    High Trypanosoma spp. diversity is maintained by bats and triatomines in Espírito Santo state, Brazil

    The aim of this study was to reevaluate the ecology of an area in the Atlantic Forest, southeast Brazil, where Chagas disease (CD) has been found to occur. In a previous study, immediately after the occurrence of a CD case, we did not observe any sylvatic small mammals or dogs with Trypanosoma cruzi cruzi infections, but Triatoma vitticeps presented high T. c. cruzi infection rates. In this study, we investigated bats together with non-volant mammals, dogs, and triatomines to explore other possible T. c. cruzi reservoirs/hosts in the area. Seventy-three non-volant mammals and 186 bats were captured at three sites within the Guarapari municipality, Espírito Santo state. Rio da Prata and Amarelos sites exhibited greater richness in terms of non-volant mammals and bats species, respectively. The marsupial Metachirus nudicaudatus, the rodent Trinomys paratus, and the bats Artibeus lituratus and Carollia perspicillata were the most frequently captured species. As determined by positive hemocultures, only two non-volant mammals were found to be infected by Trypanosoma species: Monodelphis americana, which was infected by T. cascavelli, T. dionisii and Trypanosoma sp., and Callithrix geoffroyi, which was infected by T. minasense. Bats presented T. c. cruzi TcI and TcIII/V, T. c. marinkellei, T. dionisii, T. rangeli B and D, and Trypanosoma sp. infections. Seven dogs were infected with T. cruzi based only on serological exams. The triatomines T. vitticeps and Panstrongylus geniculatus were found to be infected by trypanosomes via microscopy. According to molecular characterization, T. vitticeps specimens were infected with T. c. cruzi TcI, TcII, TcIII/V, and TcIV, T. c. marinkellei and T. dionisii. We observed high trypanosome diversity in a small and fragmented region of the Atlantic Forest. This diversity was primarily maintained by bats and T. vitticeps. Our findings show that the host specificity of the Trypanosoma genus should be thoroughly reviewed. In addition, our data show that CD cases can occur without an enzootic cycle near residential areas

    A Graph Grammar to Transform a Dataflow Graph into a Multithread Graph and its Application in Task Scheduling

    The scheduling of tasks in a parallel program is an NP-complete problem, where scheduling tasks over multiple processing units requires an effective strategy to maximize the exploitation of the parallel hardware. Several studies focus on the scheduling of parallel programs described into DAGs (Directed Acyclic Graphs). However, this representation does not describe a multithreaded program suitably. This paper shows the structure and semantics of a DCG, an abstraction which describes a multithreaded program, and proposes standards to map structures found in DAGs into segments of a DCG. A graph grammar has been developed to perform the proposed transformation and case studies using DAGs found in the literature validate the transformation process. Besides the automatic translation and precise definition of the mapping, the use of a formal language also allowed the verification of the existence and uniqueness of the out coming model.

    Interactions between Nef and AIP1 proliferate multivesicular bodies and facilitate egress of HIV-1

    BACKGROUND: Nef is an accessory protein of primate lentiviruses, HIV-1, HIV-2 and SIV. Besides removing CD4 and MHC class I from the surface and activating cellular signaling cascades, Nef also binds GagPol during late stages of the viral replicative cycle. In this report, we investigated further the ability of Nef to facilitate the replication of HIV-1. RESULTS: To this end, first the release of new viral particles was much lower in the absence of Nef in a T cell line. Since the same results were obtained in the absence of the viral envelope using pseudo-typed viruses, this phenomenon was independent of CD4 and enhanced infectivity. Next, we found that Nef not only possesses a consensus motif for but also binds AIP1 in vitro and in vivo. AIP1 is the critical intermediate in the formation of multivesicular bodies (MVBs), which play an important role in the budding and release of viruses from infected cells. Indeed, Nef proliferated MVBs in cells, but only when its AIP1-binding site was intact. Finally, these functions of Nef were reproduced in primary macrophages, where the wild type but not mutant Nef proteins led to increased release of new viral particles from infected cells. CONCLUSION: We conclude that by binding GagPol and AIP1, Nef not only proliferates MVBs but also contributes to the egress of viral particles from infected cells

    Anticorpos IgG anti-metacestódeo de Taenia solium em amostras de soro de habitantes da região centro-oeste do Brasil

    A total of 354 serum samples from inhabitants who frequent the Clinical Laboratory in Catalão, Goiás, in the central-western region of Brazil, were collected from June to August, 2002. The samples were evaluated by indirect immunofluorescence antibody tests and an enzyme linked immunosorbent assay in order to detect anti-Taenia solium metacestode IgG antibodies. Reactive and inconclusive samples were tested by Western blotting (WB). Considering WB as a confirmation, the frequency of antibodies in the serum samples of the above population was 11.3% (CI 5.09 - 17.51). The immunodominant bands most frequently recognized in WB were 64-68 kDa (97.5%) and 47-52 kDa (80%). The percentage of seropositivity to cysticercosis was significantly higher for individuals residing in areas without sewage systems (p < 0.0001). In conclusion, the results indicate a probable endemic situation of cysticercosis in this population. These results reinforce the urgent need for control and prevention measures to be taken by the local public health services.Um total de 354 amostras de soro de habitantes que freqüentaram o Laboratório Clínico em Catalão, Goiás, na região centro-oeste do Brasil, foram colhidas no período de junho a agosto de 2002. As amostras foram avaliadas pelo teste de imunofluorescência indireta e enzyme-linked immunosorbent assay (ELISA) com o objetivo de detectar anticorpos IgG anti-metacestódeo de Taenia solium. As amostras reativas e inconclusivas foram testadas pelo Western blotting (WB). Considerando WB como reação confirmatória, a freqüência de anticorpos nas amostras de soro da população estudada foi 11,3% (IC: 5,09 - 17,51). As bandas imunodominantes mais frequentemente reconhecidas no WB foram 64-68 kDa (97,5%) e 47-52 kDa (80%). A porcentagem de soropositividade para cisticercose foi significativamente maior nos indivíduos que residiam em áreas sem sistema de esgoto (p < 0,0001). Concluiu-se que os resultados indicam uma provável situação de endemicidade para cisticercose nesta população, reforçando a urgente necessidade de medidas de controle e prevenção que devem ser implantadas pelo serviço de saúde pública local

    New understanding of a coastal erosion hotspot in a bimodal wave climate.

    In previous studies addressing coastal erosion hotspots, evidence is presented to demonstrate that geology and nearshore seabed features can exert control on local beach dynamics. In this study, an episodic coastal erosion hotspot at Thorpeness, UK is examined. Antecedent wave conditions and changes in incident wave characteristics resulting from interaction with the seabed geology and/or temporal changes in nearshore bathymetry are measured and modelled. Observations of changing sea bed features derived from radar measurements and results from numerical modelling demonstrate links between nearshore bathymetry and beach erosion events. The study examines also the conditions frequently experienced at the study site whereby persistent waves from a narrow sector reduce beach levels and modify the nearshore bathymetry in such a way that the impacts from storms from an opposing direction are larger than would normally be the case

    The discovery of new drugs, especially when many samples have to be analyzed in the minimum of time, demand the improvement or development of new analytical methods. Various techniques may be employed for this purpose. In this context, this chapter gathers the collection of paper and represents the review of past work on spectrophotometric technique coupled to a continuous flow system to determine low concentrations of several chemical species in different kinds of pharmaceutical and biological samples. A short historical background of the flow-injection analysis technique and a brief discussion of the basic principles and potential are presented. Part of this chapter is devoted to describing the sample preparation techniques, principles, and figures of merit of analytical methods. Representative applications of flow-injection spectrophotometry to pharmaceutical and biomedical analysis are also described

    Prevalence of cutaneous neoplasms in dogs from the metropolitan area of Porto Alegre, RS, Brazil : 1,017 cases (2002-2007)

    O objetivo deste trabalho foi realizar um estudo retrospectivo sobre neoplasias cutâneas diagnosticadas em cães. A avaliação foi realizada pela análise dos arquivos diagnósticos do Setor de Patologia Veterinária (SPV) da Universidade Federal do Rio Grande do Sul (UFRGS), Brasil, considerando-se um intervalo de seis anos (2002 a 2007). Neste intervalo, um total de 1.869 (37,3%) amostras de pele canina foram obtidas de 5.016 amostras variadas de tecidos de cães encaminhadas ao SPV. Dentre as amostras cutâneas, 1.002 pertenciam a cães diagnosticados com um tipo de neoplasia cutânea e 15 animais apresentaram mais de uma neoplasia de pele, totalizando 1.017 (20,3%) amostras. Os resultados revelaram que 50,5% (514/1017) das neoplasias cutâneas apresentaram origem mesenquimal, 45,1% (459/1017) para epitelial e 3,9% (40/1017) para melanocítica. Mastocitoma foi o tipo neoplásico cutâneo mais frequente, diagnosticado em 228 casos (22,4%), seguido por carcinoma de células escamosas (7,5%), lipoma (7,3%), adenoma de glândula perianal (7,1%) e tricoblastoma (5,8%). Cocker Spaniel, Boxer, Poodle e Pastor Alemão foram as raças mais representadas em diversos neoplasmas. Os dados obtidos, comparados aos estudos prévios, ressaltam as variáveis raças, idade e sexo, relacionadas a alguns tumores cutâneos e salientam a importância e prevalência dos diferentes tipos de neoplasia cutânea em cães. __________________________________________________________________________________________ ABSTRACTThe aim of this study was to perform a retrospective study of cutaneous neoplasms diagnosed in dogs. The evaluation was established by analyzing the diagnostic files at the Veterinary Pathology Sector, UFRGS, Brazil, over a 6-year period (2002 to 2007). During this period a total of 1869 (37.3%) skin samples were obtained from 5016 different tissue samples of dogs submitted for examination. Among the referred skin samples, 1002 were from dogs with the diagnosis of cutaneous neoplasia and 15 dogs exhibited more than one type of skin tumor, what amounted to a total of 1017 (20.3%) cutaneous tumor samples. Results confirmed 50.5% (514/1017), 45.1% (459/1017), and 3.9% (40/1017) of respectively mesenquimal, epithelial, and melanocytic origin. Mast cell tumor was the most frequent neoplasia, diagnosed in 228 cases (22.4%), and was followed by squamous cell carcinoma (7.5%), lipoma (7.3%), perianal gland adenoma (7.1%), and trichoblastoma (5.8%). Purebred dogs such as Cocker Spaniel, Boxer, Poodle and German Sheepdog were the most representative breeds affected by various neoplasms. The data obtained, compared to data from previous studies, emphasize the variables breed, age and sex related to some skin tumors, and reinforce the importance and prevalence of different types of skin tumors in dogs

    Type 2 diabetes mellitus and the prevalence of apical periodontitis and endodontic treatment in an adult Brazilian population

    Introduction This cross-sectional study evaluated the prevalence of apical periodontitis (AP) and endodontic treatment in type 2 diabetic individuals as compared with nondiabetics from an adult Brazilian population. Methods Full-mouth radiographs from 30 type 2 diabetic and 60 age- and sex-matched nondiabetic individuals were examined, and the presence of AP lesions in untreated and root canal-treated teeth was recorded. The number of teeth and the prevalence of root canal treatment were also evaluated. Results AP was significantly more present in teeth from diabetic individuals (98/652, 15%) than in nondiabetic controls (162/1,368, 12%) (P = .05). A separate analysis of untreated and treated teeth revealed that significance was mostly because of the prevalence of AP in untreated teeth, which was 10% in diabetics and 7% in nondiabetics (P = .03). No significant difference between diabetics and nondiabetics was observed for the other parameters under study, including the prevalence of AP in root canal–treated teeth, the number of teeth in the oral cavity, the number of treated teeth per individual, the number of individuals with at least 1 AP lesion or 1 root canal treatment, and the number of teeth with AP per individual (P > .05). Conclusions AP was significantly more prevalent in untreated teeth from type 2 diabetics. This suggests that diabetes may serve as a disease modifier of AP in the sense that individuals with diabetes can be more prone to develop primary disease. However, findings do not confirm that diabetes may influence the response to root canal treatment because treated teeth had no increased prevalence of AP when compared with controls

    Dental approach of orofacial pain in head and neck cancer patients

    Orofacial pain (OFP) is an undesirable sensation frequently associated with head and neck cancer (HNC) and its treatment. OFP negatively impacts the quality of life of oncological patients. The approach to OFP diagnosis and management can differ if the patient visits a dentist or physician. The aim of this study was to present a case series of HNC with OFP managed by a dentist team and to discuss its role in the management of OFP. We recruited twenty-two adult patients with OFP and previous diagnosis of HNC referred to an academic dental clinic from 2015 to 2017. Nociceptive was more frequent than mixed and neuropathic pain, however 54,4% of the cases showed a neurological component. All types of pain were managed by dentist through removal of pain?s cause and appropriated local and systemic treatment. The intensity of pain was reduced in 86,3% of patients, and 45,4% of them reported absence of pain at the end of treatment. Dentist?s assessment plays a distinct and crucial role in the diagnosis and management of OFP in HNC patients throughout the oncological treatment

    Perfil clínico de neonatos internados em uma Unidade de Tratamento Intensivo Neonatal / Título abreviado: Perfil clínico de neonatos internados em uma UTIN

    Objetivo: Descrever o perfil de recém-nascidos internados em uma Unidade de Tratamento Intensivo Neonatal (UTIN) de um hospital público no interior de Minas Gerais. Métodos: Trata-se de um estudo transversal inédito realizado em uma cidade mineira na região do Vale do Jequitinhonha com neonatos retidos na UTIN e suas mães. Período de realização do estudo de outubro de 2016 a dezembro de 2018. Resultados: A amostra foi composta por 44 neonatos, com distribuição similar em relação ao sexo (46% do sexo feminino x 48% masculino). A maioria nasceu pré-termo (n=37; 84,1%) e com peso adequado para idade gestacional (n=35; 78,7%), grande parte, de parto normal (n=26; 59,1%), com valor de Apgar superior a 7 no 5º minuto de vida (n=36; 81,8%), e alimentados exclusivamente com leite materno (n=38; 86,4%) e por sonda (n=33; 75%). A principal causa de sua retenção na UTIN foi a síndrome respiratória (n=16; 36,4%) e o óbito ocorreu em (n=3; 6,8%) dos neonatos. Em relação às características gestacionais de suas mães, estas têm em média dois filhos, grande parte planejou a gestação atual (n=22; 52,5%), a maioria realizou mais de 6 consultas pré-natal (n=40, 50%), pelo Sistema Único de Saúde (n=33; 78,6%) e com clínico geral (n=16; 38,1%). Nenhuma relatou utilizar cigarro e drogas ilícitas na gestação. Conclusão: Observou-se que a prematuridade foi a principal causa de retenção dos neonatos na UTI sendo esta inversamente proporcional ao peso de nascimento dos bebês estudados.
