    Evaluation of trace elements mobility from soils to sediments between the Iberian pyrite belt and the Atlantic Ocean

    An environmental study was conducted in 193 stream sediments and 355 soil samples collected in the lower section of Guadiana River Basin to evaluate the trace elements transfer from one compartment into another. The objective was to evaluate the dispersion of Pb, Cu, Zn and other chemical elements resulting from upstream mines into the lower N-S sector of the Guadiana River Basin to the Atlantic Ocean. The area partly includes the Iberian Pyrite Belt, a known volcanogenic massive sulphide (VMS) province with important mining activity history. Median concentrations of the elements Cu, Pb, Zn, Al, As, Ba, Br, Ca, Mg, Mn and Na are generally higher in the studied sediments than in soils. Soils are normally acidic and mobility of elements is in these conditions higher. When the relations between upstream soils and downstream sediments were established, median values were higher in the upstream soils only for elements such as Co, Ni, K, Pb, Mn and Ti, probably the less mobile, and the Cu, Zn, Al, As, Ba, Br, Ca, Cr, Fe, Mg and Na have highest concentrations in the downstream sediments. Lead was considered the less mobile element and Zn the highly mobile of the base metals in the mining area of the lower Guadiana River. Dispersion of the metals, considering the studied soil and sediment samples is partially restricted to the mining areas or downstream sediments but close to the mine sources, where ore tailings and acid waters occur. Near the mouth, concentrations of As, Cu, Pb and Zn increase in relation with mining and other pollution sources. Interaction with salt water forces chemical elements precipitation from water and subsequent increase of their concentration in sediments. This may be the reason for the increase of those chemical elements concentrations in the sediment

    New integrative computational approaches unveil the Saccharomyces cerevisiae pheno-metabolomic fermentative profile and allow strain selection for winemaking

    During must fermentation by Saccharomyces cerevisiae strains thousands of volatile aroma compounds are formed. The objective of the present work was to adapt computational approaches to analyze pheno-metabolomic diversity of a S. cerevisiae strain collection with different origins. Phenotypic and genetic characterization together with individual must fermentations were performed, and metabolites relevant to aromatic profiles were determined. Experimental results were projected onto a common coordinates system, revealing 17 statistical-relevant multi-dimensional modules, combining sets of most-correlated features of noteworthy biological importance. The present method allowed, as a breakthrough, to combine genetic, phenotypic and metabolomic data, which has not been possible so far due to difficulties in comparing different types of data. Therefore, the proposed computational approach revealed as successful to shed light into the holistic characterization of S. cerevisiae pheno-metabolome in must fermentative conditions.     Antioxidant and Antiproliferative Activities of Heated Sterilized Pepsin Hydrolysate Derived from Half-Fin Anchovy (Setipinna taty)

    In this paper we studied the antioxidant and antiproliferative activities of the heated pepsin hydrolysate from a marine fish half-fin anchovy (HAHp-H). Furthermore, we compared the chemical profiles including the amino acid composition, the browning intensity, the IR and UV-visible spectra, and the molecular weight distribution between the half-fin anchovy pepsin hydrolysate (HAHp) and HAHp-H. Results showed that heat sterilization on HAHp improved the 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical-scavenging activity and reducing power. In addition, the antiproliferative activities were all increased for HAHp-H on DU-145 human prostate cancer cell line, 1299 human lung cancer cell line and 109 human esophagus cancer cell line. The contents of free amino acid and reducing sugar of HAHp-H were decreased (P < 0.05). However, hydrophobic amino acid residues and the browning intensity of HAHp-H were increased. FT-IR spectroscopy indicated that amide I and amide III bands of HAHp-H were slightly modified, whereas band intensity of amide II was reduced dramatically. Thermal sterilization resulted in the increased fractions of HAHp-H with molecular weight of 3000–5000 Da and below 500 Da.     A normal prion protein (PrPc) is converted to a protease-resistant isoform by an apparent self-propagating activity in transmissible spongiform encephalopathy, a neurodegenerative disease. The cDNA encoding open reading frame (ORF) of the bovine prion protein gene (Prnp) was cloned from Korean cattle by PCR, and was transfected into Chinese hamster ovary (CHO-K1) cells using lipofectamine. The gene expression of the cloned cDNA was confirmed by RT-PCR and Western blotting with the monoclonal antibody, 6H4. Cellular changes in the transfected CHO-K1 cells were investigated using parameters such as MTT, lactate dehydrogenase (LDH), and superoxide dismutase (SOD) activities, as well as nitric oxide (NO) production, and an apoptosis assay. In the MTT and LDH assays, the bovine PrnP-transfectant showed a lower proliferation rate than the wild-type (p < 0.05). Production of NO, after LPS or ConA stimulation, was not detected in either transfectants or CHO-K1 cells. In SOD assay under ConA stimulation, the SOD activity of transfectants was 10 times higher than that of CHO-K1 cells at 6 h after treatment (p < 0.05). The genomic DNA of both the transfectants and control cells began to be fragmented at 6 h after treatment with cyclohexamide. Caspase-3 activity was reduced by transfection with the bovine Prnp (p < 0.05). Conclusively, the viability of transfectants expressing exogenous bovine Prnp was decreased while the capacities for cellular protection against antioxidative stress and apoptosis were increased

    Evolution of the resonance and incommensurate spin fluctuations in superconducting YBa2_2Cu3_3O6+x_{6+x}

    Polarized and unpolarized neutron triple-axis spectrometry was used to study the dynamical magnetic susceptibility χ(q,ω)\chi^{\prime\prime}({\bf q},\omega) as a function of energy (ω\hbar\omega) and wave vector (q{\bf q}) in a wide temperature range for the bilayer superconductor YBa2_2Cu3_3O6+x_{6+x} with oxygen concentrations, xx, of 0.45, 0.5, 0.6, 0.7, 0.8, 0.93, and 0.95. The most prominent features in the magnetic spectra include a spin gap in the superconducting state, a pseudogap in the normal state, the much-discussed resonance, and incommensurate spin fluctuations below the resonance. We establish the doping dependence of the spin gap in the superconducting state, the resonance energy, and the incommensurability of the spin fluctuations. We discuss in detail the procedure used for separating the magnetic scattering from phonon and other spurious effects. In the comparison of our experimental results with various microscopic theoretical models, particular emphasis was made to address the similarities and differences in the spin fluctuations of YBa2_2Cu3_3O6+x_{6+x} and La2x_{2-x}Srx_xCuO4_4.Comment: 23 pages with 30 figures, Phys.     Deciphering the contribution of biofilm to the pathogenesis of peritoneal dialysis infections: characterization and microbial behaviour on dialysis fluids

    Infections are major complications in peritoneal dialysis (PD) with a multifactorial etiology that comprises patient, microbial and dialytic factors. This study aimed at investigating the contribution of microbial biofilms on PD catheters to recalcitrant infections and their interplay with PD related-factors. A prospective observational study was performed on 47 patients attending Centro Hospitalar of Porto and Vila Nova de Gaia/Espinho to whom the catheter was removed due to infectious (n = 16) and non-infectious causes (n = 31). Microbial density on the catheter was assessed by culture methods and the isolated microorganisms identified by matrix-assisted laser desorption/ionization time-of-flight intact cell mass spectrometry. The effect of conventional and three biocompatible PD solutions on 16 Coagulase Negative Staphylococci (CNS) and 10 Pseudomonas aeruginosa strains planktonic growth and biofilm formation was evaluated. Cultures were positive in 87.5% of the catheters removed due infectious and 90.3% removed due to non-infectious causes. However, microbial yields were higher on the cuffs of catheters removed due to infection vs. non-infection. Staphylococci (CNS and Staphylococcus aureus) and P. aeruginosa were the predominant species: 32% and 20% in the infection and 43.3% and 22.7% in the non-infection group, respectively. In general, PD solutions had a detrimental effect on planktonic CNS and P. aeruginosa strains growth. All strains formed biofilms in the presence of PD solutions. The solutions had a more detrimental effect on P. aeruginosa than CNS strains. No major differences were observed between conventional and biocompatible solutions, although in icodextrin solution biofilm biomass was lower than in bicarbonate/lactate solution. Overall, we show that microbial biofilm is universal in PD catheters with the subclinical menace of Staphylococci and P. aeruginosa. Cuffs colonization may significantly contribute to infection. PD solutions differentially impact microbial species. Os relatos sobre as dificuldades encontradas com o processo de ensino e aprendizagem dessa disciplina, assim como sobre as diferentes iniciativas propostas para superá-las, são cada vez mais frequentes. Neste artigo, um projeto voltado para o apoio ao aprendizado de conceitos de lógica e linguagem de programação é apresentado juntamente com os resultados de uma avaliação realizada sobre o ele. O projeto defende a aplicação da técnica de dojo de programação como um meio de reforçar o ensino de programação e mostra, por meio da experimentação prática desta ideia, que os resultados são positivos
