Daño cerebral por estrés oxidativo y alteraciones en el comportamiento después de la abstinencia alcohólica en ratas Lewis
Los mecanismos por los cuales ocurre la toxicidad por el alcohol no son totalmente conocidos. Un criterio de diagnóstico de la dependencia del alcohol es la aparición de síntomas de abstinencia. Durante la abstinencia alcohólica ocurre incremento de la formación de especies reactivas de oxígeno. Objetivos: Este estudio se propuso determinar la relación entre los marcadores indicadores del daño cerebral inducido por estrés oxidativo y cambios conductuales después de 2 semanas de abstinencia alcohólica en ratas. Método: Ratas Lewis machos(250-300g) se distribuyeron en dos grupos; el Grupo I recibió agua durante el experimento y en el Grupo II se les fue introduciendo gradualmente el consumo de etanol en soluciones de 10, 20, 30, y 40 % (56 días). Se realizó la prueba de tolerancia farmacológica y se determinaron variables bioquímicas relacionadas con el estado redox. Se retiró la administración de etanol de forma abrupta. Después de dos semanas de abstinencia las ratas fueron sujetas a varios ensayos conductuales. Se obtuvo tejido de cerebro y se midieron marcadores de estrés oxidativo. Resultados: Dos semanas después de la abstinencia alcohólica hubo una disminución de la actividad de las enzimas Superóxido Dismutasa y Catalasa así como de la concentración de Glutatión reducido, indicando una disminución de las defensas antioxidantes en tejido cerebral. Se constató un incremento en las concentraciones de Malonildialdehido y nitritos/nitratos a nivel cerebral. Ocurrieron alteraciones en el grupo II, relacionadas con la memoria y capacidad de aprendizaje, actividad locomotora y ansiedad, correlacionada con las variables redox. Conclusiones: El estrés oxidativo es un inductor del daño cerebral y está relacionado con afectaciones ocurridas después de dos semanas de abstinencia alcohólica, en nuestras condiciones experimentales.Palabras Claves: Abstinencia alcohólica, estrés oxidativo, cambios conductuales
Overexpression of the X-Linked Inhibitor of Apoptosis Protein (XIAP) in Neurons Improves Cell Survival and the Functional Outcome after Traumatic Spinal Cord Injury
Mechanical trauma to the spinal cord causes extensive neuronal death, contributing to the loss of sensory-motor and autonomic functions below the injury location. Apoptosis affects neurons after spinal cord injury (SCI) and is associated with increased caspase activity. Cleavage of X-linked inhibitor of apoptosis protein (XIAP) after SCI may contribute to this rise in caspase activity. Accordingly, we have shown that the elevation of XIAP resulted in increased neuronal survival after SCI and improved functional recovery. Therefore, we hypothesise that neuronal overexpression of XIAP can be neuroprotective after SCI with improved functional recovery. In line with this, studies of a transgenic mice with overexpression of XIAP in neurons revealed that higher levels of XIAP after spinal cord trauma favours neuronal survival, tissue preservation, and motor recovery after the spinal cord trauma. Using human SH-SY5Y cells overexpressing XIAP, we further showed that XIAP reduced caspase activity and apoptotic cell death after pro-apoptotic stimuli. In conclusion, this study shows that the levels of XIAP expression are an important factor for the outcome of spinal cord trauma and identifies XIAP as an important therapeutic target for alleviating the deleterious effects of SCI

Clustering of Dietary Patterns and Lifestyles Among Spanish Children in the EsNuPI Study
Dietary patterns (DPs) are known to be tied to lifestyle behaviors. Understanding DPs and their relationships with lifestyle factors can help to prevent children from engaging in unhealthy dietary practices. We aimed to describe DPs in Spanish children aged 1 to <10 years and to examine their associations with sociodemographic and lifestyle variables. The consumption of toddler and young children milk formulas, enriched and fortified milk within the Spanish pediatric population is increasing, and there is a lack of evidence whether the consumption of this type of milk is causing an impact on nutrient intakes and if they are helping to reach the nutrient recommendations. Within the Nutritional Study in the Spanish Pediatric Population (EsNuPI), we considered two study cohorts and three different age groups in three year-intervals in each of them. The study cohort included 740 children in a representative sample of the urban non-vegan Spanish population and 772 children in a convenience cohort of adapted milk consumers (AMS) (including follow-on formula, toddler’s milk, growing up milk, and fortified and enriched milks) who provided information about sociodemographics, lifestyle, and dietary habits; a food frequency questionnaire was used for the latter. Principal component analysis was performed to identify DPs from 18 food groups. Food groups and sociodemographic/lifestyle variables were combined through a hierarchical cluster algorithm. Three DPs predominated in every age group and study sample: a palatable energy-dense food dietary pattern, and two Mediterranean-like DPs. However, children from the AMS showed a predominant dietary pattern markedly related to the Mediterranean diet, with high consumption of cereals, fruits and vegetables, as well as milk and dairy products. The age of children and certain lifestyle factors, namely level of physical activity, parental education, and household income, correlated closely with the dietary clusters. Galaxy clusters and groups in the ALHAMBRA Survey
We present a catalogue of 348 galaxy clusters and groups with
selected in the 2.78 ALHAMBRA Survey. The high precision of our
photometric redshifts, close to , and the wide spread of the seven
ALHAMBRA pointings ensure that this catalogue has better mass sensitivity and
is less affected by cosmic variance than comparable samples.
The detection has been carried out with the Bayesian Cluster Finder (BCF),
whose performance has been checked in ALHAMBRA-like light-cone mock catalogues.
Great care has been taken to ensure that the observable properties of the mocks
photometry accurately correspond to those of real catalogues. From our
simulations, we expect to detect galaxy clusters and groups with both
completeness and purity down to dark matter halo masses of
for . Cluster redshifts are
expected to be recovered with precision for . We also expect
to measure cluster masses with
precision down to , masses which are
smaller than those reached by similar work.
We have compared these detections with previous optical, spectroscopic and
X-rays work, finding an excellent agreement with the rates reported from the
simulations. We have also explored the overall properties of these detections
such as the presence of a colour-magnitude relation, the evolution of the
photometric blue fraction and the clustering of these sources in the different
ALHAMBRA fields. Despite the small numbers, we observe tentative evidence that,
for a fixed stellar mass, the environment is playing a crucial role at lower

Staging Parkinson’s disease according to the MNCD classification correlates with caregiver burden
Malaltia de Parkinson; Cuidador; Símptomes no motorsParkinson's disease; Caregiver; Non-motor symptomsEnfermedad de Parkinson; Cuidador; Síntomas no motoresBackground and objective: Recently, we demonstrated that staging Parkinson's disease (PD) with a novel simple classification called MNCD, based on four axes (motor, non-motor, cognition, and dependency) and five stages, correlated with disease severity and patients’ quality of life. Here, we analyzed the correlation of MNCD staging with PD caregiver's status. Patients and methods: Data from the baseline visit of PD patients and their principal caregiver recruited from 35 centers in Spain from the COPPADIS cohort from January 2016 to November 2017 were used to apply the MNCD total score (from 0 to 12) and MNCD stages (from 1 to 5) in this cross-sectional analysis. Caregivers completed the Zarit Caregiver Burden Inventory (ZCBI), Caregiver Strain Index (CSI), Beck Depression Inventory-II (BDI-II), PQ-10, and EUROHIS-QOL 8-item index (EUROHIS-QOL8). Results: Two hundred and twenty-four PD patients (63 ± 9.6 years old; 61.2% males) and their caregivers (58.5 ± 12.1 years old; 67.9% females) were included. The frequency of MNCD stages was 1, 7.6%; 2, 58.9%; 3, 31.3%; and 4–5, 2.2%. A more advanced MNCD stage was associated with a higher score on the ZCBI (p < .0001) and CSI (p < .0001), and a lower score on the PQ-10 (p = .001), but no significant differences were observed in the BDI-II (p = .310) and EUROHIS-QOL8 (p = .133). The NER-related gene GTF2H5 predicts survival in high-grade serous ovarian cancer patients
We aimed to evaluate the prognostic and predictive value of the nucleotide excision repair-related gene GTF2H5, which is localized at the 6q24.2-26 deletion previously reported by our group to predict longer survival of high-grade serous ovarian cancer patients. In order to test if protein levels of GTF2H5 are associated with patients' outcome, we performed GTF2H5 immunohistochemical staining in 139 high-grade serous ovarian carcinomas included in tissue microarrays. Upon stratification of cases into high- and low-GTF2H5 staining categories (> and ≤ median staining, respectively) Kaplan-Meier and log-rank test were used to estimate patients' survival and assess statistical differences. We also evaluated the association of GTF2H5 with survival at the transcriptional level by using the on-line Kaplan-Meier plotter tool, which includes gene expression and survival data of 855 high-grade serous ovarian cancer patients from 13 different datasets. Finally, we determined whether stable short hairpin RNA-mediated GTF2H5 downregulation modulates cisplatin sensitivity in the SKOV3 and COV504 cell lines by using cytotoxicity assays. Low expression of GTF2H5 was associated with longer 5-year survival of patients at the protein (hazard ratio [HR], 0.52; 95% CI, 0.29 to 0.93; p=0.024) and transcriptional level (HR, 0.80; 95% CI, 0.65 to 0.97; p=0.023) in high-grade serous ovarian cancer patients. We confirmed the association with 5-year overall survival (HR, 0.55; 95% CI, 0.38 to 0.78; p=0.0007) and also found an association with progression-free survival (HR, 0.72; 95% CI, 0.54 to 0.96; p=0.026) in a homogenous group of 388 high-stage (stages III-IV using the International Federation of Gynecology and Obstetrics staging system), optimally debulked high-grade serous ovarian cancer patients. GTF2H5- silencing induced a decrease of the half maximal inhibitory concentration upon cisplatin treatment in GTF2H5 -silenced ovarian cancer cells. Low levels of GTF2H5 are associated with enhanced prognosis in high-grade serous ovarian cancer patients and may contribute to cisplatin sensitization
Hereditary Leiomyomatosis and Renal Cell Cancer Syndrome in Spain: Clinical and Genetic Characterization
Simple Summary Hereditary leiomyomatosis and renal cell cancer (HLRCC) syndrome is a very rare hereditary disorder characterized by cutaneous leiomyomas (CLMs), uterine leiomyomas (ULMs), renal cysts (RCys) and renal cell cancer (RCC), with no data on its prevalence worldwide. No genotype-phenotype associations have been described. The aim of our study was to describe the genotypic and phenotypic features of the largest series of patients with HLRCC from Spain reported to date. Of 27 FH germline pathogenic variants, 12 were not previously reported in databases. Patients with missense pathogenic variants showed higher frequencies of CLMs, ULMs and RCys, than those with loss-of-function variants. The frequency of RCCs (10.9%) was lower than those reported in the previously published series. Hereditary leiomyomatosis and renal cell cancer syndrome (HLRCC) is a very rare hereditary disorder characterized by cutaneous leiomyomas (CLMs), uterine leiomyomas (ULMs), renal cysts (RCys) and renal cell cancers (RCCs). We aimed to describe the genetics, clinical features and potential genotype-phenotype associations in the largest cohort of fumarate hydratase enzyme mutation carriers known from Spain using a multicentre, retrospective study of individuals with a genetic or clinical diagnosis of HLRCC. We collected clinical information from medical records, analysed genetic variants and looked for genotype-phenotype associations. Analyses were performed using R 3.6.0. software. We included 197 individuals: 74 index cases and 123 relatives. CLMs were diagnosed in 65% of patients, ULMs in 90% of women, RCys in 37% and RCC in 10.9%. Twenty-seven different pathogenic variants were detected, 12 (44%) of them not reported previously. Patients with missense pathogenic variants showed higher frequencies of CLMs, ULMs and RCys, than those with loss-of-function variants (p = 0.0380, p = 0.0015 and p = 0.024, respectively). This is the first report of patients with HLRCC from Spain. The frequency of RCCs was lower than those reported in the previously published series. Evaluation of Poly(N-Ethyl Pyrrolidine Methacrylamide) (EPA) and Derivatives as Polymeric Vehicles for miRNA Delivery to Neural Cells
MicroRNAs (miRNAs) are endogenous, short RNA oligonucleotides that regulate the expression of hundreds of proteins to control cells' function in physiological and pathological conditions. miRNA therapeutics are highly specific, reducing the toxicity associated with off-target effects, and require low doses to achieve therapeutic effects. Despite their potential, applying miRNA-based therapies is limited by difficulties in delivery due to their poor stability, fast clearance, poor efficiency, and off-target effects. To overcome these challenges, polymeric vehicles have attracted a lot of attention due to their ease of production with low costs, large payload, safety profiles, and minimal induction of the immune response. Poly(N-ethyl pyrrolidine methacrylamide) (EPA) copolymers have shown optimal DNA transfection efficiencies in fibroblasts. The present study aims to evaluate the potential of EPA polymers as miRNA carriers for neural cell lines and primary neuron cultures when they are copolymerized with different compounds. To achieve this aim, we synthesized and characterized different copolymers and evaluated their miRNA condensation ability, size, charge, cytotoxicity, cell binding and internalization ability, and endosomal escape capacity. Finally, we evaluated their miRNA transfection capability and efficacy in Neuro-2a cells and rat primary hippocampal neurons. I. MUFFIT: A multi-filter fitting code for stellar population diagnostics
Numerical methods and codes.-- et al.[Aims]: We present MUFFIT, a new generic code optimized to retrieve the main stellar population parameters of galaxies in photometric multi-filter surveys, and check its reliability and feasibility with real galaxy data from the ALHAMBRA survey. [Methods]: Making use of an error-weighted X2-test, we compare the multi-filter fluxes of galaxies with the synthetic photometry of mixtures of two single stellar populations at different redshifts and extinctions, to provide the most likely range of stellar population parameters (mainly ages and metallicities), extinctions, redshifts, and stellar masses. To improve the diagnostic reliability, MUFFIT identifies and removes from the analysis those bands that are significantly affected by emission lines. The final parameters and their uncertainties are derived by a Monte Carlo method, using the individual photometric uncertainties in each band. Finally, we discuss the accuracies, degeneracies, and reliability of MUFFIT using both simulated and real galaxies from ALHAMBRA, comparing with results from the literature. [Results]: MUFFIT is a precise and reliable code to derive stellar population parameters of galaxies in ALHAMBRA. Using the results from photometric-redshift codes as input, MUFFIT improves the photometric-redshift accuracy by ∼10-20%. MUFFIT also detects nebular emissions in galaxies, providing physical information about their strengths. The stellar masses derived from MUFFIT show excellent agreement with the COSMOS and SDSS values. In addition, the retrieved age-metallicity locus for a sample of z ≤ 0.22 early-type galaxies in ALHAMBRA at different stellar mass bins are in very good agreement with the ones from SDSS spectroscopic diagnostics. Moreover, a one-to-one comparison between the redshifts, ages, metallicities, and stellar masses derived spectroscopically for SDSS and by MUFFIT for ALHAMBRA reveals good qualitative agreements in all the parameters, hence reinforcing the strengths of multi-filter galaxy data and optimized analysis techniques, like MUFFIT, to conduct reliable stellar population studies.L.A.D.G. acknowledges support from the "Caja Rural de Teruel" for developing this research.
