positiva.

No entanto,
A análise ocorreu a
espécie Pinna nobilis embora pertençam a famílias diferentes.
The biomineralization process of mollusc’s shell is controlled by an extracellular
organic matrix which is produced by the calcifying epithelium and secreted into
the extrapallial space. Some studies have already been performed and give
some answers about organic matrix constituents. However almost all of them
were on species of the families Pteriidae and Pinnidae. In the present work the
study animal is the freshwater bivalve Anodonta cygnea, belonging to the
Unionidae family, from which little is known about its organic matrix. From the
extraction process it is possible to achieve two fractions of organic matrix, but
only one, the soluble fraction, was the object of this study. Different techniques
were used, including biochemical characterization by electrophoresis,
quantification and detection of the matrix directly in the shell through the
immunogold technique. Were also produced polyclonal antibodies against the
organic matrix and a complementary DNA library, constructed with the
messenger RNA of the outer mantle epithelium, which is in this animal the
calcifying epithelium, used as a template. From the electrophoretic analysis five
protein fractions were detected with molecular weights of approximately 106,
74, 59, 49 and 40 KDa. The small amount of the total protein and
glycosaminoglycans obtained from the extraction indicate the low percentage of
organic matrix when compared to the inorganic fraction which is in this case
calcium carbonate in the form of aragonite crystals, situation confirmed in other
studies. By the immunogold technique it was found that the organic matrix is
mainly intercrystalline but it can also be found within the crystals. The results
found in terms of biochemical characterization in this study with Anodonta
cygnea are similar to those described for other members of this family.
Regarding to the localization of the organic matrix in the shell, the results
obtained are very similar to the localization founded in the Pinna nobilis
although they belong to the different families. This suggests that the thousands
of years of divergent evolution should not vary greatly between families in
terms of constitution and organization of the organic matrix
Towards an interpretation of the Portuguese cultural tourist: motivations and constraints
The importance of cultural tourism for the development of tourist destinations has created a need for knowledge on both supply and demand sides, related to the motivations and constraints which can affect the decisions of the cultural tourist. The aim of this study is to develop and validate for the Portuguese tourists two scales to measure motivations to travel and its constraints, considering theories of needs and constraints, and to examine differences according to tourists’ socio-demographic characteristics. The scales were determined using a non-linear principal component analysis followed by bootstrap confirmatory factor analysis, and differences were examined using parametric tests. A six-dimensional model for motivations and a five-dimensional model for constraints were validated, both with good overall fit. Motives linked to culture, intellectual curiosity and cultural knowledge stand out with high levels of relevance. Lack of resources and other commitments are the most important constraints. Significant differences were found in almost all characteristics. The results reveal influential dimensions on travel decisions which are of utmost importance for the design of the cultural offer of destinations.info:eu-repo/semantics/publishedVersio
Through the lens of workers’ motivation: does it relate to work–family relationship perceptions?
Workers’ motivations and the work–family relationship are two subjects that have been capturing the attention of researchers and practitioners. However, to date, little is known about the link between the two subjects. Thus, this research aimed to investigate the relationships among each type of motivation conceptualized in self-determination theory and work–family conflict, work–family enrichment, and work–family balance. In addition, the current study intended to investigate the relationships among work–family conflict, work–family enrichment, and Work–family balance. Data were collected through a self-administered questionnaire. To test the hypotheses, the PROCESS macro was used. The results suggested that intrinsic motivation is negatively associated with work–family conflict and positively associated with work–family enrichment and work–family balance. Additionally, identified regulations seem to be positively associated with work–family enrichment and work–family balance. However, contrary to expectations, this study revealed a positive relationship between introjected regulation and work–family enrichment and work–family balance. Concerning external regulation (material and social) and amotivation, globally, the findings were consistent with the hypotheses, i.e., the higher the external regulation and amotivation, the higher work–family conflict and the lower the work–family enrichment and work–family balance. Moreover, the higher the work–family conflict, the lower the work–family balance, and the higher the work–family enrichment, the higher the work–family balance. Theoretical and practical implications are discussed.info:eu-repo/semantics/publishedVersio

Currently, there is insufficient knowledge about the burden of multimorbidity in patients with previous fragility fracture. The aim of this study was to evaluate the association between multimorbidity and previous fragility fracture, and to assess the effect of fragility fracture and/or multimorbidity in the perception of quality-of-life and physical function, in women 50 years of age and older. Methods: Women aged ≥50 years from the EpiReumaPt study (2011−2013), a nationwide population-based study, were evaluated. Self-reported data regarding sociodemographics, health-related quality of life, physical functioning, fragility fracture, and multimorbidity were collected using a semi-structured questionnaire. Multimorbidity was defined as 2 or more chronic non-communicable diseases. Descriptive exploratory analysis of the data was performed using hypothesis testing. Multiple logistic regression modelling was used to assess the association between multimorbidity and fragility fractures, and linear regression was used for the quality-of-life and physical function outcomes. Results: The estimated prevalence of fragility fracture in women older than 50 years was 17.5%. A higher prevalence of multimorbidity (74.6%) was found in the group of women with previous fragility fracture than in those without previous fragility fracture. Multivariate logistic regression analysis revealed that women with multimorbidity had a higher odds of fragility fracture (adjusted odds ratio, 1.38; 95% confidence interval, 1.12–1.69), compared with women with 1 or no self-reported non-communicable chronic diseases. In women with previous fragility fracture, rheumatic diseases (62.7%) and hypertension (58.6%) were the most frequently self-reported non-communicable chronic diseases. Both multimorbidity and a previous fragility fracture were independently associated with worse health-related quality of life and physical functioning. Conclusions: Women 50 years and older with multimorbidity had a significantly increased odds of fragility fracture. Fragility fracture and multimorbidity were negatively associated with quality of life and disability. This study emphasizes the need to redesign health services to care for patients to prevent non-communicable chronic diseases and fragility fracture, particularly in women 50 years and older, in whom these diseases are likely to potentiate the risk of fragility fracture.publishersversionpublishe

Genotypic and Phenotypic Characterization of Pathogenic Escherichia coli, Salmonella spp., and Campylobacter spp., in Free-Living Birds in Mainland Portugal
Birds are potential carriers of pathogens affecting humans and agriculture. Aiming to evaluate the occurrence of the top three most important foodborne pathogens in free-living birds in Portugal, we investigated 108 individual fecal samples from free-living birds and one pooled sample of gull feces (n = 50) for the presence of Escherichia coli (pathogenic and non-pathogenic), Salmonella spp. and Campylobacter spp. Virulence- and antimicrobial resistance- (AMR) associated genes were detected by PCR and Whole-Genome Sequencing (WGS), and phenotypic (serotyping and AMR profiles) characterization was performed. Overall, 8.9% of samples tested positive for pathogenic E. coli, 2.8% for Salmonella spp., and 9.9% for Campylobacter spp. AMR was performed on all pathogenic isolates and in a fraction of non-pathogenic E. coli, being detected in 25.9% of them. Ten of the tested E. coli isolates were multidrug-resistant (MDR), and seven of them were Extended-spectrum β-lactamase (ESBL) producers. Among Salmonella (n = 3) and Campylobacter (n = 9), only one strain of C. jejuni was identified as MDR. Most of the identified serotypes/sequence types had already been found to be associated with human disease. These results show that free-living birds in Portugal may act as carriers of foodborne pathogens linked to human disease, some of them resistant to critically important antimicrobials. This work was supported by funding from the European Union’s Horizon 2020 Research

desenvolvimento do modelo do GAP-SASUC. Tendo

Fibrosis of Peritoneal Membrane, Molecular Indicators of Aging and Frailty Unveil Vulnerable Patients in Long-Term Peritoneal Dialysis
Funding: Sociedade Portuguesa de Nefrologia (SPN) SPN funded a project and Ana Rita Martins, MD, Nephrology fellow, for a residence at Jiménez Díaz Foundation University Hospital, Madrid under the scope of novel serum biomarkers of CKD. iNOVA4Health research program (UIDP/04462/2020) is also acknowledged to support J.M.Peritoneal membrane status, clinical data and aging-related molecules were investigated as predictors of long-term peritoneal dialysis (PD) outcomes. A 5-year prospective study was conducted with the following endpoints: (a) PD failure and time until PD failure, (b) major cardiovascular event (MACE) and time until MACE. A total of 58 incident patients with peritoneal biopsy at study baseline were included. Peritoneal membrane histomorphology and aging-related indicators were assessed before the start of PD and investigated as predictors of study endpoints. Fibrosis of the peritoneal membrane was associated with MACE occurrence and earlier MACE, but not with the patient or membrane survival. Serum α-Klotho bellow 742 pg/mL was related to the submesothelial thickness of the peritoneal membrane. This cutoff stratified the patients according to the risk of MACE and time until MACE. Uremic levels of galectin-3 were associated with PD failure and time until PD failure. This work unveils peritoneal membrane fibrosis as a window to the vulnerability of the cardiovascular system, whose mechanisms and links to biological aging need to be better investigated. Galectin-3 and α-Klotho are putative tools to tailor patient management in this home-based renal replacement therapy.publishersversionpublishe
