    Deutsch mithilfe der Hauswirtschaftslehre lernen

    Abstract . In dieser Arbeit wird eine Lerneinheit entwickelt, die Deutsch und Hauswirtschaftslehre verbindet. Die Lerneinheit ist für A-Sprache-Lernende geeignet, die in der 8. oder 9. Klasse sind. In der Lerneinheit werden sowohl die typischen Gerichte Deutschlands, Österreichs und der Schweiz behandelt als auch Wortschatz nötig in der Küche. Die Verbindung der unterschiedlichen Fächer in der Schule wird im aktuellen Lernplan betont. Dafür sind fertige Materialien den Lehrpersonen hilfreich. Im Hintergrund der Lerneinheit steht die CLIL-Methode (Content and Language Integrated Learning), die in der Arbeit auch vorgestellt wird. Das Fremdsprachenlernen wird bei CLIL mit Inhalten aus anderen Fächern, in diesem Fall aus der Hauswirtschaftslehre, kombiniert. Die Lernsituationen sind authentisch und bieten Platz für das Sprechen, Hören, Lesen und Schreiben. Die Verknüpfung mit dem Alltag kann die Lernmotivation der Lernenden erhöhen. Die Lerneinheit zeigt, dass die Verbindung von einer Sprache und Hauswirtschaftslehre möglich ist. Beim Kochen, Backen und Essen kann Wortschatz geübt werden und gleichzeitig etwas aus der Kultur der Zielsprache kennengelernt werden. Zusammen zu arbeiten kann auch den Teamgeist in der Gruppe verbessern, was das Lernen unterstützt

    A 96-well format for a high-throughput baculovirus generation, fast titering and recombinant protein production in insect and mammalian cells

    <p>Abstract</p> <p>Background</p> <p>Baculovirus expression vector system (BEVS) has become a standard in recombinant protein production and virus-like particle preparation for numerous applications.</p> <p>Findings</p> <p>We describe here protocols which adapt baculovirus generation into 96-well format.</p> <p>Conclusion</p> <p>The established methodology allows simple baculovirus generation, fast virus titering within 18 h and efficient recombinant protein production in a high-throughput format. Furthermore, the produced baculovirus vectors are compatible with gene expression in vertebrate cells <it>in vitro </it>and <it>in vivo</it>.</p

    A multipurpose vector system for the screening of libraries in bacteria, insect and mammalian cells and expression in vivo

    We have constructed a novel tetra-promoter vector (pBVboostFG) system that enables screening of gene/cDNA libraries for functional genomic studies. The vector enables an all-in-one strategy for gene expression in mammalian, bacterial and insect cells and is also suitable for direct use in vivo. Virus preparation is based on an improved mini Tn7 transpositional system allowing easy and fast production of recombinant baculoviruses with high diversity and negligible background. Cloning of the desired DNA fragments or libraries is based on the recombination system of bacteriophage lambda. As an example of the utility of the vector, genes or cDNAs of 18 different proteins were cloned into pBVboostFG and expressed in different hosts. As a proof-of-principle of using the vector for library screening, a chromophoric Thr(65)-Tyr-Gly(67)-stretch of enhanced green fluorescent protein was destroyed and subsequently restored by novel PCR strategy and library screening. The pBVboostFG enables screening of genome-wide libraries, thus making it an efficient new platform technology for functional genomics

    Auxin Influx Carriers Control Vascular Patterning and Xylem Differentiation in Arabidopsis thaliana

    Auxin is an essential hormone for plant growth and development. Auxin influx carriers AUX1/LAX transport auxin into the cell, while auxin efflux carriers PIN pump it out of the cell. It is well established that efflux carriers play an important role in the shoot vascular patterning, yet the contribution of influx carriers to the shoot vasculature remains unknown. Here, we combined theoretical and experimental approaches to decipher the role of auxin influx carriers in the patterning and differentiation of vascular tissues in the Arabidopsis inflorescence stem. Our theoretical analysis predicts that influx carriers facilitate periodic patterning and modulate the periodicity of auxin maxima. In agreement, we observed fewer and more spaced vascular bundles in quadruple mutants plants of the auxin influx carriers aux1lax1lax2lax3. Furthermore, we show AUX1/LAX carriers promote xylem differentiation in both the shoot and the root tissues. Influx carriers increase cytoplasmic auxin signaling, and thereby differentiation. In addition to this cytoplasmic role of auxin, our computational simulations propose a role for extracellular auxin as an inhibitor of xylem differentiation. Altogether, our study shows that auxin influx carriers AUX1/LAX regulate vascular patterning and differentiation in plants.Peer reviewe

    Many particle physics models of matter admit solutions corresponding to stable or long-lived topological defects. In the context of standard cosmology it is then unavoidable that such defects will form during phase transitions in the very early Universe. Certain types of defects lead to disastrous consequences for cosmology, others may play a useful role, as possible seeds for the formation of structure in the Universe, or in mediating baryon number violating processes.     Ten-Year Mortality and Cardiovascular Morbidity in the Finnish Diabetes Prevention Study—Secondary Analysis of the Randomized Trial

    The Finnish Diabetes Prevention Study (DPS) was a randomized controlled trial, which showed that it is possible to prevent type 2 diabetes by lifestyle changes. The aim of the present study was to examine whether the lifestyle intervention had an effect on the ten-year mortality and cardiovascular morbidity in the DPS participants originally randomized either into an intervention or control group. Furthermore, we compared these results with a population-based cohort comprising individuals of varying glucose tolerance states.Middle-aged, overweight people with IGT (n = 522) were randomized into intensive intervention (including physical activity, weight reduction and dietary counseling), or control "mini-intervention" group. Median length of the intervention period was 4 years and the mean follow-up was 10.6 years. The population-based reference study cohort included 1881 individuals (1570 with normal glucose tolerance, 183 with IGT, 59 with screen-detected type 2 diabetes, 69 with previously known type 2 diabetes) with the mean follow-up of 13.8 years. Mortality and cardiovascular morbidity data were collected from the national Hospital Discharge Register and Causes of Death Register. Among the DPS participants who consented for register linkage (n = 505), total mortality (2.2 vs. 3.8 per 1000 person years, hazard ratio HR = 0.57, 95% CI 0.21-1.58) and cardiovascular morbidity (22.9 vs. 22.0 per 1000 person years, HR = 1.04, 95% CI 0.72-1.51) did not differ significantly between the intervention and control groups. Compared with the population-based cohort with impaired glucose tolerance, adjusted HRs were 0.21 (95% CI 0.09-0.52) and 0.39 (95% CI 0.20-0.79) for total mortality, and 0.89 (95% CI 0.62-1.27) and 0.87 (0.60-1.27) for cardiovascular morbidity in the intervention and control groups of the DPS, respectively. The risk of death in DPS combined cohort was markedly lower than in FINRISK IGT cohort (adjusted HR 0.30, 95% CI 0.17-0.54), but there was no significant difference in the risk of CVD (adjusted HR 0.88, 95% CI 0.64-1.21).Lifestyle intervention among persons with IGT did not decrease cardiovascular morbidity during the first 10 years of follow-up. However, the statistical power may not be sufficient to detect small differences between the intervention and control groups. Low total mortality among participants of the DPS compared with individuals with IGT in the general population could be ascribed to a lower cardiovascular risk profile at baseline and regular follow-up.ClinicalTrials.gov NCT00518167

    Diffusible repression of cytokinin signalling produces endodermal symmetry and passage cells.

    In vascular plants, the root endodermis surrounds the central vasculature as a protective sheath that is analogous to the polarized epithelium in animals, and contains ring-shaped Casparian strips that restrict diffusion. After an initial lag phase, individual endodermal cells suberize in an apparently random fashion to produce 'patchy' suberization that eventually generates a zone of continuous suberin deposition. Casparian strips and suberin lamellae affect paracellular and transcellular transport, respectively. Most angiosperms maintain some isolated cells in an unsuberized state as so-called 'passage cells', which have previously been suggested to enable uptake across an otherwise-impermeable endodermal barrier. Here we demonstrate that these passage cells are late emanations of a meristematic patterning process that reads out the underlying non-radial symmetry of the vasculature. This process is mediated by the non-cell-autonomous repression of cytokinin signalling in the root meristem, and leads to distinct phloem- and xylem-pole-associated endodermal cells. The latter cells can resist abscisic acid-dependent suberization to produce passage cells. Our data further demonstrate that, during meristematic patterning, xylem-pole-associated endodermal cells can dynamically alter passage-cell numbers in response to nutrient status, and that passage cells express transporters and locally affect the expression of transporters in adjacent cortical cells
