Comparative DNA methylome analysis of endometrial carcinoma reveals complex and distinct deregulation of cancer promoters and enhancers
BACKGROUND: Aberrant DNA methylation is a hallmark of many cancers. Classically there are two types of endometrial cancer, endometrioid adenocarcinoma (EAC), or Type I, and uterine papillary serous carcinoma (UPSC), or Type II. However, the whole genome DNA methylation changes in these two classical types of endometrial cancer is still unknown. RESULTS: Here we described complete genome-wide DNA methylome maps of EAC, UPSC, and normal endometrium by applying a combined strategy of methylated DNA immunoprecipitation sequencing (MeDIP-seq) and methylation-sensitive restriction enzyme digestion sequencing (MRE-seq). We discovered distinct genome-wide DNA methylation patterns in EAC and UPSC: 27,009 and 15,676 recurrent differentially methylated regions (DMRs) were identified respectively, compared with normal endometrium. Over 80% of DMRs were in intergenic and intronic regions. The majority of these DMRs were not interrogated on the commonly used Infinium 450K array platform. Large-scale demethylation of chromosome X was detected in UPSC, accompanied by decreased XIST expression. Importantly, we discovered that the majority of the DMRs harbored promoter or enhancer functions and are specifically associated with genes related to uterine development and disease. Among these, abnormal methylation of transposable elements (TEs) may provide a novel mechanism to deregulate normal endometrium-specific enhancers derived from specific TEs. CONCLUSIONS: DNA methylation changes are an important signature of endometrial cancer and regulate gene expression by affecting not only proximal promoters but also distal enhancers. Development of a novel flexible polymer-based biosensor platform for the thermal detection of noradrenaline in aqueous solutions
Molecularly Imprinted Polymers (MIPs) are synthesized for the neurotransmitter noradrenaline with the optimal composition and binding conditions being determined via optical batch rebinding experiments. Next, the obtained MIP polymer particles are mixed within screen-printed inks to produce mass-producible bulk modified MIPs screen-printed electrodes (MIP-SPEs). In this contribution, the supporting surface which the MIP-SPEs are screen-printed upon are explored to deviate from conventional polyester, to polyvinylchloride, tracing paper and household-printing paper. The performance of the MIP-SPEs are measured using the Heat-Transfer Method (HTM), a straightforward and low-cost detection technique based on thermal resistance. At first, the noise on the signal is minimized by adjusting the settings of the temperature feedback loop. Second, the response of the MIP-SPEs to noradrenaline is measured and compared for the different substrate materials. Sensors printed onto paper are considered in further experiments as their response to noradrenaline is the highest and advantageous material properties, including sustainability and flexibility of the material. Subsequently, dose-response curves are determined by simultaneously measuring HTM and Thermal Wave Transport Analysis (TWTA). The latter is a new thermal detection method that relies on the use of thermal waves and has the advantage of a short measurement time (2 min). With these thermal methods, it is possible to specifically detect noradrenaline in aqueous solutions and quantify it at relevant concentrations. In summary, by combining synthetic receptors with thermal measurement techniques it is possible to develop a portable sensor platform that is capable of low-cost and straightforward detection of biomolecules. Through exploring novel SPE substrates, a system is designed that is flexible and holds potential for the use in commercial biomedical devices and complex sensor architectures
Observation of two new baryon resonances
Two structures are observed close to the kinematic threshold in the mass spectrum in a sample of proton-proton collision data, corresponding
to an integrated luminosity of 3.0 fb recorded by the LHCb experiment.
In the quark model, two baryonic resonances with quark content are
expected in this mass region: the spin-parity and
states, denoted and .
Interpreting the structures as these resonances, we measure the mass
differences and the width of the heavier state to be
MeV,
MeV,
MeV, where the first and second
uncertainties are statistical and systematic, respectively. The width of the
lighter state is consistent with zero, and we place an upper limit of
MeV at 95% confidence level. Relative

Observation of associated production of a boson with a meson in the~forward region
A search for associated production of a boson with an open charm meson is
presented using a data sample, corresponding to an integrated luminosity of
of proton--proton collisions at a centre-of-mass energy
of 7\,TeV, collected by the LHCb experiment. %% Seven candidate events for
associated production of a boson with a meson and four candidate
events for a boson with a meson are observed with a combined
significance of 5.1standard deviations. The production cross-sections in the
forward region are measured to be where the first uncertainty is statistical and the

Measurements of the , , meson and baryon lifetimes
Measurements of -hadron lifetimes are reported using collision data,
corresponding to an integrated luminosity of 1.0fb, collected by the
LHCb detector at a centre-of-mass energy of Tev. Using the exclusive decays
, , ,
and the average decay
times in these modes are measured to be = 0.004 0.003 ps, =
0.006 0.004 ps, = 0.013
0.005 ps, = 0.027
0.006 ps and = 0.011
0.005 ps, where the first uncertainty is statistical and the second is
systematic. These represent the most precise lifetime measurements in these
decay modes. In addition, ratios of these lifetimes, and the ratio of the
decay-width difference, , to the average width, , in
the system, , are
reported. All quantities are found to be consistent with Standard Model

Differential branching fraction and angular analysis of decays
The differential branching fraction of the rare decay is measured as a function of , the
square of the dimuon invariant mass. The analysis is performed using
proton-proton collision data, corresponding to an integrated luminosity of 3.0
\mbox{ fb}^{-1}, collected by the LHCb experiment. Evidence of signal is
observed in the region below the square of the mass. Integrating
over 15 < q^{2} < 20 \mbox{ GeV}^2/c^4 the branching fraction is measured as
d\mathcal{B}(\Lambda^{0}_{b} \rightarrow \Lambda \mu^+\mu^-)/dq^2 = (1.18 ^{+
0.09} _{-0.08} \pm 0.03 \pm 0.27) \times 10^{-7} ( \mbox{GeV}^{2}/c^{4})^{-1},
where the uncertainties are statistical, systematic and due to the
normalisation mode, , respectively.
In the intervals where the signal is observed, angular distributions are
studied and the forward-backward asymmetries in the dimuon ()
and hadron () systems are measured for the first time. In the
range 15 < q^2 < 20 \mbox{ GeV}^2/c^4 they are found to be A^{l}_{\rm FB} =
-0.05 \pm 0.09 \mbox{ (stat)} \pm 0.03 \mbox{ (syst)} and A^{h}_{\rm FB} =

Precision measurement of violation in decays
The time-dependent asymmetry in decays is
measured using collision data, corresponding to an integrated luminosity
of fb, collected with the LHCb detector at centre-of-mass energies
of and TeV. In a sample of 96 000 decays, the
-violating phase is measured, as well as the decay widths
and of the light and heavy mass eigenstates of the
system. The values obtained are rad, ps, andps, where the first uncertainty is
statistical and the second systematic. These are the most precise single
measurements of those quantities to date. A combined analysis with decays gives rad. All
measurements are in agreement with the Standard Model predictions. For the
first time the phase is measured independently for each polarisation
state of the system and shows no evidence for polarisation
