tauschii into the MDSLs improved their drought-adaptive traits. Specifically, MNH5 and MSD345 showed higher photosynthesis rates and triose phosphate utilization than N61 under control conditions, resulting in greater accumulation of glucose and sucrose in the shoots. However, under drought stress, MNH5 and MSD345 had higher intrinsic water use efficiency than MSD53 and N61. The total antioxidant capacity and superoxide dismutase activity increased in all three MSDLs, whereas no significant changes were found in N61 in response to drought stress. Metabolome analysis identified six common drought-induced metabolites in all of the investigated genotypes. However, four metabolites (adenine, gamma aminobutyric acid, histidine, and putrescine) each specifically accumulated in an MSDL in response to drought stress, suggesting that these metabolites are important for MSDL drought resistance. In conclusion, MNH5 and MSD345 showed better adaptive responses to drought stress than MSD53 and N61, suggesting that these two MSDLs could be explored for breeding drought-resistant wheat lines

COCONUT LEAF WASTE PROCESSING WITH BIODRYING PROCESS AS RAW MATERIAL FOR BIOMASS PELLETS IN WASTE TO ENERGY

    Get PDF
Coconut leaf waste is a type of waste that contains a good enough calorific value to be used as raw material for biomass pellets. The minimum standard calorific value of biomass in Indonesia according to SNI 8675:2018 is 16.05 MJ/kg. Meanwhile, based on the research results, the calorific value of coconut leaf waste has a value of 13.34 MJ/kg. The way that can be applied to increase the calorific value is by biodrying. This study aimed to determine the effectiveness of the biodrying process in processing coconut leaf waste. This study use bioactivators in the form of tempe yeast, baker's yeast, and tapai yeast. Mass loss and moisture reduction can be achieved, respectively, 8.25-17.62% and 62.4-65.6%. The calorific value can reach the minimum standard after seven days of processing with a value of 17,995-18,623 MJ/kg

Avelumab, an Anti-Programmed Death-Ligand 1 Antibody, In Patients With Refractory Metastatic Urothelial Carcinoma: Results From a Multicenter, Phase Ib Study

    Get PDF
Purpose We assessed the safety and antitumor activity of avelumab, a fully human anti-programmed death-ligand 1 (PD-L1) IgG1 antibody, in patients with refractory metastatic urothelial carcinoma. Methods In this phase Ib, multicenter, expansion cohort, patients with urothelial carcinoma progressing after platinum-based chemotherapy and unselected for PD-L1 expression received avelumab 10 mg/kg intravenously every 2 weeks. The primary objectives were safety and tolerability. Secondary objectives included confirmed objective response rate (Response Evaluation Criteria in Solid Tumors [RECIST] version 1.1), progression-free survival, overall survival (OS), and PD-L1-associated clinical activity. PD-L1 positivity was defined as expression by immunohistochemistry on ≥ 5% of tumor cells. Results Forty-four patients were treated with avelumab and followed for a median of 16.5 months (interquartile range, 15.8 to 16.7 months). The data cutoff was March 19, 2016. The most frequent treatment-related adverse events of any grade were fatigue/asthenia (31.8%), infusion-related reaction (20.5%), and nausea (11.4%). Grades 3 to 4 treatment-related adverse events occurred in three patients (6.8%) and included asthenia, AST elevation, creatine phosphokinase elevation, and decreased appetite. The confirmed objective response rate by independent central review was 18.2% (95% CI, 8.2% to 32.7%; five complete responses and three partial responses). The median duration of response was not reached (95% CI, 12.1 weeks to not estimable), and responses were ongoing in six patients (75.0%), including four of five complete responses. Seven of eight responding patients had PD-L1-positive tumors. The median progression-free survival was 11.6 weeks (95% CI, 6.1 to 17.4 weeks); the median OS was 13.7 months (95% CI, 8.5 months to not estimable), with a 12-month OS rate of 54.3% (95% CI, 37.9% to 68.1%). Conclusion Avelumab was well tolerated and associated with durable responses and prolonged survival in patients with refractory metastatic UC

The Impact of Facilitated Quarantine on Mental Health Status of Non-Severe COVID-19 Patients

    Get PDF
It has been shown that quarantine is important to reduce the incidence and mortality during COVID-19 pandemic.1 However, quarantine has consequences towards mental health. From previous outbreaks, several negative psychological effects such as post-traumatic stress symptoms, confusion, and anger were observed.2 In Indonesia, several successful suicide cases of COVID-19 patients in quarantine facilities have been reported, and being quarantined in quarantine facilities has been suggested to be the main cause. Nevertheless, no study has confirmed whether being quarantined in quarantine facilities in fact, worsens the mental health status of COVID-19 patients

Iminosugar inhibitors of carbohydrate-active enzymes that underpin cereal grain germination and endosperm metabolism

Starch is a major energy store in plants. It provides most of the calories in the human diet and, as a bulk commodity, it is used across broad industry sectors. Starch synthesis and degradation are not fully understood, owing to challenging biochemistry at the liquid/solid interface and relatively limited knowledge about the nature and control of starch degradation in plants. Increased societal and commercial demand for enhanced yield and quality in starch crops requires a better understanding of starch metabolism as a whole. Here we review recent advances in understanding the roles of carbohydrate-active enzymes in starch degradation in cereal grains through complementary chemical and molecular genetics. These approaches have allowed us to start dissecting aspects of starch degradation and the interplay with cell-wall polysaccharide hydrolysis during germination. With a view to improving and diversifying the properties and uses of cereal grains, it is possible that starch degradation may be amenable to manipulation through genetic or chemical intervention at the level of cell wall metabolism, rather than simply in the starch degradation pathway per se

The Cardiac Atlas Project—an imaging database for computational modeling and statistical atlases of the heart

    Get PDF
Motivation: Integrative mathematical and statistical models of cardiac anatomy and physiology can play a vital role in understanding cardiac disease phenotype and planning therapeutic strategies. However, the accuracy and predictive power of such models is dependent upon the breadth and depth of noninvasive imaging datasets. The Cardiac Atlas Project (CAP) has established a large-scale database of cardiac imaging examinations and associated clinical data in order to develop a shareable, web-accessible, structural and functional atlas of the normal and pathological heart for clinical, research and educational purposes. A goal of CAP is to facilitate collaborative statistical analysis of regional heart shape and wall motion and characterize cardiac function among and within population groups
