    Seeking to enhance the bioenergy of municipal sludge: Effect of alkali pre-treatment and soluble organic matter supplementation

    The aim of this research is to enhance the mesophilic anaerobic digestion of municipal sludge from Cadiz-San Fernando (Spain) wastewater treatment plant at 20 days hydraulic retention time (HRT). Two different strategies were tested to improve the process: co-digestion with the addition of soluble organic matter (1% v/v); and alkali sludge pre-treatment (NaOH) prior to co-digestion with glycerine (1% v/v). Methane production (MP) was substantially enhanced (from 0.36 ± 0.09 L CH4 l/d to 0.85 ± 0.16 L CH4 l/d), as was specific methane production (SMP) (from 0.20 ± 0.05 L CH4/g VS to 0.49 ± 0.09 L CH4/g VS) when glycerine was added. The addition of glycerine does not seem to affect sludge stability, the quality of the effluent in terms of pH and organic matter content, i.e. volatile fatty acids (VFA), soluble organic matter and total volatile solid, or process stability (VFA/Alkalinity ratio < 0.4). Alkali pre-treatment prior to co-digestion resulted in a high increase in soluble organic loading rates (more than 20%) and acidification yield (more than 50%). At 20 days HRT, however, it led to overload of the system and total destabilization of the mesophilic anaerobic co-digestion of sewage sludge and glycerine

    Mesophilic anaerobic co-digestion of sewage sludge with glycerine: Effect of solids retention time

    The main objective of this paper is to examine the effect of the increase in organic loading rates (OLRs), by reducing the solids retention time (SRT) from 20 d to 5 d, in single-phase mesophilic anaerobic co-digestion of sewage sludge with glycerine (1% v/v). It was experimentally confirmed that anaerobic co-digestion of these biowastes under steady-state conditions can achieve an 85 ± 5% reduction in volatile fatty acids (VFA) at SRTs of between 20 and 9 d, with a methane production yield of around 0.8 l CH4/l/d. Decreases in the SRT not only allow the sludge stability and biogas production to be maintained, but also lead to an increase in the waste that could be treated and lower operating costs. Therefore, mesophilic anaerobic co-digestion of sewage sludge and glycerin at a SRT lower than 20 d is possible and preferable due to being more economical and environmentally friendl

    <p>Abstract</p> <p>Background</p> <p>To date, eight assemblages of <it>Giardia lamblia </it>have been described, but only assemblages A and B are known to infect humans. Despite the fact that the genomic, biological, and clinical differences found between these two assemblages has raised the possibility that they may be considered different species, there is relatively limited information on their phenotypic differences. In the present study, we developed monoclonal antibodies against alpha-1 and beta giardin, two immunodominant proteins produced during <it>G. lamblia </it>infection, and studied their expression and localization in WB (assemblage A) and GS trophozoites (assemblage B).</p> <p>Results</p> <p>The polyclonal antibodies generated against WB trophozoites, particularly those recognizing intracellular proteins as well as the proteins present at the plasma membrane (variable-specific surface proteins), showed cross-reactivity with intracellular proteins in GS trophozoites. The use of monoclonal antibodies against beta giardin indicated ventral disc localization, particularly at the periphery in WB trophozoites. Interestingly, although beta giardin was also restricted to the ventral disc in GS trophozoites, the pattern of localization clearly differed in this assemblage. On the other hand, monoclonal antibodies against alpha-1 giardin showed plasma membrane localization in both assemblages with the bare area of GS trophozoites also being distinguished. Moreover, the same localization at the plasma membrane was observed in Portland-1 (Assemblage A) and in P15 (Assemblage E) trophozoites.</p> <p>Conclusions</p> <p>We found differences in localization of the beta giardin protein between assemblages A and B, but the same pattern of localization of alpha-1 giardin in strains from Assemblages A, B and E. These findings reinforce the need for more studies based on phenotypic characteristics in order to disclose how far one assemblage is from the other.</p

    Perturbaciones debidas a efluentes domésticos en poliquetos asociados a bancos de bivalvos de Brachidontes rodriguezii en Mar de Plata (Atlántico sudoccidental)

    Continuous and short-term disturbances are produced on intertidal Brachidontes rodriguezii mussel beds by the sewage outfall of Mar del Plata, SW Atlantic. The effects on the associated polychaetes living on and between mussels were investigated, in both the spatial and short-term temporal scales. Multivariate methods show that the polychaete fauna was influenced by tidal level, total amount of sediment accumulated between mussels, distance from the sewage outfall, organic carbon of interstitial sediments, and sewage volume before and after the summer season. Environmental variables in the water column fitted with a spatial and temporal organic gradient. Capitella &lsquo;capitata&rsquo; sp. showed typical opportunistic behaviour in relation to organic enrichment, as did Neanthes succinea, both being numerically dominant near the sewage discharge. Cirratulus sp., Caulleriella alata, and Boccardia polybranchia were common at intermediate distances from the outfall. Syllis prolixa, S. gracilis, and Protoariciella uncinata reached their highest abundance at intermediate stations and were characteristic of the control site. Close to the outfall mussel density and dominance is low, and opportunist polychaetes successfully colonize organically enriched interstitial sediments. At intermediate distances from the outfall sediments retained by mussels reach their maximum values, and density of polychaetes is also high. The control station is characterized by monolayered mussel beds and very low diversity of polychaetes. Opportunistic polychaetes increase in response to short-term increases of sewage discharge during the summer season (November-March).Perturbaciones cont&iacute;nuas y de corto per&iacute;odo de tiempo debidas a efluentes dom&eacute;sticos afectan a los bancos de los bivalvos Brachidontes rodriguezii en Mar del Plata, Atl&aacute;ntico sudoccidental. Se investigaron sus efectos sobre los poliquetos que viven asociados dentro y sobre los bancos, tanto en la escala espacial como la temporal (corto plazo). M&eacute;todos multivariados mostraron que los poliquetos estuvieron relacionados al nivel de marea, al total de sedimentos acumulados entre los bivalvos, a la distancia al efluente, al carbono org&aacute;nico total de los sedimentos y al volumen de descarga antes y despu&eacute;s del verano. Las variables ambientales de la columna de agua estuvieron en concordancia con un gradiente de enriquecimiento org&aacute;nico espacial y temporal. Capitella &lsquo;capitata&rsquo; sp. mostr&oacute; el t&iacute;pico comportamiento oportunista en relaci&oacute;n al enriquecimiento org&aacute;nico, al igual que Neanthes succinea, siendo ambas especies num&eacute;ricamente dominantes cerca del efluente.     Time scale ovarian maturation in Greenland halibut

    In this paper possible evidence of a prolonged ovarian development phase in Greenland halibut is presented. The reproductive cycle in this species has been originally described based on the assumption that this phase should last about one year. The results of several years of analysis showed that there is more than one year between the mean age of females at the onset of the ovarian development and the mean age of actually spawning females. Two possible interpretations of this fact are discussed: the ovarian development phase (vitellogenesis) could last more than one year, and individual spawning does not necessarily occur on an annual basis as a consequence, or the incidence of non-spawning females every year could be very high. Both possibilities have important implications for the species’ reproductive potential and stock dynamics of this valuable deep water resource

    Bloom dynamics and life cycle strategies of two toxic dinoflagellates in a coastal upwelling system (NW Iberian Peninsula)

    A study of Gymnodinium catenatum and Alexandrium minutum blooms on the Galician coast was conducted from 2005 to 2007 in order to increase knowledge of the mechanisms governing recurrent blooms of these species. Considerable differences in their bloom dynamics were observed. G. catenatum blooms occurred in autumn and winter, following the pattern previously reported in the literature: they began offshore and were advected to the Galician rias when a relaxation of the coastal upwelling occurred. On the other hand, A. minutum blooms developed inside embayments in spring and summer during the upwelling season and were associated with water stability and stratification. Both the vegetative population and the cyst distribution of A. minutum were related to less saline water from freshwater river outputs, which supports a saline-gradient relationship postulated herein for this species. Dinoflagellates may produce both long-term double-walled cysts (resting) and short-term pellicle cysts. Resting cyst deposition and distribution in sediments showed that seeding occurred during the blooms of both species. However, the relationship between the cyst distribution in the sediments in Baiona Bay and the intensity and occurrence of G. catenatum blooms, suggests that the latter are not directly related to resting cyst germination. Moreover, the results presented in the present study point to other difference between the two species, such as the detection of pellicle cysts only for A. minutum. Finally we discuss how the life cycle strategies of these two species may help to explain the different mechanisms of bloom formation reported herein.VersiĂłn del editor2,277

    The giardial VPS35 retromer subunit is necessary for multimeric complex assembly and interaction with the Vacuolar protein sorting receptor

    The retromer is a pentameric protein complex that mediates the retrograde transport of acid hydrolase receptors between endosomes and the trans-Golgi network and is conserved across all eukaryotes. Unlike other eukaryotes, the endomembrane system of Giardia trophozoite is simple and is composed only of the endoplasmic reticulum and peripheral vesicles (PVs), which may represent an ancient organellar system converging compartments such as early and late endosomes and lysosomes. Sorting and trafficking of membrane proteins and soluble hydrolases from the endoplasmic reticulum to the PVs have been described as specific and conserved but whether the giardial retromer participates in receptor recycling remains elusive. Homologs of the retromer Vacuolar Protein Sorting (Vps35p, Vps26p, and Vps29p) have been identified in this parasite. Cloning the GlVPS35 subunit and antisera production enabled the localization of this protein in the PVs as well as in the cytosol. Tagged expression of the subunits was used to demonstrate their association with membranes, and immunofluorescence confocal laser scanning revealed high degrees of colabeling between the retromer subunits and also with the endoplasmic reticulum and PV compartment markers. Protein-protein interaction data revealed interaction between the subunits of GlVPS35 and the cytosolic domain of the hydrolase receptor GlVps. Altogether our data provide original information on the molecular interactions that mediate assembly of the cargo-selective retromer subcomplex and its involvement in the recycling of the acid hydrolase receptor in this parasite.Fil: Miras, Silvana.
