    Continuous Monitoring of Information on Anode Current Distribution as Means of Improving the Process of Controlling and Forecasting Process Disturbances

    За последние десятилетия система АСУТП электролиза достигла больших возможностей по улучшению сбора и обработки сигналов, которые обеспечивают основу для регулирования питания, теплового баланса и магнитогидродинамической (МГД) стабильности. Несмотря на значительное развитие, постоянно растут потребности в датчиках контроля, которые могут распознавать и реагировать на возникающие изменения. C 2008 г. на электролизерах опытного участка РА-400, расположенного в опытно-промышленном корпусе электролиза ОАО «РУСАЛ Саяногорск», эксплуатируется система непрерывного мониторинга информации о распределении тока по анодам, интегрированная в АСУТП СААТ-2. На протяжении промышленных испытаний система мониторинга постоянно совершенствовалась в части достоверности информации, обеспечения работоспособности оборудования в условиях агрессивных сред, а также при выполнении технологических и ремонтных операций на электролизере. Полученная информация о распределении тока по анодам позволила разработать алгоритмы, прогнозирующие на ранней стадии отклонения в питании глиноземом электролизера с последующим перераспределением по точкам автоматической подачи глинозема (АПГ), прогнозирование и распознавание технологических нарушений на подошве анодов («конуса»)The aluminum reduction process control system in recent decades has given great opportunities for improving the collection and processing of signals that provide the basis for the adjustment of feeding, thermal balance and magnetohydrodynamic (MHD) stability.     Functional annotation of human long noncoding RNAs via molecular phenotyping

    Long noncoding RNAs (lncRNAs) constitute the majority of transcripts in the mammalian genomes, and yet, their functions remain largely unknown. As part of the FANTOM6 project, we systematically knocked down the expression of 285 lncRNAs in human dermal fibroblasts and quantified cellular growth, morphological changes, and transcriptomic responses using Capped Analysis of Gene Expression (CAGE). Antisense oligonucleotides targeting the same lncRNAs exhibited global concordance, and the molecular phenotype, measured by CAGE, recapitulated the observed cellular phenotypes while providing additional insights on the affected genes and pathways. Here, we disseminate the largest-todate lncRNA knockdown data set with molecular phenotyping (over 1000 CAGE deep-sequencing libraries) for further exploration and highlight functional roles for ZNF213-AS1 and lnc-KHDC3L-2.Peer reviewe

    Simultaneous observations of PKS 2155-304 with H.E.S.S., Fermi, RXTE and ATOM: spectral energy distributions and variability in a low state

    We report on the first simultaneous observations that cover the optical, X-ray, and high energy gamma-ray bands of the BL Lac object PKS 2155-304. The gamma-ray bands were observed for 11 days, between 25 August and 6 September 2008, jointly with the Fermi Gamma-ray Space Telescope and the H.E.S.S. atmospheric Cherenkov array, providing the first simultaneous MeV-TeV spectral energy distribution with the new generation of gamma-ray telescopes. The ATOM telescope and the RXTE and Swift observatories provided optical and X-ray coverage of the low-energy component over the same time period. The object was close to the lowest archival X-ray and Very High Energy state, whereas the optical flux was much higher. The light curves show relatively little (~30%$) variability overall when compared to past flaring episodes, but we find a clear optical/VHE correlation and evidence for a correlation of the X-rays with the high energy spectral index. Contrary to previous observations in the flaring state, we do not find any correlation between the X-ray and VHE components. Although synchrotron self-Compton models are often invoked to explain the SEDs of BL Lac objects, the most common versions of these models are at odds with the correlated variability we find in the various bands for PKS 2155-304.Comment: Accepted for publication in the Astrophysical Journa

    We studied functional effect of rs12722489 single nucleotide polymorphism located in the first intron of human IL2RA gene on transcriptional regulation. This polymorphism is associated with multiple autoimmune conditions (rheumatoid arthritis, multiple sclerosis, Crohn's disease, and ulcerative colitis). Analysis in silico suggested significant difference in the affinity of estrogen receptor (ER) binding site between alternative allelic variants, with stronger predicted affinity for the risk (G) allele. Electrophoretic mobility shift assay showed that purified human ERα bound only G variant of a 32-bp genomic sequence containing rs12722489. Chromatin immunoprecipitation demonstrated that endogenous human ERα interacted with rs12722489 genomic region in vivo and DNA pull-down assay confirmed differential allelic binding of amplified 189-bp genomic fragments containing rs12722489 with endogenous human ERα. In a luciferase reporter assay, a kilobase-long genomic segment containing G but not A allele of rs12722489 demonstrated enhancer properties in MT-2 cell line, an HTLV-1 transformed human cell line with a regulatory T cell phenotype

    Heteroplasmic Variants of Mitochondrial DNA in Atherosclerotic Lesions of Human Aortic Intima

    Mitochondrial dysfunction and oxidative stress are likely involved in atherogenesis. Since the mitochondrial genome variation can alter functional activity of cells, it is necessary to assess the presence in atherosclerotic lesions of mitochondrial DNA (mtDNA) heteroplasmic mutations known to be associated with different pathological processes and ageing. In this study, mtDNA heteroplasmy and copy number (mtCN) were evaluated in the autopsy-derived samples of aortic intima differing by the type of atherosclerotic lesions. To detect mtDNA heteroplasmic variants, next generation sequencing was used, and mtCN measurement was performed by qPCR. It was shown that mtDNA heteroplasmic mutations are characteristic for particular areas of intimal tissue; in 83 intimal samples 55 heteroplasmic variants were found; mean minor allele frequencies level accounted for 0.09, with 12% mean heteroplasmy level. The mtCN variance measured in adjacent areas of intima was high, but atherosclerotic lesions and unaffected intima did not differ significantly in mtCN values. Basing on the ratio of minor and major nucleotide mtDNA variants, we can conclude that there exists the increase in the number of heteroplasmic mtDNA variants, which corresponds to the extent of atherosclerotic morphologic phenotype
