    Die Entwicklung der Hypophyse ist ein komplexer Prozess, an dem viele Transkriptionsfaktoren beteiligt sind. Bedeutende Funktionen haben die Bone Morphogenetic Proteins 2, 4 und 7. Ziel der Arbeit war es, an Patienten mit angeborener kombinierter Hypophyseninsuffizienz diese 3 Gene auf Mutationen zu untersuchen. Dabei wurden die Exons, Exon-Intron-Übergänge sowie die 3`- und 5`-untranslatierten Regionen sequenziert. Neu identifizierte Varianten wurden anschließend in einer gesunden Kontrollkohorte untersucht, um pathogenetisch nicht relevante Genvarianten auszuschließen. Um die funktionelle Relevanz der Varianten beurteilen zu können, wurden phylogenetische Analysen und Untersuchungen mittels Programmen zum Einfluss auf die Proteinstruktur und -funktion durchgeführt. Neben bekannten Varianten, konnten wir neue Varianten mit und ohne Aminosäureaustausch identifizieren. Eine neue Mutation im kodierenden Bereich von BMP4 erschien dabei besonders relevant. Wir konnten sie nur bei einem Patienten nachweisen, der zusätzlich zur kombinierten Hypophyseninsuffizienz Skelettveränderungen aufwies.     La importancia de la personalidad del traductor en los itinerarios de especialidad y en la selección de personal

Curs acadèmic: 2016/2017A pesar de que a simple vista no lo pueda parecer, la personalidad de los traductores influye mucho más de lo que se cree en el proceso traductor y en la toma de decisiones, como han puesto de manifiesto estudios empíricos. Para analizar cuáles son las diferencias individuales en la personalidad de los traductores en la elección del itinerario de especialidad, se ha llevado a cabo un estudio con los estudiantes de los cuatro itinerarios de traducción de la Universidad Jaume I, a fin de ver si podemos identificar los rasgos típicos de los estudiantes de cada itinerario. Además, dada la gran importancia de la personalidad en las tareas de traducción, también se ha entrevistado a responsables de tres agencias de traducción para ver si la personalidad de los traductores es un factor relevante a la hora de reclutar personal. Se ha comprobado así que sí hay algunos rasgos comunes en la personalidad de los estudiantes de cada itinerario y que realmente esta cuestión no es un factor relevante en las agencias de traducción, sino que se valora, sobre todo, el trabajo diario y la calidad de las pruebas de traducción

    Testing the aerobic stability of silage-based mixed rations using a multiple indicator approach

    Total Mixed Rations (TMR) including silages are commonly fed to cattle in many parts of the world, but they tend to deteriorate on exposure to air, especially in the warm season. The aim of the study was to develop an easy to implement protocol in order to compare the aerobic stability of different feed mixtures for lactating dairy cattle and to test the potential of TMR stabilizing products to delay spoilage. The experimental conditions were standardized to 25 °C ambient, and the sample dry matter was adjusted to 400 g kg-1 to challenge shelf life. Temperature rise in an insulated vessel, which is frequently used when testing silages, was shown to be one promising indicator of spoilage activity. Furthermore, determination of pH, scores for visual occurrence of yeasts and moulds and condensation from 0–4 were successfully applied at the end of the 72 h period. A dosage of 4.5 l propionic acid t-1 proved suitable as a positive control when compared to 1.5 and 3.0 l t-1. The commercial products tested at the recommended dose had a similar efficacy. The stabilizing effect depended principally on the original hygienic condition of the ration. Visual evaluation plus the recording of pH, in addition to continuous temperature measurement facilitated this appraisal. Early signs of spoilage, in particular, can only be assessed by visual appraisal. Thus, an evaluation of a combination of indicators of aerobic stability is recommended

    Learning How to Ask - Does a one-day training increase trauma inquiry in routine substance use disorder practice? Results of a cluster-randomized controlled trial

    Aims To examine the effectiveness of a one-day skills training program for increasing trauma inquiry in routine substance use disorder treatment. Design Cluster-randomized two-armed controlled trial, with 12 substance use disorder (SUD) organizations operating 25 counseling centers, randomly assigned to training in trauma inquiry (13 counseling centers of 8 SUD organizations) or no training (12 counseling centers of 4 SUD organizations). Setting SUD counseling centers in Northern Germany. Cases N = 5204 SUD counseling services. Intervention The professionals assigned to the intervention group received a one-day training in trauma inquiry plus a 1.5-hour refresher session 3 months later. Professionals in the control group received no training. Measures Over a 12-month period, professionals documented for each counseling service whether they asked the client about four traumatic events: physical abuse, emotional abuse, sexual abuse and neglect. Analysis Primary outcomes were rates of asking about physical abuse, sexual abuse, emotional abuse and neglect in the 6 months after training. These were compared across conditions, while adjusting for baseline probabilities in the 6 months before the intervention, using mixed-effects logistic regression. Findings In the 6 months after training, the rate of asking about physical abuse was 18% higher in the SUD counseling services of trained professionals, relative to services of untrained professionals (OR = 1.18, 95% CI = [1.01–1.37, p = .035]). No effect was found for asking about sexual abuse, emotional abuse and neglect. Conclusion A one-day training program in trauma inquiry, combined with a brief refresher session, was effective in increasing inquiries about physical abuse in routine counseling practice. The training was ineffective in increasing inquiries about sexual abuse, emotional abuse and neglect. The effectiveness of a one-day training of trauma inquiry might be increased by a longer training, or by combining it with additional elements, such as ongoing supervision

    ROCK1/2 signaling contributes to corticosteroid-refractory acute graft-versus-host disease

    Patients with corticosteroid-refractory acute graft-versus-host disease (aGVHD) have a low one-year survival rate. Identification and validation of novel targetable kinases in patients who experience corticosteroid-refractory-aGVHD may help improve outcomes. Kinase-specific proteomics of leukocytes from patients with corticosteroid-refractory-GVHD identified rho kinase type 1 (ROCK1) as the most significantly upregulated kinase. ROCK1/2 inhibition improved survival and histological GVHD severity in mice and was synergistic with JAK1/2 inhibition, without compromising graft-versus-leukemia-effects. ROCK1/2-inhibition in macrophages or dendritic cells prior to transfer reduced GVHD severity. Mechanistically, ROCK1/2 inhibition or ROCK1 knockdown interfered with CD80, CD86, MHC-II expression and IL-6, IL-1β, iNOS and TNF production in myeloid cells. This was accompanied by impaired T cell activation by dendritic cells and inhibition of cytoskeletal rearrangements, thereby reducing macrophage and DC migration. NF-κB signaling was reduced in myeloid cells following ROCK1/2 inhibition. In conclusion, ROCK1/2 inhibition interferes with immune activation at multiple levels and reduces acute GVHD while maintaining GVL-effects, including in corticosteroid-refractory settings

    Type D patients report poorer health status prior to and after cardiac rehabilitation compared to non-type D patients

    Background: Type D personality is an emerging risk factor in coronary artery disease (CAD). Cardiac rehabilitation (CR) improves outcomes, but little is known about the effects of CR on Type D patients. Purpose: We examined (1) variability in Type D caseness following CR, (2) Type D as a determinant of health status, and (3) the clinical relevance of Type D as a determinant of health status compared to cardiac history. Methods: CAD patients (n = 368) participating in CR completed the Type D Scale, the Short-Form Health Survey 36 pre- and post-CR, and the Hospital Anxiety and Depression Scale pre-CR, to assess health status and depressive and anxious symptomatology, respectively. Results: The prevalence of Type D decreased from 26.6% to 20.7% (p = 0.012) following CR, but Type D caseness remained stable in 81% of patients. Health status significantly improved following CR [F(1,359) = 17.48, p < 0.001], adjusting for demographic and clinical factors and anxious and depressive symptoms. Type D patients reported poorer health status [F(1,359) = 10.40, p = 0.001], with the effect of Type D being stable over time [F(1,359) = 0.49, p = 0.48]. Patients with a cardiac history benefited less from CR [F(1,359) = 5.76, p = 0.02]. The influence of Type D on health status was larger compared to that for cardiac history, as indicated by Cohen's effect size index. Conclusions: Type D patients reported poorer health status compared to non-Type D patients pre- and post-CR.     Mineralogical and geochemical analysis of Fe-phases in drill-cores from the Triassic Stuttgart Formation at Ketzin CO₂ storage site before CO₂ arrival

    Reactive iron (Fe) oxides and sheet silicate-bound Fe in reservoir rocks may affect the subsurface storage of CO2 through several processes by changing the capacity to buffer the acidification by CO2 and the permeability of the reservoir rock: (1) the reduction of three-valent Fe in anoxic environments can lead to an increase in pH, (2) under sulphidic conditions, Fe may drive sulphur cycling and lead to the formation of pyrite, and (3) the leaching of Fe from sheet silicates may affect silicate diagenesis. In order to evaluate the importance of Fe-reduction on the CO2 reservoir, we analysed the Fe geochemistry in drill-cores from the Triassic Stuttgart Formation (Schilfsandstein) recovered from the monitoring well at the CO2 test injection site near Ketzin, Germany. The reservoir rock is a porous, poorly to moderately cohesive fluvial sandstone containing up to 2–4 wt% reactive Fe. Based on a sequential extraction, most Fe falls into the dithionite-extractable Fe-fraction and Fe bound to sheet silicates, whereby some Fe in the dithionite-extractable Fe-fraction may have been leached from illite and smectite. Illite and smectite were detected in core samples by X-ray diffraction and confirmed as the main Fe-containing mineral phases by X-ray absorption spectroscopy. Chlorite is also present, but likely does not contribute much to the high amount of Fe in the silicate-bound fraction. The organic carbon content of the reservoir rock is extremely low (<0.3 wt%), thus likely limiting microbial Fe-reduction or sulphate reduction despite relatively high concentrations of reactive Fe-mineral phases in the reservoir rock and sulphate in the reservoir fluid. Both processes could, however, be fuelled by organic matter that is mobilized by the flow of supercritical CO2 or introduced with the drilling fluid. Over long time periods, a potential way of liberating additional reactive Fe could occur through weathering of silicates due to acidification by CO2

    Validity of Type D personality in Iceland: association with disease severity and risk markers in cardiac patients

    Type D personality has been associated with poor prognosis in cardiac patients. This study investigated the validity of the Type D construct in Iceland and its association with disease severity and health-related risk markers in cardiac patients. A sample of 1,452 cardiac patients completed the Type D scale (DS14), and a subgroup of 161 patients completed measurements for the five-factor model of personality, emotional control, anxiety, depression, stress and lifestyle factors. The Icelandic DS14 had good psychometric properties and its construct validity was confirmed. Prevalence of Type D was 26–29%, and assessment of Type D personality was not confounded by severity of underlying coronary artery disease. Regarding risk markers, Type D patients reported more psychopharmacological medication use and smoking, but frequency of previous mental problems was similar across groups. Type D is a valid personality construct in Iceland, and is associated with health-related risk markers, but not cardiac disease severity
