    Chemical Optimization of Selective Pseudomonas aeruginosa LasB Elastase Inhibitors and Their Impact on LasB-Mediated Activation of IL-1β in Cellular and Animal Infection Models

    LasB elastase is a broad-spectrum exoprotease and a key virulence factor of Pseudomonas aeruginosa, a major pathogen causing lung damage and inflammation in acute and chronic respiratory infections. Here, we describe the chemical optimization of specific LasB inhibitors with druglike properties and investigate their impact in cellular and animal models of P. aeruginosa infection. Competitive inhibition of LasB was demonstrated through structural and kinetic studies. In vitro LasB inhibition was confirmed with respect to several host target proteins, namely, elastin, IgG, and pro-IL-1 beta. Furthermore, inhibition of LasBmediated IL-1 beta activation was demonstrated in macrophage and mouse lung infection models. In mice, intravenous administration of inhibitors also resulted in reduced bacterial numbers at 24 h. These highly potent, selective, and soluble LasB inhibitors constitute valuable tools to study the proinflammatory impact of LasB in P. aeruginosa infections and, most importantly, show clear potential for the clinical development of a novel therapy for life-threatening respiratory infections caused by this opportunistic pathogen

    DNA methylation in childhood asthma : an epigenome-wide meta-analysis

    Background DNA methylation profiles associated with childhood asthma might provide novel insights into disease pathogenesis. We did an epigenome-wide association study to assess methylation profiles associated with childhood asthma. Methods We did a large-scale epigenome-wide association study (EWAS) within the Mechanisms of the Development of ALLergy (MeDALL) project. We examined epigenome-wide methylation using Illumina Infinium Human Methylation450 BeadChips (450K) in whole blood in 207 children with asthma and 610 controls at age 4-5 years, and 185 children with asthma and 546 controls at age 8 years using a cross-sectional case-control design. After identification of differentially methylated CpG sites in the discovery analysis, we did a validation study in children (4-16 years; 247 cases and 2949 controls) from six additional European cohorts and meta-analysed the results. We next investigated whether replicated CpG sites in cord blood predict later asthma in 1316 children. We subsequently investigated cell-type-specific methylation of the identified CpG sites in eosinophils and respiratory epithelial cells and their related gene-expression signatures. We studied cell-type specificity of the asthma association of the replicated CpG sites in 455 respiratory epithelial cell samples, collected by nasal brushing of 16-year-old children as well as in DNA isolated from blood eosinophils (16 with asthma, eight controls [age 2-56 years]) and compared this with whole-blood DNA samples of 74 individuals with asthma and 93 controls (age 1-79 years). Whole-blood transcriptional profiles associated with replicated CpG sites were annotated using RNA-seq data of subsets of peripheral blood mononuclear cells sorted by fluorescence-activated cell sorting. These CpG sites and their associated transcriptional profiles indicate activation of eosinophils and cytotoxic T cells in childhood asthma.     Rhinitis associated with asthma is distinct from rhinitis alone: TARIA‐MeDALL hypothesis

    Asthma, rhinitis, and atopic dermatitis (AD) are interrelated clinical phenotypes that partly overlap in the human interactome. The concept of “one-airway-one-disease,” coined over 20 years ago, is a simplistic approach of the links between upper- and lower-airway allergic diseases. With new data, it is time to reassess the concept. This article reviews (i) the clinical observations that led to Allergic Rhinitis and its Impact on Asthma (ARIA), (ii) new insights into polysensitization and multimorbidity, (iii) advances in mHealth for novel phenotype definitions, (iv) confirmation in canonical epidemiologic studies, (v) genomic findings, (vi) treatment approaches, and (vii) novel concepts on the onset of rhinitis and multimorbidity. One recent concept, bringing together upper- and lower-airway allergic diseases with skin, gut, and neuropsychiatric multimorbidities, is the “Epithelial Barrier Hypothesis.” This review determined that the “one-airway-one-disease” concept does not always hold true and that several phenotypes of disease can be defined.     Incidence of SARS-CoV-2 in people with cystic fibrosis in Europe between February and June 2020

    Background Viral infections can cause significant morbidity in cystic fibrosis (CF). The current Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) pandemic could therefore have a serious impact on the health of people with CF (pwCF). Methods We used the 38-country European Cystic Fibrosis Society Patient Registry (ECFSPR) to collect case data about pwCF and SARS-CoV-2 infection. Results Up to 30 June 2020, 16 countries reported 130 SARS-CoV-2 cases in people with CF, yielding an incidence of 2.70/1000 pwCF. Incidence was higher in lung-transplanted patients (n=23) versus non-transplanted patients (n=107) (8.43 versus 2.36 cases/1000). Incidence was higher in pwCF versus the age-matched general population in the age groups <15, 15-24, and 25-49 years (p<0.001), with similar trends for pwCF with and without lung transplant. Compared to the general population, pwCF (regardless of transplantation status) had significantly higher rates of admission to hospital for all age groups with available data, and higher rates of intensive care, although not statistically significant. Most pwCF recovered (96.2%), however 5 died, of whom 3 were lung transplant recipients. The case fatality rate for pwCF (3.85%, 95% CI: 1.26-8.75) was non-significantly lower than that of the general population (7.46%; p=0.133). Conclusions SARS-CoV-2 infection can result in severe illness and death for pwCF, even for younger patients and especially for lung transplant recipients. PwCF should continue to shield from infection and should be prioritized for vaccination

    International audienceSports and physical education takes part in secondary school pupils’ sports cultural integration. Leaning on sports and physical activities, this school subject conveys institutional values linked to these social practices. Effort, respect and health have an important place in it. Secondary school pupils can themselves invest other values in “sport”, linked to pleasure and immediacy which are characteristic of young people’s culture. The aim of this article is to discover the convergences and divergences, in terms of values, between this school sports culture and the representations of sports activities in secondary school pupils’ minds. The scale of educational values suggested by O. Reboul allows to classify and organize into a hierarchy the results of the empirical survey : they qualify the assumptions about the characteristics of a secondary school sports culture and its opposition to institutional values. They also show that the school form of sports and physical activities remains very problematic.L’éducation physique et sportive participe à l’acculturation sportive du lycéen. S’appuyant sur les activités physiques et sportives, cette matière scolaire transmet les valeurs institutionnelles liées à ces pratiques sociales. L’effort, le respect, la santé y ont bonne place. Le lycéen peut, quant à lui, investir dans le « sport » d’autres valeurs, liées au plaisir et à l’immédiateté ; valeurs caractéristiques de la culture jeune. L’objet de cet article est de découvrir les convergences et les divergences, en terme de valeurs, entre cette culture sportive scolaire et les représentations des activités sportives existant chez les lycéens. L’échelle des valeurs en éducation proposée par O. Reboul permet de sérier et de hiérarchiser les résultats issus de l’enquête empirique : ils nuancent les hypothèses sur les caractéristiques d’une culture sportive lycéenne et son opposition aux valeurs de l’Institution. Ils montrent aussi que la forme scolaire de l’EPS reste très problématique

    International audienceEn décembre 1962, le bureau politique du Parti Communiste Français, dans un souci de dynamiser son influence auprès des jeunes, regroupe ses différentes éditions « jeunes » existantes en une seule. Dès Mai 1963, Nous, les garçons et les filles, devient le mensuel des jeunes communistes. A la même époque, émerge une presse jeunesse dominée par les revues commerciales, telle Salut les Copains. Dans un contexte favorable à l’épanouissement de la société de consommation et un développement de la culture de masse, les valeurs tendent à se transformer. La culture jeune participe de cette évolution. Dès lors, N.G.F., tout au long des 69 numéros de son existence, oscille entre adhésion à cette culture jeune et un positionnement empreint de revendications politiques et d’espérances sociales. Le sport, thème récurrent de la revue, permet notamment à celle-ci de se départir de ces concurrentes en revendiquant la transmission de valeurs éloignées de la culture jeune.Ce travail, exploitant la totalité des numéros de Mai 1963 à Juillet 1969, montre que N.G.F. est dépositaire d’un sport de compétition valorisant l’effort, la persévérance et toutes autres valeurs inscrites dans le mythe olympique. Le travail récompensé du champion amateur, pour une jeunesse ouvrière en quête d’émancipation, permet une identification aux héros du stade. Ces derniers sont le plus souvent rapportés à leurs origines modestes et à l’exigence de leurs entraînements. Sur ces premiers éléments, il est possible de dire que N.G.F. défend une vision traditionnelle du sport en osmose avec la culture sportive des aînés. Pour autant, la défense d’un sport de compétition pour les femmes représente une avancée sur le plan des modèles de genre associés au sport
